The Kinetics of Protease Production by Bacillus licheniformis (Bacillus licheniformis에 의한 단백질분해효소 생산 Kinetics)

  • Carbon sources and nitrogen sources are known to be very important in protease production by microorganisms. The effects of carbon source and nitrogen source on protease biosynthesis by Bacillus licheniformis were investigated using batch cultures. As initial carbon and nitrogen concentrations of culture medium increased, the specific growth rate of Bacillus licheniformis was increased, while the specific protease production rate was decreased. From the results of batch cultures, a mathematical model which considers the effects of carbon source and cnitrogen source was proposed and the methods to increase the productivity of protease were discussed.

Regional Business Cycles in East Asia: Synchronization and its Determinants

  • This paper analyzes the dynamics and nature of regional business cycle synchronization for East Asian countries in the period of 2000:Q1-2011:Q4. Estimating a dynamic two-factor model extracts the common factor and the nation-specific factor from both the macroeconomic aggregates and plausible driving forces of regional business cycles. Evidence for regional business cycle synchronization is particularly strong for Korea, Malaysia and the Philippines, while Japan shows weak evidence of regional synchronization. On the other hand, Indonesia, Thailand, Singapore and China are decoupling from regional business cycles. The driver of monetary aggregate is the most significant determinant of regional fluctuations of macroeconomic aggregates, whereas oil price and productivity are on average important driving forces of nation-specific fluctuations of real economic activities.

  • The fiber reinforced composite materials is widely used in aircraft, space structures and robot arms because of high specific strength and high specific modulus. The on-line cure monitoring during the cure process of the composite materials has become an important research area for the better quality and productivity. In this paper, the dielectric circuit of the wheatstone bridge type for measuring the dissipation factor was designed and manufactured. Also, the dielectric sensor for the cure monitoring of the high temperature composites was developed. The residual thermal stresses of the dielectric sensor were analyzed by the finite element method and its dielectric characteristics under high temperature were evaluated. The on-line cure monitoring of the BMI resin was performed using the wheatstone bridge type circuit and developed high-temperature dielectric sensor.

  • Continuous production of hydrogen by Ca alginate-immobilized photosynthetic bacteria was studied in a packed-bed bioreactor. The dilution rate and input concentration of carbonaces substrate were selected as operating parameters. To choose the strain for immobilization, hydrogen productivities of Rhodopseudomonas caposulata 10006 and Rhodospirillum rubrum KS-301 were compared through preliminary batch cultures of their free cells: the former was found to show better hydrogen productivity in spite of its lower specific growth rate. For the continuous production of hydrogen by immobilized R capsulata, the optimum dilution rate was about 0.84 h$^{-1}$ . The Immobilized tells gave better hydrogen yield and conversion efficiency than free ones. And a kinetic parameter K'$_{m}$ was determined for the packed-bed bioreactor, being practically constant for a specific range of dilution rates.s.

  • In agricultural households cultivating vegetables and fruits, the use of various pesticides to protect crops from diseases and pests or to control weeds is widely practiced enhancing quality and productivity. However, pesticides can pose a threat to consumer health by remaining on the food surface or migrating into the food interior. Households commonly peel off skins, wash with water, or use chemical methods to remove foreign substances including residual pesticides on the food surface. In this study, we measured the washing rate by comparing the pesticide concentrations before and after washing in the leafy vegetable perilla leaves and the fruits strawberries and apples, which were intentionally exposed to pesticides. We compared washing rates using tap water, a baking soda solution, and a commercially available food-specific cleaning solution. The target pesticides for analysis were azoxystrobin, bifenthrin, boscalid, difenoconazole, flubendiamide, and indoxacarb, and the residual pesticide analysis was performed using GC-MS/MS or LC-MS/MS. The removal rates of pesticides were highest with the food-specific cleaner, followed by baking soda and tap water in order.

  • The evaluation and analysis method of information system (IS) is studied from the system perspective, the user perspective, and the management viewpoint. The detailed analysis method performs qualitative evaluation by user questionnaire or expert opinion. In this study, Measures the productivity and the effect of building administrative information systems. In the previous study, qualitative productivity and universal effect indicators were used, but in this study, quantitative productivity indicators and indicators specific to administrative complaints were selected. KONEPS, an administrative service system, used electronic contract records and information recorded in the intermediate process. The information was converted into the number of days, and the productivity based on the input manpower was calculated. The effect analysis analyzed the questionnaire related to civil affairs, which is the goal of the administrative work system. Each factor was divided into reflective structural variable and formal structural variable, and internal consistency and multi-collinearity were diagnosed. In order to verify the model, the influence of the work was set as a hypothesis, the reliability was verified according to the descriptive statistics method, the influence was measured through the regression analysis, and the model was analyzed by the multiple regression model path coefficient. Model validation methods are Chi-square (df, p), RMR, GFI, AGFI, NFI, CFI and GFI as indicators according to CFA.

