Effect of supplementation of feed with Flaxseed (Linumusitatisimum) oil on libido and semen quality of Nilli-Ravi buffalo bulls

  • Background: The current study was designed to investigate the effect of supplementation of Flaxseed (Linumusitatisimum) oil on libido and semen quality of Nilli-Ravi buffalo bulls. Methods: In this study, 12 adult healthy bulls kept at the Semen Production Unit, Qadirabad district Sahiwal, were used. These bulls were divided into three equal groups, A, B and C. Group A was kept as control, while in groups B and C supplementation of feed was provided by using flaxseed oil @125 ml/day and 250 ml/day,respectively for 12 weeks. Two ejaculates per animal were collected at 0 day then 5th, 6th, 7th, 8th, 9th, 10th, 11th and 12th week of treatment. In this way a total 216 samples were taken, and each semen sample was evaluated for color, volume, mass activity, percent motility, sperm cell concentration per ml, percentage of live sperm, and plasma membrane integrity. Libido of bulls was also evaluated before every collection. Results: Analysis of data revealed that these parameters were significantly (P < 0.01) increased in flax oil treated animals as compared to control. Conclusion: It was concluded from the present study that flax seed oil has beneficial effects on reproductive health of buffalo bull.

Effects of Egg Yolk from Hens Fed with Acanthopanax senticosus Extract on Physical Endurance and Reproductive Parameters in Rats

  • Acanthopanax senticosus, a member of the Araliaceae used as an invigorant in traditional Korean medicine, is known to relieve physical fatigue. The principal objective of this study was to evaluate the effects of eggs laid by hens fed on Acanthopanax senticosus extracts on physical endurance and reproductive parameters. Male Sprague-Dawley rats at 4 wk of age were divided into 3 groups of 15 rats each: group A (14% egg yolk powder from hens fed with Acanthopanax senticosus extracts+86% AIN-93G), group B (14% plain egg yolk powder + 86% AIN-93G) and group C (7% fat source + 93% AIN-93G), and studied for 5 wk. The rats' physical endurance was measured via forced swimming tests. According to the results, 53.3% rats of group A swam for longer than 20 min, in group B and C, only 48.9% and 46.7% of the rats achieved this. In terms of reproductive parameters, sperm motility was significantly higher in group A than in groups B and C (p<0.05). However, no differences in sperm count were detected among the groups. Additionally, the serum testosterone levels of groups A and B were higher than that of group C. Our findings suggest that rats fed with egg yolk powder from hens fed on Acanthopanax senticosus extracts may improve athletic endurance and reproductive parameters in rats.

Uptake of Mitochondrial DNA fragment into Boar Spermatoza for Sperm-Mediated Gene Transfer

  • Sperm-mediated gene transfer(SMGT) can be used to transfer exogenous DNA into the oocyte at fertilization. The main objective of this study was to assess efficiency of transferring mitochondrial DNA(mtDNA) fragment into boar spermatozoa in either presence or absence of liposome and quality of transfected spermatozoa. The mtDNA of chicken liver was isolated and purified by phenol and alkaline lysis extraction, and it was inserted to plasmid. The genome of transfected spermatozoa treated with DNase I was purified by alkaline lysis, and then amplified by the PCR analysis. After electrophoresis, DNA quantitation of each well was calculated by comparison of the band intensity with standard. As a result, exogenous DNA was composed of mtDNA fragment(1.2 kb) and plasmid(2.7 kb). On the other hand, efficiency of transfection by liposome($9.0{\pm}0.34ng/{\mu}l$) in SMGT was higher than that by DNA solution($6.9{\pm}0.53ng/{\mu}l$). However, there was no significant difference. Transfering exogenous DNA into spermatozoa was completed within 90 min of incubation. In another experiment, there were significant (p<0.05) differences between transfected spermatozoa using both DNA solution and DNA/liposome completes with unheated spermatozoa for viability ($70.8{\pm}1.80$ and $68.0{\pm}2.16%$ vs. $83.3{\pm}1.69%$, respectively) and motility($78.7{\pm}1.59$ and $79.3{\pm}2.14%$ vs. $86.7{\pm}1.59%$, respectively). This study indicates that exogenous mtDNA can be efficiently transferred into boar spermatozoa regardless of the presence of liposome, and transfected spermatozoa can also use insemination and in vitro fertilization to generate transgenic pig.

