Diversity of Chinese Indigenous Goat Breeds: A Conservation Perspective - A Review -

  • In this manuscript, a review of the diversity of Chinese indigenous goat breeds according to data from body stature and appearance, chromosome group, blood proteins, DNA molecular markers (mitochondria DNA, random amplified polymorphic DNA, microsatellite DNA, major histocompatibility complex) has been introduced. All of these provide efficient tools for the diversity analysis of Chinese indigenous goat breeds and are very important for biodiversity conservation, restoration of declining goat breeds, the priority defining in Chinese indigenous goat breeds' protection and the selection of nature preservation zones. Many Chinese indigenous goat breeds with small population size in the isolated mountains or reservoir areas are verging the potential threat of extinction, effectively lost with the rapid destroying of ecological environment. On the other hand, as a result of the introduction of modern commercial goat breeds and shortage of effective conservation, some populations, such as Small-xiang goat and Tibetan goat decrease rapidly in number of sires. In the interests of the long-term future of the goat breeds in China, conservation of goat breeds' genetic resources should be considered urgently and some conservation measures should be adopted. In addition, the continuing development of molecular biology will further enhance conservation of diversity of Chinese indigenous goat breeds.

Genetic diversity and distribution of edible scytosiphonacean algae from Ulleungdo Island, Korea

  • Despite the abundance of seaweeds from Ulleungdo Island, genetic diversity and distribution of edible brown algae from the island remain unstudied. We analyzed mitochondrial cox3 sequences from 86 specimens collected in the island and from the nearby Korean Peninsula. Our cox3 phylogeny for the first time confirmed the occurrence of fives species from Ulleungdo Island; Petalonia binghamiae, P. fascia, Planosiphon zosterifolius, and two cryptic species previously identified as Scytosiphon lomentaria. P. binghamiae was relatively homogeneous with three haplotypes. P. fascia comprised four haplotypes, which were grouped into two genetic lineages. S. lomentaria was heterogeneous with nine haplotypes and was divided into two cryptic species; one species clustered with taxa from cold waters while the other clustered with taxa from temperate and cold waters. Low genetic diversity in P. binghamiae while high genetic diversity in S. lomentaria from Ulleungdo Island are comparable to patterns observed from other species from the Korean peninsula. Ulleungdo Island, although small in size, is an ideal field laboratory to investigate genetic diversity and distributions of economic marine algae.

New Records of Three Tintinnopsis Species and Redescription of One Antetintinnopsis Species (Protozoa: Ciliophora) from Coastal Waters of Korea

  • Our study aimed to investigate the diversity of tintinnid species in Korea by collecting samples from coastal waters. As a result, we identified and redescribed three newly recorded species of the genus Tintinnopsis Stein, 1867 and one previously recorded species of the genus Antetintinnopsis Wang et al., 2021 in Korea. The loricae morphology and molecular phylogeny based on the 18S rDNA sequences of these four were analyzed. Tintinnopsis kiaochowensis Yin, 1956 is characterized by having an irregular collar with spiral turns and an obconical-shaped bowl. Tintinnopsis orientalis Kofoid and Campbell, 1929 is characterized by the inverted bell-shaped lorica with size of 121-140×86-94 ㎛. Tintinnopsis parvula Jörgensen, 1912 is characterized by its narrower collar than bowl and acute angle of the bowl (39-75°). The recorded species, Antetintinnopsis gracilis (Kofoid and Campbell, 1929) Wang et al., 2021 is characterized by a cylindrical collar narrower than the bowl width and with a size of 98-131×37-46 ㎛.

Extent of linkage disequilibrium and effective population size of the Landrace population in Korea

  • Objective: The genetic diversity of the Landrace population, a representative maternal pig breed in Korea, is important for genetic improvement. Previously, the effective population size (Ne) has been used to infer the genetic diversity of a population of interest. In this study, we aimed to use single nucleotide polymorphism (SNP) data to characterize linkage disequilibrium (LD) and the Ne of the Korean Landrace population. Methods: We genotyped 1,128 Landrace individuals from three representative Korean major grand-grand-parent (GGP) farms using the Illumina PorcineSNP60 version2 BeadChip, which covers >61,565 SNPs located across all autosomes and mitochondrial and sex chromosomes. We estimated the expected LD and current Ne, as well as ancestral Ne. Results: In the Korean Landrace population, the mean LD ($r^2$) of 3.698 million SNP pairs was $0.135{\pm}0.204$. The mean $r^2$ decreased slowly with as the distance between SNPs increased, and remained constant beyond 3 Mb. According to the $r^2$ calculations, 8,085 of 3.698 million SNP pairs were in complete LD. The current Ne (${\pm}$standard deviation) of the Korean Landrace population is approximately 92.27 [79.46; 105.07] individuals. The ancestral Ne exhibited a slow and steady decline from 186.61 to 92.27 over the past 100 generations. Additionally, we observed more a rapid Ne decrease from the past 20 to 10 generations ago, compared with other intervals. Conclusion: We have presented an overview of LD and the current and ancestral Ne values in the Korean Landrace population. The mean LD and current Ne for the Korean Landrace population confirm the genetic diversity and reflect the history of this pig population in Korea.

