Study on the Antagonism of Useful Microbes against Soybean Sprout Rotting Pathogens and Their Effect on the Growth of Soybean Sprouts (갈항미생물이 콩나물 부패균에 미치는 향균능력과 콩나물 생육 특성)

  • This study was conducted to examine the antagonism of useful microbes against soybean sprout rotting pathogens and their effect on the growth of soybean sprouts. The antagonism against soybean sprout rotting pathogens and the effect on the growth of soybean sprouts were examined by using P. areofacience 14H-3, P. fluorescens R1-12 and B. cereus Yell, bacteria were shown to inhibit mycellial growth of Rhizotonia solani strongly. The results of this study are summarized as follows. P. areofacience 14H-3 and B. cereus Yell were highly antagonistic against Rizoctonia solani, while they were especially highly antagonistic against bacterial diseases. The effect of inhibiting the proliferation of soybean sprout rotting pathogens was also examined by adding the culture solution for antagonistic bacteria to the PDA. Both P. areofacience 14H-3 and P. fluorescens Rl-12 showed the inhibition rate of 78.8%, while B. cereus Yell did 52.9%. The fresh weight and length of soybean sprouts were measured after raising them with added antagonistic microbes and culture medium. Soybean sprouts treated with B. cereus Yell showed increased higher, compared with those not treated with it. Soybean sprouts were also raised in the culture solution with antagonistic bacteria to examine the growth of soybean sprouts. Soybean sprouts treated with the culture solution of 200 times showed better growth than those not treated with it. Analyze proximate composition in soybean sprout showed that moisture, ash, total sugar did not appear difference, but in case of crude protein B. cereus Yell(8.9%) increased about 2 times than control(3.6%), but occasion of crude fat and crude fiber were P. areofacience 14H-3, P. fluorescens Rl-12 increased about each 2 times than control. In occasion of vitamin, bacterial antagonist(9.4∼10.8mg%) was more higher than control(9.9mg%).

Development of an Automatic Brown Rice Germinating System of Air-Phase Type with Intermittent Water Spraying and Anion Radiation (간헐 분무와 음이온 방사를 가진 공기노출형 자동 현미발아시스템의 개발)

  • In this study an automatic germinating system for germinated brown rice (GBR) was developed for accelerating germination and sprouting of brown rice. With the developed germinating system, the effects of anion radiation treatment on germination ratio, sprout growth, color quality and ${\gamma}$-aminobutyric acid (GABA) content of brown rice were investigated. This system can fast produce GBR by controlling the environmental conditions such as ambient temperature, relative humidity, and proper anion radiation. Air-phase germination with intermittent water spraying improved both germination ratio and sprout growth by about 2.2 and 2.3 times, respectively, compared to the conventional water soaking method. Our results strongly suggest that this germinating system that provides air-phase germination with intermittent water spraying and anion radiation can produce germinated brown rice with good color quality, high GABA content, and fast germination ratio as well as sprout growth.

  • Changes in free amino acid composition in cotyledon and axis was investigated during growth of soybean sprout. Free amino acid increased with sprout-growth, especially in axis. Arginine, proline and methionine did not appear in axis throughout. Free amino acids in total amino acids was 0.11% for soybean and 8.8% for 8 day-sprout. Free amino acid change in the course of culture was in order of Asp>His>Val>Ile>Thr>Ser>Lys>Tyr>Phe>Leu>Ala>Gly>Pro>Arg>Glu>Met. The content of free amino acid in 4 day-sprout was in order of Asp>Ser>Val>His>Ala>Ile>Lys>Thr>Arg>Leu>Glu>Phe>Tyr>Gly>Cys>Met>Pro.

