퍼플로오로알킬에틸아크릴레이트/아크릴레이트/그리시딜메타크릴레이트 공중합체의 유화중합 및 그들의 표면특성 (Emulsion Polymerization and Surface Properties of Perfluoroalkylethyl Acrylate/Acrylate/Glycidyl Methacrylate Copolymers)

  • 플로오로 함유 아크릴 공중합체를 여러 가지 조성의 아크릴 단량체[AA : 에틸 아크릴레이트(EA), 부틸아크릴레이트(BA) 혹은 메틸메타크릴레이트(MMA)], 퍼플로오로알킬에틸 아크릴레이트(PFA) 및 그리시딜메타크릴레이트(GMA)로부터 유화중합법(유화제 : 소디움 도데실 설페이트/노닐페놀에톡실레이트)으로 합성하였다. 아크릴 단량체 중에서 BA를 사용한 경우 가장 낮은 표면에너지를 가진 아크릴 공중합체를 얻을 수 있었으며, 최적량의 단량체 조성(BA: 87 wt%, GMA : 8.7 wt% 및 PFA : 4.3 wt%)에서 얻은 공중합체의 표면에너지/물에 대한 접촉각/methylene iodide에 대한 접촉각은 19.89 mN/m /$103.5^{\circ}$/$78.7^{\circ}$로서 높은 표면활성을 지닌 것을 알 수 있었다. 따라서 본 연구에서 얻은 최적의 아크릴 공중합체는 높은 발수 및 발유 표면 특성을 지닌 코팅소재로 활용할 수 있을 것이다.

열다한소탕(熱多寒少湯)이 저산소성(低酸素性) 대뇌신경세포(大腦神經細胞) 손상에 미치는 영향(影響) (Influence of Yeoldahanso-tang on the Hypoxic Damage of Cultured Cerebral Neurons from mouse and SK-N-MC cells)

  • To elucidate the neuroprotective effect of Yeoldahanso-tang(YHT) on nerve cells damaged by hypoxia, the cytotoxic effects of exposure to hypoxia were determined by XTT(SODIUM3,3'-{I-[(PHENYLAMINO) CARBONYL]-3,4-TETRAZOLIUM}- BIS (4-METHOXY-6-NITRO) BENZENE SULFONIC ACID HYDRATE), NR(Neutral red), MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) and SRB(Sulforhodamin B) asssay. The activity of catalase and SOD(Superoxide dismutase) was measured by spectrophometry, and $TNF-{\alpha}$(Tumor cell necrosis $fector-{\alpha}$) and PKC(Protein kinase C) activity was measured after exposure to hypoxia and treatment of YHTWE. Also the neuroprotective effect of YHTWE was researched for the elucidatioion of neuroprotective mechanism. The results were as follows; 1. Hypoxia decreased cell viability measured by XTT, NR assay when cultured cerebral neurons were exposed to 95% N2/5% CO2 for $2{\sim}26$ minutes in these cultures and YHTWE inhibited the decrease of cell viability. 2. H2O2 treatment decreased cell viability measured by MTT, and SRB assay when cultured cerebral neurons were exposed to 1-80 ${\mu}M$ for 6 hours, but YHTWE inhibited the decrease of cell viability. 3. Hypoxia decreased catalase and SOD activity, and also $TNF-{\alpha}$ and PKC activity in these cultured cerebral neurons, but YHTWE inhibited the decrease of the catalase and SOD activity in these cultures. 4. Hypoxia triggered the apoptosis via caspase activation and internucleosomal DNA fragmentation. Also hypoxia stimulate the release of cytochrome c forom mitochondria. YHTWE inhibited the apoptosis via caspase activation induced by hypoxia. From these results, it can be suggested that brain ischemia model induced hypoxia showed neurotoxicity on cultured mouse cerebral neurons, and the YHTWE has the neuroprotective effect in blocking the neurotoxicity induced by hypoxia in cultured mouse cerebral neurons.

