Pycnogenol, a Standardized Extract of French Maritime Pine Bark, Inhibited the Transcriptional Expression of Th2 Cytokines by Suppressing $NF-{\kappa}B$ Activation in Primary Splenocytes of C57BL/6 Mice with Murine AIDS (Murine AIDS 감염쥐 splenocytes에서 $NF-{\kappa}B$의 활성화 억제를 통한 pycnogenol의 Th2 cytokines의 mRNA 발현 조절 효과)

  • LP-BM5 murine leukemia retrovirus induces the immune dysfunction by imbalanced secretion of Th1 and Th2 cytokines in the murine AIDS model. In the present study, it was investigated whether pycnogenol (Pyc) administration could deactivate $NF-{\kappa}B$ to regulate the gene expression of Th1 and Th2 cytokines in C57BL/6 mice with murine AIDS. Treatment with Pyc for 12 weeks significantly inhibited the loss of body weight and enlargement of spleen and lymph node usually seen with AIDS. Moreover, Pyc increased the plasma level of Th1 cytokines, IL-2 and $IFN-{\gamma}$, while reducing the plasma level of Th2 cytokines, IL-6, IL-10, and $TNF-{\alpha}$. In primary culture of splenocytes, mRNA expression of Th2 cytokines was suppressed, but that of Th1 cytokines was not affected. The LP-BM5 retrovirus infection stimulated the cytoplasmic activation of $NF-{\kappa}B$ and nuclear translocation of $I-{\kappa}B$, whereas Pyc administration significantly reduced $NF-{\kappa}B$ activation and $I-{\kappa}B$ degradation. These results suggested that the inhibitory effect of Pyc on Th2 cytokines in mice with murine AIDS was dependent on suppression of the $NF-{\kappa}B$ signaling pathway and was not dependent on $INF-{\gamma}$ level, which regulates Th2 cytokines.

Effects of Danggwieumja Administration along with Samhwangseje-gamibang on NC/Nga Atopic Mice (당귀음자(當歸飮子)와 삼황세제가미방(三黃洗齊加味方) 병용이 NC/Nga 아토피 생쥐에 미치는 영향)

  • Recently Atopic Dermatitis(AD) is increasing along with allergic disease. At present, there is no infallible cure for AD. Then AD patients undergo great suffering. This study is carried out to see whether or not the administering Danggwieumja(DG) along with Samhwangseje-gamibang(SG} as a medicine for external aplication, is effective in treating atopic dermatitis. To examine the effectiveness of the above prescription, the author made an observation of diverse immune responses. through the model of NC/Nga atopic mice. Results provided evidence that the DG administration along with SG can be used as a treatment means to atopic dermatitis. The results are as follows: The extent of Clinical skin severities in 13 and 16 week old NC/Nga mice treated with DG and SG, were reduced by 50.9%, 53.9% respectively, compared to the control NC/Nga mice with no drug treatment. IgE, IL-4, IL-5, IL-6, IgM and IgG1 levels in the serum of the NC/Nga mice treated with DG and SG were significantly decreased compared to those of the untreated control mice. In contrary, to the $IFN-{\gamma}$ level, significantly increased. The spleen weight of the NC/Nga mice treated with DG and SG significantly decreased compared to those of the untreated control mice. CCR3 gene expression in the skin tissue of NC/Nga mice treated with DG and SG were highly decreased, and the IL-6 expression significantly decreased, and the $IFN-{\gamma}$ gene expression increased compared to those of the untreated control mice. Histological observation of the ear and dorsal skin tissue of the NC/Nga mice treated with DG and SG, showed that the extents of inflammation and infiltrated immune cells in the epidermal tissue and dermis, were highly reduced compared to those of the untreated control mice. In the model inducing COX-2 activity in RAW 264.7 cell, the denser DG became, the more COX-2 activity was inhibited, compared to those of the untreated control group. $IL-1{\beta}$, and $TNF-{\alpha}$, IL-6 gene expression in RAW 264.7 cell with DG, significantly decreased, compared to those of the untreated control group. According to the assessment of cell toxicity in L929 cell, the rate of cell multiplication increased by 3% in consistency to 100ppm of DG compared to the untreated control group and in more than the 200 ppm consistency, cell toxicity was occurred.

