귀리 수용성 $\beta$-glucan의 생물활성 (Biological Activities or oat soluble $\beta$-glucans)

  • 귀리를 건강 및 기능성 식품소재로서 개발하기 위하여 귀리 겉겨(OBC)로부터 수용성 $\beta$-glucan을 중심합성실험계획에 의해 분리하고, 생물활성(항균, 항산화, 암세포 생육저해능)을 검토하였다 귀리 수용성 $\beta$-glucan 시료는 250, 500$\mu\textrm{g}$/disc농도에서 항균활성을 paper disc method로 검토한 결과 항균력이 없었으며, 5%농도에서 전자공여능(electron donating ability, EDA)으로 측정한 항산화활성도 없었다. 귀리 수용성 $\beta$-glucan의 암세포에 대한 생육저해능은 제 7번 시료(추출온도 45$^{\circ}C$, 에탄올농도 15%, pH 6)가 다른 시료에 비해 유의적으로 높았는데, 1mg/$m\ell$의 시료농도에서 위암(AGS), 간암(Hep3B) 및 폐암세포주(A549)에 대하여 각각 59%, 58% 및 54%의 생육저해능을 보였다 또 제 1번 시료(추출온도 55$^{\circ}C$, 에탄올농도 5%, pH 6)의 경우도 3종의 암세포에 대하여 비교적 높은 생육저해활성을 나타내었다. 반응표면분석 결과, 독립변수들의 값의 변화에 따라 암세포의 생육저해능은 영향을 받았다.

Effects of Feeding Oxalate Containing Grass on Intake and the Concentrations of Some Minerals and Parathyroid Hormone in Blood of Sheep

  • In order to determine whether oxalate from grasses affects feed intake, blood calcium (Ca) and other blood parameters of adult sheep, two feeding trials were conducted. In Trial 1, one group of sheep received guineagrass (0.47% soluble oxalate) and another group received setaria (1.34% soluble oxalate) for 28 d. In Trial 2, one group of sheep received guineagrass while another group received the same grass treated with an oxalic acid solution (at a rate of 30 g oxalic acid/kg dry matter of hay) for 72 d. All sheep received concentrate mixtures (0.5% of body weight) throughout the experiment. In both trials, it was observed that plasma Ca concentration (11.0-11.7 mg/dl) was significantly (p<0.05) lower in sheep fed high oxalate-containing grasses than in sheep fed low oxalate-containing grasses (12.4-13.7 mg/dl). No differences (p>0.05) were observed in concentrations of magnesium, phosphorus and parathyroid hormone in plasma between the feeding of low and high oxalate-containing grasses. In addition, no differences (p>0.05) were observed in roughage dry mater (DM) intake, total DM intake or body weight of sheep. This study suggests that sheep may consume oxalate-rich forage, but Ca bioavailability may decrease with increasing oxalate levels in the ration.

Dissolved organic matter characteristics and bacteriological changes during phosphorus removal using ladle furnace slag

  • A sidestream contains the filtrate or concentrate from the belt filter press, filter backwash and supernatant from sludge digesters. The sidestream flow, which heads back into the sewage treatment train, is about 1-3% less than the influent flow. However, the sidestream can increase the nutrient load since it contains high concentrations of phosphorus and nitrogen. In this study, the removal of PO4-P with organic matter characteristics and bacteriological changes during the sidestream treatment via ladle furnace (LF) slag was investigated. The sidestream used in this study consisted of 11-14% PO4-P and 3.2-3.6% soluble chemical oxygen demand in influent loading rates. LF slag, which had a relatively high $Ca^{2+}$ release compared to other slags, was used to remove $PO_4-P$ from the sidestream. The phosphate removal rates increased as the slag particle size decreased 19.1% (2.0-4.0 mm, 25.2% (1.0-2.0 mm) and 79.9% (0.5-1.0 mm). The removal rates of dissolved organic carbon, soluble chemical oxygen demand, color and aromatic organic matter ($UV_{254}$) were 17.6, 41.7, 90.2 and 77.3%, respectively. Fluorescence excitation-emission matrices and liquid chromatography-organic carbon detection demonstrated that the sidestream treatment via LF slag was effective in the removal of biopolymers. However, the removal of dissolved organic matter was not significant during the treatment. The intact bacterial biomass decreased from $1.64{\times}10^8cells/mL$ to $1.05{\times}10^8cells/mL$. The use of LF slag was effective for the removal of phosphate and the removal efficiency of phosphate was greater than 80% for up to 100 bed volumes.