여포성숙의 대사에 관한 생화학적인 연구 II. 돼지 난소내 중여포액에 존재하는 단백질의 조성 및 동일여포액내 스테로이드 호르몬의 농도 (Biochemical studies on the metabolism of follicular maturation II. Protein composition and stenoid concentration in individually isolated medium-sized follicular fluid of pig ovary)

  • 포유류의 난소내 여포의 폐쇄기작을 알아보기 위하여 본 실험을 행하였다. 정상과 폐쇄를 형태적으로 판정한 돼지의 난소내 개개 중여포의 여표액내 스테로이드 호르몬과 단백질을 방사면역측정법과 SDS-전기영동으로 각각 분석하여 다음과 같은 결과를 얻었다. Progesterone은 여포성숙기의 여포액내 폐쇄여포에서 높은 농도를 나타내었고testosterone의 경우 황체기의 정상여포 및 여포성숙기의 폐쇄여포에서 높은 농도를 나타내었다. estradiol은 여포성숙기의 폐쇄여포에서 정상여포에 비해 현저히 낮았다. 여포액내 존재하는 단백질의 SDS-PAGE 결과, 혈청에 존재하지 않는 4종의 단백질을 확인하였다. 난소주기에 따라 112K, 141K의 단백질이 확인되었고, 폐쇄여포액내 특이하게 존재하는 23K, 24K의 단백질도 확인할 수 있었다. 이상의 결과로 보아 난소주기에 따라 여포액재에 존재하는 스테로이드 호르몬의 농도와 단백질의 조성이 서로 같지 않음을 알 수 있었으며 여포의 폐쇄기작에 있어서도 난소주기에 따라 분명한 차이가 있음을 알 수 있었다. 그러나 이들 폐쇄여포에 존재하는 단백질들의 생리적인 역할을 이해하기 위하여는 많은 연구가 수행되어야 한다고 사료된다.

  • 균일한 마그나타이트 나노 입자를 합성하기 위해 음향화학법을 이용한 공침 기술을 적용해 합성 하였다. 입자의 크기를 제어하고 응집을 막기 위해 계면활성제로 올레인산을 사용하였다. 이 방법으로 합성한 마그네타이트 나노입자를 의학적으로 이용하기 위해 나노 입자의 표면을 개질하고 자성 미소구체를 제조 하였다. 마그네타이트 입자의 표면 개질과 자성 미소 구체를 제조하기 위해 저독성, 생분해성, 생체 친화성의 특징을 갖고 있는 키토산과 $\beta$-글루칸을 사용하였다. 생체 고분자로 마그네타이트 나노 입자를 피복하기 위해 묽은 초산 용액을 이용하여 키토산을 용해하고 $\beta$-글루칸은 묽은 수산화나트륨에 용해하여 사용 하였다.나노 입자의 피복은 자성유체를 초음파를 가해주면서 용해시킨 각각의 생체 고분자 용액을 서서히 가하여 나노 입자를 피복하였다. 자성 미소구체는 2 % 키토산과 5 %의 $\beta$-글루칸 수용액을 이용해 제조 하였다. 마그네타이트를 생체 고분자에 분산 시켜 주기 위해 콜로이드 상으로 제조 하고 초음파를 조사 해주어 분무 장치를 통해 미소 구체를 제조하였다. SQUID(SuperconductingQuantum Interference Device)를 측정한 결과 피복한 나노 입자와 미소구체에서 모두 초상자성을 나타내고 있었으며 MRI(Magnetic Resonance Imaging) 측정 결과 MRI 조영제와 색전 물질로서 사용하기 적합 하다는 것을 확인 할 수 있었다.

루틴 첨가가 고지방 식이 랫드의 혈청 지질 농도의 변화에 미치는 영향 (Effect of Rutin on Major Lipid-Related Parameters in Sera of Rats Fed High Fat Diet)

  • This study was performed to investigate the changes of the serological lipid-related parameters of the rats when they were fed with the high fat diets supplemented with or without rutin for five weeks. Twenty-four Sprague-Dawley male rats ($272.2{\pm}7.2 g$ of body weight) were randomly divided into three groups: control (C) group and two treatment groups. Rats in the C group were fed with the high-fat diet containing 20% lard, 1% cholesterol and 0.5% sodium cholate (w/w) which was modified from the formula of the American Institute of Nutrition-76 (AIN-76) diet. Rats in treatment groups were fed with above diet supplemented with 0.75% rutin (R-0.75) or 1.5% rutin (R-1.5) on the weight to weight basis, respectively. The supplementation of rutin did not induce any significant difference on the final body weight, gain of body weight, the amount of feed intake and the feed efficiency of rats in both control and treatment groups. In addition the values of glucose concentration, total protein, albumin, globulin and albumin/globulin (A/G) ratio showed no significant differences among groups. The values of total cholesterol (TC) and low density lipoprotein-cholesterol (LDL-C) in sera of rats in both R-0.75 and R-1.5 groups were lower than those in C group but the significances were showed in only between R-0.75 and C group (p < 0.05 and p < 0.01, respectively). The values of high density lipoprotein-cholesterol (HDL-C) in sera of rats in both R-0.75 and R-1.5 groups were higher than those in C group but the significances were showed in only between R-1.5 and C group (p < 0.01). The values of atherogenic index(AI) of rats in both R-0.75 and R-1.5 groups were the lower than those in C group (p < 0.01 and p < 0.05, respectively). The values of triglyceride in sera of rats showed no significant differences among groups. The values of AST and ALT in sera of rats showed no significant differences among groups. Therefore the supplementation of rutin to high fat diet in rats reduced effectively the serum lipid levels such as TC and LDL-C which were regarded as to cause the cardiovascular diseases, and moreover it elevated effectively HDL-C value which was regarded to protect cardiovascular diseases.

