Variation in Ureide and Amide Content during the Growth period of some Soybean Cultivars and its Physiological Significance (대두생장중(大豆生長中) Ureide와 Amide의 소장양상(消長樣相)과 그의 생리적(生理的) 의의(意義)에 관한 연구(硏究))

  • Samples of plant tissue and xylem sap collected from pot-grown three soybean cultivars (Hwangkum, Williams 79, Wild) were analyzed for ureide and amide, and their distribution and changes in contents were measured over the growth period to study whether there are any differences in the changes of ureide contents among soybean cultivers. Application of ureide content as an indicator of $N_2$-fixation was examined. There was little evidence of soybean cultivars affecting the patterns of the change of ureide contents, while relative ureide contents of plant organ or xylem sap were significantly different among soybean cultivars. In plant stem and xylem sap, negative correlation ($r=-0.5193^{**}$, $-0.5053^*$ respectively) were found between ureide-N and amide-N content. Considering that exogeneous nitrogen caused a decrease in ureide accumulation and that nodule is the main site of ureide formation, the results demonstrated that there were differences in the effect of $N_2$-fixation among soybean cultivars. Consequently, the relative uredie content of xylem sap or plant stem could be used as a practical indicator of $N_2$-fixation in soybeans.

Several causes of non virus-induced mosaic symptom on potato leaves and its induction by herbicides (감자 이상모자이크증상의 몇 가지 발생원인 및 제초제에 의한 증상 유기)

  • In recent, non virus-induced mosaic symptoms(NVMS) on potato leaves were observed in the seed potato fields, and its incidence rate was $5{\sim}20%$ nationwide. It made difficult to rogue out virus-infected plants, and caused much arguments between seed potato production farmers and seed potato inspectors. The objectives of these experiments were to find out the causes of NVMS, and also to induce mosaic symptom(phytotoxicity) on potato plants by treatment of several herbicides. No significant correlations were found between incidence rates of NVMS and values from soil analyses; soil pH, soil EC, organic matter content, and contents of inorganic constituents($P_2O_5,\;NO_3$, Ca, Mg, K) in the soil around the potato planted. The examinations by ELISA, virus indicator plants, and TEM showed that NVMS on potato leaves was not caused by the viruses infection. But, the use of herbicides could induced the NVMS on potato leaves. The incidence rates of potato treated with pendimethalin linuron of 400 mL/10 a, pendimethalin of 200 mL/10 a, pendimethalin.oxadiazon of 300 mL/10 a, and control were 61.1%, 47.2%, 19.4%, and 1.4%, respectively. Based on these results, we confirmed that the treatment of pendimethalin alone and in mixture with other herbicides were the reason of NVMS on potato leaves. The yields among test plots were similar except dicamba treated plot, which decreased by about 23% compared to control plot. When their progenies harvested in 1999 were planted in the following season, no symptoms of mosaic were observed.

Microbiological Hazard Analysis for Strawberry Farms at the Harvest Stage to Establish Good Agricultural Practices (GAP) Model Based on Principle of HACCP (HACCP 원리에 기초하는 GAP모델 확립을 위한 딸기 농장의 수확단계에 대한 미생물학적 위해요소 조사)

  • This study assessed hazards at the harvest stage of strawberry farms which may cause risk to humans. A total of 216 samples were collected from 6 strawberry farms (soil culture farms: A, B, C; nutriculture farms: D, E, F) located in Western Gyeongnam. The collected samples were subjected for sanitary indicator bacteria (aerobic plate count, coliforms and Escherichia coli), major foodborne pathogens (E. coli O157:H7, Listeria monocytogenes, Salmonella spp., Staphylococcus aureus and Bacillus cereus), and fungi. The levels of APC and coliform in the soil culture farms were 1.0-6.9 and 0.4-4.6 log CFU/g (leaf, mL, hand or 100 $cm^2$), respectively. The samples obtained from the nutriculture farms were contaminated with the levels of 0.8-4.9, and 0.2-2.6 log CFU/g (leaf, mL, hand or 100 $cm^2$) of APC and coliform. However, E. coli was not detected in any samples. In major foodborne pathogens, S. aureus was detected at the level of ${\leq}$3.3 log CFU/hand in workers' hand samples and B. cereus was detected at the levels of 0.4-4.1 log CFU/g (hand or 100 $cm^2$) in soil, plants and workers' hygiene. L. monocytogenes, E. coli O157:H7 and Salmonella spp. were not detected. Fungi were detected at the levels of 1.0-5.2 and 0.2-4.4 log CFU/g (leaf, mL, hand or 100 $cm^2$) in soil culture and nutriculture farms, respectively.

