Chemical coagulation and sonolysis for total aromatic amines removal from anaerobically pre-treated textile wastewater: A comparative study

  • The present study primarily focuses on the evaluation of the comparative effect of chemical coagulation and ultrasonication for elimination of aromatic amines (AAs) present in anaerobically pretreated textile wastewater containing different types of dyes including azo dyes. Color and COD reduction was also monitored at the optimized conditions. The production of AAs was measured spectrophotometrically in the form of total aromatic amines (TAAs) and also verified with high performance liquid chromatography (HPLC) selectively. A composite coagulant, magnesium chloride (MC) aided with aluminium chlorohydrate (ACH) in an equal ratio (MC + ACH) was utilized during the coagulation process, which yielded 31% of TAAs removal along with 85% of color and 52% of COD reduction. At optimized power (200 W) and sonication time (5 h), an appreciable TAAs degradation efficiency (85%) was observed along with 51% color reduction and 62% COD removal using ultrasonication. The chromatographic data indicate that sulphanilic acid and benzidine types of aromatic amines were produced after the reductive cleavage of utilized textile dyes, which were effectively mineralized after ultrasonication. The degradation followed the first order kinetics with a correlation coefficient ($R^2$) of 0.89 and a first-order kinetic constant (k) of $0.0073min^{-1}$.

니오솜을 이용한 $[^{3}H]$아시클로버의 경피투과 (Transdermal Permeation of $[{^3}H]Acyclovir$ Using Niosome)

  • Niosomes are vesicles formed from synthetic non-ionic surfactants, offering an alternative to chemically unstable and expensive liposomes as a drug carrier. Non-ionic surfactant and cholesterol mixture film leads to the formation of vesicular system by hydration with sonication method. The formation of niosome was ascertained by negative staining of TEM. The entrapment efficiency of niosomal suspension was gradually increased with increasing the ratio of cholesterol to surfactant. It was found that the niosome with 6 : 4 (polyoxyethylene 2-cetyl ether: cholesterol) ratio was more stable than those with other ratios. The topical application of acyclovir(ACV) in the treatment of herpes simplex virus type 1(HSV-1) skin disease has a long history. There are an increasing number of reports, however, in which topical ACV therapy is not as effective as oral administration. Lack of efficacy with topical ACV has been hypothesized to reflect the inadequate delivery of drug to the skin. We investigated the permeation of niosome containing $[^{3}H]ACV$ in hairless mouse skin using Franz diffusion cell model. Permeation coefficient(P) of aqueous ACV was $6.7{\times}10^{-4}\;(cm/hr)$ and that of ACV in niosome was $23.4{\times}10^{-4}\;(cm/hr)$, suggesting about 3.5 times increase in the transdermal permeation.

초음파 처리 활용 실시간 투과전자현미경 관찰용 금속 시편 전사 방법에 관한 연구 (A Study on the Method of Transferring Metal Specimens for Real-time Transmission Electron Microscopy using Ultrasonic Treatment)

  • Micro-electromechanical systems (MEMS) based in-situ heating holders have been developed to enable high resolution imaging of heat treatment analysis. However, unlike the standard 3 mm metal disk specimens used in the furnace-based heating holder and general transmission electron microscopy holder, the MEMS-based in-situ heating holder requires thin specimens that can be penetrated by electrons to be transferred onto the MEMS chip. Previously, focused ion beam milling was used to transfer metal specimens, but it has the disadvantage of being expensive and the risk of specimen damage due to gallium ions. Therefore, in this study, we devised a method of transferring metallic materials by ultrasonic treatment using a transmission electron microscopy specimen made by electro jet polishing. A 3mm electropolished metal disk was placed in an appropriate solution, ultrasonicated, and then drop casted. The transfer of the specimen was successful, but it was confirmed that dislocations were formed inside the specimen due to ultrasonic treatment. This study provides a novel method for transferring metallic materials onto MEMS chips, which is cost-effective and less gallium ion damaging to the specimen. The results of this study can be used to improve the efficiency of heat treatment analysis using MEMS-based in-situ heating holders.

