Analysis of Myosin Heavy Chain Isoforms from Longissimus Thoracis Muscle of Hanwoo Steer by Electrophoresis and LC-MS/MS

  • The purpose of this study was to analyze myosin heavy chain (MHC) isoforms in bovine longissimus thoracis (LT) muscle by liquid chromatography (LC) and mass spectrometry (MS). LT muscles taken from Hanwoo (Korean native cattle) steer (n=3) used to separate myosin bands by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The peptide queries were obtained from the myosin bands by LC-MS/MS analysis following in-gel digestion with trypsin. A total of 33 and 43 queries were identified as common and unique peptides, respectively, of MHC isoforms (individual ions scores >43 indicate identity or extensive homology, p<0.05). MHC-1 (IIx), -2 (IIa), -4 (IIb), and -7 (slow/I) were identified based on the Mowse score (5118, 3951, 2526, and 2541 for MHC-1, -2, -4, and -7, respectively). However, more analysis is needed to confirm the expression of MHC-4 in bovine LT muscle because any query identified as a unique peptide of MHC-4 was not found. The queries that were identified as unique peptides could be used as peptide markers to confirm MHC-1 (14 queries), -2 (8 queries), and -7 (21 queries) in bovine LT muscle; no query identified as a unique peptide of MHC-4 was found. LC-MS/MS analysis is a useful approach to study MHC isoforms at the protein level.

Comparisons of Anthropometric and Environmental Factors, and Food Behaviors of Normocholesterolemia and Hypercholesterolemia in the Postmenopausal Women (정상콜레스테롤혈증과 고콜레스테롤혈증을 가진 폐경 후 여성의 신체적, 환경적 요인과 식행동 비교)

  • Women have a greater incidence of coronary heart disease(CHD) after menopause. It relates to hormone imbalance-induced changes in known CHD risk factors, especially hyperlipidemia. Accordingly, once women are diagnosed, the rate of morbidity and mortality is greater than for men. Thus, preventive strategies are critical to improve the quality oflife in the later years for women. The purpose of ths study was to explore the difference of anthropometric, environmental factors and food behaviors between normocholesterolemia and hypercholesterolemia within postmenopausal women. The subjects were classified as normocholesterolemia and hypercholesterolemia based on the Guideline for Korean Hyperlipidemia. The results obtained are summarized as follows. The distributions of general characteristics such as age, family income, occupation status, age at menarche, and menopausal period were not significantly different between the normocholesterolemia group and hypercholesterolemia group. The anthropometric factors such as body mass index(BMI) and waist hip ratio(WHR) were not significantly different between the normocholesteroemia group and the hypercholesterolemia group but odds ratio of hypercholesterolemia group was higher than that of normocholesterolemia but not significant. Aerobic activity more than 1hours/day was significantly related to low serum cholesterol concentration. Eating behaviors related to regular meal time, regular breakfast habit, and slow meal speed were related to low serum cholesterol concentration. Eating the chicken without skin was significantly related to low serum cholesterol concentration. Our data indicate that desirable eating habit, aerobic activity more than 1hr/day are important factors of serum cholesterol concentrations in postmenopausal women. The result of this study provide information that is important in designing appropriate dietary guidelines for hypercholesterolemia in postmenopausal women.

Physicochemical Characteristics of Toha-Jeot Added Cabbage Kimchi during Fermentation (토하젓 첨가 배추김치의 숙성 중 이화학적 특성)

