Microbial Diversity in Swamp

  • The revolution in molecular biology has given us greatly increased ability to obtain and to modify biological resources and to use them for the benefit of all humankind. The sequencing and the associated analysis of gene functions for a growing number of genomes will have an unprecedented effect on the uses of biological resources and the need for access to them. To investigate the diversity of microbial community in swamp, molecular systematic methods were applied. By amplified rDNA restriction analysis (ARDRA) and rDNA partial sequence analysis, $75\%$ of the isolates were known species. In case of uncultured analysis, almost all the selected clones were new species candidate. Especially archea and uncultured bacterial analyses, all clones were new taxon candidates. As for the eukaryotic diversity, several yeast form cultures were isolated from various samples of swamp. Among them, about $60\%$ of the isolates were easily identified. In case of a new species candidate, most strain were included in hymenomycetal yeasts.

New Records of Blastobasidae (Lepidoptera) from Korea, with Description of a New Species

  • A new species of Blasobasinae, Blastobasis spiniella Park, sp. nov., is described, and four species: Neoblastobasis spiniharpella Kuznetzov et Sinev, Hypatopa silvestrella Kuznetzov, hypatopa tianshanica Sinev, and Holcoceroides scythrella Sinev ave reported for the first time in Korea. Description, diagnosis, illustrations of the male genitalia, and collection data of these species are provided.

Universal-, Genus-specific, Species-specific Probes and Primers Design for Microbial Identification

  • MIPROBE is a web-based tool for design of universal, genus-specific, and species-specific primers and probes. The main functions of MIPROBE are collection of target gene sequences, construction of consensus sequences, collection of candidate primers and probes, and evaluation of candidates by BLAST. Biologists with little computer skills can easily use MIPROBE to design large-scale universal, genus-, and species-specific primers and probes. This software is available at http://www.miprobe.com. Also detailed descriptions of how to use the program are found at this site.

Notes on Cordyceps species Collected from the Central Region of Nepal

  • The present study was carried out to explore the Cordyceps species and other entomopathogenic fungal flora around Kathmandu Valley and a few high altitude locations of Nepal. In this paper, we report eight Cordyceps species as new to Nepal: C. gracilis, C. ishikariensis, C. liangshanensis, C. martialis, C. militaris, C. pruinosa, C. sphecocephala and C. tricentri. We also mention a few allied genera such as Beauveria, Hirsutella and Paecilomyces from Nepal. Further collections from different ecological regions of Nepal will show the richness of entomopathogenic fungal floral diversity of Nepal.

Hypoxis aurea Lour. (Hypoxidaceae): a Rare Species from Jeju Island which is Rediscovered Seventy Years after its First Collection in Korea

  • We described and illustrated a rare species in Korea, Hypoxis aurea Lour. (Hypoxidaceae) which was rediscovered about 70 years after its first collection from Jeju island in Korea. The members of the family Hypoxidaceae R. Br. are distinguished from the plants of Amaryllidaceae J. St-Hill. by having grass-like leaves, an invisible stem which is modified into a corm or a rhizome, trimerous, and radially symmetric flowers with an inferior ovary developing into a capsule on scapes. Hypoxis aurea Lour. is readily distinguishable from Curculigo orchinoides Gsertn. in Japan by beakless ovary and capsular fruit. The number of somatic chromosome is 2n=54.

Developing species-specific quantitative real-time polymerase chain reaction primers for detecting Lautropia mirabilis

  • This study aimed to develop Lautropia mirabilis-specific quantitative real-time polymerase chain reaction (qPCR) primers based on the sequence of DNA-directed RNA polymerase subunit beta gene. The PrimerSelect program was used in designing of the qPCR primers, RTLam-F4 and RTLam-R3. The specificity of the qPCR primers were performed by conventional PCR with 37 strains of 37 oral bacterial species, including L. mirabilis. The sensitivity of the primers was determined by qPCR with the serial dilution of purified genomic DNA of L. mirabilis KCOM 3484, ranged from 4 ng to 4 fg. The data showed that the qPCR primers could detect only L. mirabilis strains and as little as 40 fg of genome DNA of L. mirabilis KCOM 3484. These results indicate that this qPCR primer pair (RTLam-F4/RTLam-R3) may be useful for species-specific detection of L. mirabilis in epidemiological studies of oral bacterial infectious diseases such as periodontal disease.

우리나라 일부 해안 지역 야생화들로부터 분리한 효모들의 분자 생물학적 동정 (Identification of Yeasts Isolated from Wild Flowers Collected in Coast Areas of Korea Based on the 26S rDNA Sequences)

  • 국내 자연환경으로부터 다양한 효모들을 분리, 동정하고 나아가 이들로부터 유용물질을 생산하는 효모자원을 확보하기 위한 연구의 일환으로 우리나라 동해안, 서해안, 남해안에 서식하는 야생화들을 채집하여 이들로부터 효모들을 분리한 후 분자생물학적 방법으로 동정하였다. 동해시에서 수집한 야생화로부터는 Candida silvae 등을 포함한 15종에 속하는 27균주의 효모들을 분리하였다. 서해안의 대천시 해수욕장 주위 야생화에서는 Bulleromyces albus를 비롯한 17종 34균주가 분리, 동정되었다. 또한 남해안의 완도군 대문리 주위의 야생화들로부터는 Cryptococcus flavus를 포함하여 13종에 속하는 효모 22 균주들이 분리 동정되었다. 전체적으로 우리나라 동해안, 서해안, 남해안의 야생화로부터 모두 45종에 속하는 효모들 83균주를 분리, 동정하였다.

  • 벼 이앙후에 발생되는 잡초를 조사하여 벼와의 경쟁적 영향을 해석함에 있어 가장 적은 노력으로 전체잡초발생상태를 대표할 수 있는 최소표본크기(minium quadrat size)와 최소표본회수(minimun quadrat number)를 초종일표본변적(species-area) 및 초종일표본회수(species-quadart number) 관계식에 의해 조사하였다. 1. 초종일표본면적, 또는 초종일표본회수 관계식에서 다같이 표본면적, 또는 표본회수가 증가하게 됨에 따라 얻어지는 초종증가는 처음에는 급속도로 증가하고, 서서히 증가속도가 완만해지며, 어느 한계점을 지나면 더 이상의 초종증가는 없이 평형상태를 유지하는 특징적인 곡선을 보였다. 2. 표본추출면적, 또는 표본 추출회수를 10% 증가시킴에 따라 얻어지는 초종 증가율이 10%되는 관계를 고려한 최소표본추출 회수는 3회였다.