Studies on Vitamin Mineral Interactions in Relation to Passive Transfer of Immunoglobulins in Buffalo Calves

  • Status of blood minerals and their absorption by neonate calves as influenced by fat soluble vitamins supplementation in their respective mothers, mineral supplementation in calves themselves has been evaluated. The objective was to know the impact of antioxidant vitamin supplementation to advance pregnant buffaloes, on enhanced acquired immunity during first few hours after birth, in relation to weight gain in buffalo calves. Advance pregnant buffaloes (n = 30) consisting of average body weight of $550{\pm}15$ kg and of 4-6 parity were fed on 25 kg green (green Jawar-Sorghum bicolor), 2-3 kg wheat straw and 3-4 kg concentrate mixture individually per day. Intramuscular injections of vitamin triplex A $D_3$ E consisting of -2,500,000 IU of vit A -Palmitate; 2,500,000 IU of vitamin $D_3$ and 1,000 IU of vit E (dl-alpha tocopherol acetate) were given per dose, a month prior to parturition, twice at 15 days interval to 15 dams. Rest of the 15 pregnant buffaloes served as negative controls. Secretion of immune proteins, immunoglobulin (Ig) enhanced by 80% in colostrum. The blood serum levels of Zn, Cu, Ca, Mg were measured from birth to 90 days in calves. A significant (p<0.05) difference between the blood serum Zn levels of calves born to vitamin supplemented and non-supplemented dams was measured and a positive correlation between blood serum Zn levels and injections of vitamins was identified. Association of Zn and Cu with passive immunity status has been identified in these calves. A significant positive correlation between Zn and Cu was also identified which showed a change under the impact of vitamin supplementation in buffaloes. The study signifies the role of micronutrients supplementation in dams prior to parturition, in calf immunity development. The study indicates significant mineral - vitamins interactions during this process.

Evaluating Nutritional Quality of Single Stage- and Two Stage-fermented Soybean Meal

  • This study investigated the nutritional quality of soybean meal (SBM) fermented by Aspergillus ($FSBM_A$) and/or followed by Lactobacillus fermentation ($FSBM_{A+L}$). Both fermented products significantly improved protein utilization of SBM with higher trichloroacetic acid (TCA) soluble true protein content, in vitro protein digestibility and available lysine content, especially in $FSBM_{A+L}$. Moreover, $FSBM_{A+L}$ produced a huge amount of lactic acid resulting in lower pH as compared to the unfermented SBM or soybean protein concentrate (SPC) (p<0.05). $FSBM_A$ and $FSBM_{A+L}$ raised 4.14% and 9.04% of essential amino acids and 5.38% and 9.37% of non-essential amino acids content, respectively. The ${\alpha}$-galactoside linkage oligosaccharides such as raffinose and stachyose content in $FSBM_A$ and $FSBM_{A+L}$ decreased significantly. The results of soluble protein fractions and distribution showed that the ratio of small protein fractions (<16 kDa) were 42.6% and 63.5% for $FSBM_A$ and $FSBM_{A+L}$, respectively, as compared to 7.2% for SBM, where the ratio of large size fractions (>55 kDa, mainly ${\beta}$-conglycinin) decreased to 9.4%, 5.4% and increased to 38.8%, respectively. There were no significant differences in ileal protein digestibility regardless of treatment groups. SPC inclusion in the diet showed a better protein digestibility than the SBM diet. In summary, soybean meal fermented by Aspergillus, especially through the consequent Lactobacillus fermentation, could increase the nutritional value as compared with unfermented SBM and is compatible with SPC.

자색 고구마 농축액을 첨가하여 제조한 젤리의 품질특성 및 항산화활성 (Quality and Antioxidant Properties of Jelly Incorporated with Purple Sweet Potato Concentrate)

  • 자색 고구마 농축액의 첨가량을 0-4%로 달리하여 젤리를 제조한 후 물리 화학적 품질, 항산화특성 및 소비자 기호도를 비교하였다. 자색 고구마 농축액의 첨가 비율이 증가함에 따라 젤리의 pH와 가용성 고형분은 다소 증가하는 경향을 보였지만 유의적 차이는 발견되지 않았다(p>0.05). 수분함량은 82.03-83.82% 범위의 값을 나타내었으며 실질적인 차이는 미미하였다. 밝기를 나타내는 $L^*$값은 자색 고구마 농축액의 첨가량이 증가할수록 유의적으로 감소하였고(p<0.05), 반면 적색도를 나타내는 $a^*$값과 황색도를 나타내는 $b^*$값은 유의적으로 증가하는 경향을 나타내었다(p<0.05). 흡광도 및 경도(0.18-0.21 $kg_f$)는 자색 고구마 농축액의 첨가량이 증가함에 따라 유의적으로 증가하였다(p<0.05). 항산화특성을 나타내는 총 페놀화합물 함량과 전자공여능 또한 유의적 차이를 나타내며 증가하였다(p<0.05). 소비자 선호도 검사결과 농축액을 첨가한 젤리가 그렇지 않은 시료에 비해 높은 맛에 대한 평가를 받았으며, 전체적인 기호도 측면에서도 2% 첨가군이 가장 높게 평가되어 젤리의 관능품질과 건강 기능성 효과 등을 고려할 때 자색 고구마 농축액을 2% 첨가한 젤리가 가장 적절한 것으로 판단된다.