  • We made the Toha-jeot added cabbage kimchi with Toha-jeot, which is salt-fermented Toha shrimp in Korean traditional fermented food. The physicochemical characteristics of cabbage kimchi for 5 week fermentation at 4$^{\circ}C$ were observed. The salt content was 2.10~1.50% during the fermentation period. The salt content decreased rapidly during the first week, stayed the same level until the 4 week and then decreased after the 4 week. The pH of control kimchi and Toha-jeot added cabbage kimchi were 5.69, 5,64, respectively on preparation day. The rate of decrease of pH of Toha-jeot added cabbage kimchi was slow as the fermentation processed. The total acidity of the two kinds of kimchi was increased. The total acidity was higher from Toha-jeot added cabbage kimchi than control cabbage kimchi at the later stage of fermentation. The contents of total sugar and reducing sugar were higher from Toha-jeot added cabbage kimchi than control cabbage kimchi at the early stage of fermentation. The total sugar and reducing sugar contents of the two kinds of cabbage kimchi were decreased gradually as the fermentation processed. The ascorbic acid contents of the two kinds of kimchi were decreased slightly at the early stage of fermentation and then increased from 2 to 4 week, while there was no further change at 5 week.

  • On the basis of UV spectral changes and TBA reaction, malonaldehyde (MA) in aqueous solution receives considerable photochemical modification by UV light of short wavelengths around $260m{\mu}$. When aflatoxin is added in the solution, UV light of long wavelengths around $360m{\mu}$ induces such changes quite rapidly and although the rate of change is rather slow, it is also true even with ordinary laboratory illumination(fluorescent). The modification is irreversible in nature and the role of aflatoxin in this reaction is identified as a photosensitizer. The mechanism involved in this modification is apparently due to the parallel dimerization of MA molecules, but not by head to tail combination of the molecules.

  • In order to culture the placenta of Capsicum annuum L. in synthetic medium, the experiment was initiated to find out the proper medium for callus formation and growth rate. Linsmaier and Skoog RM 1964 media was more effective than Murashige and Skoog or white media. The addition of 2, $4-D(10^{-5}M)\;and\;kinetin(10^{-6}M)$ into Linsmaier and Skoog RM 1964 media was very effective in callus formation. The growth rate was the most rapid from 2 weeks to 3 weeks after inoculation, and then it showed slow rate.

Micronutrients as Supportive Care for Gastrointestinal Cancer Patients; Benefits and Concerns

  • Cancer is the first leading cause of death in Korea and the second leading cause of death in the USA. There is extensive research into prevention of cancer and the support of oncology patients with diet or dietary supplements. In vitro and in vivo animal studies have indicated that antioxidants, including beta-carotene, alpha-tocopherol, and ascorbic acid, can yield anti-cancer effects in addition to providing protection against oxidative damage. Although many observational studies have shown that consuming fruits and vegetables can reduce the risk of some cancers, the results of several large-scale human intervention trials testing the benefits of a single or combined higher-dose of individual micronutrients have been inconsistent. Cancer can cause profound metabolic and physiological changes which may affect patients' nutrient requirements. Although the optimal route of nutrient delivery is through diet, cancer patients often suffer symptoms that disrupt their food intake, including anorexia, premature satiety, altered taste and smell, and changes in bowel mobility. In particular, micronutrient deficits can slow postoperative healing, contribute to depression symptoms, and decrease immune competence. Cancer patients are generally motivated to take dietary supplements to improve responses to treatment and quality of life. The Physician's Health Study II (PHS II) randomized controlled trial reported recently that daily multivitamin supplementation significantly, albeit modestly, reduced the risk of total cancer. Although evidence of multivitamin use benefits is limited in cancer patients, taking dietary supplements with constituents in the range of the recommended daily allowance according to the Dietary Reference Intake (DRI) recommendation is generally considered to be safe.

Chemical Characteristics of Taurine Added Kimchi during Fermentation at Low Temperature (타우린 첨가 김치의 저온 저장 중 화학적 특성)

  • The present study was carried out to elucidate the changes in the chemical characteristics of taurine added Kimchi during fermentation for 46 days at $6^{\circ}C$. Chinese cabbage was brined in a 10% salt solution for the control Kimchi and in a 10% salt solution containing 5% taurine for taurine added Kimchi (Taurine I, II, and III). One and three percent (w/w, based on Chinese cabbage) of taurine were added to make Taurine II and Taurine III, respectively. Reducing sugar and vitamin C contents for all the samples decreased after the 46-day fermentation. Taurine was not detected in the control, and the taurine contents, from the largest to the smallest, during the fermentation period were Taurine III, Taurine II and Taurine I. Amino nitrogen contents in all the samples studied proportionally increased during the 46-day fermentation. It is suggested that taurine does not affect the chemical characteristics of Kimchi during fermentation at low temperature ($6^{\circ}C$).

