A thennostable chitosanase was purified from Bacillus sp. KFB-C108, by fractionation of 30 to 70% saturation with ammonium sulfate, DEAE-Toyopearl chromatography, Butyl-Toyopearl chromatography, and TSK-Gel HW-55F gel filtration. The purified enzyme showed a single band on sodium dodecyl sulfate polyacrylamide gel electrophoresis, and the molecular weight was estimated to be 48 kDa. The enzyme degraded soluble chitosan and colloidal chitosan, but did not degrade glycol chitosan, chitin, and the other compounds investigated. There was no effect on the chitosanase activity by treatment with chelating agents, alkylating agents, and various metals investigated, and only cobalt ions inhibited the activity. Optimum temperature and pH were $55^{\circ}C$ and 6.5, respectively. The enzyme was stable after heat treatment at $80^{\circ}C$ for 10 min or $70^{\circ}C$ for 30 min and fairly stable in several organic solvents as well. Chitosan was hydrolyzed to $(GlcN)_4$as a major product by incubation with the enzyme.
The purpose of this study was to analyze myosin heavy chain (MHC) isoforms in bovine longissimus thoracis (LT) muscle by liquid chromatography (LC) and mass spectrometry (MS). LT muscles taken from Hanwoo (Korean native cattle) steer (n=3) used to separate myosin bands by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The peptide queries were obtained from the myosin bands by LC-MS/MS analysis following in-gel digestion with trypsin. A total of 33 and 43 queries were identified as common and unique peptides, respectively, of MHC isoforms (individual ions scores >43 indicate identity or extensive homology, p<0.05). MHC-1 (IIx), -2 (IIa), -4 (IIb), and -7 (slow/I) were identified based on the Mowse score (5118, 3951, 2526, and 2541 for MHC-1, -2, -4, and -7, respectively). However, more analysis is needed to confirm the expression of MHC-4 in bovine LT muscle because any query identified as a unique peptide of MHC-4 was not found. The queries that were identified as unique peptides could be used as peptide markers to confirm MHC-1 (14 queries), -2 (8 queries), and -7 (21 queries) in bovine LT muscle; no query identified as a unique peptide of MHC-4 was found. LC-MS/MS analysis is a useful approach to study MHC isoforms at the protein level.
Chelating agents to sand lance fish sauce for the prevention of precipitate formation were applied. The precipitates consisted of crude protein (74.4%), ash (18.7%), and other components (6.9%). Sand lance sauce was mainly composed of glutamic acid (3.69 mg/g), alanine (2.96 mg/g), and lysine (2.64 mg/g). However, there was an increase in the amount of hydrophobic amino acids, phenylalanine, isoleucine, and leucine, in the precipitates. Sodium ions were not detected in the precipitate; rather, the main elements were Mg ($1.98{\times}10^4\;{\mu}g/g)$, K ($1.36{\times}10^4\;{\mu}g/g)$, and Ca ($6.66{\times}10^2\;{\mu}g/g)$. In HPLC analysis, fish sauce was composed of 2 main peaks with molecular weights of 85.5 and 528.4 Da, respectively. However, the precipitate contained one peak with a molecular weight of 1,513.5 Da. The addition of 0.2% malic acid and citric acid caused 55 and 70% prevention of the precipitate, respectively. Citric acid was the most effective chelating agent and efficiently prevented precipitation in the fish sauce.
A novel superabsorbent composite was prepared by intercalation polymerization of acrylic acid (AA) and 2-acrylamido-2-methyl-1-propane sulfonic acid (AMPS) in the presence of montmorillonite (MMT), using ammonium persulfate (APS) as an initiator and $N,N'$-methylenebisacrylamide (MBA) as a cross linker. The superabsorbent composite was characterized by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), X-ray diffraction (XRD) and thermogravimetric analysis (TGA). Maximum absorbency of the composite in distilled water and 0.9% sodium chloride solution was 722 and 108 g/g, respectively. The composite was used for removal of heavy metal ions from aqueous solutions. Maximum amount of adsorption for $Ni^{2+}$, $Cu^{2+}$ and $Pb^{2+}$ was 211.0, 159.6 and 1646.0 mg/g, respectively, and the adsorption was in accordance with both Langmuir and Freundlich model. The composite could be regenerated and reused in wastewater treatment.
Biosynthesis of nanoparticles has acquired particular attention due to its economic feasibility, low toxicity and simplicity of the process. Extracellular synthesis of nanoparticles by bacteria and fungi has been stated to be brought about by enzymes and other reducing agents that may be secreted in the culture medium. The present study was carried out to determine the underlying mechanisms of extracellular silver nanoparticle synthesis by Pseudomonas hibiscicola isolated from the effluent of an electroplating industry in Mumbai. Synthesized nanoparticles were characterized by spectroscopy and electron microscopic techniques. Protein profiling studies were done using Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis (1D-SDS PAGE) and subjected to identification by Mass Spectrometry. Characterization studies revealed synthesis of 50 nm nanoparticles of well-defined morphology. Total protein content and SDS PAGE analysis revealed a reduction of total protein content in test (nanoparticles solution) samples when compared to controls (broth supernatant). 45.45% of the proteins involved in the process of nanoparticle synthesis were identified to be oxidoreductases and are thought to be involved in either reduction of metal ions or capping of synthesized nanoparticles.