활성탄관에 포집된 극성유기용제의 탈착효율에 관한 연구 (A Study on Desorption Efficiency for Polar Solvents Collected on Charcoal Tube)

  • This study was performed to evaluate factors affecting desorption of organic solvents collected on charcoal tube and to find out the optimum condition. Desorption efficiency for polar analytes was improved when several polar desorption solvents such as methanol, dimethylformamide(DMF), 2-(2-butoxyethoxy)ethanol were added to carbon disulfide($CS_2$). The best improvement was achieved when 10% dimethylformamide(DMF) in $CS_2$ was used as desorption solvent. During storage of polar analytes, recovery was greatly reduced. Especially, the recovery of cyclohexanone was decreased to 18.1 % after a month storage at $34^{\circ}C$. After two weeks storage, recovery of polar analytes was sharply decreased. Water adsorbed on charcoal interfered the recovery of polar analytes but didn't interfere that one of nonpolar solvent, toluene. When 10% DMF in $CS_2$ was used as desorption solvent, the effect of water on recovery was decreased, comparing with Desorption efficiency increased when analyte loading increased, and usage of 10% DMF in $CS_2$ decreased the loading effect. Increasing volume of desorption solvent was not effective to improve desorption efficiency of analytes when 10% DMF was used. Continuous shaking and sonication is not helpful to increase the desorption efficiency of analytes except cyclohexanone using 10% DMF. When silica gel used as adsorbent, methanol was better desorbent than dimethylsulfoxide. Analytes adsorbed on silica gel showed high recovery in low concentration and less affected by humidity. On the basis of this study, the following conclusions have been drawn. To improve the recovery of polar organic materials in air samples, it is necessary to analyze samples as soon as possible after they were collected. Otherwise, samples must be stored at low temperature. Using two components of desorption solvents, such as 10% DMF in $CS_2$, the effects of loading and humidity decreased for polar analytes such as methyl ethyl ketone and methyl isobutyl ketone. When work place has high humidity with low concentration of polar organic solvents, silica gel can be used as adsorbent, because it produces quantitative recovery for polar analytes at this condition. But it should be noted that high humidity makes breakthrough easy in silica gel samples.

LC/MS/MS 분석을 위한 microcystins 전처리 단계별 효율성 연구 (Evaluation of Pre-treatment Stages for Microcystins Analysis using LC/MS/MS)

  • We investigated the most effective pre-treatment processes and LC/MS/MS condition for microcystins analysis. With a step-by-step pre-treatment, efficiencies of several established methods were compared. At the level of cell burst, sonication method was found to be the most efficient. As a mycrocystins first extraction solvent, 5% acetic acid showed the highest efficiency. An isolation and recovery rate of mycrocystins of ODS Sep-Pak $C_{18}$ cartridge was higher than HLB SPE cartridge. As a final elution solvent from cartridge, 100% MeOH had a better efficiency than others. Using a LC/MS/MS, effective analytical methods were established. C18 reverse column was used and gradient elution was performed with using acetonitrile, 0.1% formic acid as a mobile phase. We analysed to 0.8 mL/min flow rate fit to the $5{\mu}m$ particle size column and $55^{\circ}C$ housing temperature. The validity of established analytical method was evaluated that MDL as average $0.050{\pm}0.014{\mu}g/L$ and LOQ as average $0.160{\pm}0.045{\mu}g/L$ had a good sensitivity over 40 magnification rather than $2{\mu}g/L$ detection limit of HPLC.

