Performance Evaluation of Hydrogen Generator for Fuel Cell Unmanned Aircraft (연료전지 무인기 탑재용 수소발생기의 성능평가)

  • Performance of a hydrogen generator for a fuel cell unmanned aircraft was evaluated as the change of temperature environment. Sodium borohydride ($NaBH_4$) was used as a hydrogen source due to its high hydrogen content and good storability. The hydrogen gas was generated by the hydrolysis reaction using a catalytic reactor. Reaction chambers were set up with the range of temperatures from -20 to $60^{\circ}C$. The hydrogen generation rate and temperatures changes of reactor and separator were measured at the $NaBH_4$ concentrations of 20 and 25wt.%. As a result, the hydrogen generation rate was decreased as the repeated reaction cycles. It showed that the hydrogen generation rate was stable at low temperature, while at high temperature the hydrogen generation rate was rapidly decreased. The performance degradation was mainly caused by the catalyst loss and $NaBO_2$ deposition on the catalyst surface.

  • 6-[1-(4-Ribitylanilino)ethyl]-1,3,7-trimethyllumazine (2), which is related to pteridine moiety of methanopterin, was synthesized. 4-Ribitylaniline derivative was prepared from D-ribose and N-benzoyl-4-bromoaniline (7) as the starting materials through several steps. 6-Acetyl-1,3,7-trimethyllumazine (4) was obtained from the reaction of 4-amino-1,3-dimethyI-5-nitrosouracil (3) with 2,4-pentanedione by Timmis reaction. Compound 4 was converted to 6-(1-chloroethyl)-1,3,7-trimethyllumazin (6) by the reduction with sodium borohydride and followed by chlorination with thionyl chloride. The nucleophilic displacement reaction of compound 6 with 4-(2,3:4,5-di-O-iso-propylidene-D-ribityl)aniline (13) in anhydrous DMF yielded 6-[1-{4-(2,3:4,5-di-O-isopropylidene-D-ribityl)anilino}ethyl]-1,3,7-trimethyllumazine (14). The target molecule 2 was obtained by the hydrolysis of compound 14 in the presence of an acid catalyst.

  • Time-dependent inactivation of E. coli GTP cyclohydrolase I with a 2',3'-dialdehyde derivative of GTP (oGTP) was directed to the active site of the enzyme, and was dependent on the concentration of oGTP. The kinetics of inactivation were biphasic with a rapid reaction occurring immediately upon exposure of the enzyme to oGTP followed by a slow rate of inactivation. The $K_i$ value of oGTP for the enzyme was 0.25 mM. Inactivation was prevented by preincubation of the enzyme with GTP, the substrate of the enzyme. At 100% inactivation, 2.3 mol of [8.5'-$^3H$]oGTP were bound per each enzyme subunit, which consists of two identical polypeptides. The active site residue which reacted with the affinity label was lysine. oGTP interacted selectively with the ${\varepsilon}$-amino group of lysine in the GTP-binding site to form a morpholine-like structure which was stable without sodium borohydride treatment. However, triphosphate group was eliminated during the hydrolysis step. To identify the active site of the enzyme, [8.5'-$^3H$]oGTP-labeled enzyme was cleaved by endoproteinase Lys-C, and the $^3H$-labeled peptide was purified by HPLC. The amino acid sequence of the active site peptide was Pro-Ser-Leu-Ser-Lys, which corresponds to the aminoterminal sequence of the enzyme.

  • Hepatotoxic cyanobacteria, Microcystis species, were collected from the Nakdong River and we could isolate hepatotoxins, microcystin-LR and microcystin-RR, which are also strong inhibitors of protein phosphatase 1 and 2A. From the microcystins, several microcystin derivatives were synthesized and tested on the mouse toxicity in order to establish the structure-activity relationship. Esterification od carboxyl groups of Glu and MeAsp residue produced nontoxic compounds. However, when we reduced the Mdha residue with sodium borohydride into Ala residue, toxicity was still maintained. Also, the change of guanidyl moiety of Arg residue in microcystin-LR into dimethylpyrimidyl moiety did not change the toxicity of microcystins as well. Thus the carboxyl groups seem to play important roles in binding with protein phosphatase 1 and 2A, whereas Mdha residue and the guanidyl moiety of Arg residue do not.

  • New procedures for the reduction of carboxylic acids, carboxylic acid salts, tertiary amides, and sulfoxides with sodium borohydride and triphenyl borate in tetrahydrofuran were developed. Thus carboxylic acids were reduced quantitatively in 6∼12 h at $25^{\circ}C$. Alphatic acid salts were quantitatively reduced to the corresponding alcohols in 6h at $25^{\circ}C$ whereas aromatic acid salts required 24h at $65^{\circ}C$. Tertiary amides were reduced to the corresponding amines in 88∼100% yields in 3∼6h at room temperature. Most sulfoxides examined were reduced to the corresponding sulfides in 98∼100% yields in 1∼6h at $25^{\circ}C$ and completely reduced at $65^{\circ}C$; however, diphenyl sulfoxide required 48h in a more vigorous condition.

