This study examined the extent of improvement of food safety knowledge and practices of employee through food safety training. Employee knowledge and practice for food safety were evaluated before and after the food safety training program. The training program and questionnaires for evaluating employee knowledge and practices concerning food safety, and a checklist for determining food safety performance of restaurants were developed. Data were analyzed using the SPSS program. Twelve restaurants participated in this study. We split them into two groups: the intervention group with training, and the control group without food safety training. Employee knowledge of the intervention group also showed a significant improvement in their score, increasing from 49.3 before the training to 66.6 after training. But in terms of employee practices and the sanitation performance, there were no significant increases after the training. From these results, we recommended that the more job-specific and hand-on training materials for restaurant employees should be developed and more continuous implementation of the food safety training and integration of employee appraisal program with the outcome of safety training were needed.
The gene product of dctD (DctD) activates transcription from the dctA promoter regulatory region by the $\sigma^{54}$ -holoenzyme form ofRNA polymerase ($E\sigma^{54}$ ) in Rhizobium meliloti and R. leguminosarum. The Escherichia coli integration host factor (IHF) stimulated DctD-mediated activation from the dctA promoter regulatory region of R. leguminosarum but not R. meliloti. In the absence of UAS, IHF inhibited DctD-mediated activation from both of these promoter regulatory regions. IHF also inhibited activation from R. leguminosarum dctA by nitrogen regulatory protein C (NtrC), another activator of $E\sigma^{54}$ but not by one which lacks a specific binding site in this promoter regulatory region. IHF, however, stimulated NtrC-mediated activation from the R. meliloti dctA promoter. Upon removal of the UAS, IHF inhibited NtrC-mediated transcription activation from the R. meliloti dctA promoter regulatory region. These data suggest that IHF likely faciliates productive contacts between the activators NtrC or DctD and $E\sigma^{54}$ to stimulate activation from dctA promoter.

This study was conducted to develop a selection marker for the identification of the Bacillus licheniformis K12 strain in microbial communities. The strain not only demonstrates good growth at moderate temperatures but also contains enzymes that catalyze the decomposition of various polymer materials, such as proteases, amylases, cellulases, lipases, and xylanases. To identify molecular markers appropriate for use in a microbial community, a search was conducted to identify variable gene regions that show considerable genetic mutations, such as recombinase, integration, and transposase sites, as well as phase-related genes. As a result, five areas were identified that have potential as selection markers. The candidate markers were two recombinase sites (BLK1 and BLK2), two integration sites (BLK3 and BLK4), and one phase-related site (BLK5). A PCR analysis performed with different Bacillus species (e.g., B. licheniformis, Bacillus velezensis, Bacillus subtilis, and Bacillus cereus) confirmed that PCR products appeared at specific locations in B. licheniformis: BLK1 in recombinase, BLK2 in recombinase family protein, and BLK3 and BLK4 as site-specific integrations. In addition, BLK1 and BLK3 were identified as good candidate markers via a PCR analysis performed on subspecies of standard B. licheniformis strains. Therefore, the findings suggest that BLK1 can be used as a selection marker for B. licheniformis species and subspecies in the microbiome.
Transgenic animals are very useful for scientific, pharmaceutical, and agricultural purposes. In livestock, transgenic technology has been used forthe genetic alteration of farm animals, the production of human proteins inlarge quantities in the milk of transgenic farm animals, and the generation of animals with organs suitable for xenotransplantation. To date, the transfer of foreign genes into farm animals has been performed mainly by microinjection of DNA into the pronuclei of fertilized eggs. However, the overall success rate of transgenic animals in livestock so far has been disappointingly low, eg., the efficiency is 0∼5% in swine, and less than 1% in sheep and cattle, compared with the rate in mice where 5% microinjected develop into transgenic animals. Recently, McGreath et al. (2000) have succeeded in producing the gene targeted sheep by the use of nuclear transfer from cultured somatic cells transfected with a foreign gene in vitro. However, we may need plenty of time until currently employ this method for gene transfer to farm animals. We have been studying to exploit the method for improving production efficiencies of transgenic animals with emphasis of its application to farm animals. The present paper describes three approaches that we have made in our laboratory to improve production efficiencies of transgenic animals, based on the DNA microinjection method. 1. Co-injection of restriction enzyme with foreign DNA into the pronucleus for elevating production efficiencies of transgenic animals. 2. Efficient selection of transgenic mouse embryos using EGFP as a marker gene. 3. Phenotypes of tansgenic mice expressing WAP/hGH-CAG/EGFP fusion gene produced by selecting transgenic embryos. 4. Efficient site-specific integration of the transgene targeting an endogenous lox like site in early mouse embryos.
