The neurotoxin of Clostridium botulinum type B was purified from a liquid culture. The purification steps consist of ammonium sulfate precipitation of whole culture, treatment of Polymin P(0.15%, v/v), gel filtration on Sephadex G-100 at pH5.6 and DEAE-Sephadex charomatography at pH8.0. The procedure recovered 17% of the toxin assayed in the starting culture. The toxin was homogeneous by sodium dodecyl sulfate(SDS)-polyacrylamide gel electrophoresis and had a molecular weight of 163,000. Subunits of 106,000 and 56,000 molecular weight were found when purified toxin was treated with a disulfide-reducing agent and electro phoresed on SDS-polyacrylamide gels.
$\omega$-Phenylalkylamine salt의 존재하에서 sodium dodecyl sulfate (SDS)의 임계 미셀농도(CMC)를 전기전도도법으로 구하였다. 유기염의 alky기의 길이가 길어짐에 따라 CMC는 감소하는 경향을 보여주었다. 유기첨가제의 존재하에서 $18^{\circ}C-50^{\circ}C$의 온도 범위에서 SDS의 미셀화 과정에 수반되는 열역학적 파라메터를 구하였다. 이 온도 범위에서 SDS의 미셀화 자유에너지 (${\Delta}G_m^{\circ}$)는 음의 값을 나타내며 미셀화 엔트로피(${\Delta}S_m^{\circ}$)는 큰 양의 값을 나타내었으며 미셀화 엔탈피(${\Delta}H_m^{\circ}$)는 낮은 온도($18^{\circ}C$)에서는 양의 값을 나타내지만 높은 온도($>25^{\circ}C$)에서는 음의 값을 나타내었다. 미셀화는 자발적인 상전이 현상임을 알 수 있었다. 이들 열역학적 파라메터들은 유기염의 소수성 부분의 길이가 길어짐에 따라 감소하는 경향을 보여주었다. 비전도도 값의 차(${\Delta}\kappa$)가 유기염의 mol 비가 증가할수록 큰 값을 나타내었다. 그러므로 유기염의 소수성 부분의 길이가 길어질수록 유기염들은 더 쉽게 SDS 미셀내의 palisade층까지 깊이 침투하여 안정한 혼합 미셀을 형성함을 알 수 있었다.
SDS-PAGE를 이용한 일반적인 단백질 분리는 단백질을 분자량에 따라 분리하는 방법이며 가장 보편적이고, 간단한 방법 중의 하나이다. 본 연구는 SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis)를 두 번에 걸쳐 동일한 분리 원리로 이차원으로 전개하는 방법의 전기영동을 시도하여 기존의 일차원 전기영동 분석법과 비교하였으며, 그 결과 향상된 분리능이 본 연구의 이차원 전기영동법에서 확인되었다. Gel에서 분리된 단백질들은 MALDI TOF MS를 이용하여 동정하여 서로 다른 단백질임이 확인되었으며, 이러한 duplex SDS-PAGE 분리법은 상대적으로 적은 비용으로 단백질 분리능을 용이하게 향상시킬 수 있는 경제적인 분석법으로 이용될 수 있을 것이다.
The leukocyte NADPH oxidase of neutrophils is a membrane-bound enzyme that catalyzes the production of $O_2^-$ from oxygen using NADPH as an electron donor. Dormant in resting neutrophils, the enzyme acquires catalytic activity when the cells are exposed to appropriate stimuli. During activation, the cytosolic oxidase components $p47^{phox}$ and $p67^{phox}$ migrate to the plasma membrane, where they associate with cytochrome $b_{558}$, a membrane-bound flavohemoprotein, to assemble the active oxidase. The oxidase can be activated in a cell-free system; the activating agent usually employed is an anionic amphiphile such as sodium dodecyl sulfate (SDS). Because $p47^{phox}$ can translocate by itself during activation, the conformational change in $p47^{phox}$ may be responsible for the activation of NADPH oxidase. We show here that the treatment of $p47^{phox}$ with SDS leads to an increase in the reactivity of the sutbydryl group of cysteines toward N-ethylmaleimide, indicating that the conformational change occurs when $p47^{phox}$ is exposed to SDS. We propose that this change in conformation results in the appearance of a binding site through which $p47^{phox}$ interacts with cytochrome $b_{558}$during the activation process.
