The aim of the present study was to assess the oxygen transfer rate and oxygen uptake rate in antibiotic fermentation. As a model study, cultures of Streptomyces kanamyceticus in a complex medium were analyzed to evaluate the oxygen transfer and uptake rates using oxygen balance technique. Quantitative evidence for the effect of oxygen transfer rate on the volumetric antibiotic production was clearly demonstrated. The oxygen uptake rates and the specific oxygen requirements were significantly changed with culture time. Those phenomena were indicative of biological turnover in the antibiotic fermentation.
In order to achieve high-level expression of interferon-${\alpha}1$ (IFN-${\alpha}1$) during fed-batch fermentation of recombinant E. coli, effects of oxygen supply and induction temperature on the expression of recombinant proteins were evaluated. Supplementation of oxygen and its transfer into cells is one of the most important parameters involved in the design and operation of mixing-sparging equipment for bioreactors. Generally, higher oxygen supply stimulates cell growth of aerobic microorganism and consequently the amount of products is increased. In this study, the optimum aeration strategy for the higher production of IFN-${\alpha}1$ during fed-batch fermentation of recombinant E. coli was surveyed. The growth of the cells was also monitored with four different concentrations of dissolved oxygen (DO; limiting, 20%, 35%, 50%) conditions. The DO was controlled by varying aeration rates of air and pure oxygen. Oxygen uptake rate (OUR) and specific oxygen uptake rate (SOUR) were evaluated and compared for the enhanced growth and induction of the cells and IFN-${\alpha}1$, respectively. We confirmed that increased DO by additional oxygen supply, up to 35%, can improve the production of IFN-${\alpha}1$ during the fermentation.
The growth kinetics of Streptomyces noursei NRRL 5126 was investigated under different aeration and agitation combinations in a 5.0 l stirred tank fermenter. Poly-${\varepsilon}$-lysine biosynthesis, cell mass formation, and glycerol utilization rates were affected markedly by both aeration and agitation. An agitation speed of 300 rpm and aeration rate at 2.0 vvm supported better yields of 1,622.81 mg/l with highest specific productivity of 15 mg/l.h. Fermentation kinetics performed under different aeration and agitation conditions showed poly- ${\varepsilon}$-lysine fermentation to be a growth-associated production. A constant DO at 40% in the growth phase and 20% in the production phase increased the poly-${\varepsilon}$-lysine yield as well as cell mass to their maximum values of 1,992.35 mg/l and 20.73 g/l, respectively. The oxygen transfer rate (OTR), oxygen utilization rate (OUR), and specific oxygen uptake rates ($qO_2$) in the fermentation broth increased in the growth phase and remained unchanged in the stationary phase.
The objectives of this research are to evaluate and compare the oxygen transfer coefficients($K_{La}$) in both a general bubbles reactor and a micro-nano bubbles reactor for effective operation in sewage treatment plants, and to understand the effect on microbial kinetic parameters of biomass growth for optimal biological treatment in sewage treatment plants when the micro-nano bubbles reactor is applied. Oxygen transfer coefficients($K_{La}$) of tap water and effluent of primary clarifier were determined. The oxygen transfer coefficients of the tap water for the general bubbles reactor and micro-nano bubbles reactor were found to be 0.28 $hr^{-1}$ and 2.50 $hr^{-1}$, respectively. The oxygen transfer coefficients of the effluent of the primary clarifier for the general bubbles reactor and micro-nano bubbles reactor were found be to 0.15 $hr^{-1}$ and 0.91 $hr^{-1}$, respectively. In order to figure out kinetic parameters of biomass growth for the general bubbles reactor and micro-nano bubbles reactor, oxygen uptake rates(OURs) in the saturated effluent of the primary clarifier were measured with the general bubbles reactor and micro-nano bubbles reactor. The OURs of in the saturated effluent of the primary clarifier with the general bubbles reactor and micro-nano bubbles reactor were 0.0294 mg $O_2/L{\cdot}hr$ and 0.0465 mg $O_2/L{\cdot}hr$, respectively. The higher micro-nano bubbles reactor's oxygen transfer coefficient increases the OURs. In addition, the maximum readily biodegradable substrate utilization rates($K_{ms}$) for the general bubbles reactor and micro-nano bubbles reactor were 3.41 mg COD utilized/mg active VSS day and 7.07 mg COD utilized/mg active VSS day, respectively. The maximum specific biomass growth rates for heterotrophic biomass(${\mu}_{max}$) were calculated by both values of yield for heterotrophic biomass($Y_H$) and the maximum readily biodegradable substrate utilization rates($K_{ms}$). The values of ${\mu}_{max}$ for the general bubbles reactor and micro-nano bubbles reactor were 1.62 $day^{-1}$ and 3.36 $day^{-1}$, respectively. The reported results show that the micro-nano bubbles reactor increased air-liquid contact area. This method could remove dissolved organic matters and nutrients efficiently and effectively.
