Ohmic Heating Characteristics of Fermented Soybean Paste and Kochujang (된장 및 고추장의 Ohmic heating 특성)

  • Ohmic heating is a food processing operation in which heat is internally generated within foods by the passage of alternating electric current. The process enables highly viscous paste foods such as Kochujang, and fermented soybean paste to be heated very fast. In order to develope the novel pasteurization process of paste foods, static Ohmic heating system was built, and heating characteristic during Ohmic heating under various conditions were studied. Electric conductivities of Kochujang and fermented soybean paste at room temperature were 1.865 S/m and 2.510 S/m, respectively and increased linearly with increasing temperature. Specific heating rate was highly dependent on the frequency. The highest heating rate was achieved at 5 KHz for Kochujang and 20 KHz for fermented soybean paste. Uniform heating throughout the sample was achieved during Ohmic heating with low frequency electrical currents, however above 5 KHz frequency, surface temperature was several degrees higher than the bulk.

Study of the Indicators of Dampness-Phlegm Pattern Identification Based on Tentative Korean Standard Differentiation of the Symptoms and Signs for Stroke (한국형 중풍 변증 표준시안의 습담 변증 지표에 대한 연구)

  • Objectives : This study was done to investigate clinical frequency and correlation among the indicators of dampness-phlegm pattern identification settled by tentative Korean Standard Differentiation of the Symptoms and Signs for Stroke. Methods : The subjects were 147 hospitalized patients with stroke, and a list of registry was made for each of them. Among the five types of pattern identification, fire-heat, dampness-phlegm, blood-stasis and deficiency of Qi and of Yin, those that have shown a high frequency in dampness-phlegm type were categorized as the dampness-phlegm pattern group. Frequency of dampness-phlegm indicators was compared with those from the non-dampness-phlegm pattern group. Correlations among dampness-phlegm indicators were also studied. Results : 1. Dampness-phlegm pattern group included 26 patients out of 147. 2. Among the indicators of dampness-phlegm pattern. those, in order of highest frequency, were 'tiredness or sluggishness', 'white Coated tongue' and 'sputum'. 3. In comparing dampness-phlegm pattern group with non-dampness-phlegml group, the indicators such as 'lightheadedness', 'nigrescence', 'sputum', 'dermatic dysaesthesia' showed significantly high frequency. 4. Among the indicators, 'sputum' and 'tiredness or sluggishness', 'sputum' and 'yellow coated tongue', and 'white coated tongue' and 'yellow coated tongue' showed significant correlation. 5. In investigation of the correlation of scale in symptoms, various results such as positive correlation and negative correlation were obtained. Conclusion : In this study, more sensitive indicators of dampness-phlegm pattern identification were found. Based on these results, it is suggested that a more practical Korean Standard Differentiation of the Symptoms and Signs of Stroke would be established through continuous clinical studies by giving weight on each specific type of pattern identification.

The effects of antibodies and complement in macrophage-mediated cytotoxicity on metacercariae of the lung fluke, Paragonimus westeymani (폐흡충(Paragonimus Tuestermani) 피낭유충에 대한 대식세포의 세포독성에 있어서 항체 및 보체가 미치는 영향)

  • Paragonimus westermani is a tissue migrating parasite in the early stage until arriving at lung, and most of the parasites spend their life spans there. Considerable immune responses including activation of macrophages are taken place during the residence of parasites in the host. However, concerning the immunologic defense mechanisms of the host against this parasite, only a few document is available so far. In this study, the cytotoxic effect of peritoneal macrophages under the presence of antibody and/or complement against metacercariae of F. westermani was investigated in vitro. Metacercarlae were collected from the crayfish, Cambaroides similis and hatched out in Tyrode solution (pH 7.4). Plastic adherent cells from normal or infected rat (Wistar) peritoneal exudates were used as experimental macrophages. Polyclonal antibodies were obtained from infected rats and a cat. Cat IgG was fractioned with ion exchange chromatography. Fresh rabbit complement was used according to experimental scheme. Various combinations of peritoneal macrophages, normal or infected rat serum, complement and cat IgG were incubated at $36^{\circ}C$ in 5% $CO_2$ incubator for 6, 14, 24 and 48 hours. The results obtained were as follows: 1. P. westermani infection activated peritoneal macrophages non-specifically and this activation induced increases of cell adherence and cytotoxicity on metacercariae. 2. In the presence of infected rat serum the antibody.dependent cell-mediated cytotoxicity of peritoneal macrophages on metacercariae was significantly increased and showed a peak at 6-hour incubation. But the cytotoxic effect was markedly reduced after inactivation of complement and heat.labile IgE antibody by the heating of infected serum at 56$^{\circ}C$ for 30 minutes. 3. The highest cytotoxic effect (100%) of concomitant incubation with IgG and complement showed 24 hours after incubation, although cell adherence was relatively low at 6-hour incubation and 0% at 24-hour incubation. 4. Coordinative functions of complement with serum and IgG were effective in cell adherence and in cytotoxicity, but it is not clear the independent role of complement on the macrophage- mediated cytotoxicity in this study- With these results it is assumed that P. westermani infection can induce the non-specific activation of peritoneal macrophages, and strum antibodies including IgE antibody might enhance the cytotoxicity by macrophages,

