Preparation and Characterization of Piroxicam/Poloxamer Solid Dispersion Prepared by Melting Method and Solvent Method

  • Solid dispersions of piroxicam were prepared by melting method using poloxamer as a carrier. The results of DSC and XRD studies showed that the amorphous farm of piroxicam coexisted with the crystalline form in the solid dispersions. However, the ratio of crystalline form of piroxicam in the solid dispersion prepared by melting method decreased in comparison with the same ratio of the solid dispersion prepared by solvent method. As the ratio of poloxamer in the solid dispersion increased, the ratio of the amorphous form of piroxicam in the solid dispersion increased. The dissolution rate of piroxicam from the solid dispersions was significantly higher than that from piroxicam powder. In comparison to the solid dispersion prepared by solvent method, the dissolution rate of piroxicam from the solid dispersion prepared by melting method was higher. As the ratio of poloxamer in the solid dispersion prepared by melting method increased, the initial dissolution rate decreased, however, the total amount dissolved at the end of the study increased.

  • Sodium sulfate is commonly added in reactive dyebath in order to increase substantivity of the reactive dye to cellulose fiber by reducing repulsion between anionic dye and fiber. While sodium sulfate is mostly used in solid form, it is inconvenient to dissolve a large amount of powder sodium sulfate. Furthermore, if there is undissolved salt in dyebath it might cause unlevel dyeing. In this study, sodium sulfate in liquid or solid form was used in dyeing of cellulose fabric with reactive dyes of three primary color and the effect of type or amount of sodium sulfate on dyeing and fastness properties was investigated. When the amount of sodium sulfate rose to 30-50 g/l, K/S value of the dyed fabric markedly increased; further rise in sodium sulfate concentration resulted in slow increase in K/S value. For light color, optimum amount was about 30 g/l in solid form and 50-100 g/l in liquid form while, for medium to deep color, it was 50 g/l and 100-150 g/l in solid and liquid form, respectively. When using each optimum amount of salt in solid or liquid form for medium color, shape of dyeing curve as well as exhaustion was similar to each other. On the whole, similar color fastness results were obtained regardless of type or amount of sodium sulfate.

방사성폐기물 아스팔트 고화체 안정화 특성연구 (A Development of the Stabilization Technology for the Solid Form of Radioactive Waste)

  • 본 연구에서는 방사성폐기물 아스팔트고화체를 장기간 저장시 형태유지가 되지 않는 단점을 보완하기 위하여 폴리에틸렌을 고화보조제로 아스팔트에 첨가하여 전처리의 분쇄 공정 없이 입상수지를 직접 고화처리 함으로서 시멘트 고화체처럼 형태안정성을 지니는 아스팔트 고화체를 제조하는 방법을 개발하고 이 방법에 의해 제조된 고화체의 침출특성을 평가하여 실제 방사성 폐이온 교환수지 처리에 적용함을 그 목적으로 하였다. 실험결과 고화보조제인 폴리에틸렌은 아스팔트와 폐수지가 혼합된 고화체내에서 가교역활을 하며 PE 함유량이 10 wt% 이상일 때 장기간 노출실험에서도 고화체 형태를 그대로 유지하였다. 최적조업조건은 폐수지의 함유량이 무게비로 40wt% 이고 PE의 함유량이 10wt%이며 이때 고화체내부에 수지입자의 밀도도 균일하게 분포됨을 확인하였다. 실험은 폐이온교환수지에 아스팔트를 혼합하고 여기에 폴리에틸렌(PE) 필름을 고화보조제로 첨가함으로서 고화체의 균일성, 수분과 접촉시 고화체의 침출을 최저로 하는 안정된 고화체를 개발하였다.

