Theoretical Study on the Absorption Spectrum of a Chromophore in Liquid (용액상 색소분자의 흡수스펙트럼에 대한 이론적 연구)

  • Molecular motion influencing the absorption spectrum of a chromophore in liquid is theoretically described by a quantum mechanical time correlation function. In the present paper, we developed a theoretical method to calculate such a quantum mechanical time-correlation function from a classical time-correlation function using semi-classical approximations. The calculated time-correlation function was combined with the second order cumulant expansion method to calculate the absorption spectrum of nile blue in acetonitrile. Reasonably good agreement with experimental spectrum was obtained. From the comparison with experimental spectrum, we concluded that the time scale of solvation dynamics of the system should be longer then 1ps and the first shell of solvent is the major contribution to the solvation dynamics.

Studies on toxic substances of Gingko leaves inducing contact dermatitis in rabbit (은행엽(銀杏葉)의 가토(家兎) 피부(皮膚) 접촉성염증(接觸性炎症) 유발(誘發) 독성물질(毒成物質)에 관(關)한 연구(硏究))

  • Gingko biloba is one of the oldest trees on earth. It can be traced 300 million years or more into past. In recent years, this tree has become increasingly popular because of its longevity. Amount of comsumption of Gingko leaves in Korea is increased to produce drugs for blood circulating system's disorder. The workers who work at the company of product drug with Gingko leaves complain pruritus at belt position of west, however toxic substance from extracts of Gingko biloba was carried out to determine what kind of the toxic substance from extract could induce a contact dermatitis by means of histopathological study. Dried leaves of Korean G. biloba were exhaustively extracted with chloroform. The aqueous chloroform residue was extracted with hexane. After evaporation, the hexane extract was passed through a silica gel column chromatography, it was separated into 9 fractions by means of gradient solvent (benzene: acetone/ 98~93 : 2~7%). The 4th and 7th fraction were idendified as bilibol and gingkolic acid by silica gel thin layer chromatography and NMR spectrophotometer. The 4th and 7th fractions of Korean Gingko leaves have an irritant substances which can be induced ear swelling and increased thickness of stratum cornium of rabbit ear exposed with the fractions (3 time/3 day, $100{\mu}l$ of 50mg/ml ethyl alcohol). In the 4th fraction, the substance have induced the surface of ear to swell and mass of unknown cell like inflammatory cell were infiltrated within the stratum corneum, and the 7th fraction have the surface of ear to loss the hairs, and sebaceous gland and follicle of hair were infiltrated with inflammatory cells. and other fractions at all did not induced a certain contact dermatitis and also Deutschen drug of Gingko (named as Gerigeron) have induced the contact dermatitis such as dematitis of Korean Gingko extract.

  • Styrene-butadiene rubber (SBR) is widely used in tire treads due to its excellent abrasion resistance, braking performance, and reasonable cost. Depending on the polymerization method, SBR is classified into solution-polymerized SBR (SSBR) and emulsion-polymerized SBR (ESBR). ESBR is less expensive and environmentally friendlier than SSBR because it uses water as a solvent. A higher molecular weight is also easier to obtain in ESBR, which has advantages in mechanical properties and tire performance. In ESBR polymerization, a surfactant is added to create an emulsion system with a hydrophobic monomer in the water phase. However, some amount of surfactant remains in the ESBR during coagulation, making the polymer chains in micelles clump together. As a result, it is well-known that residual surfactant adversely affects the physical properties of silica-filled ESBR compounds. However, researches about the effect of residual surfactant on the physical properties of ESBR are lacking. Therefore, in this study we compared the effects of remaining surfactant in ESBR on the mechanical properties of silica-filled and carbon black-filled compounds. The crosslinking density and filler-rubber interaction are also analyzed by using the Flory-Rehner theory and Kraus equation. In addition, the effects of surfactant on the mechanical properties and crosslinking density are compared with the effects of TDAE oil (a conventional processing aid).

Emulsifying Properties and Oxidative Stability of Purified Surface-Active Substances from Defatted Rapeseed Cake Extract (탈지 유채박 중 표면활성정제물의 유화특성 및 산화 안정도에 관한 연구)

  • Surface-active substances in defatted rapeseed cake were obtained using a supercritical fluid extraction method. Then, it was purified by removing sinapine in the extract through a series of steps using a mixed solvent: diethyl ether and ethyl acetate (1:1, v/v). Emulsifying properties of purified surface-active substances were investigated, including fat globule size, zeta potentials and creaming stability and its antioxidant activity in emulsion systems were also studied by peroxide value and $^1H$-NMR spectrum. It was found that fat globules in emulsions with purified surface-active substances were much smaller than ones with the unpurified. In addition, as pH of the emulsion lowered and with increasing NaCl concentration in the emulsion, they were observed to increase, which led to worse creaming stability. These properties were reflected in changes of zeta potentials of emulsions. The oxidative stability was better in emulsions with purified surface-active substances than ones with Tween 20 or commercial lecithin, possibly resulted from the existence of sinapic acid in the extract. It was concluded that purified surface-active substances from defatted rapeseed cake could be simultaneously used as emulsifier and antioxidant agent in emulsion system.