  • This experiment was carried out to investigate the effects of monosaccharides extracted by saturated portland cement solution on the cement setting in comparision with the inhibitory index (I) of each lignocellulosic-cement system. The wood species which have been widely reforested in Korea, Populus alba-grandulosa, Larix leptolepis, Abies holophylla, Pinus koraiensis, Pinus rigida, Pinus densiflora and agricultural wastes of rice husk and rice stalk were used at this study. The wood meal, 0.50g on dry weight basis, through 0.83 mm(20 mesh) and retained on 0.35mm (40 mesh) screen was extracted by 25 ml saturated portland cement solution and the pH of saturated portland cement solution Was 12.7. To eliminate cation exsisting in the extracted solution, the cation exchange column was used (Fig. 4). Afterwards the extracted monosaccharides were reduced into alditols with sodium borohydride and analyzed by the gas-liquid chromatography for xylan, mannan, arabinan, galactan, gluean. The heat of cement hydration for lignocellulosic-cement system was measured in Dewar flask (Fig. 2). And then the inhibitory indices were calculated from maximum hydration temperature, time and maximum slops of hydration curve of ligno cellulosic-cement systems. The results obtained were as follows; (1) The inhibitory index of pines-Pinus rigida (I=29.33) and Pinus densiflora (I=35.76), were lower than that of poplar-Populus alba-glandulosa (I=41.48), and the index of Larix ieptoiepis (I=73.00) was the highest among eight lignocellulosic-cement systems, and accordingly both Pinus rigida and Pinus des(flora were seemed to be good wood species for wood-cement composite manufacture. (2) In case of Pinus rigida, the inhibitory index was 29.33 and the ratio of the hexoses to the pemoses was 6.04 and in case of Larix leptolepis, the index and the ratio were 73.00 and 35.19, respectively. Therefore the inhibitory index increased with increasing the ratios of the hexoses to the pentoses. (3) The richer amount of xylose and mannose in species caused decreasing the slops of the hydration curve of the lignocellulosic-cement system, prohahly due to the chemical adsorption of the acetyl groups in the hemicellulose on the surface of cement grains. (4) The amoun of xylose and mannose were significant to the inhibitory index of each lignocellulosic-cement system but any specific relation between the amount of glucose and inhibitory index was not found.

Properties and Activities of Nireogenase System of Azospirillum amazonensa Kp1 (Azospirillum amazonense Kp1의 질소고정효소계의 활성 및 특성)

  • The maximum nitrogen fixation activity of the associative, microaerobic and acid tolerant bacteria, Azospirillum amazonense Kp1 was obtained with 0.2Kpa of $O_{2}$ and showed a reversible inhibition by the higher concentrations. Ammonium treatment caused a gradual inhibition of the activity up to 350mM. The nitrogenase systems were purified by gradient chromatography on DEAE-52 cellulose, heat treatment and preparative PAGE. The MoFe protein showed molecular weight of 210,000 including two nonidentical subunits with apparent molecular weights of 55,000 and 50,000 and an isoelectricpoint of 5.2 and contained 2, 24 and 28 atoms of Mo, Fe and acid labile S per molecule. The Fe protein revealed molecular weight of 66,000 including two types of subunits with molecular weights of 35,000 and 31,000 and an isoelectric point of 4.6, and contained 4 atoms of Fe and 6 atoms of S per molecule. The maximum specific nitrogenase activity attained 2,200 and 1,700nM $C_2H_4mg^{-1} min^{-1}$, respectively for MoFe and Fe proteins at pH7 and $35^{\circ}C$. The activity was lost after 10 and 30 days under the cold room ($4^{\circ}C$) condition for Fe and MoFe proteins, respectively.

  • This study uses beacons and smartphone Global Positioning System (GPS) receivers to establish a location-based disaster/hazard prediction system. Beacons are usually installed indoors to locate users using triangulation in the room, but this study is differentiated from previous studies because the system is used outdoors to collect information on registration location and temperature and humidity in hazardous areas. In addition, since it is installed outdoors, waterproof, dehumidifying, and dustproof functions in the beacons themselves are required, and in case of heat and humidity, the sensor must be exposed to the outside, so the waterproof function is supplemented with a separate container. Based on these functions, information on declining and vulnerable areas is identified in real time, and temperature/humidity information is collected. We also propose a system that provides weather and fine-dust information for the area concerned. User location data are acquired through beacons and smartphone GPS receivers, and when users transmit from declining or vulnerable areas, they can establish the data to identify dangerous areas. In addition, temperature/humidity data in a microspace can be collected and utilized to build data to cope with climate change. Data can be used to identify specific areas of decline in a microspace, and various analyses can be made through the accumulated data.

