  • Fe (III) impurities in clay could be microbially removed by inhabitant dissimilatory Fe (III) reducing microorganisms. Insoluble Fe (III) in clay particles was leached out as soluble reductive form, Fe (II). The microorganisms removed from 10 to 45% of the initial Fe (III) when each sugar was supplemented to be in ranges of 1 - 5 % (w/w; sugar/clay). The microorganisms reduced 2.1 - 12.8 mol of Fe (III) per 100 mol of carbon in sugars metabolized when sugars such as glucose, maltose, and sucrose were used as sole carbon source. Bacillus sp. IRB-W and Pseudomonas sp. IRB-Y were isolated from the enrichment culture of the clay. The isolates were considered to participate in metabolizing organic compounds to fermentative intermediates with relatively little Fe (III) reduction at initial Fe (III) reduction process. By the microbial treatment, the whiteness of the clay was increased form 63.20 to 79.64, whereas the redness was obviously decreased form 13.47 to 3.55. This treatment did not cause any unfavorable modifications in mineralogical compositions of the clay.

Immunoradiometric Assay using Monoclonal Antibody Against Human Serum Transferrin Receptor for Diagnosis of Iron Deficiency (사람 혈청 트란스페린수용체의 단클론 항체를 이용한 방사면역측정과 철영양상태의 진단)

  • The soluble transferrin receptor(TfR) in human serum has been shown recently to be a truncated form of intact membrane bound receptor containing most of the extracellular domain. We purfied the transferin-free TfR from human serum by immounoaffinity chromatography which produced the single protein identity in high resolution gel chormatography. The monoclonal antibodies(MAb) against purifed serum TfR were produced by fusion of spleen cells o fimmunized Balb/c mice and SP2 cells. Ten hybrids producing MAb specific for serum TfR were identifed and determine their iostypes. A immunoraddiometric assay (IRMA) for serum TfR was established using two monoclonal IgG1 antibodies as the coating and indicator antibodies on the bosis of their suitability in sandwich IRMA of serum TfR. The mean serum TfR levels in the 15 normal male, 15 normal female, and 19 iron-deficient subjects were 5.4$\pm$0.98, 4.6$\pm$0/76, and 18.0$\pm$12.8mg/1, respectively, and the difference in mean values between normal and iron deficient subjects was significant(p=0.0005). There existed the inverse logarithmic relationship(r=-0.9336, p<0.0001) between the serum TfR and ferritin levels.

Expression of a Carboxy-Terminal Deletion Mutant of Recombinant Tadpole H-Chain Ferritin in Escherichia coli

  • In order to study the role of the protein shell in both iron uptake and iron core formation of ferritin, we constructed a deletion mutant of the ferritin gene and expressed the mutant gene in Escherichia coli, This mutant was obtained by introducing an amber mutation at position Pro-157 and a deletion of the 19 amino acid residues at the carboxy-terminus of the recombinant tadpole H-chain ferritin. The deleted amino acids correspond to E-helix forming the hydrophobic channel in the protein. E. coli harboring the plasmid pTHP157, which contains the deleted gene, was grown at $23^{\circ}C$ in the presence of 0.1 mM IPTG, and the induced protein appeared to be partly soluble. Nondenaturing polyacrylamide gel electrophoresis showed that the expressed mutant H-chains coassemble into holoprotein, suggesting that E-helix is not necessary for assembly of the subunits as reported for human H-chain ferritin. Its ability in iron core formation was proven in an Fe staining gel, the result disagreeing with the observation that the hydrophobic channel is necessary for iron core formation in human H-chain ferritin.

A Study on Nutritional Characteristics of Common Korean Dietary Fiber Rich Foods (한국인 상용 식이섬유 급원 식품의 영양 생리적 특성)

  • The effects of common Korean dietary fiber-rich foods(dried sea mustard, Lentinus edodes and Korean radish leaves) on total gut transit time of diet, serum cholesterol level and mineral apparent absorption ration and tissue level were investigated. Twenty eight male were divided into 4 groups and were fed a control diet(containing 2% cellulose) and three fiber supplementary diet mixed with 10% of the above dietary fiber rich foods for 4 weeks, respectively. Body weight gain and food efficiency ration were not affected by the supplementary diet of fiber but the total gut transit time of diet was shortened in all dietary fiber groups. The Lentinus edodes reduced significantly serum total cholesterol and increased the HDL-cholesterol/total cholesterol ratio at 4th week without and effect on calcium and iron apparent absorptions and tissue levels of the minerals. Radish leaves decreased significantly iron apparent absorption ration with a tendency to increase iron retention in spleen and did not alter serum cholesterol level. Serum cholesterol concentration and calcium and iron apparent absorptions were not significantly altered by the ingestion of sea mustard, while it had more soluble dietary fiber than mushroom and radish leaves.

  • Magnetic separation was carried out in order to improve the magnetite grade of the spent iron oxide catalyst, that was composed with magnetite, ceria and soluble alkaline salt. The recovery of magnetite from the spent iron oxide catalyst was over 99%, and the magnetite contents was upgraded to about 80% from 70% via wet type magnetic separation at 500 Gauss. This improvement was due to the removal of alkaline salt by water instead of the magnetic separation.

