The Mycobiota of Air Inside and Outside the Meju Fermentation Room and the Origin of Meju Fungi

  • The fungi on Meju are known to play an important role as degrader of macromolecule of soybeans. In order to elucidate the origin of fungi on traditional Meju, mycobiota of the air both inside and outside traditional Meju fermentation rooms was examined. From 11 samples of air collected from inside and outside of 7 Meju fermentation rooms, 37 genera and 90 species of fungi were identified. In outside air of the fermentation room, Cladosporium sp. and Cladosporium cladosporioides were the dominant species, followed by Cladosporium tenuissimum, Eurotium sp., Phoma sp., Sistotrema brinkmannii, Alternaria sp., Aspergillus fumigatus, Schizophyllum commune, and Penicillium glabrum. In inside air of the fermentation room, Cladosporium sp., Aspergillus oryzae, Penicillium chrysogenum, Asp. nidulans, Aspergillus sp., Cla. cladosporioides, Eurotium sp., Penicillium sp., Cla. tenuissimum, Asp. niger, Eur. herbariorum, Asp. sydowii, and Eur. repens were collected with high frequency. The concentrations of the genera Aspergillus, Eurotium, and Penicillium were significantly higher in inside air than outside air. From this result and those of previous reports, the origin of fungi present on Meju was inferred. Of the dominant fungal species present on Meju, Lichtheimia ramosa, Mucor circinelloides, Mucor racemosus, and Scopulariopsis brevicaulis are thought to be originated from outside air, because these species are not or are rarely isolated from rice straw and soybean; however, they were detected outside air of fermentation room and are species commonly found in indoor environments. However, Asp. oryzae, Pen. polonicum, Eur. repens, Pen. solitum, and Eur. chevalieri, which are frequently found on Meju, are common in rice straw and could be transferred from rice straw to Meju. The fungi grow and produce abundant spores during Meju fermentation, and after the spores accumulate in the air of fermentation room, they could influence mycobiota of Meju fermentation in the following year. This could explain why concentrations of the genera Aspergillus, Eurotium, and Penicillium are much higher inside than outside of the fermentation rooms.

Genome-based identification of strain KCOM 1265 isolated from subgingival plaque at the species level

  • The aim of this study was to identify strain KCOM 1265 isolated from subgingival plaque at the species level by comparing 16S ribosomal RNA gene (16S rDNA) and genome sequences. The whole genome of strain KCOM 1265 was extracted using the phenol-chloroform extraction method. 16S rDNA was amplified using polymerase chain reaction and sequenced using the dideoxy chain termination method. Pairwise genome comparison was performed using average nucleotide identity (ANI) and genome-to-genome distance (GGD) analyses. The data showed that the percent similarity of 16S rDNA sequence of strain KCOM 1265 was 99.6% as compared with those of Fusobacterium polymorphum ATCC 10953T and Fusobacterium hwasookii KCOM 1249T. The ANI values of strain KCOM 1265 with F. polymorphum ATCC 10953T and F. hwasookii KCOM 1249T were 95.8% and 93.0%, respectively. The GGD values of strain KCOM 1265 with F. polymorphum ATCC 10953T and F. hwasookii KCOM 1249T were 63.9% and 49.6%, respectively. These results indicate that strain KCOM 1265 belongs to F. polymorphum.

  • Antimicrobials were one of the great invention of modern era. However, the abuse of antimicrobial both in human and animals has led to a high rate of occurrence of antimicrobial resistant microbes. Disease treatment caused by antimicrobial resistant microbes including superbacteria has emerged as critical issue worldwide. Communication and cooperation among researchers in diverse fields are needed to solve the resistance to antimicrobials. Culture Collection of Antimicrobial Resistant Microbes (CCARM) has taken a leadership role an intermediary among various research fields by providing certified antimicrobial resistant microbes with their information since 1999. CCARM collects antimicrobial resistant microbes from clinical, agricultural animals and products, and environmental fields, and classifies and stores them according to their origins, species and antimicrobial resistance mechanisms. CCARM is performing the roles (collection, deposit, preservation, distribution, service, and consulting) of Biological Resource Center designated by Organisation for Economic Co-operation and Development.

  • 치악산의 개미상을 조사하기 위해서 1997년부터 1998년까지 2년간 3개의 코스에서 746 colony를 채집하였다. 해부현미경하에서 관찰 동점한 결과 4아과 23속 45종이 확인되엇으며, 이들 중 10종은 강원도의 미기록 종이었으며, 특히 Myrmica sp.6은 한국의 미기록종으로 추가된다. 이로 인해 강원도에 분포하는 개미종은 4아과 28속 73종이 된다.

구실바위취의 신조합명 및 계통 유연관계 (A new combination for Saxifraga octopetala (Saxifragaceae) and its phylogenetic relationship)