This paper deals with the influence of chemical oxygen demand to nitrogen ratio ((COD/N) ratio) on the performance of an membrane bioreactor. We aim at establishing relations between COD/N ratio, organisms' distribution and sludge properties (specific resistance to filtration (SRF) and membrane fouling). It is also essential to define new criteria to characterize the autotrophic microorganisms, as the measurements of apparent removal rates of ammonium seem irrelevant to characterize their specific activity. Two experiments (A and B) have been carried on a 30 L lab scale membrane bioreactor with low COD/N ratio (2.3 and 1.5). The obtained results clearly indicate the role of the COD/N ratio on the biomass distribution and performance of the membrane bioreactor. New specific criteria for characterising the autotrophic microorganisms activity, is also defined as the ratio of maximum ammonium rate to the specific oxygen uptake rate in the endogenous state for autotrophic bacteria which seem to be constant whatever the operating conditions are. They are about 24.5 to 23.8 $gN-NH_4{^+}/gO_2$, for run A and B, respectively. Moreover, the filterability of the biological suspension appear significantly lower, specific resistance to filtration and membrane fouling rate are less than $10^{14}m^{-2}$ and $0.07\;10^{12}m^{-1}.d^{-1}$ respectively, than in conventional MBR confirming the adv < antage of the membrane bioreactor functioning under low COD/N ratio.
A quantitative physiological approach has been employed to estimate the metabolic parameters such as specific uptake rates of nutrients and specific production rate in continuous culture of Pseudomonas elodea for gellan gum production. The estimated values of metabolic parameters are used for process improvement. During the exponential growth phase, the specific growth rate was 0.16hr-1 in batch culture. The gellan gum concentration increased up to 0.7g dry weight/100g broth and the apparent viscosity of the culture broth was about 4,500 cp.(72hrs culture). The ratio of specific uptake rate of carbon to that of nitrogen were found to be optimum at about 3.0mg-carbon/mg-nitro-gen. With the improved medium, the maximum gellan production rate, 0.6g dry weight/1/hr, was obtained at D=0.14 hr-1. The shear stresses of culture broth were fairly well correlated with shear rates by using Casson equation and at highly viscous culture broth, oxygen transfer coefficient was greatly reduced.
Growth kinetic parameters for mass cultivation of Chinese Hamster Ovary (CHO) cells are estimated by measuring oxygen uptake rates. It Is found that there is strong correlation between cell growth and oxygen consumption, showing that correlation factor is 0.83. Derived linear model predicts actual cell density very well. It tells that oxygen uptake rate can play important role in indirectly measuring cell density when conventional method of estimating cell density is no longer meaningful due to heavy cell clumpings. Cell yield per oxygen consumption, $Y_{\chi}o$ and mass transfer coefficient for oxygen, Ka are also estimated as 1.26$\times$10$^4$cells/mmole $O_2$ consumed and 1.01/h, respectively. Average specific growth rate over all runs is 2.891/day for CHO cells with producting 2 grams of tPA per day under continuous perfusion operations.