Molecular Cloning, Characterization and Expression of a Novel Trehalose-6-phosphate Synthase Homologue from Ginkgo biloba

  • In many organisms, trehalose acts as protective metabolite against harsh environmental stresses, such as freezing, drought, nutrient starvation, heat and salt. Herein a cDNA (designated as GbTPS, GenBank Accession Number AY884150) encoding a trehalose-6-phosphate synthase homologue was isolated and characterized from the living fossil plant, Ginkgo biloba, which is highly tolerant to drought and cold. GbTPS encoded an 868-amino-acid polypeptide with a predicted isoelectric point of 5.83 and molecular mass of 97.9 kD. Amino acid sequence alignment revealed that GbTPS shared high identity with class II trehalose-6-phosphate synthase homologues (67% identical to AtTPS7), but had only 17% and 23% of identity with OstA from Escherichia coli and ScTPS1 from S. cerevisiae, respectively. DNA gel blot analysis indicated that GbTPS belonged to a small multi-gene family. The expression analysis by RT-PCR showed that GbTPS expressed in a tissue-specific manner in G biloba and might involve in leaf development. GbTPS was also found to be induced by a variety of stresses including cold, salt, drought and mannitol.

  • Recently, the eddy current testing(ECT), alternating current field testing, magnetic flux leakage testing and remote field testing have been used as a nondestructive evaluation method based on the electromagnetic induction phenomenon. The eddy current testing is now widely accepted as a NDE method for the heat exchanger tube in the electric power industry, chemical, shipbuilding, and military. The ECT system mainly consists of the synthesizer module, analog module, analog-to-digital converter, power supplier, and data acquisition and analysis program. In the previous study, the synthesizer module and the analog module which is essential to the ECT system were primarily developed, and in this study the data acquisition and analysis program were developed. The operation system for this program is based on the Windows 7, and optimized for the Korean users, and the specific feature of this program using setup wizard enables inspector to make a setup easily for acquisition and analysis of ECT data. In this paper, the configuration and functions of eddy current data acquisition and analysis program will be introduced.

Characteristics of Yersinia enterocolitica Isolated from Foods (식품에서 분리한 Yersinia enterocolitica의 특성조사)

  • The incidence of Yersinia enterocolitica in raw meat was determined over 10 month period. Y. enterocolitica was isolated from 8.5% of beef, 17.0% of pork and 25.6% of chicken. Overall prevalence of Y. enterocolitica in raw meat was 17.5%. Seasonal difference was observed in isolation rate in which pork and chicken samples collected in the second half of the year twice was more than that of the first half of the year. Serotypes of Y. enterocolitica isolates were O:5 (17.3%), O:8 (8.6%), O:3 (6.2%), O:1,2 (1.2%), and others. The antibiotics susceptibility tests showed Y. enterocolitica isolates were resistant to carbenicillin, ampicillin, erythromycin, penicillin and bacitracin. In contrast it showed sensitivity to polymyxin B, kanamycin, ciprofloxacin, gentamicin, trimethoprim/sulfamethoxazole, nalidixic acid, and tetracycline. PCR with specific primers derived from ail gene of Y. enterocolitica was applied to detect and confirm pathogenic Y. enterocolitica. About 10% of the isolated Y. enterocolitica proved to be pathogenic and most of them were found in pork. However, proper cooking and meat process can kill and remove all Y. enterocolitica in meat, because the organism is very sensitive to heat.

  • An attempt has been made to optimize elongated rotating arc plasma $NH_3$ scrubber. Among diverse semiconductor processes, diffusion and implantation process inevitably produce $NH_3$ as byproduct and efficient dry process for the decomposition of $NH_3$ is required. Plasma process does not produce NOx that is commonly produced in combustion process and there is no problem of deactivation, usually experienced in catalyst process. However, plasma process uses electrical energy and needs to be optimized to achieve feasibility of application. In this work, mode control of rotating arc is presented as tentative solution for the possible optimization of the process. Based on existing rotating arc, scale-up and following mode mapping was tried. Proposed reactor design was evaluated in the $NH_3$ decomposition process and revealed that optimization scheme is at hand. In the experiment of full scale scrubber including heat exchanger, the process gave more stable and efficient process of $NH_3$ decomposition.