Total impurity analysis of a primary standard solution is one of the essential procedures to determine an accurate concentration of the standard solution by the gravimetry. Bi impurity is determined in Pb standard solutions by inductively coupled plasma mass spectrometry (ICP-MS). The direct nebulization of the Pb standard solution produces a significant amount of the Pb matrix-induced molecular ions which give rise to a serious spectral interference to the Bi determination. In order to avoid the spectral interference from the interferent $^{208}PbH^+$, the hydride generation method is employed for the matrix separation. The Bi hydride vapor is generated by reaction of the sample solution with 1% sodium borohydride solution. The vapor is then directed by argon carrier gas into the ICP after separation from the mixture solution in a liquid-gas separator made of a polytetrafluoroethylene membrane tube. The presence of 1000 ${\mu}$g/mL Pb matrix caused reduction of the bismuthine generation efficiency by about 40%. The standard addition method is used to overcome the chemical interference from the Pb matrix. Optimum conditions are investigated for the hydride-generation ICPMS. The detection limit of this method is 0.5 pg/mL for the sample solutions containing 1000 ${\mu}$g/mL Pb matrix.
Electrokinetic technique was considered in removing arsenic from the abandoned mining tails. In order to estimate the removal characteristics of arsenic, the sequential extraction analysis and desorption experiment were carried out prior to the application of electrokientic process. The result of sequential extraction analysis indicated that the water soluble and exchangeable fraction, easily leachable to ground water, were very low as much as about 2.5% and the fraction except residual (38.3%), possibly extractable under very acidic or alkalic environment, was about 59%. In the result of desorption test using four different kinds of electrolytes, the mixture of citric acid and sodium dodecyl sulfate (SDS) showed the highest desorption efficiency as much as 77.3%. The removal efficiencies of arsenic from mining tailings by electrokinetic process under the different electrolyte environments were slightly low and resulted in the following order: citric acid + SDS (18.6%) > 0.1 $NHNO_3$ (8.1%) > HAc (7.4%) > Distilled water(6.6%). Also, arsenic in soil matrix was moved favorably in the direction of anodic rather than cathodic region, which is opposite trend with cationic metal ions generally existing in soil, because anionic form of arsenic is dominated in acidic soil caused by the movement of acid front form anode.
The adsorptive stripping voltammetric determination method of trace germanium (IV) using mercaptoacetic acid as a ligand was studied. Optimal conditions were found to be 0.25 M NaCl solution (pH 6.0) containing mercaptoacetic acid concentration of $5.0{\times}10^{-6}M$. The peak potential appeared at - 1.402 V vs. Ag/AgCl. Effects of sodium chloride concentration, mercaptoacetic acid concentration, and accumulation time for the complex of Ge(IV)-Mercaptoacetic acid on the peak current were studied. Amberlite IRC-718 chelating resin was applied to the separation of Ge(IV) from other metal ions.

This study was undertaken to apply the yin-yang theory in action potential. In order to apply the yin-yang theory in action potential, nature of yin and yang, interrelation of yin and yang and action potential in cell were reviewed. According to the yin-yang theory, inner cellular space corresponds to yin, but outer cellular space corresponds to yang. If we classify ions in intracellular fluid or extracellular fluid by nature of yin and yang, potassium(K+) corresponds to yang within yin(陰中之陽), protein(Pr-) corresponds to yin within yin(陰中之陰) in intracellular fluid, and sodium(Na+) corresponds to yang within yang(陽中之陽), chloride(Cl-) corresponds to yin within yang(陽中之陰) in extracellular fluid. Double donnan equilibrium and equilibrium potential were caused by intracellular anion(Pr-) and extracellular cation(Na+) are related with mutual rooting of yin and yang(陰陽互根) and opposition of yin and yang(陰陽對立). The influx and efflux of ion through cell membrane means waxing and waning of yin and yang(陰陽消長), the change of membrane potential means yin-yang conversion(陰陽轉化) during action potential.
A halophilic bacterium was isolated from fermented seafood. The 16S rDNA sequence identity between the isolate and Halomonas subglaciescola AJ306801 was above 95%. The isolate that did not grow in the condition without NaCl or in the condition with other sodium (Na$\^$+/) or chloride ions (Cl$\^$-/) instead of NaCl was named H. subglaciescola DH-l. Two mutants capable of growing without NaCl were obtained by random mutagenesis, of which their total soluble protein profiles were compared with those of the wild type by two-dimensional electrophoresis. The external compatible solutes (betaine and choline) and cell extract of the wild type did not function as osmoprotectants, and these parameters within the mutants did not enhance their growth in the saline environment. In the proton translocation test, rapid acidification of the reactant was not detected for the wild type, but it was detected for the mutant in the condition without NaCl. From these results, we derived the hypothesis that NaCl may be absolutely required for the energy metabolism of H. subglaciescola DH-l but not for its osmoregulation, and the mutants may have another modified proton translocation system that is independent of NaCl, except for those mutants with an NaCl-dependent system.