Relief of the negative effects of heat stress on semen quality, reproductive efficiency and oxidative capacity of rabbit bucks using different natural antioxidants

  • Objective: The potential of extra virgin olive oil (EVOO), betaine (BET), and ginger (GIN), as natural antioxidants, in reducing negative effects of heat stress on physiological responses, antioxidant capacity, semen quality and fertility of bucks under heat stress were investigated. Methods: Forty adult Animal Production Research Institute line rabbit bucks were distributed randomly into four experimental treatments of ten rabbits each. The first treatment was fed the commercial pellet diet (CPD) without supplementation and served as a control. The other three treatments were fed CPD supplemented with EVOO (300 mg), BET (1,000 mg), and GIN (200 mg) per kg diet for 3 consecutive months during the summer season. Results: Supplementation of EVOO, BET, or GIN improved (p<0.05) the sexual desire, progressive motility, vitality, intact acrosome and membrane integrity, sperm cell concentration, sperm outputs and fertility. Seminal plasma total proteins, globulin, total antioxidant capacity, glutathione and glutathione S-transferase, and initial fructose increased (p<0.05), while total lipids, aspartate and alanine aminotransferases and malondialdehyde decreased (p<0.05) compared with the control. In comparing the natural antioxidants treatments, GIN evoked the largest improvement. Conclusion: The inclusion of GIN (200 mg/kg diet) appeared to improve the sexual desire, semen quality and oxidative stress of bucks. This may be a beneficial supplement for the management of rabbit bucks used in natural mating or artificial insemination.

A comparison of five sets of overlapping and non-overlapping sliding windows for semen production traits in the Thai multibreed dairy population

  • Objective: This study compared five distinct sets of biological pathways and associated genes related to semen volume (VOL), number of sperm (NS), and sperm motility (MOT) in the Thai multibreed dairy population. Methods: The phenotypic data included 13,533 VOL records, 12,773 NS records, and 12,660 MOT records from 131 bulls. The genotypic data consisted of 76,519 imputed and actual single nucleotide polymorphisms (SNPs) from 72 animals. The SNP additive genetic variances for VOL, NS, and MOT were estimated for SNP windows of one SNP (SW1), ten SNP (SW10), 30 SNP (SW30), 50 SNP (SW50), and 100 SNP (SW100) using a single-step genomic best linear unbiased prediction approach. The fixed effects in the model were contemporary group, ejaculate order, bull age, ambient temperature, and heterosis. The random effects accounted for animal additive genetic effects, permanent environment effects, and residual. The SNPs explaining at least 0.001% of the additive genetic variance in SW1, 0.01% in SW10, 0.03% in SW30, 0.05% in SW50, and 0.1% in SW100 were selected for gene identification through the NCBI database. The pathway analysis utilized genes associated with the identified SNP windows. Results: Comparison of overlapping and non-overlapping SNP windows revealed notable differences among the identified pathways and genes associated with the studied traits. Overlapping windows consistently yielded a larger number of shared biological pathways and genes than non-overlapping windows. In particular, overlapping SW30 and SW50 identified the largest number of shared pathways and genes in the Thai multibreed dairy population. Conclusion: This study yielded valuable insights into the genetic architecture of VOL, NS, and MOT. It also highlighted the importance of assessing overlapping and non-overlapping SNP windows of various sizes for their effectiveness to identify shared pathways and genes influencing multiple traits.

소 체외성숙 난자에 ICSI에 의한 수정율의 향상에 관한 연구 (Studies on the Improvement of Fertilization and Development Rates Using ICSI with In Vitro Matured Bovine Oocytes)

  • 본 연구는 저정자증 또는 불임축의 수태율 증진과 불임해결에 적용할 목적으로, 일차적으로 소 정자의 PVP, HA의 농도, 신선 및 동결정자, 정자의 농도, 활력, 정자의 수정능획득법 및 난자의 투명대 부착과 미부착별로 현미조작에 의해 난자의 세포질내 단일정자의 주입했을 때 수정율 및 체외발생율을 조사하였다. 1. 소 난자의 ICSI시 PVP 농도를 0.01, 0.02, 0.03, 0.05%별로 처리한 정자를 micropipette내에 흡입하여 세포질내 주입하였을 때 수정율과 분할율은 각각 72.7∼90.9% 및 38.5∼54.5%로서 0.02%농도에서 비교적 높은 수정율과 분할율을 나타냈다. 2. 소 난자의 ICSI시 HA농도를 0.01, 0.02%, 0.02%의 PVP + HA별로 micropipette내에 흡입하여 세포질내 주입하였을 때 수정율과 분할율은 각각 72.7%와 81.8% 및 45.5%와 54.5% 및 83.3%와 37.5%였다. 3. 소 난자에 신선 및 동결정자를 이용하여 ICSI법으로 수정시 수정율은 각각 93.3%, 86.1%였으며, 분할율은 60.0%, 46.7%로서 신선정자를 이용하였을 때가 동결정자를 이용했을 때보다 높은 수정율과 분할율을 나타냈다. 4. Heparin, BFF 및 His법으로 수정능획득 처리한 정자로 IVF 및 ICSI시 수정율은 각각 61.9%, 52.6%, 45.0% 및 85.7%, 78.9%, 65.0%였으며, 분할율은 각각 23.8%, 15.8%, 10.0% 및 61.9%, 52.6%, 50.0%로서, 수정능획득 처리에 있어서 heparin법이 다른 수정능획득 처리법에 높은 수정율과 분할율을 나타났다. 5. 투명대의 부착과 미부착별 난자로 IVF 및 ICSI 법으로 수정시 수정율은 각각 63.2%, 47.8%와 84.2%, 78.3%였으며 분할율은 각각 15.8%, 8.7%와 57.9%, 34.8%였다. 6. 소 체외성숙 난자에 IVF법과 ICSI법으로 수정시켰을 때 수정율은 각각 63.3%, 64.6%와 88.2∼90.0%였으며, 분할율은 각각 26.7%, 29.2%와 52.9%, 67.5%로서 ICSI법으로 수정시켰을 때 수정율과 분할율이 크게 향상되었다.