마이크로파, 초음파 및 볶음 전처리가 오가피의 열수 추출특성에 미치는 영향 (Effects of Microwave, Ultrasound and Roasting Pretreatments on Hot Water Extraction of Acanthopanax senticosus)

  • 오가피(Acanthopanax senticosus)로부터 영양 및 기능성 성분의 열수추출 수율을 증대시킬 수 있는 방법을 개발하기 위하여 추출 전처리 및 방법의 효과를 검토하였다. 오가피를 분쇄, 체질$(8\~20\;mesh)$하고 초음파(20 kHz), 마이크로파 (2,450 MHz) 또는 볶음$(180^{\circ}C)$ 처리를 10분 동안 실시한 후 물을 용매로 상압$(100^{\circ}C)$ 추출과 가압$(121^{\circ}C)$ 추출을 각각 2시간 동안 실시하였다. 전반적으로 전처리는 성분들의 추출수율과 유리기 소거능을 증가 시키는 경향을 보였다. 특히, 가용성 고형물, 총당 및 페놀성 물질의 추출수율은 마이크로파와 초음파에 의해, 단백질의 추출수율과 유리기 소거능은 볶음처리에 의해 더욱 증가됨을 보였다. 상압추출보다 가압추출에 의해 가용성 고형물, 총당, 단백질 및 페놀성 물질의 추출수율이 증대되는 경향을 보였다. 이로써 초음파, 마이크로파 및 볶음 전처리 및 가압추출은 오가피의 추출효율 향상에 유효한 방법인 것으로 판단된다.

초임계이산화탄소 내에서 공용매 및 초음파를 이용한 고농도이온주입 포토레지스트의 제거 (Stripping of High-Dose Ion-Implanted Photoresist Using Co-solvent and Ultra-sonication in Supercritical Carbon Dioxide)

  • 초임계이산화탄소와 공용매의 혼합물을 사용하여 반도체 웨이퍼 기판으로부터 고농도이온주입 포토레지스트(HDIPR)를 제거하였다. 또한 고압 셀 내부에 초음파 장치를 부착하여 웨이퍼 표면에 물리적 힘을 제공함으로서 세정용액의 HDIPR에 대한 스트리핑 성능을 현저히 향상시키고, 제거 시간을 단축시켰다. 공용매의 종류 및 농도, 공정 온도, 압력 변화에 따른 HDIPR 스트리핑 특성을 조사하였으며, 웨이퍼 표면의 제거 전후의 상태 및 성분을 scanning electron microscopy 과 energy dispersive X-ray spectrometer를 이용하여 분석하였다. 10 w/w% 함량의 아세톤 공용매를 이용하여 공정압력 27.6 MPa과 온도 343 K 의 조건에서 3분의 초음파 처리시간을 거쳐 HDIPR을 완전히 제거할 수 있었다.

미세조류 Dunaliella bardawil에서 오존과 과산화수소에 의한 ${\beta}$-carotene의 축적과 식용기름을 이용한 추출 (Induction of ${\beta}$-carotene by Ozone and Hydrogen Peroxide and Extraction Using Vegetable Oil from Microalga Dunaliella bardawil)

  • Dunaliella bardawil의 적절한 생장과${\beta}$-carotene의 효과적인 축적 조건을 확립하기 위하여 염 농도, pH, 광량, 교반속도, 오존처리, 과산화수소 첨가 등의 영향을 비교하였고, 식용 식물성 기름을 이용한 ${\beta}$-carotene의 추출조건을 비교하였다. D. bardawil의 비생장 속도는 1N NaCl, pH 8.0 광량 80${\mu}$$E/\m^2/s$ , 70rpm 으로 배양 시에 0.168/hr로 최대를 나타냈다. 흰색 형광등을 250${\mu}$$E/\m^2/s$ 로 조사한 경우 ${\beta}$-carotene isomer 축적은 cis isomer가 trans isomer에 비해 높은 증가율을 보였다. ${\beta}$-carotene의 축적을 위하여 50${\mu}$M 과산화수소를 배양 2일 경과 후에 첨가한 경우 대조군의 축적율이 150${\mu}$g/ml인데 비해 324${\mu}$g/ml로, 200ppb로 2시간 오존 처리했을 때 약 1.8배 증가한 270${\mu}$g/ml이었다. 식용기름에 의한 37${\circ}C$에서의 추출효율은 올리브유, 참기름, 미강유, 옥수수유, 대두유의 순서로 나타났으며, 올리브유를 수층 부피의 9배로 첨가하고 50${\circ}C$에서 1시간 동안 sonication한 결과 96.95의 추출효율을 나타내었다.