  • In order to investigate the effects of glutathione against ceroidosis on flounder, Paralichthys olivaceus, a serial experiment was carried out at indoor tanks in Cheju province, the southern island of R.O.Korea, from October 1 to November 4 of 1991. The choosed group of flounder for this experiment, ranged from 92 to 122g in body weight, already showed a distinct histological ceroidosis in liver, spleen, kidney about six months before this experiment. They were fed on formulated diets supplemented with glutathione of 0mg, 1mg, 2mg, 5mg, 10mg per kg of diet per day. Some flounders were fed on the decomposed diet as a control continuosuly. Two individuals fed on the experimental diets were cought at an interval 7 days for 35 days and analyzed the values of blood indexs. 1. While the values of RBC, Hb, Ht, MCHC, MCV of flounder fed on the decomposed diet and the glutathione non-supplemented diet were not changed compared with the beginning levels of experiment, those fed on the glutathione supplemented diets, showed markedly increased values from 7th day of experiment and from 21th day of experiment reached to the values of healthy flounder. 2. The variation of TP, Glu, Alp, BUN, TG, T-cho, GOT and GPT were not observed in the groups fed on the control diet and glutathione non-supplemented diet. In contrast, those values in the groups fed on the glutathione supplemented diets were markedly decreased from 7th day and become constant from 21th day. 3. The effect of glutathione supplementation with high concentration in diets showed much better improvements of symptoms concequently, compared with those of low concentration diets.

  • This study was designed to investigate the effects of korean pueraria radix extract in Pb administered rats. Forty-two male Sprague-Dawley rats weighing $100{\pm}10g$ were used for this experiment and divided into following 6 groups; control group, 3% pueraria radix group(3% pue.), 100 ppm and 200 ppm alone pb group, 100 ppm and 200 ppm alone pb group with 3% pue. Tissue weight of liver, lung, stomach, heart, kidney and spleen of pb exposed rats were reduced by 3% extracts of pueraria radix group. The Pb content in the rats tissue of pb alone administered group was lower than in the rats tissue of pb group with 3% pue. GPT and GOT were increased in pb-administered group and lower in the 3% extracts of pueraria radix group. LDH was lower in the 3% extracts of pueraria radix-pb group than in the pb group. ChEase was higher in the 3% extracts of pueraria radix group than in the pb alone administered group.

Effects of Dietary Probiotic Mixture on Caecal Microflora, Immune Response, Egg Quality and Production of Layerunder Heat Stress (혼합 생균제가 열 스트레스에 노출된 산란계의 생산성, 계란품질, 면역반응 및 맹장 미생물에 미치는 효과)

  • This study was conducted to investigate the effects of probiotic mixture on fecal ammonia, caecal microorganism, immune response, egg quality and production in layer under heat stress (HS).A total of four hundred 50 week olds Hy-Line Brown layers were randomly divided into four groups of 100 heads each: C (control, room temperature $25^{\circ}C$), HS (heat stress $33^{\circ}C$), PM (HS plus probiotic mixture 500, 750 mg/kg of diets). Egg production, egg quality, spleen weight, blood IgG and lymphocyte concentrations were increased in the PM group compared to the HS group, while mortality, the heterophil:lymphocyte (H:L) ratio, and corticosterone levelswere significantly decreased. Lactobacillus was increased in the PM group compared to the HS group, but E. coli, coliform bacteria and aerobic bacteria were significantly reduced. Fecal ammonia production was significantly increased in the HS group compared to the PM group. In conclusion, the results of this study that these mixed probiotics can reduce the heat damage of the summer laying hens and can be an effective nutritional strategy to reduce odor generation from feces, and to improve egg quality and laying production through immune response and caecal microbial balance.

Effects of inverse lighting and extreme heat diet on short chain fatty acid and blood lipid profile in extreme heat stress-exposed broilers (폭염 브로일러 닭의 혈액지질 및 짧은 사슬지방산에 대한 폭염사료와 역전점등 효과)

  • This study was carried out to investigate the effects of feeding the broilers that are exposed to extreme heat stress by control of inverse lighting times with night restricted feeding of extreme heat diet(EHD1, 2: extreme heat diet) containing different amount of soy oil, molasses, amino acids and vitamin C on short chain fatty acid and blood lipid profile. 300 broiler chickens(Abaica strain) were randomized into four dietary treatment groups according to a randomized block design on the day they were hatched. The four dietary treatment groups were: T1(EHD 1, 10:00~19:00 Dark, 19:00~10:00 Light), T2(EHD 2, 10:00~19:00 Dark, 19:00~10:00 Light), T3(EHD 1, 09:00~18:00 Dark, 18:00~09:00 Light), T4(EHD 2, 09:00~18:00 Dark, 18:00~09:00 Light). The body weight gain of the broilers was highest in T2, and high in order T1, T4, T3(p<0.05). Weights of the lymphoid organ, thymus and bursa of Fabricius were high in T1, T2 as compared to T3, T4 but spleen was lower in T4 than T1, T2, T3(p<0.05). Blood triglyceride, total cholesterol and glucose were higher in T1, T2 than T3, T4(p<0.05). LDL-C was high in orderT4, T3, T2, T1 but HDL-C showed the opposite trend(p<0.05). Blood concentrations of IgG, IgG and IgM were higher in T1, T2 than inT3, T4, but the corticosterone concentration decreased significantly in them. In T1 and T2, Lactobacillus in the feces increased, but total aerobic bacteria, E.coli, coliform bacteria was decreased rather significantly, compared with those in T3 and T4(p<0.05). Concentrations of acetic acid, propionic acid and total SCFA in cecum were high in order T2, T1, T3, T4, but butyric acid, isobutyric acid, valeric acid, isovaleric acid were lower in T1, T2 than in T3, T4 (p<0.05).

