Natural Com Starch(NCS) spray system has been widely used In multiply board mill for the better bonding strength between board layers. However the unsuitable NCS spray system could lead to a drop in plybond efficiency of multiply board. It could also result in the accumulation of unretained-starch in the white water system, which could not only rise in BOD and COD level but also reduce the effects of rosin-alum sizing system and other additives. In this study, the simple and rapid method by UV-spectroscopy for measuring the starch contents in the white water was proposed, and the quantitative analysis for dissolved starch content in white water was carried out in two kinds of board production process. It could be confirmed that about 150 ppm of oxidized starch was retained in white water dewatered from undertop ply of multiply board process, and about 470 ppm of natural starch was dissolved in white water originated from unsuitable spraying system in the multiply board production lines for cup base paper.
Direct synthesis of cyclodextrin (CD) from extruded insoluble corn starch without liquefaction procedure using cyclodextrin glucanotransferase (CGTase) was carried out. Increased CD production rate and yield were achieved in heterogeneous enzyme reaction system containing extruded corn starch compared with those of conventional system employing liquefied or partially cyclized starch. At extruded starch concentration of 100 g/l the CD concentration and conversion yield were reached up to 54 g/l and 0.54, respectively. High purity of $\alpha \beta \gamma$-CDs without accumulation of undesirable malto-oligosaccharides was produced, furthermore, the residual extruded starch was easily separated by centrifugation from reaction mixture, whlch will facilitate the purification procedure. Granular structure of extruded starch was observed by SEM to investigate enzyme reaction mechanism. Supplemental addition of $\alpha$-amylase enhanced slightly the initial CD production rate, but it decomposed produced CD at the late stage. Various! extruded raw starches, such as, corn, rice, and barley were also suitable substrates for CD production.

The fungal species of Rhizopus oryzae 2062 has the capacity to carry out a single stage fermentation process for lactic acid production from potato starch wastewater. Starch hydrolysis, reducing sugar accumulation, biomass formation, and lactic acid production were affected with variations in pH, temperature, and starch source and concentration. A growth condition with starch concentration approximately 20 g/ L at pH 6.0 and 30$^{\circ}C$ was favourable for starch fermentation, resulting in a lactic acid yield of 78.3%∼85.5% associated with 1.5∼2.0 g/L fungal biomass produced in 36 h of fermentation.

Two comparative poplar clones (I-214: Populus euramericana, Peace: P koreana x P. trichocarpa) were exposed to two $CO_2$ concentrations (350 or 2,000 ${\mu}L\;L^{-1}\;CO_2)$ for 21 days. When both poplar clones were compared at growth conditions, the net photosynthetic rate $(P_N)$ in $CO_2-enriched$ (2,000 ${\mu}L\;L^{-1}\;CO_2=C_{2,000})$ plants become about $50-60\%$ higher than that of 350 ${\mu}L\;L^{-1}\;CO_2(=C_{350})$ plants on 7 days treatment. But the enhancement of $P_N$ by high $CO_2$ was not maintained throughout all the experimental period. At 21 days, there was no difference of photosynthetic rates between $C_{350}\;and\;C_{2,000}$ plants. In contrast with photosynthesis, the response of leaf conductance to the elevated $CO_2$ concentration was very different between I-214 and Peace. During all experimental period, leaf conductance $(g_s)$ of $C_{2,000}$ plants is $50\%$ lower than that of the $C_{350}$ plants for I-214, while there is no difference of $g_s$ between the plants of $C_{350}\;and\;C_{2,000}$ on for Peace. The results of gs in Peace indicate that decreased photosynthetic rate after 21 days in $C_{2,000}$ on plants for two poplar clones is possibly due to non-stomatal factors. To investigate the non-stomatal factors, starch accumulation and ribulose-1,6-bisphosphate carboxylase (RuBPCase) were measured. We found significant accumulation of starch in two poplar clones exposed to high $CO_2,$ especially starch of I-214 in $C_{2,000}$ become 3.5 times higher than in $C_{350}$ plants at 21 days. This suggests that high proportion of photosynthates was directed into starch. After 21 days, the activity of ribulose-1, 6-bisphosphate carboxylase of $C_{2,000}$ plants become decreased in $40-50\%$ compared with that of the $C_{350}$ plants. Two poplar clones show the same trend to RuBPCase declines under high $CO_2$ concentration, although the decline is more significant for I-214. The results reported here suggest that starch accumulation and decreased RuBPCase activity in $C_{2,000}$ plants can be partly ascribed to the loss of photosynthetic efficiency of high $CO_2-grown$ poplar plants.

