Optimization of ginseng hairy roots culture and its ginsenoside analysis

  • Hairy root culture of ginseng is industrially prospected because the cultivation period of ginseng is relatively long. In this study, the effect of medium concentration and sucrose concentration on hairy root culture of ginseng was evaluated. The optimization of ginseng hairy roots transformed by Agrobacterium rhizogene were performed liquid medium. The MS(Murashinge & Skoog basal medium) concentration was selected with 1/2 strength MS and the optimal sucrose concentration was determined at 2-3%(w/v). At the optimum culture condition, The yield (the ratio of weight of grown hairy root cultures to weight of fresh ginseng hairy roots) and production rate of ginseng root were 19.42 times and 5.73 g/l-day. The major ginsenosides were Rb group, Re and Rg1. The produced total ginsenoside content in the solid medium was 9.87 (mg/g) and increased 1.34 times in the liquid medium (13.23 mg/g). In solid culture, the contents of ginsenosides Rb, Re and Rg1 were 2.14, 3.65 and 1.87 mg/g, respectively. In liquid culture, the contents of ginsenosides Rb, Re and Rg1 were 3.54, 4.12 and 2.63 mg/g, respectively.

Characteristics of Hg, Pb, As, Se Emitted from Medium Size Waste Incinerators (중형폐기물 소각시설의 수은, 납, 비소, 셀렌 배출특성)

  • The aim of this study is to evaluate the emission characteristics of mercury, lead, arsenic, and selenium from medium size municipal solid waste incinerators(MSWIs) in Korea. The concentrations of mercury, lead, arsenic, and selenium emitted from medium size MSWI stack were $2.67\;{\mu}g/Sm^3,\;0.38\;mg/Sm^3,\;1.33\;{\mu}g/Sm^3,\;0.28\;{\mu}g/Sm^3$, respectively. The concentration levels of mercury, lead, arsenic in flue gas from medium size MSW incinerator stacks selected were nearly detected under the Korea criteria level. Removal efficiencies of mercury, lead, arsenic, and selenium in waste heat boiler(WHE) and cooling tower(CT) were $90.36\%,\;69.76\%,\;43.04\%,\;40.64\%$, respectively. In general, the removal efficiencies of mercury and lead in WHE were higher than those of arsenic and selenium in WHE. Emission gas temperature reduction from waste heat boiler(WHB) and cooling tower(CT) can control mercury and lead of medium size MSWIs. To evaluate the relationship between mercury, lead, arsenic, selenium of fly ash and those of flue gas, it was carried out to correlation analysis of each metal concentration in the fly ash and in the flue gas from medium size MSWIs. From the correlation analysis, the coefficients of mercury, lead, arsenic, and selenium were 0.61, -0.38, 0.87, 0.28, respectively. The results of correlation analysis revealed that it should be highly positive to the correlation coefficients of mercury and arsenic in the fly ash and those of the flue gas emitted from medium size MSWIs. As it were, the concentrations of mercury and arsenic of flue gas from medium size MSWIs are high unless mercury and arsenic in fly ash are properly controlled in dust collection step in medium size MSWIs. It was also concluded that mercury, lead, arsenic, and selenium from MSWIs stacks could be controlled by waste heat boiler(WHE) and dust collecting step in medium size MSWIs.

Establishment of a novel plant regeneration system from suspension-derived callus in the halophytic Leymus chinensis (Trin.)