  • This paper proposes a new forging work-in-process control system that guarantees independency of a specific device and provides cost-effectiveness. Until now, much research has been studied on improving the process productivity through efficient work-in-process or inventory controls. Especially, incorporating various IT technologies such as barcode, RFID, and image recognition has been done. However, those approaches cause many problems due to the characteristics of the forging work-in-process control environment. To overcome the limitations of the existing approaches, this paper proposes a novel forging work-in-process control system. The proposed system in this paper identifies and precisely manages positions of objects by using GPS information of smart mobile devices. Therefore, identification tags as well as specific devices for reading the tags do not been required. It resolves the problems of the previous approaches and enhances the productivity of the overall forging work-in-process control process.

Study on minerals status of dairy cows and their supplementation through area specific mineral mixture in the state of Jharkhand

  • Background: Deficiency of macro and micro-minerals in the ration of dairy cows adversely affects growth, milk production and reproduction efficiency. It is essential to examine mineral concentrations in feeds offered to dairy cows in practical farms. Methods: Two villages from each taluka were selected at random for taking representative samples of feeds, forages and hair. Within the village, help was sought from village milk producers and district animal husbandry officer for identification of 4 to 5 farmers and collection of representative samples. All the samples were processed and analyzed for chemical composition as well as major macro and micro-minerals, using Inductively Coupled Plasma-Optical Emission Spectrometer. Results: Ca content in wheat straw (0.29%), crushed maize (0.02%) and wheat bran (0.12%) was found to be below the critical level (0.30%). The P content in concentrate ingredients was high (0.26-0.96%), but low in dry roughages (0.06-0.12%). Cereal straws (0.14%) and grains (0.12%) were deficient in Mg. Feeds and forages were found to be adequate in K (1.50%). Cereals straws were found to be deficient in S (0.11%). Greens were good source of Cu (12.02 ppm). Wheat straw was found to be low in Zn (18 ppm), but high in Mn (225 ppm) and Fe (509 ppm). Local grasses and azolla green were found to be rich source of Co (>1.00 ppm). Se (0.63 ppm) was present in appreciable quantities in most of the feedstuffs. Conclusions: From the present study, it was apparent that the feeds and forages available in the state of Jharkhand may not meet the requirements for Ca, P, Mg, Cu, Zn and Co in order to sustain a milk production of ~10 kg/day. Therefore, it is necessary to supplement these deficient minerals through area specific mineral mixture in the ration of dairy cows for improving productivity and reproduction efficiency.

Operational Strategy for Increasing Ethanol Production in Repeated Fed-batch Ethanol Fermentation Using Saccharomyces cerevisiae (Saccharomyces cerevisiae 를 이용한 반복 유가식 ethanol 발효에서 ethanol 생산량을 증가를 위한 운전 전략)

  • We designed the optimal operational strategy in repeated fed-batch ethanol fermentation using Sacchromyces cerevisiae ATCC 24858 in views of ethanol yield, specific ethanol production rate, and ethanol productivity, when the aeration rate were controlled at 0.0 and 0.33 vvm. Coincidentally, the time intervals of withdrawal-fill of culture medium (24 and 36 h) were investigated. Ethanol yield and ethanol productivity when the aeration was carried out at 0.33 vvm were superior to those when the aeration was not carried out. Additionally, those parameters when the time interval of withdrawal-fill of culture medium was 24 h were superior to those when time interval of withdrawal-fill of culture medium was 36 h. The total ethanol production reached at the greatest value, 703.8 g-ethanol, when the aeration was carried out at 0.33 vvm and the time interval of withdrawal-fill of culture medium was 24 h. In this study, we verified experimentally the necessity of designing the operational strategy for increasing ethanol production in terms of aeration rate and time interval of withdrawal-fill of culture medium in the repeated fed-batch ethanol fermentation.

Relationship between Cell Size and Specific Thrombopoietin Productivity in Chinese Hamster Ovary Cells during Dihydrofolate Reductase-mediated Gene Amplification

  • When parental Chinese hamster ovary (CHO) cell clones that are capable of producing thrombopoietin (TPO) were subjected to high methotrexate (MTX) concentrations, clonal variations in cell growth were apparent. In the clones that had no significant enhancement in specific TPO productivity (q$\_$Tpo/)when a higher level of MTX was administered, their growth was not depressed significantly nor their cell size changed significantly. On the other hand, those clones that showed a significant-enhancement in q$\_$Tpo/ at higher a MTX dosage, cell growth was depressed initially but recovered during successive sub-cultures. Furthermore, their cell size increased, which suggested that changes in cell size may be indicative of an enhanced q$\_$Tpo/. When the enhancement of the q$\_$Tpo/ of 9 clones after a high MTX dosage was plotted against the extent of the increase of their size, there was a linear correlation (γ$^2$=0.80, p<0.001, ANOVA), which suggested that an enhancement of q$\_$Tpo/ after high MTX administration can be measured by the increase in their cell size. Taken together, our data demonstrate that the selection of amplified CHO cell clones with enhanced q$\_$Tpo/ can be done upon their increased size and growth pattern. This facilitates the development of highly productive recombinant CHO cell lines.
