Monitoring and evaluation of provincial classical swine fever immunization implementation with an E2 subunit vaccine in Jeju Island, South Korea

  • Purpose: Accidental vaccination with a live attenuated low-virulence strain of Miyagi (LOM) vaccine led to the reemergence of classical swine fever virus (CSFV) in Jeju province, South Korea in 2014. To control the continual outbreaks of LOM-derived CSFV, the provincial government launched a provincial mass vaccination project using a CSF-E2 subunit vaccine. We conducted this study to assess the herd immunity level and outcomes of E2 vaccine-based immunization in breeding and growing herds on Jeju Island during 2020-2021. Materials and Methods: A large-scale vaccination trial using the Bayovac CSF-E2 vaccine investigated its efficacy in breeding and growing herds under farm application conditions (10 CSFV-affected and three CSFV-naïve swine farms). Results: The level of herd immunity in each farm was classified into three (S1-S3) and six (G1-G6) profiles in breeding and growing herds, respectively. Immunity monitoring revealed a remarkable improvement in the herd immunity status in all farms. The majority (10/13) of farms, including CSFV-free farms, showed the S1G1 immunity profile in 2021, indicating the appropriate implementation of the advised vaccination regime. Moreover, there were significant decreases in Erns seropositivity from 100% to 50% and 25.9% to 4.3% at farm and pig levels, respectively. In particular, all farms were confirmed as CSFV free in the growing-finishing herds. Conclusion: Our large-scale trial demonstrated the effectiveness of the E2 subunit vaccine in establishing herd immunity stabilization and eliminating CSFV circulation in the affected farms and highlighted the need for a provincial vaccination policy to regain the CSF-free status on Jeju Island.

Tangeretin Improves Glucose Uptake in a Coculture of Hypertrophic Adipocytes and Macrophages by Attenuating Inflammatory Changes

  • Obesity is characterized by a state of chronic low-grade inflammation and insulin resistance, which are aggravated by the interaction between hypertrophic adipocytes and macrophages. In this study, we investigated the effects of tangeretin on inflammatory changes and glucose uptake in a coculture of hypertrophic adipocytes and macrophages. Tangeretin decreased nitric oxide production and the expression of interleukin (IL)-6, $IL-1{\beta}$, tumor necrosis $factor-{\alpha}$, inducible nitric oxide synthase, and cyclooxygenase-2 in a coculture of 3T3-L1 adipocytes and RAW 264.7 cells. Tangeretin also increased glucose uptake in the coculture system, but did not affect the phosphorylation of insulin receptor substrate (IRS) and Akt. These results suggest that tangeretin improves insulin resistance by attenuating obesity-induced inflammation in adipose tissue.

Effects of Liquid Pig Manure on Yield and Nutrition Supply of Green Manure Crops in Jeju

  • The effects of liquid pig manure (LPM) on the yield and nutrient supply of green manure crops, such as hairy vetch alone and hairy vetch + rye mixture, were investigated. We used three treatments: no-fertilizer (NF); LPM, $4.0t10a^{-1}$ ($N-P_2O_5-K_2O=4.0-0.024-4.0kg10a^{-1}$); and chemical fertilizer (CF; $N-P_2O_5-K_2O=4.0-0-4.0kg10a^{-1}$). The green manure crops, namely, Vicia villosa Roth. (hairy vetch) and Secale cereale L. (rye). were sown on March 11 and harvested on June 24, 2014. Yield of green manure crops significantly increased in hairy vetch + rye mixture plot compared with the hairy vetch alone plot. In hairy vetch alone plot, yield of green manure crops was increased by application of LPM and CF, but there were no significant differences. Yield of green manure crops in hairy vetch + rye fresh mixture plot significantly increased by the LPM and CF treatments, but no difference was noted between the LPM and CF treatments. Nitrogen supply by the green manure crops was higher in hairy vetch + rye mixture plot than hairy vetch alone plot. However, there was no significant difference. Phosphorus and potassium supply significantly increased in hairy vetch + rye mixture plot compared with hairy vetch alone plot. By treating LPM and CF, the nitrogen, phosphorus and potassium supply were higher than those in no fertilizer. In conclusion, LPM could replace CF. When planting green manure crops, the hairy vetch + rye mixture was more effective than hairy vetch alone to effectively use green manure crops.