  • The establishment of cell suspension culture and plant regeneration of the halophytic Leymus chinensis (Trin.) are described in this study for the first time. Callus induction solid medium containing Murashige and Shoog (MS) basic salt, $2.0\;mg\;l^{-1}$ 2,4-dichlorophenoxyacetic acid (2,4-D), and $5.0\;mg\;l^{-1}$ L-glutamic acid with $30.0\;g\;l^{-1}$ sucrose and $4.0\;g\;l^{-1}$ gelrite for solidification induced the highest rate of cell division in Type 1 callus among calli of various types. Liquid medium with the same hormone distribution was therefore, used for cell suspension culture from Type 1 callus. Over a 30 d suspension culture at 100 rpm, great amounts of biomass were accumulated, with 71.07% average daily increment and 22.32-fold total fresh weight increment. Comparison of before and after suspension culture, the distribution of different size callus pieces and the maintenance of callus type were basically unaltered, but a slight increase in relative water contents was observed. To induce the potential of plant regeneration, the directly transferring on plant regeneration solid medium containing MS basic salt, $0.2\;mg\;l^{-1}$ $\alpha$-naphthalene acetic acid (NAA), $2.0\;mg\;l^{-1}$ kinetin (Kn), and $2.0\;g\;l^{-1}$ casamino acid and indirectly transferring were simultaneously performed. Even now growth rates of suspension-derived callus on solid medium were approximately half of those of Type 1 callus, but faster somatic embryogenesis was observed. Rooting of all regenerated shoots was successfully performed on half-strength MS medium. All plants appeared phenotypically normal.

Mass production of potato shoots by liquid culture (액체배지를 이용한 조직배양 감자줄기의 대량증식)

  • A study was conducted to investigate in vitro micro propagation of four potato cultivars of Daese, Jasim, Chubaek and Haryeng in MS medium and PM medium (a medium containing half concentration of $NH_4NO_3$ and $KNO_3$, two fold concentration of $CaCl_2$ as compared to MS medium). During $1{\sim}2$ weeks of culture, Daese and Jasim showed better shoot elongation on the solid MS medium than Chubaek and Haryeng whereas Chubaek and Haryeng did better shoot elongation on the solid PM medium. But no difference was observed after 4 weeks of culture. As compared to shoot elongation on the solid medium, it was delayed at early stage of culture in the liquid medium without shaking. Shoot formation ratio of potato (above 4 cm of shoot length) began to increase significantly after 1 week of culture and kept on increasing until 4 weeks. The four cultivars showed the different patterns of shoot growth in bioreactor. The PM medium with a quarter salt strength was effective for the regeneration of axillary buds as well as shoot elongation of Jasim and Chubaek. Daese showed vigorous regeneration of axillary buds in the PM medium with a half salt strength. On the other hand, Haryeng showed slower growth than the other three cultivars.

The effect of medium change after pretreating microspores, medium addition, and volume of under solid medium in double layer culture on the production of embryos in isolated microspore culture of hot pepper (Capsicum annuum L.) (고추의 소포자 배양 시 전처리 후 배지의 교환, 배지의 첨가 및 2층배양 시 하층고체 배지의 양이 배의 생산에 미치는 영향)

  • The effect of the addition of the fresh medium, volume of under solid medium in double layer culture as well as the medium change after pretreating microspores on the production of embryos in microspore culture of hot pepper (Capsicum annuum L.) has been studied. When cultured after heat pre-treatment, changing pretreatment media with fresh culture media proved to be more effective for embryo production rather than supplementing additional culture media. Heat-pretreating for 3 days turned out more effective for embryo production than pretreating for 1 or 2 days. In the case of anther pretreatment, the addition of fresh medium after culture was not effective for embryo production. In pretreating microspores, however, supplementing additional fresh culture media greatly improved embryo yield and quality. The best time point of media addition was 4 days after culture commenced, and the most effective number of times of media addition was one time addition. Moreover, the effective volume of added medium in double layer culture for embryo production was 1.5 ml. The addition of media more than 1.5 ml reduced both embryo yield and quality. Double layer medium was more effective for embryo development than liquid medium. When the volume of under solid medium increased ranging from 3 ml to 7 ml, more cotyledonary embryos were produced in either 5 ml or 7 ml compared to 3 ml, even though the total number of embryos were highest in 3 ml. These results can be used as an important data for establishing an efficient microspore culture system for producing high frequency of normal embryos in hot pepper.