  • Sodium chloride plays an important role as the main condiment at daily meal. It is well known that humans require sodium chloride as an essential nutrient to keep the homeostasis of electrolytes. The amounts of salt intake may be a reflection of geography, culture and food habit rather than necessity. Lee has reported (1962) that Koreans ingest high amounts of sodium chloride in their meals, with an intake of excess carbohydrate (80-90% of total Calories) and low protein in their diet. This includes large amounts of rice, Kimchi and other fermented soybean products common in the Korean diet. This investigation was designed to study the dietary relations of sodium chloride to other nutrients in the Korean diet. Twenty four albino male rats, weighing from 290-300g, were divided into four dietary groups according to the amounts of carbohydrate, protein and fat in the basal diet. Each diet contained a rice powder as a carbohydrate source. Diet I was a control diet, Diet II, low protein, Diet III, low protein and low fat diet and Diet IV, low fat diet. All rats were provided with 3% sodium chloride solution. Diet and salt solution were given ad libitum. The experiment was carried out for 9 weeks during which time the body weight, the food intake, and 3% sodium chloride solution consumption were determined. At the 9th week, the urine was collected the blood sample from the artery of each rat for the analysis of sodium and potassium and other chemical studies. The rats were sacrificed and the kidney, adrenal, liver and spleen were measured, and observed changes of the pathological tissue in the kidney and adrenal. The results were summarized as follows: 1) The growth rate was higher in Diet I than in the other experimental diets (II, III and IV) after 4 weeks. There was no significant difference found between the experimental Diets II, III and IV. 2) The daily food intake was greater in the experimental diets II, III and IV than in the control diet. However, there was no difference among the high carbohydrate diets Diet II, III and IV. 3) The daily water (3% sodium chloride solution) intake was also greater in the Diets II, III and IV, than in the control diet. However, there was no difference between Diets II, III and IV. 4) The concentration of sodium and potassium in the blood were within the normal range in all diets. 5) The amount of sodium chloride in the urine was significantly greater in Diets II, III and IV than in the control diet. Diets II, III, IV had a larger amount of sodium solution consumption. 6) Observation of pathological tissue in the experimental diets found a cell proliferation in the glomerlulus of the kidney, while such change was not found in the control diet.

Effect of Antioxidants added perilla Oil Diet on Serum and Tissue in Rats (항산화물첨가 들깨기름식이가 흰쥐의 혈청과 조직에 미치는 영향)

  • This study was done to determine the effect of antioxidants added perilla oil diet on the content of cholesterol, vitamin E, and lipid peroxide in serum and tissue of rats. Four groups of experimental diets, such none added perilla oil diet, ascorbic acid added perilla oil diet, vitamin E added perilla oil diet, EDTA added perilla oil diet were fed ad libitum to the 4 weeks white male rats of Sprague-Dawley strain. The results obtained are summarized as follow: 1) The body weight gain in all experimental diet groups was higher than the control gorup and EDTA added diet group was lower than the other experimental diet group, while food intake in vitamin E added diet group was the highest and vitamin C added diet group was the lowest in the control group. 2) Total cholesterol levels in serum of all experimental diet groups were lower than that of the control group and especially the level of total cholesterol in none added diet group and vitamin C added diet group were significantly lower than that of control group. 3) HDL-cholesterol levels of all experimental diet groups were lower than that of the control group and especially none added diet group was significantly lower than that of control group. 4) The activities of glutamic oxaloacetic transaminase (GOT) in serum of all experimental diet group except EDTA added diet group were higher than that of the control group and especially none added diet group was significanly higher than that of the control group. The activites of glutamic pyruvic transaminase (GPT) in serum of all experimental diet groups except vitamin C added group were higher than that of control group. 5) Vitamin E levels in serum of none added diet group and vitamin C added diet group were lower than that of the control group and vitamin E added diet group and EDTA added diet group were higher than that of the control group. 6) Vitamin E levels in liver of all experimental diet groups were higher than that of control group and especially none added diet group and vitamin E added diet group were significantly higher than that of the control group. 7) Lipid peroxide in serum of all experimental diet group were lower than that of control group and especially EDTA added diet group. 8) Lipid peroxide in liver and spleen of all experimental diet groups were higher than that of the control group and lipid peroxide in kidney of all experimental diet groups except EDTA added diet group were higher than that of the control group. Four these results, as vitamin C, vitamin E and EDTA added diets have an effect to lipid peroxide by antioxidants, it could be suggested that perilla oil diet has required to add antioxidant because it has not sufficient vitamin E for antioxidant and intake and overtake level of perilla oil diet should be studied to go ahead.