지역별 기업가정신 추이 및 결정요인 분석 (Regional Entrepreneurship Trend in Korea and Its Determinants)

  • 본 논문은 2000년대 한국의 16개 시도의 기업가정신 추이를 분석하고 지역별 기업가정신의 결정요인을 분석했다. 최근 2008년의 기업가정신은 광역시를 중심으로 과거보다 크게 위축된 것으로 나타났다. 또한 기업가정신의 결정요인 분석을 통해 연구개발투자, 재산권 보호, 산업집중도, 지방정부 재정지출의 GDP 대비 규모가 기업가정신의 주요한 결정요인임을 확인했다. 연구개발투자 및 재산권 보호의 확대는 기업가정신을 촉진하고 지방정부 규모의 확대는 기업가정신을 저해하는 것으로 나타났다. 산업집중도의 경우 전체 기업가정신에 미치는 영향은 통계적으로 유의하지 않지만, 특정지역의 산업구조가 특정 산업에 편중될수록 개인사업체의 기업가정신은 위축되는 반면 회사법인사업체의 기업가정신은 확대되는 것으로 분석됐다.

Geographic Genetic Contour of a Ground Beetle, Scarites aterrimus (Coleoptera: Carabidae) on the Basis of Mitochondrial DNA Sequence

  • The Scarites aterrimus (Coleoptera: Carabidae), is one of the carabid beetles dwelling exclusively on coastal sandy dunes. Habitat deterioration and equivalent activity have greatly concerned population declines in several species dwelling on the coastal sandy dunes. As a first step to establish long-term conservation strategy, we investigated the nation-wide magnitude and nature of genetic diversity of the species. As a first step, we sequenced a portion of mitochondrial COI gene, corresponding to "DNA Barcode" region (658 bp) from a total of 24 S. aterrimus individuals collected over nine sandy dunes belonging to four Korean provinces. The sequence analysis evidenced moderate to low magnitude of sequence diversity compared with other insect species distributed in Korean peninsula (0.152% to 0.912%). The presence of closely related haplotypes and relatively high gene flow estimate collectively suggest that there had been no historical barriers that bolster genetic subdivision. Population decline was postulated on the basis of several missing haplotypes that are well found in the species with a large population size. This interpretation is consistent with field observation of small population size in the coastal sandy dune habitats. The highest genetic diversity estimates were found in the coastal sand dune population of Seogwipo, Jeju Island, justifying a prior attention to the population, in order to sustain overall genetic diversity of the species. Further scrutinized study might be required for further robust conclusion.

Factors influencing efficiency of somatic embryogenesis of Gentiana kurroo (Royle) cell suspension

  • In this paper, we would like to show unexpected morphogenic potential of cell suspensions derived from seedling explants of Gentiana kurroo (Royle). Suspension cultures were established with the use of embryogenic callus derived from seedling explants (root, hypocotyl and cotyledons). Proembryogenic mass proliferated in liquid MS medium supplemented with $0.5mg\;l^{-1}$ 2,4-D and $1.0mg\;l^{-1}$ Kin. The highest growth coefficient was achieved for root derived cell suspensions. The microscopic analysis showed differences in aggregate structure depending on their size. To assess the embryogenic capability of the particular culture, 100 mg of cell aggregates was implanted on MS agar medium supplemented with Kin ($0.0-2.0mg\;l^{-1}$), $GA_3$ ($0.0-2.0mg\;l^{-1}$) and AS ($80.0mg\;l^{-1}$). The highest number of somatic embryos was obtained for cotyledon-derived cell suspension on $GA_3$-free medium, but the best morphological quality of embryos was observed in the presence of $0.5-1.0mg\;l^{-1}$ Kin, $0.5mg\;l^{-1}$ $GA_3$ and $80.0mg\;l^{-1}$ AS. The morphogenic competence of cultures also depended on the size of the aggregate fraction and was lower when size of aggregates decreased. Flow cytometry analysis reveled luck of uniformity of regenerants derived from hypocotyl suspension and 100% of uniformity for cotyledon suspension.

