"A La Recherche" of Functions for the Spore Protein SASP-E from Bacillus subtilis

  • We previously observed that Bacillus subtilis spores from sspE mutants presented a lower germination capacity in media containing high salt concentrations (0.9M NaCl). This deficiency was attributed to the absence of SASP-E (gamma-type small-acid-soluble protein), rich in osmocompatible amino acids released by degradation. Herein we observed that, in addition, this mutant spore presented a reduced capacity to use L-alanine as germinant (L-ala pathway), required longer times to germinate in calcium dipicolinate ($Ca^{2+}$-DPA), but germinated well in asparagine, glucose, fructose, and potassium chloride (AGFK pathway). Moreover, mild sonic treatment of mutant spores partially recovered their germination capacity in L-ala. Spore qualities were also altered, since sporulating colonies from the sspE mutant showed a pale brownish color, a higher adherence to agar plates, and lower autofluorescence, properties related to their spore coat content. Furthermore, biochemical analysis showed a reduced partition in hexadecane and a higher content of $Ca^{2+}$-DPA when compared with its isogenic wild-type control. Coat protein preparations showed a different electrophoretic pattern, in particular when detected with antibodies against CotG and CotE. The complementation with a wild-type sspE gene in a plasmid allowed for recovering the wild-type coat phenotype. This is the first report of a direct involvement of SASP-E in the spore coat assembly during the differentiation program of sporulation.

  • Characteristics and roles of protease released during the germination of Dictyostelium discoideum spores were investigated. When geat activated, the spores germinated, progressively releasing the protease into the extracellular medium. The protease activity exhibited high at pH 2.5. When cyclogeximide was added to culture, complete germination (emergence) and protease release were stopped. Addition of purified nonspecific protease to culture speeded up germination. These results suggest that excreted protease may play a role in removal of the spore wall.

Antifungal Activity of Narceine Methyl Ester and Narceine Isolated from Corydalis longipes Against Some Phytopathogenic Fungi

  • Narceine methyl ester and narceine are potent alkaloids which were isolated from Corydalis longipes were found effective in vitro at very low concentration, i.e., $100{\sim}500\;ppm$ against spore germination of some test plant pathogenic fungi (Alternaria solani, A. tagetica, Cercospora abelmoschi, Curvularia maculans, Erysiphe cichoracearum, E. pisi, Fusarium udum, Helminthosporium oryzae, H. penniseti, Ustilago cynodontis). Among the test, phytopathogens the spores of F. udum, C. maculans and H. penniseti were highly sensitive at 200 ppm. However, spores of E. pisi, A. solani and A. tagetica were less sensitive at low concentration followed by other test fungi. Most of the fungi showed zero or nearly zero percent spore germination at 400 and 500 ppm.

Isolation and Analysis of Temperature Sensitive Mutants in Aspergillus nidulans (Aspergillus nidulans 온도감수성 돌연변이주의 분리 및 분석)

  • About 40 temperature-sensitive mutants have been isolated as a preliminary step to study the spore germination, the cell cycle, and the control of macromolecular synthesis in Aspergillus nidulans. To obtain temperature-sensitive mutants rapidly and effectively, the selective enrichment method using antifungal antibiotic nystatin was developed. Based on the data which had applied to the concentration of auxotrophic mutants by the earlier investigators, the optimal concentration and the time of treatment at the nonpermissive temeprature were determined as 50 to 100 units per ml and 4.5 hr., respectively. Out of 41 ts mutants assigned to the strain symbol PK, thirteen that seemed to be arrested at the earlystage of spore germination were subjected to the further cytological and genetic analysis. Elght of these mutants are able to form germ tube and five not. Staining with acid fuchsin for the 5PK strains shows that one irreversible mutant, PK6 strain able to form germ tube, accumulate mitotic spindle, being arrested in mitosis. Another PK15 and PK23 strain have more than one intact nucleolus without germ tube formation at the restrictive temperature. the temperature-senstive mutation in PK12 strain, the onlystrain which is able occurred in certain gene specific for the germination of spore. All of the ts markers are recessive and complement each other in heterokaryon between two different ts markers at the restrictive temperature.

Effect of the Mixture of Two Plant Alkaloids Isolated from Corydalis longipes Against Balsam Powdery Mildew on Detached Leaves and Pea Powdery Mildew in Field

  • N-Methylhydrasteine hydroxylactam and 1-methoxyberberine chloride, both alkaloids, extracted from Corydalis longipes have been assayed for their activities against two powdery mildews. The spore germination of Erysiphe cichoracearum on detached leaf of balsam(Impatiens balsaminia) following pre- and post-inoculation treatments by their mixture has shown high efficacy against the pathogen at 100, 200 and 300 ${\mu}g/ml$. The mixture was also effective at both pre- and post-inoculation treatments at 500, 1000, 1500 ${\mu}g/ml$ doses against E. pisi causing pea powdery mildew in pea(Pisum sativum) under field conditions. The significant efficacy of the mixture of two compounds against spore germination on detached leaves of balsam and also under field conditions in pea warrants its inclusion in trials against some other diseases under field conditions.

  • Medicinal plants play important roles in controlling plant diseases as one of the safest and ecofriendly methods. These plants have been used in the form of crude extracts as well as active principles in vitro and under field conditions to control plant diseases. Among the active principles, alkaloids have shown significant antifungal activity. We have investigated the effect of two alkaloids viz., (-)-corydahnine and (-)-isocorypahnine isolated from Corydalis chaerophylla, against spore germination of some plant pathogenic and saprophytic fungal spores. Significant inhibition of spore germination at 100 ${\mu}g/ml$ was seen against Curvularia penniseti, Curvularia sp. and Colletotrichum gloeosporioides by (-)-corydahnine but (-)-isocorypalmine was also effective against fungi included in the experiment.