Anti-aging and Anti-oxidative Effect of Gongjinhugwon-dan in Early Stages of Aging Rats (공진흑원단(拱辰黑元丹)이 초기노화(初期老化) 흰 쥐의 항노화(抗老化) 및 항산화(抗酸化)에 미치는 영향)

  • 1. Objectives Purpose of this study is to prove anti-aging and anti-oxidative effects of Gongjinhugwon-dan decoction. 2. Methods The SD rats used in this experiment were 6, 18, and 36 weeks old. Each age group was again divided into three groups. These nine groups consisted of 8 rats each. One group was given no treatment, another group was dosed $200{\mu}l$ of normal saline daily, and the last group was dosed $200{\mu}l$ of 1 % Gongjinhugwon-dan and saline mixture. At the conclusion of the experiment, the age groups were relabelled accordingly (10 weeks, 22 weeks, and 40 weeks). After 4 weeks, change of weight and liver markers were measured. Serum LDL cholesterol, total bilirubin, albumin, glucose, GOT and GPT levels were observed in order to check the hematological modification. Also, each organ tissue was biopsied in order to measure the SOD activity and the glutathione content change. 3. Results & Conclusions Aging did not cause any significant change in GOT and LDH, but GPT and albumin levels showed increase after GHD intake. Serum GPT was lower in the experimental group. Serum total bilirubin of the 40 w GHD group was significantly increased. The populations of dendritic cells in the spleens of the GHD groups were significantly increased. The levels of GSH in the liver of the 40 w GHD group and in the kidney of 22w-GSD were significantly increased in comparison with those of the normal groups. The degenerative change of brain tissue was decreased in the 40 w GHD group compared with those of the 40w normal group and the 40 w saline group. These results suggest that anti-oxidative GSH concentration of liver and kidney in rats treated with GHD showed significant increase in the 40 w GHD group. GHD was effective on increasing anti-oxidative substance in liver and dendritic cells in spleen, thus helping immune system and preventing cell mutation and degenerative change of brain tissues. Further studies and clinical investigation with GHD is needed.

Effects of Sopoongsangagambang Administration along with External Spray Treatment on Atopic Dermatitis Development in NC/Nga Mice (소풍산가감방(消風散加減方)과 외치방(外治方) 병용이 NC/Nga 아토피 생쥐에 미치는 영향)

  • Although the parallel prescription of Sopoongsangagambang (SG) administration along with external treatment such as spraying or ointment application on the skin is clinically used for the treatment of atopic dermatitis (AD), molecular mechanism underlying its effectiveness is unknown. Thus in the present study, diverse immune responses in terms of chemical mediators related to AD were investigated using an atopic mouse model NC/Nga after SG administration and external treatment (ET), and major findings are summarized as follows. 1. The clinical severities in 16 and 20 week old NC/Nga mice with SG and ET treatment were decreased to 72.2% and 62.3% respectively compared to the control NC/Nga mice with no drug treatment. 2. IgE, IL-4, IL-5, IL-6, IL-13, IgM, IgG1 and IgG2a levels in the serum of SG and ET treated NC/Nga mouse group were significantly decreased compared to the untreated control mice. In contrast, $IFN-{\gamma}$ showed a significant increase in the experimental group compared to the untreated control group. 3. The spleen weight of SG and ET treated NC/Nga mice was significantly decreased compared to the untreated control group. 4. The B/T ratio in the lymph node of SG and ET treated NC/Nga mice was increased compared to the untreated control group. $CD4^+\;and\;CD8^+$ cell numbers in the lymph node of SG and ET treated NC/Nga mice were significantly increased compared to the untreated control group, but $CD69^+\;and\;CD11a^+$ cells were significantly decreased. 5. mRNA expression levels of IL-4, IL-5, and CCR3 in the skin tissues of SG and ET treated NC/Nga mice were significantly decreased, and expression levels of IL-6, IL-13, $CD69^+/CD3{\varepsilon}^+\;and\;CD19^+/CD44^+$ in the skin tissues of SG and ET treated NC/Mga mice were significantly decreased compared to the untreated control group. $IFN-{\gamma}$ mRNA expression levels were increased compared to the untreated control group. 6. Histological observation of the ear and neck skin tissues showed that the extents of inflammation and infiltrated immune cells in the epidermis and dermis of SG and ET treated NC/Nga mice were highly reduced compared to the untreated control group. 7. Lymphokine assay showed a significant decrease in IL-4 levels in SG and ET treated NC/Nga mice compared to the untreated control group, but the levels of $IFN-{\gamma}$ secretion were significantly increased drug treated NC/Nga mice.