Effects of Ultrasound Stimuli on Acceleration of Brown Rice Germination (초음파 자극이 현미발아 촉진에 미치는 영향)

  • The effects of ultrasound stimuli on the germination and sprout growth of brown rice were investigated. Ultrasound was applied to brown rice at the frequencies of 28, 40, and 60 kHz before germination test and it was germinated in three methods (Type I, II and III). Type I was to soak brown rice into water for 60 hours. Type II was to expose brown rice to air for 48 hours after soaking them into water for 12 hours. Type III was a repetitive method of water-soaking and air exposure for 12 hours respectively. The most effective method for the germination was Type III without ultrasound. However, Type I was a best method after ultrasound treatment. As power of ultrasound increased, sprouts grew faster after brown rice were treated in 40%, 70%, and 100% power (0.137, 0.241, and 0.344 $W/cm^2$) at 40 kHz. The good treatments for fast sprout growth of brown rice at each frequency were the 28 kHz-10min group, the 40 kHz-5min group, and the 60 kHz-20min group of Type I. The best effective treatment was the 40 kHz-5min group at 0.344 $W/cm^2$ and at that condition the time required for sprout growth of 2.5 m was 51.9 hours. The ultrasound stimuli was very effective in the beginning of the rice germination, and the germination ratio was more than 95% in all ultrasound treatments.

Selction of Useful Chemicals Reducing Soybean -Sprout Rot (콩나물 부패경감에 유용한 약제 선발)

  • This study was carried out to select useful chemicals to control pathogens inciting soybean-sprout rot. Pathogen strains were isolated from decaying soybean seedlings. The isolated strains were identified as Pseudomonas spp. (Pse. strains) through microbiological test, however, no rot-inciting fungus was isolated. Eight food additives were tested with different concentrations in controlling pathogens and harmfulness. Five chemicals(over 5% cone.) including sodium hypochlorite apparently inhibited the growth of Pse. strains on media plate, however, sodium hypochlorite was discarded due to its severe germination inhibition. Propionic acid and acetic acid inhibited the growth of Pse. strains more effectively than calcium propionate and sodium propionate relatively. As no harmful effect on seed germination and no growth retard of soybean sprouts were observed by those chemicals with lower concentration(0.5∼1.0%), these chemicals were considered to be applicable to sprout rot control judged by the effectiveness and permissible concentration as food additives.

Effect of Recycled Coir Organic Substrates on Vegetable Crop Growth (코이어배지의 재활용이 채소 작물의 생육에 미치는 영향)

  • The present study was investigated the effect of recycled coir organic substrates on the growth of different vegetable crops. The recycled coir had better physical and chemical properties than the new coir. The growth of tomato plant was better on the coir substrate that had been used for 2 years than that on the new coir substrate. The average number of tomato fruits was 108 on the new coir substrate, while it was 179 and 165 on the coir substrate used for 1 and 2 years, respectively. The growth of cherry tomato plant was also better on the coir substrate used for 2 years than that on the new coir substrate. The average number of cherry tomato fruits was 43 on the new coir substrate, while it was 206 and 164 on the coir substrate used for 1 and 2 years, respectively. The growth of brussel sprout was better on the coir substrate used for 3 years than that on the new coir substrate and the average number of brussel sprout leaves was 26.8 on the new coir substrate, while it was 34.3 on the coir substrate used for 3 years. The growth of Korean cabbage improved on the coir substrate used for 1 years compared to the new coir substrate and the number of leaves was 15.1 on the new coir substrate, while it was 24.3 on the coir substrate used for 1 year. Thus, used coir can be recycled to improve vegetable yields compared to using new coirs.

Correlations between the Growth Period and Fresh Weight of Seed Sprouts and Pixel Counts of Leaf Area

  • Purpose: This study was carried out to predict the growth period and fresh weight of sprouts grown in a cultivator designed to grow sprouts under optimal conditions. Methods: The temperature, light intensity, and amount of irrigation were controlled, and images of seed sprouts were acquired to predict the days of growth and weight from pixel counts of leaf area. Broccoli, clover, and radish sprouts were selected, and each sprout was cultivated in a 90-mm-diameter Petri dish under the same cultivating conditions. An image of each sprout was taken every 24 hours from the 4th day, and the whole cultivating period was 6 days, including 3 days in the dark. Images were processed by histogram inspection, binary images, image erosion, image dilation, and the overlay image process. The RGB range and ratio of leaves were adjusted to calculate the pixel counts for leaf area. Results: The correlation coefficients between the pixel count of leaf area and the growth period of sprouts were 0.91, 0.98, and 0.97 for broccoli, clover, and radish, respectively. Further, the correlation coefficients between the pixel count of leaf area and fresh weight were 0.90 for broccoli, 0.87 for clover, and 0.95 for radish. Conclusions: On the basis of these results, we suggest that the simple image acquisition system and processing algorithm can feasibly estimate the growth period and fresh weight of seed sprouts.