  • A simultaneous and rapid gas chromatographic determination of organophosphorus, organo-chlorine, carbamate, and pyrethroid pesticides in polished rice was developed. The pesticides were extracted from samples with acetone by automated soxhlet apparatus and this extract was evaporated to dryness. The residue was dissolved in hexane, the solvent was applied to a Sep-Pak florisil catridge, was eluted with 50% ethyl acetate in n-hexane, and was injected to dual GC-ECD/NPD system. This simple method affords a high recovery of hydrophilic pesticides, allows rapid analysis, and is cheap to perform. Except for dichlorvos and captan, recoveries of 46 pesticides were over 60%.

Beta-carotene and Lutein Contents in Green Leafy Vegetables (녹색잎 채소류의 ${\beta}$-Carotene과 Lutein 함량)

  • Beta-carotene and lutein contents in 7 different green leafy vegetables(perilla leaf, mugwort, chwi, lettuce, spinach, leek, and crown daisy) were analyzed by HPLC. The isocratic separation was performed on a ${\mu}-Bondapak$ $C_{18}$ column with a solvent system of acetonitrile : dichloromethane : methanol = 70:20:10. To check the reliability of the method applied, precision and recovery tests were performed. Perilla leaf showed the highest ${\beta}-carotene$ content(12,570 ${\mu}g$ / 100 g), followed by mugwort and chwi, all of those have ${\le}10,000\{\mu}g\{\beta}-carotene$ per 100 g vegetables. Green lettuce, spinach, leek, crown daisy and reddish brown lettuce contained 9,869, 6,689, 5,664, 3,601 and 3,299 ${\mu}g\{\beta}-carotene/100 g$, respectively, Lutein content was the highest in perilla leaf($13,718{\mu}g/100 g$) followed by chwi($11,989{\mu}g/100 g$), mugwort($11,522{\mu}g/100 g$), green lettuce($10,307{\mu}g/100 g$) and spinach($10,115{\mu}g/100 g$). ${\beta}-carotene$ contents in perilla leaf, mugwort, chwi and green lettuce were 47.8~49.6% of total carotenoids, and ${\beta}-carotene$ contents in the other green leafy vegetables analyzed were 37.7~41.4% Vitamin A contents of green leafy vegetables analyzed by HPLC were 2~6 times higher than the vitamin A values shown in food composition tables except crown daisy.

Development of Drug-Loaded PLGA Microparticles with Different Release Patterns for Prolonged Drug Delivery

  • For the prolonged delivery and sustained release rates of low molecular weight drugs, poly(lactic-co-glycolic acid) (PLGA) microparticles containing the drug SKL-2020 have been investigated. On increasing polyvinyl alcohol (PVA) concentration (from 0.2% to 5%), the size of microparticles decreased (from $48.02{\mu}m$ to $10.63{\mu}m$) and more uniform size distribution was noticeable due to the powerful emulsifying ability of PVA. A higher drug loading (from 5% to 20%) caused a larger concentration gradient between 2 phases at the polymer precipitation step; this resulted in decreased encapsulation efficiency (from 34.19% to 25.67%) and a greater initial burst (from 61.71% to 70.05%). SKL-2020-loaded PLGA microparticles prepared with different fabrication conditions exhibited unique release patterns of SKL-2020. High PVA concentration and high drug loading led to an initial burst effect by rapid drug diffusion through the polymer matrix. Since PLGA microparticles enabled the slow release of SKL-2020 over 1 week in vitro and in vivo, more convenient and comfortable treatment could be facilitated with less frequent administration. It is feasible to design a release profile by mixing microparticles that were prepared with different fabrication conditions. By this method, the initial burst could be repressed properly and drug release rate could decrease.

