Host Dependent Oviposition and Development of Azuki Bean Weevil (Callosobruchus chinensis L.) in Different Leguminous Seeds (기주에 따른 팥바구미(Callosobruchus chinensis L.)의 산란 선호성 및 성장)

  • To investigate the oviposition preference and development of azuki bean weevil (Callosobruchus chinensis L.), the following six different leguminous seeds were used in this study: red bean (Vigna angularis (Willd.) Ohwi & Ohashi), black soybean (Glycine max (L.) Merr.), soybean (Glycine max (L.) Merr.), seoritae (Glycine max (L.) Merr.), small black bean (Rhynchosia nulubilis) and kidney bean (Phaseolus vulgaris var. humilis Alef.). In the study of oviposition preference, the numbers of eggs per leguminous seed on red bean, black soybean, soybean, seoritae and small black bean were 1.23, 0.61, 0.69, 1.05 and 1.13, respectively. The maximum daily number of eggs was observed at 48 hours and the minimum was at 96 hours. According to each host leguminous seed, developmental time for each host seed was different. The shortest adult emergence time was on red bean (25.27 days). The other five leguminous seeds increased or doubled the adult emergence time. Adult emergence rates feeding on red bean, seoritae, black soybean, soybean, small black bean were 83.33%, 28.23%, 27.87%, 20.44%, and 11.59%, respectively. Emergence rate on red bean was four times higher than the rate on other seeds. The longevity of emerged female adults was almost all longer than that of males. The male adults weighed the lowest of feeding on small black bean. Female adults weighed the lowest of feeding on soybean. Adult weights were the heaviest for both males and females feeding on red bean. As a result, hosts of azuki bean weevil could decrease oviposition rate, emergence rate, adult longevity, and adult weight but increase emergence time. Especially in kidney bean, adult was not completely emerged. No eggs were laid. These results suggest that there might be emergence inhibitors in kidney bean. These imformation might be used to control damages caused by azuki bean weevils.

  • A bacterial disease of small red bean (Phaseolus angularis) was observed on field-grown plants in Suwon in year 2003. Leaf symptoms initially appeared as water-soaked spots that gradually enlarged, became flaccid and necrotic and were often bordered by a small zone of lemon yellow tissue. In the case of severe infection, dead leaves were defoliated. Pod symptoms consisted of the lesions that were generally circular, slightly sunken and dark reddish brown. Isolation made from diseased leaves on yeast extract dextrose calcium carbonate agar yielded nearly pure cultures of a yellow-pigmented bacterium typical of a xanthomonad. Three bacterial strains were purified and used for further tests. Pathogenicity of strains was confirmed on 3-week-old small red bean plants sprayed with bacterial suspensions containing $10^8 cfu/ml$ of phosphate buffered saline. The representative Xanthomonas strains isolated from small red bean were compared with X. axonopodis pv. phaseoli and X. axonopodis pv. phaseoli var. fuscans type strains for fatty acid profiles, biochemical tests and metabolic fingerprints using Biolog GN2 microplate, showing that all outcomes were indistinguishable between our isolates and reference strains. Two of three strains produced a melanin-like brown pigment extracellularly on King's medium B agar. These results suggest that this new small red bean disease observed in Suwon is bacterial fuscous blight caused by X. axonopodis pv. phaseoli and X. axonopodis pv. phaseoli var. fuscans.

Molecular Structural Properties of Legume Starches (두류 전분의 분자구조적 특성)

  • Molecular structural properties of legume starches were investigated. In intrinsic viscosity and degree of Polymerization of amylose and amylopectins, cow pea and mung bean were high, but kidney bean was low. Low molecular weight fractions for kidney bean starch were much eluted by gel chromatography. In the elution profiles of their amylose by Sepharose 2B-CL, molecular weight of kidney bean amylose was smaller than that of other amylose Molecular weights of cow pea and mung bean amyloses were large, but that of kidney bean amylose was small and red bean amylose was medium. The elution profiles by Sephadex G-50 after debranching amylopectins with pullulanase showed similar patterns.

  • To investigated the anticarcinogenic activity of 70% ethanol extracts from various cereal in vitro, antimutagenic activity, inhibitory effect of DNA strand scission and tumor promotion were examined. The antimutagenic activity of the beans such as black bean and small red bean was generally higher than that of cereals examined. However inhibitory activity of 70% ethanolic extracts against DNA strand scission induced mitomycin C showed that millet, job's tear, black bean and soy bean among cereals and beans tested in this study inhibited effectively the DNA strand scission. Antioxidative activity of some cereal extracts determined by using linoleic acid model system showed that Job's tear, millet and black bean were higher antioxidative activity than other cereals and beans. Conventional short-term antipromoter assay system using activation of Epstein Barr virus (EBV) clearly demonstrated that sorghum, buckwheat, black bean and small red bean have inhibitory effects on promotion in cellular carcinogenesis.

Effect of Mixed Soybeans Materials on Qulity Characteristics of Traditional Soybean Paste(Doenjang) during Aging (혼합콩이 전통된장의 숙성 중 품질에 미치는 영향)

  • The effects of five colored soybean (blue, red, yellow, white, and black) varieties on the quality characteristics of soybean paste (Doenjang) were investigated for proximate composition, salt content, pH, color, organic acid content, amino acid content, and mineral content after 80 days of aging. Soybean paste (Doenjang) samples were divided into TDS (traditional Doenjang prepared with soybean) and TDM (traditional Doenjang prepared with mixed soybeans, kidney bean, black soybean, kind of small bean, sword bean, green bean, red bean, at a radio of 1:1:1:1:1(w/w)). Moisture content was 49.66% for TDM and 49.52% for TDM. Protein and salt contents of the TDS sample were higher than those of TDM. The "L" values of the TDB sample increased the aging period was decreased. In contrast, the "L" and "$\alpha$" values of TDM did not show any significant differences. The main organic acids detected were malic acid > lactic acid > tartaric acid in the TDS and TDM samples. Glutamic acid contents were 2.83 g/100 g for the TDS sample and 2.15 g/100 g for the TDM. The levels of K, Mg, and Ca of the TDS sample were higher than those of TDM. No significant difference occurred between TDS and TDM regarding their quality characteristics after 80 days of aging. Further studies needed include sensory evaluation and functionality of soybean paste made from mixed soybean materials.

