Effect of Various Additives and Solvents on Thermostability of Cyclodextrin Glucanotransferase from Bacillus stearothermophilus (여러 첨가물의 용매가 Bacillus stearothermophilus가 생산하는 Cyclodextrin Glucanotransferase의 열안정성에 미치는 영향)

  • The influence of ethylene glycol, glycerol, sorbitol and sucrose on the thermostability of Bacilus stearothermophzlus cyclodextrin glucanotransferase (CGTase) was investigated. Glycerol, sorbitol and sucrose had effect on thermostability of the CGTase. The effects appeared to be strongly dependent on concentration of additives. The thermostability of CGTase also was assayed in organic solvents such as n-butanol, l, &dioxane, n-octane. The therrnostability of CGTase increased in l, 4-dioxane and n-octane. Particularly, in n-octane, the CGTase retained the 81% of the initial activity after incubation at $75^{\circ}C$ for 90 min.

  • We immobilized Thermomyces lanuginosus lipase to catalyze transesterification reaction in DMF. This lipase was selected after screening among other commercial lipases. We found that prepared immobilized lipase is particularly useful for preparation of 6-O-acylsucrose with higher conversion rate even in 10 g scale. Several solvents were evaluated for selective transesterification reaction. We noticed that the immobilized lipase retained more than 80 % activity after 5 cycles of 96 h reaction. A general method was also developed to purify the products using simple crystallization and precipitation process. Furthermore, 6-O-vinyladipoylsucrose was subjected to synthesis of the corresponding polymer by radical initiator. The sucrose branched polymer can be used further for evaluation of its biodegradability and other biological applications.

  • 연속반응조에서의 수소생산에 대한 수리학적체류시간(희석율, D)의 영향을 $1\%$ sucrose를 함유한 $37{\pm}1^{\circ}C$ 조건에서 조사하였다. 실험결과 수리학적체류시간(희석율)의 각각의 조건에 따라 생성된 가스중 수소성분은 $50~71\%$의 범위로 발생되었다. $H_2/CO_2$ 비율은 희석율이 증가할 때 $H_2/CO_2$ 비율도 증가하였다. 최대수소생성 수율은 희석율 0.14 l/h까지는 증가하다가 이후에는 감소하였고, 수소생성 수율은 0.81 l/g sucrose이었다. Acetate 생성 수율은 butyrate생성 수율 보다 희석율 조건변화에 민감하게 변화하였다. propionate 및 solvents는 희석율 변화에 영향을 받지 않았다 biomass 수율은 희석율이 0.2 l/h 까지는 증가하였으나, 그 이상의 조건에서는 감소하였다.

Effect of Heating Condition, Additives and Solvents on Rheology of Squid Meat Paste Products (오징어 연제품의 물성 개선을 위한 탄력증강제 및 용매처리 효과)

  • The squid had not been utilized for gel products because of its lower gel forming ability. The objectives of this study were as followed; 1) the optimum heating condition on squid meat paste products and 2) the optimum added level for jelly strength of squid meat paste products. Optimum heating conditions of squid meat kamaboko were as followed; setting(pre-heating) at 15$^{\circ}C$ or 55$^{\circ}C$ for 2 hours and heating at 9$0^{\circ}C$ for 60 minutes. The additives examined were as follows; 20mM EDTA, 10mM PMSF, 5 $\mu$mol/100g TGase, 0.2% potassium bromate, 2% collagen, 2% sucrose ester of stearic acid and 1% egg shell powder. The effects of additives on jelly strength were observed as follow, in descending order; 10mM of PMSF>5 $\mu$mo1/100g of TGase>0.2% of potassium bromate>20mM of EDTA. But sucrose ester of stearic acid and 1% egg shell powder were no effect. The solvents examined were as follows; n-amyl alcohol, n-butyl alcohol, n-hexyl alcohol, ethyl alcohol, ethylene glycol, propylene glycol and glycerin glycol. It showed that high jelly strength as 787gㆍcm for 3% of n-butyl alcohol and 749gㆍcm for 3% of n-amyl alcohol. To adding 5% of n-butyl alcohol and n-amyl alcohol, gave the highest jelly strength and water holding capacity(WHC). Effect of alcohol on jelly strength appeared higher value at added 5% of n-butyl alcohol than n-amyl alcohol, and flying squid product was higher than jumbo squid product.