흰쥐에 경구 투여시 히드록시프로필-베타-시클로덱스트린과 니트렌디핀 고체분산에 의한 생체이용률 증가 (Enhancement of Nitrendipine Bioavailability in Rats by its Solid Dispersion with $Hydroxypropyl-{\beta}-Cyclodextrin$ after Oral Administration)

  • Nitrendipine, a slightly soluble calcium channel blocking agent forms a solid dispersion system with $hydroxypropyl-{\beta}-cyclodextrin$, which exhibits better dissolution characteristics than the uncomplexed drug. The dissolution rate of nitrendipine was markedly increased in solid dispersion system in pharmacopeial disintegration media at pH 1.2 and pH 6.8. Four different dosage forms of nitrendipine were administered to rats: (a) nitrendipine in the solution of PEG 400; (b) nitrendipine solid dispersion system with $hydroxypropyl-{\beta}-cyclodextrin$ in a molar ratio of 1:2 by solvent evaporation method and administered in capsule form; (c) physical mixture of nitrendipine with $hydroxypropyl-{\beta}-cyclodextrin$ in a molar ratio of 1:2 and administered in capsule form; (d) nitrendipine alone administered in capsule form. Relative bioavailability after the oral administration of various dosage forms to rats with a dose of 10 mg/kg equivalent to nitrendipine was compared with that of nitrendipine in the solution of PEG 400. The AUC of solid dispersion was significantly bigger than that of nitrendipine powder. $T_{max}$ of solid dispersion was significantly shorter and $C_{max}$ was higher than that of nitrendipine powder. These results indicate that the bioavailability of nitrendipine could be improved markedly by inclusion complexation. An interesting correlation also appears to exist between the in vitro dissolution data and the area under the plasma concentration-time curves.

반고상 A356 합금 슬러리의 미세조직에 따른 유동특성에 관한 연구 (Effects of Microstructure Morphology on Fluid Flow Characteristics of A356 Commercial Alloy in Semi-Solid Slurry)

  • The rheocasting characteristics are strongly influenced by the microstructural morphology such as particle size, form factor and contiguity. In this study, the effect of structural morphology on fluid flow characteristics of A356 semi-solid alloy was investigated with a vacuum suction fluidity test. Semi-solid metal slurry was made by the mechanical stirring, the liquidus casting, and H-NCM to be analysed. H-NCM could obtain uniform and fine globular microstructures of 0.9 form factor and 55 ${\mu}m$ particle size. Inoculation was found to be effective for reducing particle size, however, for H-NCM it should be avoided due to the cause of increasing contiguity. The fluidity test indicated that the non-stirring method had higher fluidity and smaller liquid segregation in the same solid faction of 0.4 than the stirring method, for smaller particle size and higher form factor. It was observed that liquid segregation decreased as the particle size is smaller and form factor is higher. The results of die-casting experiment were a good agreement with those of fluidity test.

비결정성 세푸록심 악세틸 고체분산체의 제조 및 평가 (Preparation and Evaluation of Non-Crystalline Cefuroxime Axetil Solid Dispersion)

  • Cefuroxime axetil is a cephalosporin antibiotic having a high activity against a wide spectrum of Grampositive and Gram-negative microorganisms. It is a cephalosporin antibiotic which exist as 2 diastereoisomers: diastereoisomer A and B. It shows polymorphism of three forms: a crystalline form having a melting point of about $180^{\circ}C$, a substantially amorphous form having a high melting point of about $135^{\circ}C$ and a substantially amorphous form having a low melting point of about 70^{\circ}C$. The crystalline form of cefuroxime axetil is slightly soluble in water because diastereoisomer A has lower solubility than B in water. Substantially amorphous form of which there are no difference in solubility between diastereoisomer A and B has better solubility than crystalline form, but it forms a thicker gel than crystalline form upon contact with an aqueous medium. Based on this reason, cefuroxime axetil is not readily absorbable in the gastrointestinal tract, rendering its bioavailability on oral administration very low. The object of this study was to develop an improved non-crystalline cefuroxime axetil composition having a high physicochemical stability and bioavailability. A non-crystalline cefuroxime axetil solid dispersant showing no peak on a Differential Scanning Calorimetry (DSC) scan is prepared by dissolving cefuroxime axetil and a surfactant in an organic solvent; suspending a water-insoluble inorganic carrier in the resulting solution; and spray drying the resulting suspension to remove the organic solvent, said solid dispersant having an enhanced dissolution and stability of cefuroxime axetil and being useful for the preparation of a pharmaceutical composition for oral administration. Tablet was formulated with this cefuroxime axetil solid dispersant, disintegrants and other ingredients. It disintegrated and dissolved easily and dynamically in dissolution medium, so showed a good dissolution profile.