AndroMed를 이용한 흑우 동결 정액으로 체외수정란 생산 효과 (Effect of Production In Vitro Embryo with Frozen-thawed Semen using AndroMed Extender in Korean Black Cow Semen)

  • The aim of present experiment was to examine commercial synthetic extender(AndroMed) for semen cryopreservation of Korean Black Bull. Semen was collected from a Korean Black Bull using an artificial vagina and transported to the laboratory. The semen was diluted 1:1 by AndroMed. The pellect was diluted to final sperm concentration of $5{\times}10^5/ml$ by doubling in every 10 minutes at $4^{\circ}C$ cold chamber. The semen was equilibrated for 1 hr at cold chamber and packed to 0.5 ml straw. The semen straws were located above 5 cm of liquid nitrogen for 5 minutes, above 5 cm for 10 minutes and above 10 cm for 10 min. And then the frozen straw was plunged to $LN_2$. The presented straws were examined the viability and motility after thawed at $37^{\circ}C$ water bath. Hanwoo semen was used as KPN (Korea Proven Bull Number) in this experiment. The survival rates was significantly higher in fresh semen than frozen semen ($80{\pm}14%\;and\;43{\pm}11%$). However, the motility rates was similar (80.7% and 66.4%). The survival and motility rates were higher in 5cm, 10 min treatment group than the other two groups in straw-located height and duration above $LN_2$ ($50{\pm}14%$ and 70.7% vs, 33.18% and $65{\pm}7%$ vs, 30.14% and 65.7%, respectively). The development rates to cleavage was higher in Black Cow than Hanwoo semen (62.2%, 64.4%), However, The development rates to blastocyst was higher in Hanwoo than Black cow semen (25.9%, 23.0%). In conclusion. The present results that acceptable fertilization and cryopreservation could be obtained by in vitro fertilization with frozen-thawed semen using a synthetic semen extender (AndroMed).

칡소 동결 정액 생산을 위한 스티로폼상자와 액체질소 이용 방법 (The Use of Styrofoam Box for Chikso (Korean Brindled Cattle) Semen Cryopreservation with Liquid Nitrogen)

  • 가축유전자원으로서 동결정액을 생산하는 가장 쉬운 방법은 스티로폼박스를 이용한 간이동결법으로 알려져 있다. 본 연구에서는 스티로폼 동결박스를 제작하여 가축의 동결정액 생산에 활용하는 방법을 검토하였으며 칡소 동결정액 생산을 최적화 할 수 있는 방법을 제시하고자 박스의 크기, 액체질소와 노출된 정액 스트로와 거리, 노출시간 및 생산량을 검토하였다. 2가지 동결박스를 비교하여 자료를 확보하였으며 내부 크기는 세로×가로×높이가 23.5×30.5×22.5 cm와 25.5×46.5×26.5 cm로 측정되었다. 액체질소를 5cm 높이로 채우고 액체질소 위 2, 5 및 8cm 높이에서 동결하여 융해 후 생존성을 조사하였다. 칡소 정액을 동결할 경우, 액체질소와의 노출시간은 모두 10분이 적절하였으며 25.5×46.5×26.5 cm 크기의 상자가 높은 생존율을 보여주었다(60.4±5.3% 대 67.2±3.1%). 동결 상자의 최적화 공간은 정자 동결에 가장 중요한 요소로 판단되며 1회 동결 시 최대 생산 가능한 칡소 동결정액은 60개 이상으로 증가시킬 수 있었다. 이러한 정보를 활용하면, 축종에 따라 동결 정액 생산량 결정하고 목적에 맞는 용기를 활용하여 효율적인 동결정액 생산이 가능할 것으로 판단된다.

