Effects of Sowing Date for Seed Quality of Sprout-soybean (나물콩 재배시기가 종실 품질에 미치는 영향)

  • These studies were conducted to clarify the effects of sowing date on seed quality characters in soybean-sprouts. Six sprout-soybean cultivars were planted on three sowing dates in 2000 and 2001. Percentages of purple spot seed stain, seed coat-cracking and immatured seeds were higher in the May 25 sowing than later sewings. Pungsannamulkong showed the lowest percentages of seed coat-cracking and immatured seeds, while Hannamkong showed the highest percentages. The protein contents decreased as the sowing dates were delayed. Tawonkong and Pungsannamulkong showed the highest protein content on May 21 and June 15 sewings, and on July 5 sowing, respectively. The oil contents was not affected by the sowing dates, however, Somyeongkong and Tawonkong showed the highest and the lowest oil contents, respectively. The aglycone contents of isoflavone were the highest in the lune 15 sowing followed by July S sowing and May 25 sowing in that order. Somyeongkong showed the highest contents of isoflavone and aglycone, while Tawonkong showed the lowest contents.

Effect of Benzyladenopurine Treatment Time during Imbibition on Growth of Soybean Sprouts and its Cost (BA 처리시기에 따른 콩나물의 생장과 처리비용 분석)

  • Benzyladenopuyine (BA) commonly used for soybean sprout production is of high price. The study was carried out to determine the effect of BA treatment time during seed imbibition on growth and development of soybean sprouts and to analyse its treatment cost. The soybean seeds of 4 cultivars were soaked in 4 ppm BA solution during the first 5.5 hour imbibition (EFHI), the second 5.5 hour imbibition (SFHI) immediately after 0.5 hour aeration, or whole 11.0 hour imbibition (WFHI) intervened by aeration. On the 6th day after culture, the soybean sprouts were classified into 4 categories on the base of hypocotyl length; > 7cm, 4 to 7cm, < 4 cm and not germinated, and their morphological characters, fresh and dry weights were measured. Water absorption of the seeds was sharply reduced after it was almost done for the first 5.5 hours. The percentage of sprouts with hypocotyls of longer than 4cm was higher in FFHI treatment than in the other two ones. Regardless of BA imbibition time and periods, the lateral roots were not observed. WFHI treatment showed shorter hypocotyl and root lengths but thicker hypocotyl and hook diameters than FFHI and SFHI treatments. All component fresh and dry weights except cotyledon fresh weight were nearly same. Treatment cost of BA was the lowest in SFHI treatment. It is concluded that BA treatment during SFHI is the best time because its treatment time did not affect sprout growth but its treatment cost.

Effect of Seed Imbibition into Water and Acetic Acid Solution on its Floating Rate and Growth of Soybean Sprouts (수침(水浸)과 Acetic Acid 처리에 따른 콩나물의 생장과 형태 변화)

  • Soybean sprout decay during its culture should be one of serious problems. The study was carried out to clarify the effect of water imbibition and acetic acid treatments on growth and morphological characters of the soybean sprouts. The soybean seeds of 3 cultivars (cv. Eunhakong, Pungsannamulkong and Junjery) imbibed in pure water or 0.l% acetic acid for 3 minutes before soaked for 6 hours into 1 ppm BA solution and aerated for 3 hours immediately before 6 day culture. On the 6th day, the sprouts were classified into 4 categories on the base of hypocotyl length; > 7cm, 4 to 7cm, > 4cm and not germinated and seed floating rate, their morphological characters, fresh fraction weights and productivity were measured. The best water imbibition for seed cleaning was to soak the seeds for 5 minutes and then to aerate soak them for 40 minutes. In Pungsannamulkong and Junjery, percentage of the sprouts with hypocotyls of longer than 4 cm was higher in water imbibition than in acetic acid treatment for seed disinfection although in Eunhakong there was no significant difference between the two treatments. Eunhakong and Junjery had greater lateral root formation rate and its number per sprout in water imbibition than in acetic acid treatment but Pungsannamulkong showed reverse result. Eunhakong and Pungsannamulkong, furthermore, had more total fresh weight in acetic acid treatment than in water imbibition but Junjery showed reverse result, although there was no significant difference between the two treatments in productivity of mass production system.