  • 본 연구에서는 간혹 흰바위취(Micranthes manchuriensis)와 동일종으로 취급되기도 하는 한국 고유종 구실바위취(Saxifraga octopetala)의 분류학적 지위를 알아보고자 수행되었다. 구실바위취의 계통학적 위치와 종의 경계를 확인하기 위해 두 종의 기준표본에 대한 형태적 검토와 핵 리보솜 DNA의 ITS 지역 염기서열에 대한 집중적인 계통분석을 시행하였다. 총 65개 구실바위취 개체는 ITS 계통수에서 하나의 무리를 이루면서 Micranthes 분기군(clade)에 포함되었고, 톱바위취 및 흰바위취와 가까운 계통 유연관계를 나타내었다. 중국과 러시아에서 채집된 다수의 흰바위취 개체 역시 독자적인 분기군을 형성하면서 M. nelsoniana var. pacifica 및 M. fusca와 자매군을 이루었다. 구실바위취의 실체가 모호했던 것은 흰바위취 개체와 톱바위취의 화서를 함께 포함하고 있는 복합표본인 Wilford의 채집품(흰바위취의 기준표본)을 Nakai가 잘못 관찰하였기 때문으로 생각되었다. 구실바위취와 흰바위취의 높은 형태적 유사성에도 불구하고 이들은 지하 포복경의 특징에 있어서 차이를 보였다. ITS 계통수에서 구실바위취가 Micranthes 내에 자리하는 것으로 나타나 구실바위취는 Micranthes 내에서 종의 지위를 유지하여야 할 것으로 판단되었고, 이에 따라 신조합명(Micranthes octopetala)을 발표하였다.

한국산 꽃구름버섯속의 분류학적 연구 (Taxonomic Study on Korean Stereum)

  • 현재까지 국내에 보고된 꽃구름버섯은 도합 5종으로서 그중 4종은 흰테꽃구름버섯(Stereum gausapatum), 꽃구름버섯(S. hirsutum), 갈색꽃구름버섯(S. ostrea), 및 배착꽃구름버섯(S. ochraceo-flavum)이며, 나머지 1종은 거북꽃구름 버섯속에 속하는 너털거북꽃구름버섯(Xylobolus spectabile)으로서 낭상체의 형태적 특징에 의하여 거북꽃구름버섯속(Xylobolus)에 속하여 왔으나, Boidin과 Chamuris는 배양학적 특징에 의하여 후자를 꽃구름버섯속(Stereum)으로 분류하고 있다. 본 연구에서는 "너털거북꽃구름버섯"(X. spectabile)을 꽃구름버섯속에 재분류하여 분류체계를 개선하고 우리말 이름을 "너털꽃구름버섯"(S. spectabile)으로 개칭하는 바이다. 또한 서울대학교 표본실(Seoul National University Fungal Collection, SFC)에 보관되어 있는 꽃구름버섯을 대상으로 조사한 결과 5종의 꽃구름버섯종들이 미기록종으로 동정되었으며, 이들 미기록종은 Stereum subtomentosum(갈색털꽃구름버섯, 신칭), S. peculiare(껍질 꽃구름버섯, 신칭), S. sanguinolentum(유혈꽃구름버섯, 신칭), S. striatum(줄무늬꽃구름버섯, 신칭), 및 S. complicatum(복합꽃구름버섯, 신칭)이었다. 이로서 한국산 꽃구름버섯 종들은 도합 10종에 이르며 본 연구를 통하여 국내 꽃구름 버섯의 동정을 위하여 새로운 검색표를 제시하였다.

The photochemical reactions of iron species in rain and snow in Higashi-Hiroshima, Japan

  • This paper describes the concentrations of total dissolved iron (tFe) and $Fe^{2+}$ in rainwater and snow, the relationship of Fe species with other metals and ions in bulk rainwater, and the $Fe^{2+}$ generation mechanism in aqueous samples in rainwater of time series collection. Volume weight mean concentrations of tFe and $Fe^{2+}$ were 3.22 and $1.25{\mu}gL^{-1}$ in bulk rainwater, and 50.1 and $43.5{\mu}gL^{-1}$ in snow, respectively. $Fe^{2+}$ was significant fraction to the tFe, accounted for 3.25-93.4% of the tFe in rainwater and 87% in snow. We also investigated temporal variations of tFe, $Fe^{2+}$, other metals and ions in rainwater of time series collection during rain event. Although the concentration range of tFe was different from those of other species, a decreasing trend of tFe from the beginning of the rain event was similar with other species. However, though $Fe^{2+}$ did not show such a decreasing trend, $Fe^{2+}$/tFe was in good correlation with solar radiation. From the results of multiple linear regression analysis and thermodynamic calculations (Mineql+), $Fe^{2+}$ in our samples may be generated from photochemical reduction of $Fe^{3+}$ species (such as $Fe(OH)^{2+}$,$Fe(OH)^{2+}$ and Fe-oxalate) at daytime.

Taxonomic Study on Six Yeast Species Unlisted in the National Species List of Korea

  • More than five hundreds of yeast species (including 9 variants) encompassing 142 genera and 48 classes of 2 phyla exist in Korea. However, only 173 species have been cataloged in the National Species List of Korea (NSLK), the backbone reference to claim sovereign rights over biological resources, as of December 2021, due to the lack of taxonomic descriptions, although some of these species are extensively used in industry. The present pilot study investigated the taxonomy of strains belonging to the six most widely used or frequently isolated yeast species (Meyeromyma guilliermondii, Saccharomyces cerevisiae, Saccharomycopsis fibuligera, Wickerhamomyces anomalus, Candida tropicalis, and Papiliotrema flavescens) to include these species in the NSLK. Strains with diverse habitats and geographic origins were retrieved from the National Institute of Biological Resources culture collection. These strains clustered in the same clade as the type strains of the designated species according to phylogenetic analysis of the D1/D2 sequences. Moreover, we described the cell morphology and physiological characteristics of representative strains of each species. This study suggests that these six species are indigenous to Korea and can be accordingly listed in the NSLK.

  • A total of 82 genera and 271 species of soil-inhabiting fungi including saprobic, nematode-trapping, and arbuscular mycorrhizal but plant pathogenic fungi published hitherto in South Korea are listed with the information on geographic location, habitat, vegetation when available, and relevant literatures.