Batch test methods have developed for a long time to measure kinetic and stoichiometric parameters which are required to perform steady state design and mathematical modelling of activated sludge processes. However, at various So/Xo ratios, abnormal behaviors of ordinary heterotrophic organism in batch tests have been reported in many researches. Thus, in this research, abnormal behaviors of heterotrophs in batch tests were investigated at various So/Xo conditions by measuring and interpreting oxygen utilization rate. As So/Xo ratio increased, the calculated values of maximum specific growth rates, ${\mu}_{H,max}$ and $K_{MP,max}$, increased. However, at a certain point of So/Xo (around 10mgCOD/mgMLAVSS), ${\mu}_{H,max}$ and $K_{MP,max}$ values started to decrease. According to this observation, three prominent behaviours of heterotrophs were identified at various So/Xo conditions. (1) At low So/Xo region (below 5 mgCOD/mgMLAVSS), the oxygen utilization rate of heterotrophs in batch tests were almost stable and consequently yielded lower maximum specific growth rate. (2) At high So/Xo region (up to 5~10 mgCOD/mgMLAVSS), oxygen utilization rate incresed sharply with time and indicated more upward curvature than the predicted OUR with conventional activated sludge model, which consists of single hetetrotrophs group. Thus, in this region, competition model of two organisms, fast-grower and slow-grower, seemed to be appropriate. (3) At extremely high So/Xo region (over 10mgCOD/mgMLAVSS), significant oxygen utilization rate was still observed even after depletion of readily biodegradable COD. This might be caused by retarded utilization of intermediates which were generated by self inhibition mechanism in the process of RBCOD uptake.
Recently, entomopathogenic nematodes have received a considerable attention as biologicalcontrol agents. For in vitro cultivation, storage and transportation of nematodes, oxygen supply isextremely impotant due to its limited solubility and mass transfer problem. The oxygen uptake rates(OURs) of four different Steinernema species were measured in a 5L bioreactor at varying temperatures.The OURs of the Steinernema spp. were below 0.5 x mmolO'||'&'||' . min in the range of 13-17$^{\circ}$C. TheOURs (mmo102/L - min) of S. glaseri Dongrae and S. carpocapsae Pocheon strains were 0.4 x lo-', 0.75x lo-\ulcorner at 21$^{\circ}$C, 1.5 x lo-\ulcorner, 3.2 x 10-2 at 25"C, and 2.8 x lo-', 5.8 x lo-\ulcorner at 29"C, respectively. However,the OURs were not significantly altered by the agitation speed of 50-150 rpm. The specific oxygenuptake rates (qol) of S. glaseri NC, S. glaseri Dongrae, S. glaseri Mungyeong and S. carpocapsaePocheon strains were 0.3 x 0.5 x 0.3 x and 0.2 x mmolO~/cell min at 25"C,respectively. As the nematode size and temperature were increased, the qo, was also increased.the qo, was also increased.

To improve the ability of oxygen transfer in Biological Fluidised Bed(BFB) processes, air lift aeration system was introduced, experimental investigations were performed for the oxygen transfer in reactor, the fluidisation as to Biomass Volatile Solids variation and the relationship between substrate removal rate and oxygen utilization. The experiments for this purpose were executed for the synthetic wastewater by continuous type reactor at $20^{\circ}C$ using reticulated polypropylene sheets as media. The obtained results showed that the oxygen transfer by air lift aeration would be more effective than any other aeration systems used in BFB reactor. Also, it has observed that the critical biomass concentration in reactor took a range of 20 to 23g/l. Applying cages to BFB reactor, biomass would be maintained uniformly in the bed and the fluidisation characteristics of media could be improved. Varying F/M ratio from 0.36 to 0.73, BOD removals were 91% or more. Therefore, this process was suited to the treatment of which F/M ratios are variable and specific oxygen uptake rates ($K_r$) were 0.23 to 0.26g $O_2/g\;VSS{\cdot}day$ at range of 15 to 20g BVS/l.