Mathematical modeling of growth of Escherichia coli strain RC-4-D isolated from red kohlrabi sprout seeds (적콜라비 새싹채소 종자에서 분리한 Escherichia coli strain RC-4-D의 생장예측모델)

  • This study was conducted to develop a predictive model for the growth of Escherichia coli strain RC-4-D isolated from red kohlrabi sprout seeds. We collected E. coli kinetic growth data during red kohlrabi seed sprouting under isothermal conditions (10, 15, 20, 25, and $30^{\circ}C$). Baranyi model was used as a primary order model for growth data. The maximum growth rate (${\mu}max$) and lag-phase duration (LPD) for each temperature (except for $10^{\circ}C$ LPD) were determined. Three kinds of secondary models (suboptimal Ratkowsky square-root, Huang model, and Arrhenius-type model) were compared to elucidate the influence of temperature on E. coli growth rate. The model performance measures for three secondary models showed that the suboptimal Huang square-root model was more suitable in the accuracy (1.223) and the suboptimal Ratkowsky square-root model was less in the bias (0.999), respectively. Among three secondary order model used in this study, the suboptimal Ratkowsky square-root model showed best fit for the secondary model for describing the effect of temperature. This model can be utilized to predict E. coli behavior in red kohlrabi sprout production and to conduct microbial risk assessments.

A New Sprout-soybean Cultivar, "Hoseo" with Early maturity, Small Seed Size and High Sprout yielding (극소립·조숙 고수율 나물콩 신품종 "호서")

  • A new sprout-soybean cultivar, "Hoseo" was developed at the Honam Agricultural Research Institute (HARI) in 2007. Hoseo was selected from a cross between Camp and Nattosan. The preliminary, advanced, and regional yield trials to evaluate the performance of Iksan 57 were carried out from 2003 to 2007. This cultivar has a determinate growth habit with purple flower, grayish brown pubescence, yellow seed coat, grayish brown hilum, rhomboid leaflet shape and small seed size (7.4 g/100 seeds). The maturity date of "Hoseo" is 12 days earlier than the check variety, "Pungsan". It has good seed quality for soybean-sprout and resistance to lodging. This cultivar has resistance to soybean mosaic virus (SMV) and necrotic symptom (SMV-N). The average yield of "Hoseo" was 2.51 ton per hectare in the regional yield trials (RYT) for double cropping carried out for three years from 2005 to 2007.

Effects of heat treatments on the microbial reduction and germination rates of red radish sprout seeds (Raphanus sativus) (열처리 방법이 적무 새싹종자(Raphanus sativus)의 미생물 저감화 및 발아율에 미치는 영향)

  • This study was conducted to investigate the effects of various heat treatments on the microbial reduction and germination of red radish seeds for the development of effective and economical sterilization methods of improving microbial safety without reducting the germination rate. Hydrothermal treatment was conducted at 60, 65, 70, 75, 80, and $90^{\circ}C$ for 30 and 60 seconds, and dry heat treatment was performed at 70, 80, 90, and $100^{\circ}C$ for 5 minutes. In the seeds that underwent the hydrothermal treatment, time had little effect on the microbial reduction. There was no significant microbial reduction over time. However, there was significant microbial reduction as temperatures increased (p<0.001). The total plate count (TPC) was reduced by more than 3 logs, and Listeria monocytogenes was not detected at temperatures above $70^{\circ}C$. In the seeds that were subjected to the dry heat treatment, the TPC and the population of the L. monocytogenes were significantly reduced as the temperatures increased (p<0.001). After treatment at $100^{\circ}C$ for 5 minutes, the TPC and the L. monocytogenes were reduced by 3 logs. As with the microbial reduction, time had little effect on the germination. There were no significant changes in the germination after the hydrothermal treatment over time; but at the temperatures above $75^{\circ}C$, the germination rate significantly decreased as the temperature increased (p<0.001). When the seeds were soaked after the hydrothermal treatment, their germination was stimulated. The dry heat treatment at temperatures of $80^{\circ}C$ and higher significantly decreased the germination rate as the temperature increased (p<0.001). Dry heat treatment before the germination of the seeds soaked in distilled water for three hours significantly decreased the germination at temperatures greater than $90^{\circ}C$ (p<0.05). This study showed that appropriate heat treatments can increase the microbiological safety and germination of red radish seeds.