나린진이 고지방 및 고콜레스테롤 식이 랫드의 혈청 성분 농도의 변화에 미치는 영향 (Effect of Naringin on Major Biochemical Parameters in Sera of Rats Fed High Fat and Cholesterol Diet)

골관절염 랫드 모델에서 계피의 연골보호 효과 (Chondroprotective Effects of Cinnamomum cassia Blume in a Rat Model of Osteoarthritis)

  • 본 연구는 내측반월판 절제와 전십자인대 단열로 유발된 랫드 골관절염 모델을 이용하여 계피 (Cinnamomum cassia Blume, 육계(肉桂)) 추출물의 연골손상 방지에 대한 효과를 평가하였다. 골관절염유발 랫드 48마리를 군당 8마리씩 6군으로 음성 대조군과 골관절염 대조군, 체중당 diclofenac 2 mg 투여군, 계피 추출물 25 mg과 50 mg, 100 mg 투여군으로 각각 분류하여 수술 1주 후부터 12주동안 투여하였다. 연골 소실과 관절의 불안정성은 계피 추출물과 diclofenac 투여군이 골관절염 대조군과 비교할 때 유의하게 감소하였다(p < 0.05). 병리조직학적 평가에서 연골의 퇴행은 계피 추출물 투여량에 따라 용량 의존적으로 개선됨이 확인되었다(p < 0.01). 계피 추출물 투여군에서 관절구조 퇴행성 변화의 감소와 Safranin-O 염색 정도의 용량 의존적인 증가를 보여 연골 퇴행이 억제됨을 확인하였다. Diclofenac이 골관절염 진행에 있어 활성화된 caspase-3와 절단된 poly(ADP-ribose) 중합효소에 유도된 세포자멸사 표지율 증가에 별 다른 영향을 주지 않았지만 계피 추출물 투여군에서는 이들 세포자멸 표지자에 대한 반응세포는 유의하게 감소되었다(p < 0.05). 그러나, diclofenac과 계피 추출물 투여군은 5-bromo-2-deoxyuridine의 섭취에는 영향을 주지 않았다. 이러한 결과에서 계피추출물이 항염활성과 항세포 자멸 활성을 통해 관절연골에 보호 효과가 있음을 보였다.

천연기념물 한국 산양의 혈액 및 혈액 화학적 정상표준범위 (Hematologic and biochemical reference intervals for natural monument Korean goral (Naemorhedus caudatus))

  • Korean goral (Neamorhedus caudatus) is registered as a natural monument number 217 by South Korea Cultural Heritage Administration. It is also recognized as the endangered species I by Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES). In this study, blood samples of Korean goral were collected to make reference intervals. Blood sampling was conducted on 19 numbers of Korean gorals (ten females, nine males), which were reared in Korean Goral Restoration Center. For total samples, the reference intervals of hematological parameters were: white blood cell $7.69{\sim}10.87K/{\mu}{\Lambda}$; hematocrit 36.73~46.18%; red blood cell $10.72{\sim}12.86K/{\mu}{\Lambda}$; hemoglobin 12.79~15.14 g/dL; mean corpuscular volume 33.15~36.75 fL; mean corpuscular hemoglobin 11.53~12.23 pg; mean corpuscular hemoglobin concentration 32.64-35.91 g/dL and red blood cell distribution width 39.2~43.46%. For total samples of each parameters, the following results were obtained for serum biochemistry: glucose 111.81~153.77 mg/dL; blood urea nitrogen 22.35~28.91 mg/dL; creatine 1.22~1.84 mg/dL; phosphate 4.57~6 mg/dL; calcium 8.7~9.1 mg/dL; total protein 6.53~6.92 g/dL; albumin 3.1~3.48 g/dL; globulin 3.26~3.62 g/dL; alanine aminotransferase 56.7~158.56 U/L; aspartate aminotransferase 230.35~473.06 U/L; alkaline phosphatase 178.06~332.47 U/L; gamma-glutamyl transpeptidase 131.6-~181.24 U/L; total bilirubin 1.47~2.12 mg/dL; cholesterol 46.48~71.52 mg/dL; amylase 16.3~26.03 U/L; sodium 150.43~153.88 mmol/L; potassium 3.98~4.6 mmol/L and chlorine 109.48~113.26 mmol/L. The ranges of values were similar campared to previous studies except in the case of RDW value, which showed higher range than the RDW value of a previous study. The reference intervals from this study will be useful data for treatment and management of gorals.