Speciation of Chromium in Water Samples with Homogeneous Liquid-Liquid Extraction and Determination by Flame Atomic Absorption Spectrometry

  • A novel method was developed for the speciation of chromium in natural water samples based on homogeneous liquid-liquid extraction and determination by flame atomic absorption spectrometry (FAAS). In this method, Cr(III) reacts with a new Schiff's base ligand to form the hydrophobic complex, which is subsequently entrapped in the sediment phase, whereas Cr(VI) remained in aqueous phase. The Cr(VI) assay is based on its reduction to Cr(III) by the addition of sodium sulfite to the sample solution. Thus, separation of Cr(III) and Cr(VI) could be realized. Homogeneous liquid-liquid extraction based on the pH-independent phase-separation process was investigated using a ternary solvent system (water-tetrabutylammonium ion ($TBA^+$)-chloroform) for the preconcentration of chromium. The phase separation phenomenon occurred by an ion-pair formation of TBA and perchlorate ion. Then sedimented phase was separated using a $100\;{\mu}L$ micro-syringe and diluted to 1.0 mL with ethanol. The sample was introduced into the flame by conventional aspiration. After the optimization of complexation and extraction conditions such as pH = 9.5, [ligand] = $1.0{\times}10^{-4}\;M$, [$TBA^+$] = $2.0{\times}10^{-2}\;M$, [$CHCl_3$] = $100.0\;{\mu}L$ and [$ClO_4$] = $2.0{\times}10{-2}\;M$, a preconcentration factor (Va/Vs) of 100 was obtained for only 10 mL of the sample. The relative standard deviation was 2.8% (n = 10). The limit of detection was sufficiently low and lie at ppb level. The proposed method was applied for the extraction and determination of chromium in natural water samples with satisfactory results.

Effect of Continuous Exposure to Reactive Oxygen Species on ${\gamma}$-Glutamyltranspeptidase Expression and Activity in HepG2 Cells (HepG2 세포에서 지속적인 활성 산소 노출이 ${\gamma}$-Glutamyltranspeptidase 발현과 활성에 미치는 영향)

  • The adverse health effects of a number of environment pollutions are related to the formation of free radicals. Induction of antioxidant defensive system in the response to an oxidative attack is an essential element of the cell to survive. CYP2E1 is easily induced by organic solvents and induces continuous formation of reactive oxygen species (ROS). ${\gamma}$-Glutamyltranspeptidase (${\gamma}$GT) plays an important role in glutathione metabolism and xenobiotic detoxification. To evaluate the characteristic of oxidative stress which induces GGT expression and to understand human antioxidant defensive response against oxidative stress induced by CYP2E1, we studied regulation of ${\gamma}$GT enzyme expression in response to various oxidative stresses in human HepG2 cells. The ${\gamma}$GT activity was not modified after exposure of acute oxidative stress inducing agents (ferric nitrilotriacetate, cumene hydroperoxide, ADP-Fe, O-tetradecanoylphorbol-13-acetate, tumor necrosis factor-alpha). To induce continuous exposure of cells to ROS, HepG2 cells were transfected by human CYP2E1 gene transiently. The CYP2E1 activity was verified with chlorzoxazone hydroxylation. Transfection of CYP2E1 showed continuous 60% increase in intracellular ROS and 240 % increase in microsomal ROS. CYP2E1 overexpressing cells showed increased ${\gamma}$GT activity (2.5-fold). The observed enhancement of ${\gamma}$GT activity correlated with a significant increase of ${\gamma}$GT mRNA (2.1-fold). Treatment with antioxidant strongly prevented the increase in ${\gamma}$GT activity. The CYP2E1 overexpression did not modify toxicity index and increased glutathione levels. These results show that continuous exposure of cells to ROS produced by CYP2E1 up-regulates ${\gamma}$GT; This may be one of the adaptive antioxidant responses of cells to oxidative insult. Present study also suggests that the induction of ${\gamma}$GT could be used as a marker of oxidative stress induced by exposure to organic solvents.

Human Risk Assessment of Perchloroethylene Considering Multi-media Exposure (다매체 노출을 고려한 Perchloroethylene의 인체위해성평가연구)

  • Objectives: Perchloroethylene (PCE) is a volatile chemical widely used as a solvent in the dry-cleaning and textile processing industries. It was evaluated as Group 2 "probably carcinogenic to humans" by the Integrated Risk Information System (IRIS) of the United State Environmental Protection Agency (U.S. EPA) in 2012. In order to provide a scientific basis for establishing risk management measures for chemicals on the national priority substances list, aggregate risk assessment was conducted for PCE, included in the top-10 substances. Methods: We conducted the investigation and monitoring of PCE exposure (e.g., exposure scenario, detection levels, and exposure factors, etc.) and assessed its multi-media (e.g., outdoor air, indoor air, and ground water) exposure risk with a deterministic and probabilistic approach. Results: In human risk assessment (HRA), the level of human exposure was higher in the younger age group. The exposure level through inhalation at home was the highest among the exposure routes. Outdoor air or uptake of drinking water represented less than 1% of total contributions to PCE exposure. These findings suggested that the level of risk was negligible since the Hazard Index (HI) induced by HRA was below one among all age groups, with a maximum HI value of 0.17 when reasonable maximum exposure was applied. Conclusion: In conclusion, it was suggested that despite low exposure risk, further studies are needed considering main sources, including occupational exposure.