Somatic Embryogenesis and Plant Regeneration from Poinsettia (Euphorbia pulcherrima L.) Stem Explants (포인세티아 줄기조직배양에 의한 재분화체 제조.)

  • Conditions for somatic embryogenesis and plant regeneration from stem tissues of Euphorbia pulcherrima were esta-blished. Explants from leaf, petiole, stem were examined for their embryogenesis on MS solid medium supplemented with plant growth hormones in combination at various concentrations. From leaf or petiole explants, callus was indu-ced well but never proceeded to the embryonic stage in our expermental conditions. From stem explants, however, multiple shoots following callus induction emerged in about 6 to 8 weeks on MS agar medium supplemented with 1.5 mg/L of benzyladenine. Upon transfer, roots were developed on hormone-free MS solid medium.

  • The present paper is concerned with the investigation of Rayleigh waves in a homogeneous transversely isotropic magnetothermoelastic medium with two temperature, in the presence of Hall current and rotation. The formulation is applied to the thermoelasticity theories developed by Green-Naghdi theories of Type-II and Type-III. Secular equations are derived mathematically at the stress free and thermally insulated boundaries. The values of Determinant of secular equations, phase velocity and Attenuation coefficient with respect to wave number are computed numerically. Cobalt material has been chosen for transversely isotropic medium and magnesium material is chosen for isotropic solid. The effects of rotation, magnetic field and phase velocity on the resulting quantities and on particular case of isotropic solid are depicted graphically. Some special cases are also deduced from the present investigation.

  • A critical angle method was used to measure the index of refraction of a solid medium when an air gap between the prism and the medium is present. The gap effect was analyzed both numerically and experimentally. Since the total internal reflection is severely disturbed by the large gap, determination of the critical angle and the resulting refractive index becomes ambiguous and inaccurate. By using an index matching fluid, we could determine the index of refraction with an uncertainty of ${\pm}2{\times}1^{-3}$ even when the gap is as large as 1 ${\mu}m$.

  • The present paper investigates the heat transfer characteristics in a cylinder packed with porous medium of solid spheres for various parameters such as mass flow rate, sphere diameter, length of the porous medium, and gas temperatures. Pressures and temperatures at the inlet and outlet regions were measured by using static pressure gages and R-type thermocouples. The modified relationship based on the Ergun equation is suggested for the estimation of pressure drops. In addition, the useful empirical correlation for thermal efficiency is obtained in the current study. Thermal efficiency is expressed in terms of non-dimensional time, sphere diameter, porosity, and pressure drops. It is also found that the pressure drop through the cylinder becomes larger as the gas temperature does higher at the inlet region, whereas it substantially decreases when the inlet flow rate decreases.

Evaluation of Absorption and Release, Survival Efficiency and Recovery Rate of Transport Medium according to the CLSI M40-A2 Standard (CLSI M40-A2 기준에 따른 수송배지의 흡수 및 배출, 생존 효율, 회수율 평가)

  • The absorption and release capacities, survival efficiency, and recovery rates of five kinds of transport media were evaluated based on the swab elution method (Quantitative) of CLSI (Clinical and Laboratory Standards Institute) M40-A2. Liquid media showed mostly better results than semi-solid media in the three evaluations. The flocked swabs had better ability to absorption and discharge bacteria than the standard swabs. The liquid medium (S4) had the best survival efficiency. Pneumococcal strains with poor growth had a higher survival efficiency and recovery rate in liquid media (S4, S5). The results of microbial recovery showed that S. pyogenes met all the CLSI standards in all media. S. pneumoniae was inadequate in the semi- solid media (S2, S3) and all the remaining media met the criteria. H. influenzae was unsuitable in semi-solid media (S1, S3) and met the criteria in semi-solid medium (S2) and liquid medium (S4, S5). The viability of the H. influenzae, pneumococcal strain causing respiratory disease, was poor in most media. Overgrowth of P. aeruginosa was observed at room temperature. The combination of liquid medium and flocked swab confirmed the best results in the three evaluation methods.