Solubilization isotherms of 1-chlorobenzene (MCB) and 1, 2-dichlorobenzene (DCB) were investigated in ionic surfactant solutions such as sodium dodecyl sulfate (SDS), cetylpyridinium chloride (CPC), and dedecyltrimethylammonium chloride (DMAC). The solubilization extent of DCB was much higher than that of MCB because of the main driving force of solubilization Is hydrophobic interactions between chlorobenzenes and hydrophobic interior of ionic micelles and DCB is more hydrophobic than MCB. CPC showed highest solubilization capacity because of longest hydrophobic tails. Simultaneous solubilization of MCB and DCB decreased slightly the extent solubilization of both MCB and DCB because the solubilization locus in the micelles is same.
A fast and sensitive protein staining method in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) using both an acidic dye, Coomassie Brilliant Blue R-250 (CBBR) and a basic dye, Neutral Red (NR) is described. It is based on a counter ion-dye staining technique that employs oppositely charged two dyes to form an ion-pair complex. The selective binding of the free dye molecules to proteins in an acidic solution enhances the staining effect of CBBR on protein bands, and also reduces gel background. It is a rapid staining procedure, involving fixing and staining steps with short destaining that are completed in about 1 h. As the result, it showed two to fourfold increase in sensitivity comparing with CBBR staining. The stained protein bands can be visualized at the same time of staining.
Clostidium botulinum type B가 생성하는 독소를 정제할 수 있는 방법을 연구하였다. 정제과정은 독소를 ammonium sulfate로 배양액에서 침전시켜 추출한후 Polymin P를 처리하여 핵산 및 기타 단백질을 최대한 제거한 후 Sephaex G-I00에서 gel fiItration을 시키고 DEAE-Sephadex로 이온교환 크로마토그래피를 시켰다. 이러한 과정으로 정제된 독소의 회수율은 17%였으며 SDS-polyacrylamide gel electrophoresis 결과 하나의 선을 나타내 동질성을 증명하였다. 정제된 독소의 분자량은 163,000이였으며 $\beta$-mercaptoethanol을 사용하여 환원시킨 결과 분자량 106,000과 56,000의 하위 단위체로 분리되었다.
SDS(sodium dodecyl sulfate) 존재하에서 NP-40EO[nonylphenol-(ethylene oxide)40]와 요오드와의 상호작용에 따른 $Ca^{2+}$의 영향을 UV-visible spectrophotometer를 이용하여 수용액 중에서 조사하였다. SDS 부재시보다 상호작용 피크의 흡광도는 줄어들었으며, CMC 이상에서 $Ca^{2+}$ 첨가에 따른 상호작용 피크의 강도는 크게 증가하였다. 상호작용 피크의 감소는 SDS 삽입에 따른 혼합미셀 단위 표면적당 요오드와 상호작용할 수 있는 EO 수의 감소에 기인하며, 상호작용 피크 강도의 증가는 $Ca^{2+}$ 존재하에서 미셀 구조가 더욱 조밀해진데 따른 전자주게-받게 겹침 증가로 불 수 있다. 이러한 현상들은 수용액 중에서 상대적으로 자유로운 배향을 갖는 선형 EO 사슬이 유사 크라운 에테르 구조를 형성하여 $Ca^{2+}$ 이온과 착물을 형성할 수 있는 가능성을 보여준다.
분자량이 다른 두 가지 폴리(4-비닐피리딘) (Mw=50 kg/mol 및 200 kg/mol)을 알킬기의 탄소수(m)를 변화시키면서 N-알킬화시켜 이온성 고분자를 합성하였다. 알킬화제로서 디메틸 설페이트(m=1) 및 브롬화 알칸(m=5, 8, 12, 16 및 22)을 사용하였다. 합성한 이온성 고분자의 조성은 NMR 분광분석법 및 원소분석법을 사용하여 결정하였다. 그 결과로써 거의 완전한 4차 알킬화 반응에 의해 전해질 고분자가 얻어졌음을 알 수 있었다. 합성한 전해질고분자의 수용액에 도데실 황산 소듐(SDS)을 첨가 시 발생되는 탁도 변화를 조사하여 임계응집농도(CAC)를 결정하였으며, 이러한 CAC가 고분자의 분자량, N-알킬기의 길이 및 NaCl의 농도 변화에 어떻게 의존하는가를 조사하였다. 결과로써 폴리(4-비닐피리딘)의 분자량이 클수록 또한 알킬 곁사슬의 길이가 길수록 더 적은 양의 SDS 첨가로도 응집체가 형성되었음을 알 수 있었다.