ISSR 표지자에 의한 동강할미꽃(Pulsatilla tongkangensis)의 유전다양성과 구조 (Genetic diversity and structure of Pulsatilla tongkangensis as inferred from ISSR markers)

  • 본 연구는 ISSR 표지자를 이용하여 동강할미꽃 3개 지역, 5개 집단, 총 177 개체로부터 유전적 특성을 구명하기 위하여 수행되었다. 6개 ISSR primer의 56개 표지자에서의 종 수준에서의 유전다양성은 P=94.6, SI=0.377, h=0.240로, 제한된 분포와 작은 집단크기를 고려할 때 상당한 수준으로 평가되었다. AMOVA 분석 결과 전체 유전변이의 약 12%가 지역 간에, 약 13%가 지역 내 집단 간에 존재하는 것으로 나타나, 지역 및 집단에 따른 분화정도는 상대적으로 낮은 것으로 평가되었다. 이는 인근집단 간에 유전자 교류가 비교적 원활히 이루어지기 때문으로 판단되었다. 동강할미꽂 집단 간 분화정도는 주로 지리적 거리에 의해 영향을 받는 것으로 나타났으며, 유집분석과 주좌표분석을 통해서도 이를 확인할 수 있었다. 크기가 작고 고립된 삼척집단(SC)에서는 많은 유전자좌에서의 표지자 밴드의 빈도가 현저히 다르거나 다른 방향으로 고정되어 유전적 부동이 진행되고 있음을 알 수 있었다. 앞으로 동강할미꽃의 집단크기가 계속 감소하면 유전다양성이 심각하게 훼손될 뿐만 아니라 집단 전체의 절멸로 이어질 우려가 있다. 따라서 동강할미꽃의 보전을 위해서는 무엇보다 고유의 서식처 환경을 보호하여 집단크기가 감소하거나 집단 간의 연결성이 훼손되지 않도록 해야 할 것이다. 현지에서의 종자산포나 이식 등의 보전 조치는 생태적, 유전적 특성을 고려하여 신중히 이루어져야 한다. 또한 집단의 유전적 구조 변화 등에 관한 기초연구와 함께 동강할미꽃의 효율적 보전을 위한 통합적인 체계 구축이 시급한 과제 중의 하나이다.

강릉 연안에서 형망의 갈퀴 간격 및 망목 크기에 따른 어획 생물의 종조성 및 군집 분석 (Species composition and cluster analysis of the communities caught by dredge in relation to tooth spacing and mesh size in the coastal waters of Gangneung, Korea)

  • The dredge gear is dragged along the bottom of the sea to catch targeted edible bottom dwelling species. Species composition and ecological index of the catches of dredge were estimated around Gangneung coastal fishing ground by dredge with different mesh size and tooth space from July to December 2013. Eight different types of dredge including four different tooth space (24.7 mm, 29.9 mm, 34.9 mm, 40.1 mm) and four different mesh size(15.5 mm, 32.7 mm, 51.1 mm, 60.0 mm) were used in the experiment. During the experiment, total catches were collected 31 species as sipunculida 1 species, mollusca 13 species, annelida 3 species, arthropoda 8 species, echinodermata 4 species and others 2 species. The dominant genus were mollusca and echinodermata while the dominant species were Megangulus venulosus, Pseudocardium sachalinensis, Schaphechinus brevis. The richness index was ranged 1.29-1.72, evenness index was 0.6-0.65 and diversity index was 1.65-1.83 according to the tooth space and mesh size of dredge. Richness index, diversity index were high at tooth space 34.9 mm dredge and ecological index showed decreasing tendency with the increasing of mesh size of dredge. Cluster and MDS analysis, based on a Bray-Curtis and similarity matrix of fourth root transformed data of number of species and wet weight, showed division into four different groups as four different tooth space (Group A), four different mesh size Group B (51.1 mm, 60.0 mm), Group C (32.7 mm) and Group D (15.5 mm).

Genetic Diversity among Tea (Camellia sinensis) Accessions Based on Random Amplified Polymorphic DNA (RAPD) Patterns

  • Genetic diversity of 45 tea accessions from Korea, Japan, China and Taiwan was investigated by using RAPD analysis. Out of the eighty primers screened, twenty primers generated 99 polymorphic bands with a polymorphic rate 87.0%. The size of the amplified fragments ranged from about 3,138 bp to 520 bp. By cluster analysis, all of the 45 accessions can be grouped into five groups. Over 90% of the 32 Korean accessions belonged to group II, III, IV and V. Moreover, newly developed Korean cultivars (accession no. 13, 14 and 15) belonged to very different group compared with any other Korean accessions. Among the Korean accessions, the minimum genetic similarity 0.500 was obtained between accession no. 17 and 37 and the largest genetic similarity 0.912 between no. 20 and 21.