Effect of Aqueous Chlorine Dioxide and Citric Acid on Reduction of Salmonella typhimurium on Sprouting Radish Seeds (이산화염소수 및 구연산처리에 따른 무(Raphanus sativus L.) 새싹과 종자의 미생물 제어 효과)

  • The effect of citric acid-aqueous chlorine dioxide ($ClO_2$) treatment of radish seeds artificially contaminated with Salmonella typhimurium was studied. Radish seeds were inoculated by immersion, in more than 106 log CFU/g seed, of a suspension of S. typhimurium, dried, and stored sealed at $4^{\circ}C$ Radish seeds soaked in 200 ppm aqueous ClO2 solution for 10 min showed a bacterial reduction of 1.08 log CFU/g seed, and the lowering of microbial burden noted in seeds soaked in 2% (w/v) citric acid solution for 10 min was 2.89 log CFU/g seed. Next, radish seeds were exposed to 0.5% (v/v) glycerol solution for 10 min either before or after treatment with 200 ppm aqueous ClO2 or 2% (w/v) citric acid for 10 min. Glycerol exposure after citric acid treatment reduced bacteria by 3.46 log CFU/g seed, and glycerol treatment after aqueous $ClO_2$ treatment reduced the microbial burden by 2.14 log CFU/g seed. Both glycerol treatments yielded better elimination of S. typhimurium than did a single treatment with either citric acid or aqueous $ClO_2$. Radish seeds inoculated with S. typhimurium were treated throughout the entire growth period. Although radish seed treatment was effective, treatment by citric acid and aqueous $ClO_2$ after sprouting was not effective to eliminate S. typhimurium.

Change of asparagine content in soybean sprouts by variety, root growth, and cultivation period (콩나물의 품종, 뿌리발생 및 재배일수에 따른 asparagine 함량 변이)

  • The varietal difference, effect of root amounts and cultivation duration on the asparagine content of soybean sprouts, which is known to have the excellent detoxifying effect of ethanol, were investigated for developed varieties and indigenous sprout lines to establish cultivation methods of increasing the asparagine content and to develop soybean varieties having high asparagine content. Some of the research results obtained are summarized as follows; 1. Range and mean of asparagine content of 174 germplasm were 0.38~1.67% and 0.99%, respectively, on fresh weight basis. 2. Developed sprout-soybean varieties showed somewhat higher asparagine content of 1.29% than that of indigenous sprout lines of 0.96% on fresh weight basis. 3. No significant difference in asparagine content among the seed size groups was recognized though the highest asparagine content, 1.02% on fresh weight basis was observed in the seed size of 8.1~10.0g/100 seeds. 4. Among the seed coat color groups, soybean of brown seed coat color showed the highest asparagine content (1.15%) on fresh weight basis. No difference was observed among other groups of seed coat color. 5. Range of asparagine content of 174 varieties was 4.08~6.24% on dry weight basis. Soybean varieties that showed high asparagine content were Dawonkong, Sobacknamulkong, Sowonkong, and Somyungkong, with the contents of 6.24%, 6.21%, 5.95%, and 5.85%, respectively. 6. Amount of roots which have the highest asparagine content out of sprout parts was greatly increased up to 10 days of sprout cultivation. 7. Highly significant difference in asparagne content of sprout was recognized between those grown for five and ten days, with the asparagine content of 0.68% and 1.21%, respectively, on fresh weight basis. 8. Asparagine content of hypocotyle part was also highly proportional to days to cultivation; it increased from 2.91% at 2 days of cultivation to 15.68% on fresh weight basis at 14 days of cultivation.