Effect of Clay on the Growth and Changes of Mineral Contents of Soybean Sprout (점토광물이 콩나물의 생장과 미네랄 함유량에 미치는 영향)

  • The objective of this research was to investigate the effect of various clay materials on the growth and change in mineral composition of soybean sprout. Further effect of sericite concentration on growth of soybean sprout was optimized. The experiments were carried by using three different viz., loess supernatant, tap water and sericite supernatant. From the obtained results it was found that a significant growth in soybean sprouts was observed while using sericite supernatant this growth was 14.71% and 50% more total growth than that of loess supernatant and tap water. In addition, using sericite supernatant the length of root of soybean sprouts was reached 17.30% and 36.50% longer than that of tap water and loess supernatant. The plausible reason for this growth using sericite supernatant may be due to the presence of various mineral($K^+$, $Ca^+$) in sericite. Further the mineral contents of sericite supernatant were 11.56% and 13.48% more than that for loess supernatant and tap water. Among the various sericite concentration used it was found that 100g/L sericite supernatant showed significant growth rate compared with other fractions. From the obtained results comparing with all materials 100g/L sericite concentration was economic for the cultivation of soybean sprout.

Elucidation of Cause of Cotyledon Black-Decay of Soybean Sprout by Bean Bug, Riptortus clavatus

  • Cotyledon Black Decay (CBD) on soybean sprout mimics the black spot due to microbial infection. CBD, not visible or predictable at seedlot state, for some reason, shows up exclusively on cotyledon of soybean sprout during sprouting process. Such an incidence rate fluctuated from 0.8 to 19.5% over three years from 2004. We suspected some pod-infecting anthracnose fungi and/or pod-blight pathogen, or pod-sucking bean bug, one of the major pests of soybean, might have involved, of which we ruled out fungal pathogen because it was preventable through heat treatment, a proven method for seedlot disinfestation. The healthy seeds artificially fed by bean bug for one to seven days were sprouted, and 6 to 41% of the soybean sprout revealed the CBD mimic to those occurred in soybean sprout from previous commercial seedlot screening experiments. This finding is the first report to confirm that bean bug damage to pod at $R_8$ stage is directly responsible for the CBD, which did not concur with any other deleterious effects on sprouting such as reduction in hypocotyls elongation and rooting except unsightly sprout quality. However, earlier feeding either at green pod or greenish yellow pod stage ($R_6$ -early $R_7$ stage) resulted in rather severe damages, which strikingly reduced hypocotyls growth to about one forth to about two third, as well as the reduction in rates of seed germination.

Difference of Hypocotyl and Growth Characters of Soybean-sprouts by Sowing Date in Sprout-soybean Cultivar (나물용 콩 파종기에 따른 콩나물의 하배축 및 생장특성 차이)

  • This study was conducted to investigate the effects of sowing date on soybean-sprout growth characters and for improving cultural techniques to produce high quality soybean-sprout. Six sprout-soybean cultivars were planted on three sowing dates in 2000 and 2001. The growth of hypocotyl in soybean sprouts was accelerated at higher temperature and the differences among cultivars became greater at later culture days. The longest of hypocotyl and root lengths of soybean sprouts was obtained from July 5 sowing, followed by May 25 and June 15 sowing. Doremikong and Hannamkong had the thickest and the thinness hypocotyls, respectively. Total length of soybean-sprouts was longer in seeds harvested from later sowing, respectively. Eunhakong produced the longest soybean-sprouts, while Tawonkong produced the shortest. Percentage of rotten seed and imperfectly germinated seed harvested at later sowing was lower. Somyeongkong and Tawonkong showed the highest and the lowest percentages of rotten seeds, respectively, while Pungsannamulkong and Hannamkong showed the lowest and the highest percentages of imperfectly germinated seeds. The soybean-sprouts yields were higher in seeds harvested in 2001 and later sowing. Although Eunhakong and Doremikong showed higher soybean-sprout yield, Pungsannamulkong and Somyeongkong showed stable soybean-sprout yield regardless of the sowing dates.