  • This study was performed to evaluate anesthetic and cardiovascular effects of xylazine/fentanyl/azaperone and medetomidine/midazolam as preanesthetics and their combinations with antagonists in halothane-anesthetized dogs. Eight clinically healthy dogs($4.54{\pm}2.16kg$) were used at the interval of more than 14 days between experiments in turn for propionyl promazine(PP 0.3mg/kg, IM), xylazine/fentanyl/azaperone(XFA 2mg/kg, 0.0137mg/kg, 0.11mg/kg, IM), medetomidine/midazolam(MM 0.02mg/kg, 0.3mg/kg, IM), combination of XFA and their antagonists (yohimbine 0.05mg/kg, naloxon 0.0005mg/kg, IV) and combination of MM and their antagonist(atipamezole 0.08mg/kg IM). The sedation induction times in XFA($2.56{\pm}1.01min$) and MM($5.44{\pm}2.07min$) groups were significantly better than that of PP group($10.75{\pm}2.38min$)(p < 0.05). The thiopental sodium dose required for tracheal intubation in XFA($2.38{\pm}3.38mg/kg$) and MM($3.91{\pm}3.47mg/kg$) groups were significantly less than that of PP group($12.57{\pm}2.13mg/kg$)(p < 0.05). All time indices expressing the recovery(pedal reflex recurrence time, extubation time, arousal time, standing time and walking time) were significantly shorter in the combination groups of XFA or MM with their antagonists than in PP, XFA and MM groups(p < 0.05). The suppressions of cardiovascular function of XFA and MM were more than that of PP. Heart rate and cardiac output were recovered by the antagonists of XFA and MM, but mean arterial pressure were not recovered by the antagonists. PP induced apnea in 4 out of 8 dogs, but XFA in none and MM in one. The present study suggested that for rapid sedation, prevention of apnea after intubation and rapid recovery after halothane cessation, combinations of xylazine/fentanyl/azaperone or medetomidine/midazolam with their antagonists are recommendable as preanesthetic method in gas anesthetised dogs with normal cardiovascular function.

국내 유통 중인 먹는샘물 원수의 이온류 수질 특성에 관한 연구 (The Study on the Ion Water Characteristics of Raw Water in the Domestic Natural Mineral Water)

  • The goal of this study is to provide basic data to establish a foundation for the provision of safe drinkable water. The raw water of natural mineral water was analyzed to determine the quantities of anions (F-, Cl-, NO3-N-, and SO42- ) and cations (Ca2+, K+, Mg2+, and Na+) during the former and latter half of 2016. Analysis of the current quality of the raw water of natural mineral water among domestic manufacturers showed average anions contents of 0.46mg/L of fluorine, 8mg/L of chlorine ion, 1.5mg/L of nitrate nitrogen, and 12mg/L of sulfate ion. While the fluorine content was greater than the water quality criterion of 2.0mg/L at four points, the fluorine level was overall stable. The average cations contents included 21.3mg/L of calcium, 1.0mg/L of potassium, 3.4mg/L of magnesium, and 9.6mg/L of sodium. The chemical characteristics were compared among the major ions, and the results are presented in a piper diagram. The content ratio of cations was in the order of Ca2+> Na+>Mg2+>K+, whereas that of anions was in the order of SO42->Cl->NO3-N->F-. While the cations were slightly scattered, the anions were generally concentrated except for at a few points. The Ca-Na-HCO3 type was dominant overall in water sources from diorite, gneiss, and granite, while the Na-Mg-Ca-HCO3-Cl type was dominant in basalt sources. Mineral water manufacturers source their water under various conditions, including in-hole casing, excavation depth, and contact state of bedrock; even within the same rocky area, some differences in the water quality type can occur. When the depth of the water source was taken into account, the mean anions contents of F-, Cl-, NO3-N-, and SO42- were similar, with no significant differences according to depth. Of the cations, K+ and Na+ showed no significant differences across all the tubular wells, whereas Ca2+ and Mg2+ decreased in content with depth.