Floristic Diversity of Serpentine Area in Andong, Korea (안동 사문암지대의 식물다양성)

  • This study was carried out to investigate the flora, vegetation and soil properties of serpentine area in Andong, Korea. The vascular plants identified during the seven-round field surveys were a total of 359 taxa: 88 families, 239 genera, 311 species, 6 subspecies, 33 varieties, 6 forms and 3 hybrids. 249 taxa were newly discovered in this region. The plant composition of serpentine area is the deciduous broad-leaved and conifer-mixed forest which is the common one in the middle part of the Korean peninsula. Four taxa of Korean endemic plants such as Clematis brachyura, Populus ${\times}$ tomentiglandulosa, Paulownia coreana and Aster koraiensis were collected. The vascular plants on the red list according to IUCN evaluation basis were found to be seven taxa: Near Threatened (NT) species of Hypericum attenuatum, Polygala tenuifolia and Senecio argunensis, Least Concern (LC) species of Penthorum chinense, Potentilla discolor and Acorus calamus, and Not Evaluate (NE) species of Scorzonera austriaca ssp. glabra. The floristic regional indicator plants found in this area were 19 taxa comprising two taxa of grade IV, five taxa of grade III, four taxa of grade II, and nine taxa of grade I. The naturalized plants were identified as 34 taxa and the percentage of naturalized index (NI) was 9.5 %, and urbanization index (UI) was 10.6 %. Forest soils contained high content of nickel and cadmium. The soil layer consists of loam and silt loam from the surface to a depth of 20 cm and loam and silt clay from a depth of 20 cm to 40 cm.

Hydrogeochemical Research on the Characteristic of Chemical Weathering in a Granitic Gatchment (水文化學的 資料를 통한 花崗岩質 流域의 化學的 風化特性에 關한 硏究)

  • This research aims to investigate some respects of chemical weathering processes, espcially the amount of solute leaching, formation of clay minerals, and the chemical weathering rate of granite rocks under present climatic conditions. For this purpose, I investigated geochemical mass balance in a small catchment and the mineralogical composition of weathered bedrocks including clay mineral assemblages at four res-pective sites along one slope. The geochemical mass blance for major elements of rock forming minerals was calculated from precipitation and streamwater data which are measured every week for one year. The study area is a climatically and litholo-gically homogeneous small catchment($3.62Km^2$)in Anyang-shi, Kyounggi-do, Korea. The be-drock of this area id Anyang Granite which is composed of coarse-giained, pink-colored miner-als. Main rock forming minerals are quartz, K-Feldspar, albite, and muscovite. One of the chracteristics of this granite rock is that its amount of Ca and Mg is much lower than other granite rock. The leaching pattern in the weathering profiles is in close reltion to the geochemical mass balance. Therefore the removal or accumulation of dissolved materials shows weathering patterns of granite in the Korean peninsula. Oversupplied ions into the drainage basin were $H^+$, $K^+$, Fe, and Mn, whereas $Na^2+$, $Mg^2+$, $Ca^2+$, Si, Al and $HCO-3^{-}$ were removed from the basin by the stream. The consumption of hydrogen ion in the catchment implies the hydrolysis of minerals. The surplus of $K^+$ reflects that vegetation is in the aggravation stage, and the nutrient cycle of the forest in study area did not reach a stable state. And it can be also presumed that the accumulation of $K^+$ in the top soil is related to the surplus of $K^+$. Oversupplied Fe and Mn were presumed to accumulate in soil by forming metallic oxide and hydroxide. In the opposite, the removal of $Na^+$, Si, Al resulted from the chemical weathering of albite and biotite, and the amount of removal of $Na^+$, Si, Al reflected the weathering rate of the bedrock. But $Ca^2+$ and $Mg^2+$ in stream water were contaminated by the scattered calcareous structures over the surface. Kaolinite is a stable clay mineral under the present environment by the thermodynamical analysis of the hydrogeochemical data and Tardy's Re value. But this result was quite different from the real assemblage of clay miner-als in soil and weathered bedrock. This differ-ence can be explained by the microenvironment in the weathering profile and the seasonal variation of climatic factors. There are different clay forming environments in the stydy area and these differences originate from the seasonal variation of climate, especially the flushing rate in the weathering profile. As it can be known from the results of the analysis of thermodynamic stability and characteristics of geochemical mas balance, the climate during winter and fall, when it is characterized by the low flushing rate and high solute influx, shows the environmental characteristics to from 2:1 clay minerals, such as illite, smectite, vermiculite and mixed layer clay minerals which are formed by neoformation or transformation from the primary or secondary minerals. During the summer and spring periods, kaoli-nite is a stable forming mineral. However it should consider that the other clay minerals can transformed into kaolinite or other clay minerals, because these periods have a high flushing rte and temperature. Materials which are directly regulated by chemical weathering in the weathered bedrock are $Na^+$, Si, and Al. The leaching of Al is, however, highly restricted and used to form a clay mineral, and that of Si falls under the same category. $Na^+$ is not taked up by growing veget ation, and fixed in the weathering profile by forming secondary minerals. Therefore the budget of $Na^+$ is a good indicator for the chemical weathering rate in the study area. The amount of chemical weathering of granite rocks was about 31.31g/$m^2+$/year based on $Na^+$ estimation.