Tris-buffer에 첨가되는 당의 종류가 동결ㆍ융해정자의 운동성에 미치는 영향 (Effect of Kind of Sugar Added in Tris-buffer on Motility of Post-thaw Spermatozoa in Canine)

  • 본 연구는 개의 동결정액 제조 시 동결보호 희석액 내에 첨가되는 당의 종류와 조합이 동결융해 후 정자의 생존율 및 운동성에 미치는 영향과 정자동결 시 straw size가 생존율에 미치는 영향에 대하여 조사하고자 실시하였다. 희석액은 Tris-citric acid extender (Tris-buffer)의 기본용액에 20% Egg-yolk, 8% glycerol, 1% Equex STM paste등을 첨가하였으며, 당 성분으로는 monosaccharide(fructose 및 xylose) 및 disaccharide(trehalose)로 구분하여 최종 70 mM의 농도로 첨가 이용하였다. 본 연구에서는 control (fructose, xylose, trehalose), two combination (Fru+Tre, Fru+Xy1, Tre+Xy1) 및 three combination (Fru+Tre+Xy1)으로 구분하여 Tris-buffer에 첨가하였다. Fru+Tre+Xy1처리구에서 CASA분석 후 운동성이 fructose, trehalose, xylose, Fru+Tre, Fru+Xy1, Tre+Xy1 처리구에 비해 가장 높았다 (69% vs 58, 61, 50, 65, 20, 54%). 또한 전진운동율은 Fru+Tre 처리구가 fructose, trehalose, xylose, Fru+Xy1, Tre+Xy1, Fru+Tre+Xy1 처리구에 비해 가장 높았다 (59% vs 47, 55, 42, 13, 49, 44%). 이러한 결과를 바탕으로 Tris-buffer에 F겨+Tre를 첨가하여 실험을 수행하였다. 예비동결 10분에서 0.25$m\ell$ straw를 이용하였을 때 10분 예비동결에 0.5 $m\ell$, 5분 예비동결에 0.25 및 0.5 $m\ell$ straw 처리구보다 유의적으로 높은 생존율을 얻었다 (80+0.0 vs. 65+7, 68+16, 58+8%). 본 연구결과 70 mM Fru+Tre (two combination)가 첨가구가 가장 높은 전진운동율을 얻었으며, 0.25 $m\ell$ straw에 10분간 예비동결을 실시하는 것이 가장 높은 동결융해 후 생존율을 얻을 수 있었다.

Royal Jelly 첨가가 동결융해 후 개 정자의 활력도 및 생존성에 미치는 영향 (Effect of the Addition of "Royal Jelly" on Post-thaw Viability and Longevity of Canine Spermatozoa)

  • This study was conducted to evaluate whether \"Royal jelly\" (RJ) added to Tris-buffer dilute contributed to supporting post-thaw viability and longevity of frozen canine spermatozoa. Two Japanese spitzs (2 to 4 years of age) were used as a semen donor. Semen was collected by manual masturbation and separated into 3 fractions. Only the sperm-rich fraction having sperm motility of more than 70%, containing sperm concentration of 2~4$\times$10$^{8}$ cells/ml and having dead or abnormal spermatozoa of less than 15% was used for the experiment. Each ejaculated semen was centrifuged at 400 $\times$ g for 5 min and then diluted in a Tris-buffer supplemented with 20 ml egg yolk (Ext I), 4% glycero1 and 1% Equex STM Paste (Ext II) or g1ycero1, Equex STM paste and RJ of various concentrations (Ext II-RJ). After freezing and thawing, viability of spermatozoa in Ext II -RJ containing 1% RJ immediately after thawing (67.5$\pm$9.6) was significantly lower than that of Ext II , Ext II -RJ containing 0.01 or 0.1% RJ (77.5$\pm$12.5, 78.7$\pm$8.2 and 80.0$\pm$6.3). However, Ext II-RJ containing 0.1% RJ yielded higher viability than Ext II, Ext II-RJ containing 0.01% at or 1% 1 h after thawing (69.5$\pm$8.1 vs. 55.0$\pm$12.9, 57.5$\pm$9.6 and 41.5$\pm$12.6; P<0.05). At 1 h after thawing, the viability of spermatozoa thawed in 7$0^{\circ}C$ (68.8$\pm$12.5) was significantly higher than that of spermatozoa thawed in 38$^{\circ}C$ (48.8$\pm$16.3), although there was no difference in the viability between both groups immediately after thawing (77.5$\pm$9.6 and 81.3$\pm$8.1). Post-thaw viability and longevity of post-thaw spermatozoa in Ext II-RJ containing 0.1% RJ was higher in those in Ext II at 1 h (65.0$\pm$12.9 vs. 42.5$\pm$12.6), 2 h (52.5$\pm$12.6 vs. 27.5$\pm$17.1) and 3 h (40.0$\pm$14.1 vs. 20.0$\pm$12.1) after thawing. These results indicated that addition of 0.1% af to Tris-buffer enhanced post-thaw viability and longevity of canine spermatozoa and this additive can be used for increasing the possibility of collision between spermatozoa and ova during insemination.emination.