In Vitro Antifungal Activities of 13 Fungicides against Pepper Anthracnose Fungi (고추 탄저병균에 대한 13종 살균제의 살균 활성)

  • For the effective control of pepper anthracnose, in vitro antifungal activities of 13 commercial fungicides were tested on spore germination, mycelial growth, and sporulation of Colletotrichum gloeosporioides, anthracnose fungus. Among them, captan, chlorothalonil, dithianon, fluazinam and folpet completely inhibited the spore germination of C. gloeosporioides at the concentration of 0.8 ${\mu}g/ml$. They were followed by mancozeb and propineb, showing more than 80% inhibition of spore germination at 20 ${\mu}g/ml$. The mycelial growth of C. gloeosporioides was strongly inhibited by fluazinam and nuarimol. Except for nuarimol, most of the fungicides represented more antifungal activity on the spore germination than the mycelial growth of C. gloeosporioides. Azoxystrobin and metominostrobin, Strobilurin fungicides, were only moderately active against the spore germination and the mycelial growth of C. gloeosporioides, but they were effective antisporulant against C. gloeosporioides. From these results, control of pepper anthracnose will have achieved by preventive spray of the commercial fungicides.

  • With microtiter plate, the assay method was developed for detecting the fungicidal activity of new compounds against spore germination, spore adhesion and mycelial growth of Colletotrichum sp. JC24 cal1Sing red pepper anthracnose. Also, the effects of some commercialized fungicides on fungal development like above mentioned were investigated by measuring the optical density of mycelia grown into wells of microtiter plate. For the standardization of assay method, some factors, such as the treatment of MTT and/or propanol, inodulum density and incubation period, affecting on mycelial optical density were investigated. For obtaining precise and consistent mycelial optical density, it was necessary the treatment of MTT for 12 hrs and propanol for 1 hr. inoculum density adjusted to $1\times10^5$ spores/mL and incubation period for 36 hrs at $25^{\circ}C$. For fungicidal activities, 6 protective fungicides, 6 ones inhibiting sterol biosynthesis, and one inhibiting respiration were used in this study. While mancozeb, chlorothalonil and dithianon among 6 protective fungicides inhibited strongly spore germination, adhesion, and mycelial growth at $6.25{\mu}g/mL$, propineb, iminoctadine and fluazinam inhibited intermediately spore germination and mycelial growth at $100{\mu}g/mL$. Washing above 3 fungicides with new PD broth, their activity against spore adhesion decreased. With hexaconazole, tebuconazole and myclobutanil, the tendency of the activity against fungal differentiation of the early infection stage was similar to the latter group of protective fungicides, showing the decrease of the inhibitory activity against spore adhesion by washing 2 hrs after incubation. However, kresoxim-methyl inhibited spore adhesion distinctly, depending on the applied concentrations. Based on these results, it might be able to assess the fungicidal activity of many compounds against spore germination, adhesion and mycelial growth by the use of microtiter plate in vitro. Using the assay developed in this report, it was possible to investigate the inhibitory activity of some commercialized fungicides, too.

  • In order to establish an appropriate spray schedule to reduce the infection, 11 kinds of chemicals were tested for their properties on the inhibition of spore germination at the surface of apple fruits and the duration of the inhibitory effect after spray of each chemical was examined from late June to early September with basically 10 day intervals. Actual control efficacy of each chemical by the 8 successive spray and the patterns of waterborne spore dispersals during that periods were also examined. Combining those results with the meteorological observation data, actual control efficacy of each chemical in the given periods could be estimated. It was revealed that folpet, Brodeaux mixture, mancozeb, oxine copper and imminoctadine-triacetate could be used at any time during the possible infection periods. Captan and dithianon could also be used except the rainy season due to the short duration of inhibitory efficacy against spore germination under heavy rain. However, the usefulness of propineb, benomyl and chlorothalonil against the apple white rot could not be demonstrated in this experiment. Thiram, even though has not been used for apple white rot, can also be used before or after the rainy season to control not only white rot but also alternaria blotch.

In vitro Anti-fungal Activity of Various Hydroxylated Fatty Acids Bioconverted by Pseudomonas aeruginosa PR3

  • The in vitro anti-fungal activity of hydroxylated fatty acids obtained from microbial conversion by Psuedomonas aeruginosa PR3 using ricinoleic acid(RA), eicosadienoic acid(EDA) and conjugated linoleic acid(CLA) as substrates, was investigated. Bioconverted hydroxylated fatty acids showed different anti-fungal activities potentials against the range of phytopathogenic fungi such as Botrytis cinerea, Rhizoctonia solani, Fusarium oxysporum, Sclerotonia sclerotiorum, Colletotricum capsici, Fusarium solani and Phytophthora capsici. RA and EDA showed up to 50% fungal mycelial inhibition at the concentration of $5{\mu}l\;ml^{-1}$. RA, EDA and CLA also exhibited anti-fungal activities with minimum inhibitory concentration(MIC), ranging from 500 to $1000{\mu}g\;ml^{-1}$. Screening was also carried out using varied concentrations of bioconverted RA and EDA for determining the anti-fungal effect on the spore germination of different fungi. Bioconverted RA and EDA showed a considerable degree of spore germination inhibition.