Distribution of Foodborne Pathogens from Garlic Chives and Its Production Environments in the Southern Part of Korea (남부지방 부추와 재배환경의 식품매개병원균의 분포)

  • Recently, foodborne illness outbreaks linked to fresh produce are being increasingly reported in the United States, the EU, and Korea as well. Some of this increase may be due to improved surveillance, increase in consumption, change in consumers' habits, and complex distribution systems. Garlic chive is a green, fresh-cut vegetable consumed year-round as a nutrition-rich herb in Korea. It is also prone to contamination with foodborne pathogens during pre-harvest, as amendment with high amounts of livestock manure or compost to soil is required in its cultivation. Our aim in this study was to evaluate microbial contamination of garlic chives, garlic chives cultivation soil, compost, and irrigation water in the southern part of Korea. Samples were collected in A, B, and C regions in 2019 and 2020, and 69, 72, 27, and 40 of garlic chives, soil, compost, and irrigated water, respectively, were analyzed for the presence of sanitary indicator bacteria (total aerobic bacteria, coliforms and Escherichia coli), Bacillus cereus, Staphylococcus aureus, pathogenic E. coli, E. coli O157:H7, Listeria monocytogenes, and Salmonella spp. In A, B, and C regions, levels of total aerobic bacteria, coliform, B. cereus, and S. aureus on all samples were between 1.14 and 8.83 log CFU/g, 0.43 and 5.01 log CFU/g, 0.41 and 5.55 log CFU/g, and 1.81 and 6.27 log CFU/g, respectively. B. cereus isolated from garlic chives and environmental samples showed β-hemolysis activity. Incidence of S. aureus in garlic chive and its production environments in 2020 was different from 2019. In this study, B. cereus and S. aureus were the only pathogenic microorganisms detected in all samples. As a result, this work suggests that continuous monitoring in the production and pre-harvest environment is required to improve hthe hygiene and safety of garlic chive.

Species Diversity Analysis of the Aquatic Insect in Paddy Soil

  • The aquatic insect collected at six areas (each 2 for mountain area, plain field, and urban area) from 2009 to 2011 were classified to analyze the distribution and diversity of species. Frequency (number of aquatic insect: N), number of species (S), similarity index (C), richness index (R1, R2), variety index (V1, V2), evenness index (E1, E2, E3, E4, E5), and dominance index (D1) were investigated. Total N and S were 143 and 84, respectively. C matrix of 153 combinations was constructed with the average of 0.542. The average C of 3 years (0.659) was 9.9% P, more higher than the average C of 6 areas (0.560). The average values of the index of 18 plots were 2.28, 0.17, 1.24, 1.08, 0.07, 0.06, 0.01, 0.87, 0.31, 0.93 for R1, R2, V1, V2, E1, E2, E3, E4, E5, D1, respectively. The order in the coefficient of variation (CV) of the indicator for 18 plots was N (70.0%) > E3 (54.9%) > E1 (49.6%) > R2 (40.5%) > S (35.3%) > R1 (33.7%) > E2 (28.4%) > E5 (15.9%) > V1 (11.1%) > E4 (6.3%) > V2 (5.1%) > D1 (4.8%). The correlation matrix with 66 combinations between the indexes was constructed with statistical significance for 33 combinations. However, R1, V1, E2 and D1 were the proper indexes to represent species diversity of aquatic insect based on the correlation matrix and the theory of statistical independence. The richness index was highest in mountain, variety index in urban area, and evenness index in plain field. However, the dominance index was lowest in urban area.