  • This study was investigated traditional food utilization actual conditions of Gwangju and Chollanamdo. Frequency of main meal ice plain white rice, boiled rice and cereals, bean-mixed rice, gruel Dakjuk, winter squash porridge, sesame porridge, noodles by noodles cut out with a kitchen knife, noodles with assorted mixtures, soup with dough flakes order frequency much have. It is Ssukgatmuchim, squid debt saliva, Jabanmuchim's order that season, hard-boiled food is beef boiled in soy sauce, mackerel radish hard-boiled food, order of bean curd hard-boiled food, panbroiling ate often by order of Kimchi panbroiling, red pepper anchovy panbroiling, pork panbroiling. Standardization of the Recipe for the Korean Traditional Drink "Omigalsu" (오미갈수(五味渴水)의 전통적 조리법 표준화 연구)

  • Omija, the major material of omigalsu, has liver protective and antioxidant effects, while mung bean has detoxification effects. A series of studies were conducted to standardize the traditional recipe for omigalsu to develop traditional functional drinks made from Omija extract and mung bean juice. Study 1 was designed to determine the optimal conditions for Omija extraction and mung bean juice. A higher water temperature and longer immersion time was associated with higher, free sugar and organic acid contents of omija extract; however, sensory evaluation revealed that the optimal extraction conditions for the highest acceptability, proper taste and red tone were $23^{\circ}C$ and 18 hrs of immersion. Conversely, the pH of the mung bean juice produced by varying the immersion time (5 hr, 11 hr, 17 hr) was found to be neutral, containing small levels of organic acid and free sugar, and showing a yellow tone. The results of the sensory evaluation also showed that the optimal conditions for taste, flavor and yellowness of mung bean juice was 5-hour-long immersion. Study 2 was designed to determine the optimum mixing ratio of omigalsu concentrate. Sensory evaluation revealed that the contents of sugar and total free sugar were highest when the mixing ratio among omija extract, mung bean juice and sugar was 1:1:20%, indicating that these conditions produced the most attractive color and highest overall acceptability. Study 3 was designed to determine the optimum dilution magnification for omigalsu. Sensory evaluation during summer revealed that the omigalsu produced by mixing 54 g of omigalsu concentrate into 200 cc water of $4^{\circ}C$ or $80^{\circ}C$ was most preferred, while during winter. Overall, the optimum dilution magnification for omigalsu was 4.7~5.4.

A Fluorometric Assay for Trypsin Inhibitor (트립신 저해단백질의 형광측정법)

  • A fluorometric method is described which permits the assay of trypsin inhibitor contained in a sample in an extremely small amount, utilizing a novel reagent generally called fluorescamine. The fluorometric assay with an enzyme kinetic approach has been found to be at least 100 times more sensitive than the well-known Kunitz's spectrophotometric method, considerably taster and less complicated, when it was demonstrated with the anti tryptic activities of very dilute extracts from soybean, red-bean and mung bean. Details of experimental procedure as well as theoretical considerations will be discussed.

Screening of Antimutagenic Activities from Cereals and Beans Including Rice (쌀을 포함한 곡류 및 두류의 항변이원 활성의 검색)

  • We have established the quantitative method for assay antimutagenic activity from natural products using SOS chromotest technique. Establishment of the method in this study makes it possible to numerize antimutagenic activities from samples in term of the sample amount required for 50% inhibition to mutagenic activity induced by the chemical mutagen under the standard assay condition. Antimutagenic activities of rices from different cultivars as well as other cereals were assayed through this method. The results revealed that antimutagenic activities of mutant cultivar, Suwon 393(Hyangdo) and Sanghaehyanghyulla(Jado), were higher than Chuchung which mainly consumed for steamed rice, and also indicated that antimutagenic activities of cereals, such as job'tear, buckwheat, small red bean, black bean were generally higher than that of brown rice.

A New Small Redbean Cultivar 'Geomguseul' with Lodging Resistance and High-Antioxidant Activity (쓰러짐에 강하고 항산화 활성이 높은 팥 '검구슬')

  • A new small red bean cultivar, 'Geomguseul', was artificially crossed between 'Chilbo' and 'SA8412-3-1-4-3-3-2-3' in 1998, fixed excellent agronomic characters by pedigree breeding method, and selected for the further trials with the name of 'Miryang 10'. It was prominent and showed good result, such as high grain quality, lodging resistance, and high-antioxidant activity in the regional adaptation yield trials (RYT) for three years from 2009 to 2011 and released for the public consumption as the name of 'SA9801-24-4-3-1' in 2011. 'Geomguseul' has a semi-determinate growth habit, yellow flower, green embryonic axis, black seed coat, white hilum, and small spherical seed (12.2 grams per 100 seeds). The average yield of 'Geomguseul' was 2.01 MT/ha in the regional yield trials (RYT) carried out for three years from 2009 to 2011 which was a little higher than that (1.98 MT/ha) of the check variety, 'Chungju'.