  • The enantioselective hydrolysis of racemic styrene oxide in organic solvents was conducted using a recombinant E. coli expressing protein-engineered Mugil cephalus epoxide hydrolase (McEH). The volumetric total activity of the recombinant E. coli was enhanced 2.2-fold by IPTG induction at a mid-exponential growth phase. Among organic solvents with different log P values, isooctane was chosen based on the high activity and the enantioselectivity of McEH. Some lyoprotectants such as skim milk or sucrose enhanced the McEH activity. Enantiopure (S)-Styrene oxide with a 98% ee was obtained from the racemic styrene oxide with a 53.6% yield based on its theoretical yield in isooctane.

Effect of pH on Continuous Hydrogen Fermentation (연속반응실험에서 수소생성에 대한 pH 영향)

  • The influences of pH on hydrogen production were also investigated over the pH range from 4.1 to 8.0 at HRT 10 hours. The hydrogen content for the produced gas was changed from 41 to 71% with corresponding pHs throughout this experiment. The produced hydrogen/carbon dioxide ratio was not vary significantly up to 6.0, then steepenly increased with increases in the pH. The maximal hydrogen yield was found to be 3.16 $\ell$/g sucrose at pH 5.0. Acetate production yield increased with increased pH, but butyrate production yield decreased with increased pH. Biomass yield increased with increased pH.

  • 철 농도변화에 따른 대사산물 생성에 대한 반응은 중온조건에서 회분식 실험으로 수해하였다. 철의 농도가 40mg/$\ell$까지는 철 농도의 증가에 따라 수소생성율도 증가하였다. acetate 생성율은 철의 농도가 증가함에 따라 감소하였다. Butyrate, ethanol 및 butanol은 각각 철 농도가 200,200 및 400mg/$\ell$에서 최대 생성율을 나타내었다. 철의 농도가 200mg/$\ell$이상의 농도엘 때에 propionate, i-butyrate 및 valerate 생성율이 철의 농도가 200mg/$\ell$ 이하의 경우 보다 낮았다. 미생물농도의 경우 철 농도가 300mg/$\ell$까지는 농도가 증가함에 따라 미생물농도도 증가하였다.

  • In the area of the destruction-free NIR analysis of fruit and vegetables development has not yet progressed as far as in grain and similar products. One reason for that is, that in contrast to grains, in fruit and vegetables water appears as the outstanding main-component making up typically 80% by weight of the fruit. Of the M absorption spectrum of pure water the bands at 1450, 970 and 760 nm are the first, second and third overtones respectively of O-H stretch while those at 1940 and 1190 are combination bands involving O-H stretch and O-H bend. The choice of band for spectrometry is governed by considerations of sensitivity and selectivity. The overtone bands are satisfactory for use in moisture measurements from 0 to 4 % depending on path length. Measurements in fruits and vegetables at wavelength areas that are also important for the determination of carbohydrates (sucrose, glucose, fructose) often lead to total absorption in the presence of significant water even if short path lengths are possible. In this work model systems are used containing different carbohydrates in solvents like heavy water (D$_2$O) or dimethylsulfoxide (DMSO) that do not contain O-H functional groups.

Effect of Extraction and Evaporating Conditions on the Free SuEar in Contents Ginseng Extract (추출 및 농축조건에 따른 인삼엑기스중 유리당 함량변화)

  • This studies were conducted to investigate the effect of extraction and evaporating conditions on the free sugar in ginseng extracts. Raw ginseng roots were extracted with water or various concentration of water-ethanol solutions. Then the extracts were evaporated by boiling or vacuum evaporated at 50 $^{\circ}C$ and 70 $^{\circ}C$. The results are as follows Sucrose and maltose contents were 7.95% and 12.8% in water extracts and 19.96%, 0.34-0.93% in 90% ethanol extracts, respectively. On the otherhand, glucose and fructose contents were not affected by the solvents. Evaporating conditions, such as temperature, were rarely effected to the sugar contents.

  • Adventitious root culture was established in the Jawahar variety of Withania somnifera using MS basal medium supplemented with 0.5 (mg/l) IAA and 2.0 (mg/l) IBA. Root tips from germinated seedlings, MS0 maintained plants and adventitious roots were maintained in suspension medium (1/2 MS basal medium supplemented with 3% sucrose) for a period of 1 to 6 months. The weight gain in roots was noted and the withanolides were extracted from the dry roots using solvents petroleum ether, 50% ethanol and chloroform. The withanolides in the chloroform fractions of all root samples analyzed were compared using thin layer chromatographic analysis. Withanolide content in adventitious root sample was found to be superior compared to other roots at any given point of time during the 6month growth period.HPLC analysis of in vitro adventitious roots showed the presence of a new compound.