A study on the soil conditioning behaviour according to mixing method in EPB shield TBM chamber (EPB 쉴드 TBM 챔버 내 혼합방법에 따른 배토상태거동에 대한 연구)

  • This paper is a study to improve the efficiency of mixing technology in the shield TBM chamber. Currently, the number of construction cases using the TBM method is increasing in Korea. According to the increasing use of TBM method, research on TBM method such as Disc Cutter, Cutter bit, and Segment also shows an increasing trend. However, there is little research on the mixing efficiency in chamber and chamber. In order to improve the smooth soil treatment and the behavior of the excavated soil, a study was conducted on the change of the mixing efficiency according to the effective mixing bar arrangement in the chamber. In the scale model experiment, the ground was composed using plastic materials of different colors for ease of identification. In addition, the mixing bar arrangement was different and classified into 4 cases, and the particle size distribution was classified into single particle size and multiple particle size, and the experiment was conducted with a total of 8 cases. The rotation speed of the cutter head of all cases was the same as 5 RPM, and the experiment time was also carried out in the same condition, 1 minute and 30 seconds. In order to check the mixing efficiency, samples at the upper, middle (left or right), and lower positions of each case were collected and analyzed. As a result of the scaled-down model experiment, the mixing efficiency of Case 4 and Case 4-1 increased compared to Case 1 and Case 1-1, which are actually used. Accordingly, it is expected that the mixing efficiency can be increased by changing the arrangement of the mixing bar in the chamber, and it is considered to be effective in saving air as the mixing efficiency increases. Therefore, this study is considered to be an important indicator for the use of shield TBM in Korea.

Characteristics of Pinus densiflora-Dominant Community on the Mountain Ridges of the Nakdong-Jeongmaek - Focusing on the Baekbyeongsan, Chilbosan, Baegamsan, Unjusan, Goheonsan - (낙동정맥 마루금 일대의 소나무우점군락 특성 - 백병산, 칠보산, 백암산, 운주산, 고헌산을 중심으로 -)

  • Since the position of Nakdong-Jeongmaek stretches from south to north, it shows clear changes of the climate compared to other mountain ranges. This study aimed to analyze the characteristics of vegetation in Pinus densiflora-dominant community (PDDC), which typically appeared on the ridge of this area, by each region and community. For this, the PDDCs in five representative areas of Nakdong-Jeongmaek (Baekbyeonsan, Chilbosan, Baegamsan, Unjusan and Goheonsan) were investigated. Communities were divided into four groups based on the characteristics of the growing environment for indicator species among the low vegetation, while the pinus densiflora was dominant on the canopy according to TWINSPAN. In community I, Fraxinus sieboldiana and Rhododendron schlippenbachii were dominant and there was a typical vegetation on the mountain ridge. In community II that was located on the upper slope, there was abundant inflow of sunlight to the inside of the forest which resulted from the disturbance, and it formed the secondary forest where deciduous broadleaf tree species such as Rhus cotinus appeared. Compared to the other communities, community III had a wider range of growing environment and various species of deciduous broadleaf trees appeared, so that changes in the vegetation structure were expected in the future. Finally, with the appearance of Salix hallaisanensis and Betula davurica, community IV are presumed to be located at a relatively high region with bountiful soil moisture. Reviewing by regions, PDDCs in Unjusan and Goheonsan were near big cities such as Pohang-si and Ulsan-si, so that the artificial disturbance resulted in high level of species diversity on a stage of vegetation development and the ages of Pinus densiflora were relatively young. On the other hand, PDDCs showed a stable vegetation status with low level of species diversity in other regions, and the ages of Pinus densiflora were relatively old.

Spatial and Temporal Variation of Macroinvertebrates according to Physical Factors in Gongji Stream Area (공지천 수계에서 물리적인 요인에 따른 저서성 대형무척추동물 군집의 시.공간적인 변동)

  • Our study is purposed to understand effect on spatio-temporal variability of macroinvertebrate community 11 sampling times at 4 sites between two streams (Shinchon stream and Gongji stream in Chuncheun City) from May 2011 to October 2013. In this study, the possible physical factors on spatio-temporal variability of macroinvertebrate community were discussed. After stream improvement project, the effects of anthropogenic disturbance in study sites appeared as increased water temperature more than $30^{\circ}C$ and the difference of water temperature between Shinchon stream and Gongji stream was by maximum $9^{\circ}C$ on 2012. The monsoon rainfall decreased number of species, individuals and biodiversity index of macroinvertebrate community, particularly, in Shincheon stream compare to Gongji stream. Dominant species of macroinvertebrate taxa was caddisfly in Shincheon stream and Diptera in Gongji stream but on August 2013, it was dominated by Diptera, Chironomidae spp. in all study sites. The spatio-temporal variability of macroinvertebrate community in the streams may be useful as bio-indicator influencing anthropogenic factors such as soil erosion (landslide or cultivation) or monsoon rainfall.