A novel suspension-phase enzyme reaction system for the intermolecular transglycosylation of L-ascorbic acid into 2-O-${\alpha}$-D-glucopyranosyl L-ascorbic acid supplementing extrusion starch as the glycosyl donor was developed using cyclodextrin glucanotransferase from Thermoanaerobacter sp. A high conversion yield compared to the conventional soluble-phase enzyme reaction system using cyclodextrins and soluble starch was achieved. The optimal reaction conditions were 2,000 units of cycIodextrin glucanotransferase, 20 g/l of L-ascorbic acid, and 50 g/l of extrusion starch at $50^{\circ}C$ for 24 h. The new suspension-phase enzyme reaction system also exhibited several distinct advantages other than a high conversion yield, including a lower accumulation of oligosaccharides and easily separable residual extrusion starch by centrifugation or filtration in the reaction mixture, which will facilitate the purification of 2-O-${\alpha}$-D-glucopyranosyl L-ascorbic acid. The new suspension-phase enzyme reaction system seems to be potentially applicable as the industrial process for the production of thermally and oxidatively stable 2-O-${\alpha}$-D-glucopyranosyl L-ascorbic acid.
The synergistic effect of glucoamylase and a -amylase on the hydrolysis of raw corn starch in an agitated bead reaction system was studied by investigating the changes of sugar profiles, the granular structure, particle size distribution, and X-ray diffraction pattern of residual raw corn starch. The enzymatic hydrolysis of raw corn starch was greatly enhanced by synergistic effect of glucoamylase and $\alpha$ -amylase. Even though the sugar profiles were mainly determined by the mixing ratio of glucoamylase and $\alpha$-amylase; raw starch was mainly converted to glucose directly without accumulation of any significant amount of oligosaccharides. The cavity formation and fragmentation phenomena of raw corn starch granule subjected to enzyme reaction were analyzed by means of SEM and the particle size distribution. The X-ray diffraction pattern of raw starch was not changed at the initial stage of reaction but slightly changed at the late stage of hydrolysis, which may be caused by the preferential degradation of amorphous region by enzymatic reaction, not by the destruction of microcrystalline structure of raw corn starch.
Changes in the level of metabolites in leaves and pods were examined with respect to the seed chemical composition in black soybean. There was no further increase in pod length after 42 days after flowering (DAF). Pod weight, however, persistently increase until 73 DAF, thereafter the weight was slightly lowered. The seed storage protein, however, increased drastically as the increasing rate of pod weight was lessened at 61 DAF. The accumulation of seed storage proteins was occurred conspicuously as the increasing rate of pod weight was slowed down. The chlorophyll content both in leaves and pods was drastically decreased after 50 DAF. The beginning of drastic reduction in chlorophyll content was occurred concomitantly with the reduction of soluble protein content in leaves. The sugar content in leaves showed similar tendency with chlorophyll and soluble protein content. The starch level in leaves, however, showed different changing pattern during seed development. The starch content in leaves was increased persistently until 66 DAF, thereafter the content was decreased drastically to about $55\%$ of maximal value at 66 DAF. Total phenolics content in leaves and the anthocyanins content in seeds were stable without noticeable increase until 66 DAF. The contents were increased dramatically after 66 DAF showing the synchronized pattern with the decrease in starch level in leaves. The levels of the selected metabolites in leaf and seed suggested that the accumulation of chemical components of black soybean seed is launched actively at 66 DAF. The profile of storage proteins was nearly completed at 61 DAF because there was no large difference in densitometric intensity among protein subunits after 61 DAF. In soybean, chemical maturation of seed begins around 61 to 66 DAF at which most metabolites in vegetative parts are decreased and remobilized into maturing seeds.

To study the biochemical and physiological role of the plastidic glucose transporter (pGlcT) in carbohydrate metabolism, we characterized transgenic plants with mutations in the pGlcT gene (GT), gt-1 and gt-2, as well double mutants of GT and the maltose transporter (MEX1) and GT and the triose phosphate/phosphate translocator (TPT), GT and the cytosolic fructose-1,6-bisphosphatase gene (cFBP), and MEX1 and TPT, gt-1/mex2, gt-1/tpt-2, gt-1/cfbp-1, mex1-1/tpt-2, respectively. Compared to the wild type, all mutants except the gt-1/cfbp-1 mutant lines displayed higher starch accumulation and higher levels of maltose. Starch accumulation is due to a decrease in starch turnover, leading to an imbalance between the rates of synthesis and degradation. Sucrose levels of gt alleles were higher than those in wild-type plants during the light period, suggesting possible nightly supplementation via the maltose transport pathway to maintain proper carbohydrate partitioning in the plant leaves. The gt plants displayed less growth retardation than mex1-1 mutant and gt-1/mex2 double mutant displayed accumulativesevere growth retardation as compared to individual gt-1 and mex1-1 mutants, implying that the maltose transporter-mediated pathway is a major route for carbohydrate partitioning at night. The gt-1/tpt-2, mex1-1/tpt-2 and gt-1/cfbp-1 double mutants had retarded growth and low chlorophyll content to differing degrees, indicating that photosynthetic capacity had diminished. Interestingly, the gt-1/tpt-2 line displayed a glucose-insensitive phenotype and higher germination rates than wild type, suggesting its involvement not only in carbon partitioning, but also in the sugar signaling network of the pGlcT and TPT.
The production cost of biodiesel from microalgae is still not competitive, compared with that of petroleum fuels. The genetic improvement of microalgal strains to increase triacylglycerol (TAG) accumulation is one way to reduce production costs. One of the most promising approaches is the isolation of starch-deficient mutants, which have been reported to successfully increase TAG yields. To date, such a stable mutant is not available in an oleaginous marine microalga, despite several advantages of using marine species for biodiesel production. Algae in the genus Dunaliella are known to tolerate high salt concentration and other environmental stresses. In addition, the cultivation processes for large-scale outdoor commercialization have been well established for this genus. In this study, Dunaliella tertiolecta was used to screen for starch-deficient mutants, using an iodine vapor-staining method. Four out of 20,016 UV-mutagenized strains showed a substantial reduction of starch content. A significantly higher TAG content, up to 3-fold of the wild-type level, was observed in three of the mutants upon induction by nitrogen depletion. The carotenoid production and growth characteristics of these mutants, under both normal and oxidative stress conditions, were not compromised, suggesting that these processes are not necessarily affected by starch deficiency. The results from this work open up new possibilities for exploring Dunaliella for biodiesel production.