Fermentative Characteristics and Anti-Proliferative Activity against Mouse Carcinoma Cell Line of Kimchi prepared with Functional Cabbage (기능성 배추 김치의 발효 특성과 암세포 증식저해능)

  • To compare functional Chinese cabbage('Amtak' baechu; F1 hybrid cultivar between Brassica rapa and B. perkinensis, AB) with general Chinese cabbage ('Chunkwang' baechu; general spring cultivar, CB), two kinds of kimchi(ABK and CBK) prepared with AB and CB cultivar were fermented at $10^{\circ}C$ for 10 days. Their fermentative characteristics and anti-proliferative activities against mouse carcinoma cell lines were investigated. General kimchi(CBK) showed mature pH on the $6^{th}$ day of fermentation, whereas functional kimchi(ABK) reached pH on the $9^{th}$ day. CBK also exhibited acidity of mature stage on the $6^{th}$ day, but ABK reached mature acidity on the $9^{th}$ day. Although ABK and CBK were salted in the same condition, ABK had lower salinity than CBK, throughout the fermentation time. The highest total bacterial and lactic bacterial counts of CBK showed on the $8^{th}$ day of fermentation, but ABK showed the highest total bacterial and lactic bacterial counts on the $10^{th}$ day. The texture of ABK was harder than CBK for fermentation time. This seems to be corrleated with the slower fermentation rate of ABK. ABK showed significantly higher anti-proliferative activity (54.6% cell viability of control) in B16BL6 at $1,000{\mu}g/mL$. ABK was also higher in anti-proliferative activity than CBK throughout the fermentation time. However, there was no significant difference in the anti-proliferative activity of ABK between the fermentation times. In conclusion, fermentation of ABK showed a better texture, due to the slow fermentation rate and more anti-proliferative activity against mouse carcinoma cell line than those of CBK.

Evaluation of the Relationship between Freezing Rate and Quality Characteristics to Establish a New Standard for the Rapid Freezing of Pork

  • This study evaluated the effect of freezing rate on the quality characteristics of pork loin to establish an objective standard for rapid freezing. To generate various freezing rates, three air flow rates (0, 1.5, and 3.0 m/s) were applied under three freezing temperatures (-20℃, -30℃, and -40℃). Based on the results, freezing rates ranged from 0.26-1.42 cm/h and were graded by three categories, i.e, slow (category I, >0.4 cm/h), intermediate (category II, 0.6-0.7 cm/h) and rapid freezing (category III, >0.96 cm/h). Both temperature and the air flow rate influenced the freezing rate, and the freezing rate affected the ice crystal size and shear force in pork loin. However, the air flow rate did not affect thawing loss, drip loss or the color of pork loins. In the comparison of freezing rates, pork belonging to category II did not show a clear difference in quality parameters from pork in category I. Furthermore, pork in category III showed fresh meat-like qualities, and the quality characteristics were clearly distinct from those of category I. Although the current standard for rapid freezing rate is 0.5 cm/h, this study suggested that 0.96 cm/h is the lowest freezing rate for achieving meat quality distinguishable from that achieved with conventional freezing, and further increasing the freezing rate did not provide advantages from an energy consumption perspective.

  • In this study, in order to promote informatization of Meat Processed Food Industry, we standardize three processes of sales management, purchaser logistics and production management, develop standard modules, and develop integrated business functional design in ERP system development. This study reduces the cost of introducing the production equipment information standardization module in the food industry and the specialized IT companies that are slow in informatization, and maximizes the effectiveness and provides reliability by being a database where research standardization can be compared and analyzed. By applying the results of this study, it is possible to establish efficient processes of sales, buyer logistics, production, improvement of quality, and reduction of production cost from the receipt of food raw materials / subsidiary materials to finished products.