수화특성차이(水和特性差異)를 이용(利用)한 인산부생석고(燐酸副生石膏)로부터 정제석고(精製石膏)의 회수(回收) (Refinement of Phosphogypsum by Selective Dehydration & Hydration)

  • 본 연구는 불순물을 포함하는 인산부생석고로부터 정제석고를 얻을 목적으로 수행되었다. 인산부생석고를 일정조건에서 탈수하고 수화시켜 석고성분만을 선택적으로 미립화 시킨 다음 325mesh로 습식사분하여 사하산물로 정제석고를 얻었다. 이 과정에서 석고의 회수율에 미치는 탈수온도, 탈수시간, 탈수율, aging시간, 수화농도, 수화온도, 초음파조사시간, 첨가제 등의 영향을 조사하였다. 실험결과, 인산부생석고를 $140^{\circ}C$에서 6시간 동안 건조하여 완전히 무수석고 상태가 되도록 탈수 한 다음, 슬러리 농도 $3{\sim}10%,\;20{\sim}30^{\circ}C$의 수화온도, 수화시간 2시간의 조건에서 수화하여 사분하면 석고회수율이 95% 이상이 됨을 알았다. 회수된 석고의 품위는 회수율이 높을수록 높아지는 경향을 보였으며 회수율이 90%이상 일 때 품위는 $94{\sim}96%$를 보였다. 이 밖에도 석고회수율을 높이기 위해서는 탈수된 석고가 공기와 접촉하는 시간이 16시간을 넘어서는 아니 되며, 구연산나트륨을 0.005M 정도 첨가하는 것과, 수화반응 초기에 $5{\sim}10$분 동안 초음파를 조사하는 것이 매우 유용한 수단임을 알았다.

최종 하수처리장 슬러지의 추가감량을 위한 슬러지 전처리 연구 (Studies of Pretreatment Mehtods for Additional Reduction of Sewage Sludge)

  • 본 연구는 부산시 남부하수처리장의 탈수슬러지를 이용하여 전처리에 따른 분해 및 혐기성 상태에서의 최종 메탄 및 이산화탄소 발생량을 평가하기 위해 Biochemical Methane Potential(BMP) 실험을 실시하였다. 초기 1 % TS 농도의 탈수슬러지를 각각 30분 동안 5M NaOH를 이용하여 알칼리 전처리를 실시하였고 초음파 전처리는 30분 동안 조사밀도를 1.5 W/mL로 조절하여 실시하였으며 알칼리와 초음파 전처리를 병합한 동시 전처리를 시행하였다. 그에 따라 SCOD 값이 초기 33.1 mg/L에서 최대 494 mg/L로 증가하였다. 또한 BMP 실험을 실시한 결과 31.2~84.2 mL $CH_4/g$ VS, 9.2~13.5 mL $CO_2/g$ VS로 나타났다. 영양배지와 상등액 슬러지를 주입여부에 따른 BMP 실험을 실시한 결과 각각 73.1, 73.8 mL $CH_4/g$ VS, 11.2, 13.6 mL $CO_2/g$ VS으로 나타났으며 모델값과는 큰 차이를 보이지 않았다. BMP 실험을 종료 후 VS를 분석한 결과 초기 62 %에서 33.8~45.4 %로 감소하였다. 알칼리, 초음파 전처리보다는 동시 전처리가 메탄발생량과 SCOD 값을 증가시켰으며 VS 감소율이 증가하여 혐기성 소화율이 향상되었다.