Effects of Aeration Temperature and Period after BA Treatment on Growth and Lateral Root Formation of Soybean Sprouts (BA 처리 직후의 Aeration 온도와 기간이 콩나물의 생장과 세근발생에 미치는 영향)

  • Treatment effect of benzyladenopurine (BA) used to block the lateral roots formed on soybean sprouts should be influenced by its applying methods. This study was done to check the effects of temperature and period from seed imbibition into 2 ppm BA solution to the first watering for sprout culture on growth and morphology of soybean sprouts. Imbibed three cultivar (cv. Pungsannamulkong, Sowonkong and Junjery) seeds for 5 houys into 2 ppm BA solution were placed under different temperatures (AT; 20, 30, $40^{\circ}$) and periods (AP; 0, 1, 2, 3, 4 hours). On the 6th day, the soybean sprouts were classified by 4 categories on the base of hypocotyl length; >7cm, 4 to 7cm), < 4cm and non-germination to calculate their composition rates, number of lateral roots, lengths of hypocotyl and root diameters at middle and hook of hypocotyl, and fraction dry weights were measured. Germination and growth responses of the cultivars were changed by AT and AP treatments. The responses, lateral root formation and fresh weights were, however, mainly affected by the cultivars used rather than Af treatment. Rate of the sprouts which formed lateral roots was decreased with increased periods to 4 hours, but their number per sprout was not different between the treatments of longer than 3 hours. Lengths of hypocotyl and root organ and total fresh weights were the highest in an hour AP treatment although longer than 3 hour AP treatments did not showed the significant difference in the lengths. Conclusionally AP treatment was more important than Af one in seed aeration for soybean sprout culture immediately after imbibition into BA solution, and was done at least for 3 hours.

Lateral Root Formation and Growth of Soybean Sporouts Treated with Various Solutions (다양한 물질처리에 의한 콩나물의 세근형성 및 생장)

  • Lateral roots of soybean sprouts should deeply affect their quality and marketability. The study was done to compare the effects of ocher, chitosan, corn kennel, tea (Thea sinensis L.) and hard rubber tree (Eucommia ulmoides Oliver) leaf extracts on lateral root formation, growth and morphological characters of the sprouts. Seeds of three cultivars, Pungsannamulkong, Sowonkong and Junjery, were imbibed for 5 hours into their 10% solutions and cultured for 6 days. The cultured sprouts were classified into 4 categories to calculate their composition rates on the base of hypocotyl lengths;>7 cm (A), 4 to 7 cm (B), < 4 cm (C) and not germinated (D), and their morphological characters, fresh and dry weights were measured. Composition rate of A was the lowest in Junjery of the three cultivars, while that of C showed reverse result compared to A. This results was the most distinct in hard rubber tree leaf extracts (HRTLE) of the five treatments. In HRTLE treatment, lateral root formation rate were formed in almost of Sowonkong although reduced in order of Pungsannamulkong and Junjery. However, there was no significant difference between the other treatments. Lateral roots per sprout were the lowest in HRTLE treatment of the 5 treatments. In all treatments except the chitosan treatment, the roots were most formed in Sowonkong but least in Junjery. Sprout length adding hypocotyl and root was the shortest in Junjery compared to the other two cultivars. and was the longest in tea leaf extract treatment but the shortest in HRTLE treatment. The result in total fresh weight of sprouts was similar to that of the sprout length.