자유 곡면으로 구성되는 3차원 구조물에 대한 자동 요소 분할 (Automatic Mesh Generation for Three-Dimensional Structures Consisting of Free-Form Surfaces)

  • This paper describes an automatic finite element(FE) mesh generation for three-dimensional structures consisting of free-form surfaces. This mesh generation process consists of three subprocesses: (a) definition of geometric model, i.e. analysis model, (b) generation of nodes, and (c) generation of elements. One of commercial solid modelers is employed for three-dimensional solid and shell structures. Node is generated if its distance from existing node points is similar to the node spacing function at the point. The node spacing function is well controlled by the fuzzy knowledge processing. The Delaunay method is introduced as a basic tool for element generation. Automatic generation of FE meshes for three-dimensional solid and shell structures holds great benefits for analyses. Practical performances of the present system are demonstrated through several mesh generations for three-dimensional complex geometry.

양파(Allium cepa L,) 멀칭재배시 질소비료 추비방법이 생육, 수량 및 저장성에 미치는 영향 (Effect of Topdressing Methods of Nitrogen Fertilizer on Growth, Yield and Storage of Onion(Allium cepa L.) in Mulch-Cropping System)

  • In order to fad out an efficient way of topdressing nitrogen fertilizer in mulch-cropping system of onion(Allium cepa L.), solid, slow-release, and liquid forms of nitrogen fertilizers were allied to cv. 'Changnyungdaego' various number of times at different time, with 5 topdress applications of solid fertilizer serving as a control. Whole basal application of conventional solid fertilizer and 2 slow-release fertilizers were labor-saving and showed improved storage quality of bulbs, but resulted in poor plant growth and considerably low yield due to fertilizer shortage from early April. This suggests that topdress application is necessary. Liquid form of nitrogen fertilizer was more effective for plant growth and yield and saving labor than the solid form. Early applications was effective for increasing yield and storage quality of onion bulbs harvested. Thus two applications of liquid form of nitrogen fertilizer in February and March at rome month interval are recommended in mulch crowing system of onion.

  • The thickness ranges and conditions to form the supersaturated solid solution in Al-Cr alloys were investigated with various rapid solidification conditions. Al-Cr alloys, rapidly solidified by using the small droplet chill quenching method, were examined by means of micro-vickers hardness, lattice parameter, thermal analysis and microscopic observation. The results obtained were as follows; 1. With the increase of solidification rate, the solidified structures were changed to intermetallic compound + solid solution, incompletely supersaturated solid solution, completely supersaturated solid solution, in turn. 2. The minimum solidification rate required to form completely supersaturated solid solution was $2.5{\times}10^{-2}cm/sec$, $3.6{\times}10^{-2}cm/sec$ and $6.0{\times}10^{-2}cm/sec$ for Al-1.0wt%Cr, Al-1,2wt%Cr and Al-1.5wt%Cr, respectively. 3. The maximum distance from the chill surface required to form completely supersaturated solid solution was 5mm, 1.3mm and 0.3mm for Al-1.0wt%Cr, Al-1.2wt%Cr and Al-1.5wt% Cr, respectively.

Drug Polymorphism and its Importance on Drug Development Process

  • Polymorphism has been recognized to be a critical issue throughout the drug product development process. Most of solid phase drugs have polymorphism, which has generated a great deal of interest and the field has been evolving rapidly. Preferably, thermodynamically most stable form of a drug substance is selected to obtain consistent bioavailability over its shelf life and various storage conditions. Moreover, it has the lowest potential for conversion from one polymorphic form to another. However, metastable or amorphous forms may be used intentionally to induce faster dissolution rate for rapid drug absorption and higher efficacy. For pharmaceutical industry, polymorphism is one of the key activities in form selection process together with salt selection. This article introduces the main features in the investigation of solid form selection especially polymorphic behavior with thermodynamic backgrounds, physicochemical properties with solubility, dissolution, and mechanical properties, and characterization techniques for proper analysis. The final form can be recommended based on the physicochemical and biopharmaceutical properties and by the processability, scalability and safety considerations. Pharmaceutical scientists especially in charge of formulation need to be well aware of the above issues to assure product quality.