Studies on Preparation of Dysprosium-165 Metallic Macroaggregates for the Treatment of Rheumatoid Arthritis (류마티스 관절염 치료용 디스프로슘-165금속 응집입자($^{165}Dy-MA$)의 제조에 관한 연구)

  • Irradiation of 20mg of natural $Dy(NO_3)_3$ in a neutron flux of $2{\times}10^{13}n/cm^2$ sec for 4 hours gave 5.76 Ci of $^{165}Dy$(specific activity, 610mCi/mg Dy) with high radionuclidic purity (>99.9 %). $^{165}Dy-MA$ was prepared in a quantitative yield by reacting the aqueous solution of $^{165}Dy(NO_3)_3$ with sodium borohydride solution in 0.2N NaOH. Coulter particle analyzer exhibited mean particle size of $2.6{\mu}m$ (range $1{\sim}6{\mu}m$), Even though the $^{165}Dy-MA$ suspension in saline was stored at $37^{\circ}C$ for 24 hours or autoclaved at $121^{\circ}C$ for 30minutes, there was no significant change in particle size and leakage problem indicating the prepared $^{165}Dy-MA$ is sufficiently stable. In-vivo retention studies were carried out by administering $^{165}Dy-MA$ into the knee joint space of normal rabbits. Gamma camera analysis showed high retention in joint space of normal rabbits. Gamma camera analysis showed high retention in joint space even at 24 hours after administration (> 99.9%). The ease with which the $^{165}Dy-MA$ can be made in the narrow size range and their high invitro and vivo stability make them attractive agents for radiation synovectomy.

  • This research was carried out to investigate the effects of Maillard reaction products and nondialyzable melanoidins on the nitrite-scavenging. Nitrite-scavenging reactions were done at the different pH conditions(pH 1.2, 4.2 and 6.0). Maillard reaction products and nondialyzable melanoidins, produced from the glucose-amino acids(lys., gly., arg., his.)model systems, had a great of nitrite-scavenging effects. Nitrite-scavenging effects of Maillard reaction products and nondialyzable melanoidins were also pH dependent, being higher at pH 1.2 and lower at pH 6.0. By the treatment of Maillard reaction products and nondialyzable melanoidins with sodium borohydride, nitrite-scavenging effects were remarkably decreased at pH 1.2.

  • A method is described for the determination of organotins in water samples by GC/MS. Optimized derivatization methods for ethylation and hydrogenation of organotins were surveyed according to various reaction conditions such as time, pH and concentration of reagents. The organotins were extracted with n-hexane in presence of 0.1% tropolone and hydrogenated with sodium borohydride. Extraction recoveries of organotins with hydrogenation were in the range of 61-112%. After ethylation, organotins in water samples were extracted by liquid-liquid extraction (LLE) and solid-phase extraction (SPE). Using LLE, extraction recoveries were in the range of 74-113%. The recoveries ranged from 61-97% in the case of SPE with styrene-divinylbenzene copolymers. Method detection limits of hydrogenated and ethylated organotins ranged from 0.05 to 0.5 ng/ml and from 0.02 to 0.05 ng/ml, respectively.

  • This paper describes the characteristics of hydrogen generation from solid-state $NaBH_4$ and fuel cell operation for fuel cell aircraft. The solid-state $NaBH_4$ was used for a high hydrogen storage density, and was reacted with hydrochloric acid to generate hydrogen. The hydrogen generation rate for the solid-state $NaBH_4$ reaction was measured at various conditions. As a result, the hydrogen generation rate was increased with the feed rate and concentration of hydrochloric acid, while not be affected by the reaction temperature. A fuel cell was connected with the solid-state $NaBH_4$ hydrogen generator. The stable power output was obtained at the gradual and sudden increases of electric loads.

Determination of Selenium in Milk by ICP-OES (ICP-OES를 이용한 우유의 Selenium 분석)

  • The purpose of this paper was to determine the quantity of selenium in milk by inductively coupled plasma optical emission spectrometry. The sample was digested in teflon vessel containing nitric-hydrogen peroxide acid mixture. After digestion, the sample is treated with additional hydrochloric acid. Total selenium was reduced with sodium borohydride and concentrated hydrochloric acid in a simplified hydride generation(HG) manifold. The optimum conditions of HG are 2 M for HCl, 1.5% for NaBH$_4$, 1.2 mL/mim for sample flow. Recovery rates by the standard addition method were 88.0% at 10 ppb and 92.2% at 10 ppm. The relative standard deviations were 4.8 and 3.2%, respectively. This method showed a good accuracy and precision. And so it was highly suitable for determination of small quantity of selenium in milk.