Chromium concentration and half-life in rats biological samples after intravenous administration of soluble trivalent and hexavalent chromium compounds (흰쥐에 정맥 주사한 수용성 크롬의 분포 및 배설)

  • Chromium exposure can be in the forms of environmental pollution and occupational exposure. The harmful effects of chromium on the body greatly differ depending on its valence or solubility. Accordingly, the recommended permissible exposure limit for each chromium compound is different. This study investigated the increase or decrease of distribution and excretion of total chromium exposed simultaneously the soluble Cr+6 and Cr+3 compounds. There were no difference of total chromium concentration in plasma, red bloods cells, urine, organs between simultaneously injected and individually injected soluble Cr+6 and Cr+3 compound. The chromium clearances in urine also showed that there were two phases in the two groups. In the first phase, biological half lives of the total chromium of the two groups have been similar within 24 hr., but in the second phase, biological half life of the group injected simultaneously was 62.7 hr. and was less than that of the other group's 188.3 hr. The average concentration of total chromium in plasma was same with the control, and that of RBCs was $0.218nmol/m{\ell}$ and was slightly increased in comparison with $0.121nmol/m{\ell}$ of the control, which was not statistically significant. As a result, there were no differences of distribution and excretion of chromium between the group exposed simultaneously and the other group exposed separately the soluble Cr+6 and Cr+3 compounds. The biological half life of chromium of the former group in urine was less than that of the other group.

Iron deficiency in Retired Workers exposed to Mineral dust with Chronic Obstructive Pulmonary Disease (만성폐쇄성폐질환을 동반한 광물성분진 노출 이직근로자의 철 결핍)

  • Objective: Chronic obstructive pulmonary disease(COPD) is characterized by persistent airflow limitations associated with chronic inflammatory response due to noxious particles or gases in the lung. Iron deficiency is associated with chronic inflammation, such as COPD. The aim of this study was to evaluate the relationship among iron deficiency, iron homeostasis, and inflammation in retired miners with COPD. Methods: The serum levels of ferritin, soluble transferrin receptor(sTfR), and transferrin saturation(TSat) as biomarkers for iron deficiency and high-sensitivity C-reactive protein(hsCRP) as a biomarker for inflammation and hepcidin as a biomarker for iron homeostasis were measured in 93 male subjects. Iron deficiency was defined as any one or more of (1) sTfR>28.1 nmol/L, (2) TSat<16%, and (3) ferritin< $12{\mu}g/L$. Results: Iron deficiency was found 28% of the study subjects. Median levels of serum hsCRP was significantly increased related to airflow limitation of COPD(GOLD 1, $0.09{\mu}g/dL$ vs. GOLD 2, $0.17{\mu}g/dL$ vs. GOLD $3{\leq}$, $0.30{\mu}g/dL$, p=0.010), and was positively correlated with hepcidin(p=0.009). Mean level of serum hepcidin was lower in COPD subjects with iron deficiency(p=0.004) and serum levels of hepcidin was negatively correlated with %$FEV_1$ predicted(p=0.030). Conclusions: These results suggest that high serum levels of hepcidin are related to severe airflow limitation or inflammation and can decrease iron availability, regardless of iron status.

A Study on Iron Binding Peptides from Casein Hydrolysates (Casein 가수분해물 소재 철분결합 Peptide에 관한 연구)

  • When casein was hydrolyzed by trypsin, alcalase, neutrase, protamax, and S. aureus type V8, peptides $(100\;{\mu}g/mL)$ which were produced by trypsin and alcalase solubilized $6.42\;and\;2.37\;{\mu}g/mL)$ of added irons at pH 6, respectively, while peptides which were produced by other proteases solubilized less than $1\;{\mu}g/mL$. Peptides produced by trypsin and alcalase were fractionated to 10 fractions on a reverse phase column and each fraction was tested for its iron solubilizing ability at pH 6. Among peptides produced by trypsin, fraction 5 showed the highest iron solubilizing ability $(2.33\;{\mu}g/mL)$. In the case of alcalase, fraction 7 showed the highest iron solubilizing ability $(1.56\;{\mu}g/mL)$. To isolate iron binding peptides from peptides produced by trypsin and alcalase, immobilized iron affinity chromatography which irons were chelated to imino diacetic acids in chelating sepharose fast flow were utilized. Our results showed that immobilized iron affinity chromatography was an effective method to isolate iron binding peptides produced by either trypsin or alcalase from milk casein.

Desalting System for Excavated Metal Objects Using High Temperature, High Pressure Deoxygenated Water

  • We propose a new method to remove salts that have permeated into excavated metal objects. This method cleans the excavated metal objects by using high temperature, high pressure deoxvgenated water containing inhibitors for corrosion of metals. The method greatly reduces the washing time compared with previous methods. Waste water from the method does not need treatment, nor do chemicals need to be removed from the metal objects. Furthermore, this method is applicable to some kind of metals(for example iron objects, bronze objects). We measured quantitatively the soluble salts dissolved from actual metal objects and found that there was a large difference between soluble chloride ions and sulfate ions.

  • A new organic reagent, 1-isonicotinoyl-2-furfurylidene hydrazine was synthesized from isonicotinic acid hydrazide and furfural, gives precipitate with copper(II), mercury(II) and argent(I), whereas, it gives a water soluble yellow complex with iron(III). Copper complex of the reagent is soluble in EtOH MtOH, pyridine, dioxane and dimethylformamide with green yellow coloration. The complex has a maximum absorption at 385 m.$\mu$ and molar ratio of copper; reagent was estimated as 1:1 by continuous variation method, slop method and chelate titration method. Molar extinction coefficient (9600) and apparant formation constant of this complex was spectrophotometrically determined. K=1.7 * $10^{7}$ (Babko's method) K=2.1 * $10^{7}$ (Anderson's method). This reagent reacted with copper so sensitive that it would be available for determination of Cu (II).