2단계 고정화 효소반응기를 활용한 Cyclodextrin의 연속생산 (Continuous Production of Cyclodextrin in Two-Stage Immobilized Enzyme Reactor Coupled with Ultrafiltration Recycle System)

  • The two-stage enzyme reactor, packed with cyclodextrin glucanotransferase (CGTase) immobilized on Amberite IRA 900, coupled with ultrafiltration membrane was investigated for continuous production of cyclodextrin (CD). 5% (w/v) of soluble starch was partially cyclized, in the 0.1 l first-stage immobilized enzyme reactor, up to CD conversion yield of 10% (w/w) at retention time of 0.56hr and 1.5 units of immobilized CGTase/1g of carrier. In the second stage main immobilized enzyme reactor capacity of 1.5 l, the maximum CD conversion yield of 39% (w/v) was achieved at retention time of 2.8hr and 0.47 unit of CGTase/1 g of carrier. Unreacted residual dextrin was fractionated with ultrafiltration membrane, and then, recycled into the second-stage main bioreactor to increase the CD conversion yield. The most suitable membrane size and the volume concentration ratio (concentrate: filterate) for recycling of unreacted residual dextrin were found to be 5K dalton and 4:6, respectively. CD conversion yield was increased about 3~4% upon co-immobilization of pulluanase along with CGTase. Spent Amberite IRA 900 can be reutilized consecutively more than 3 times for immobilization of CGTase after regeneration.

Utilization of Ruminal Epithelial Cells by Ruminococcus albus, with or without Rumen Protozoa, and Its Effect on Bacterial Growth

  • Effects of supplementation with ruminal epithelial cells on fiber-degrading activity and cell growth of Ruminococcus albus (R. albus, strain 7) was tested using a basal substrate of rice straw and formulated concentrate. Cultures of R. albus alone and R. albus with rumen protozoa were grown at $39^{\circ}C$ for 48 h with an 8.4% crude protein (CP) substrate, 33% of the CP supplemented with either ruminal epithelial cells or defatted soybean meal. The ruminal epithelial cells had lower amounts of rumen soluble and degradable protein fractions as compared to defatted soybean meal, as determined by an enzymatic method, and the same was found with amino acid composition of protein hydrolysates. Ruminal epithelial cells were directly utilized by the R. albus, and resulted in greater growth of cell-wall free bacteria compared to defatted soybean meal. The effect of epithelial cells on bacterial growth was enhanced by the presence of rumen protozoa. In consistency with cultures of R. albus and R. albus with rumen protozoa, fermentative parameters such as dry matter degradability and total volatile fatty acid did not differ between supplementation with ruminal epithelial cells or defatted soybean meal.

Ruminal Degradation of Sugarcane Stalk

  • The number of protozoa and VFA content in the rumen fluid, in situ disappearance and turnover rate were examined with four rumen-fistulated cattle given either sugarcane stalk or Ruzi grass hay in order to clarify the manner of rumen digestion of sugarcane stalk. Cattle were given either sugarcane stalk or Ruzi grass hay at 1.0% of body weight level with commercial concentrate feed. Feeding sugarcane stalk reduced acetate content and increased propionate and butyrate contents in rumen fluid. While rapidlysoluble fraction of sugarcane stalk was 42%, the insoluble but potentially degradable fraction was only 17%. This clearly showed that sugarcane stalk mainly consisted of water soluble fraction (i.e. sugar) and tough fiber (i.e. bagasse). The ruminal degradation rate of both Ruzi grass hay and sugarcane stalk was lower in the animal given sugarcane stalk in comparison with those given Ruzi grass hay. While the turnover rate of liquid phase was about 50% higher in the animals given sugarcane stalk than in the animals given Ruzi grass hay, that of the solid phase was about 40% lower in the animals given sugarcane stalk. The effective degradability of DM of sugarcane stalk was higher than that of Ruzi grass hay. Sugarcane would be a promising roughage for ruminants in the tropics especially, in the dry season.

타닌접착제를 위한 시베리아산 낙엽송 수피 온수추출물의 한외여과 특성 (Ultrafiltration Characteristics of Hot-Water Extracts from Siberian Larch Barks for Tannin-based Adhesives)

  • Hot-water soluble extracts were prepared from medium-sized barks of Siberian larch (Larix gmelineii). The extracts were fractionated by ultrafiltration method for producing uniform quality of adhesives. Molecular weight distribution of the extracts was ranged of 100 to 300.000. pH of the extracts affected amounts of flux, and the range of pH 6~pH 8 was proper for wood adhesives, because of easy concentration and fast gelation time. Removal of particles greater than $0.45{\mu}m$ from the extracts increased both filtration speed (flux) and yields of solids in the filtrates. In ultrafiltration process. operating pressure, filtrate temperature, and flow rate significantly increased with the increase of individual condition. Ultrafiltration using PM10 membrane was very effective to fractionate and concentrate the extracts. Removal of large particles greater than 0.45 m from the extracts increased filtration speed(flux) and yields of solids in the filtrates. A gelation time was accelerated with the increasing pH of the extracts and its concentration. The Stiasny precipitate(26%) from the filtrate obtained by PM 10 membrane was very lower than that(78%) of the retentates. This ultrafiltration method was efficient for obtaining high yield purified phenolic compounds.