GIS has been regarded as one of important tools or methodologies for various geoscience applications. Recently, spatial data integration schemes for site-specific or field-specific thematic mapping are newly developed and utilized. However, these kinds of approaches are somewhat insufficient quantitative assessment of integrated layers towards known targets in-detailed . Moreover, GIS analysis scheme is rarely extended to scientific approaches. In this study, simple approach of Multi-Buffer Zone Analysis , related to GIS analystical aspect, is addressed and an actual application for predicting or favorable mapping of mineral occurrences, one of GIS-based geoscientific approaches, is performed, As for geo-processing in GIS itself, this scheme can be regarded as extension or adaptiation of cell-based buffering or proximity analysis to geoscientific data interpretation. This study is based on rationale that surface geological pattern around primitives such as a point, a line, or a polygon in GIS, representing significant geological features, can be efficiently utilized to delineate complex geological behaviors or events, especially handling multiple dta sets originated from multiple sources such as airborne geophysical/radiometric exploration, field survey, and even a classified image of remote sensing. Conclusively, this methodology associated wit GIS is though to be helpful to analyze the spatial pattern of multiple data, pointing given sources, and is expected to effectively utilize for exploratory analysis of cell-based resultant layer integrated with complex or different data sources.
The main aim of this article is to identify the fundamental reason for changes in the geography of Korean foreign direct investment (FDI) in China. More specifically, the article argues that changes in the industrial sectors and the location of FDI resulted from the transition of strategies for Korean FDI in China since the late 1990s is not based on ownership specific advantages, internalization advantages and locational advantages (OIL), but on changes in site where both the needs of Korea and China are articulated, i.e. the strategy for business integration, centering on market, institution and technology. In other words, changes in the location of Korean FDI in China have been the result of changes in the Chinese market structure, institutional changes in Chinese inward FDI incentives and regulations and the accumulation of Korean technology capability since 2000. In addition, by investigating production networks in China, this article attempts to identify the relationships between changes in Korean FDI location and changes in market, institution and technology. Therefore, the empirical evidence provided by the case study of CDMA (code division multiple access) mobile communications FDI since 2000 in China is used to identify the impact of market, institution and technology on the location of Korean FDI in China.
This study aims to critically consider the local cultural policies of the government and local governments by village mural. Village murals, which began to be painted in the late 1990s, have been a nationwide cultural phenomenon for decades. In particular, it was consistently used as an important element of various local cultural policies implemented by the government and local governments. The government and local governments have been working on the project for two purposes: 'improving the residential environment' and 'activating tourism' but it has largely focused on 'activating tourism'. This study grasped the concrete status of major village murals across the country through field surveys, one of the case study methods. Also, through the analysis of official documents, the government and local governments operating in the village mural project were analyzed for the policy basis and administrative management plan. As a result of on-site surveys, most of the village murals across the country had problems such as uniform subject matter, low level representation, and poor follow-up management. This is partly based on the experiences and abilities of organizations and artists who participated in the village mural project, but fundamentally, it is due to the projects of the government and local governments that focused on short-term visual tourism rather than long-term improvement of the residential environment. It was confirmed that the projects of the government and local governments were carried out through vertical integration and that there were problems in the actual work, such as requiring specific topics, subject matter, designs and colors. As a result, the village murals 'objectify' the residents and 'romanticized' the place, resulting in a unified visual representation. Therefore, this study suggests that the administration and accounting should be strictly managed, but in actual work, the vertical integration structure should be eased by simplifying various documents and procedures, as well as rich cultural understanding of the target place, selection of various topics, and original subject matter and expressions.
The genus Mycobacterium includes crucial animal and human pathogens such as Mycobacterium tuberculosis, Mycobacterium leprae, and Mycobacterium bovis. Although it is important to understand the genetic basis for their virulence and persistence in host, genetic analysis in mycobacteria was hampered by a lack of sufficient genetic tools. Therefore, many functional vectors as molecular genetic tools have been designed for understanding mycobacterial biology, and the application of these tools to mycobacteria has accelerated the study of mechanisms involved in virulence and gene expression. To overcome the pre-existing problems in genetic manipulation of mycobacteria, this paper reports new vector systems as effective genetic tools in Mycobacterium smegmatis. Three vectors were developed; pKOTs is a suicide vector for mutagenesis containing a temperature-sensitive replication origin (TSRO) and the sacB gene encoding levansucrase as a counterselectable marker. pMV306lacZ is an integrative lacZ transcriptional fusion vector that can be inserted into chromosomal DNA by site-specific recombination. pTnMod-OKmTs is a minitransposon vector harboring the TSRO that can be used in random mutagenesis. It was demonstrated in this study that these vectors effectively worked in M. smegmatis. The vector systems reported here are expected to successfully applicable to future research of mycobacterial molecular genetics.

This research has a meaning to provide basic data for eco-friendly way of district unit plans and ecological landscape planning by evaluation of biotope preservation value at the level of district unit and designating land development of the site, the whole area of Sinseo-dong (Dong-gu, Daegu metropolitan city) for research site. The summary of analysis result is as follows. As a result of classification of biotope types on the research site, it is divided into 11 biotope groups such as a residential biotope group and 51 specific biotope types which is subordinate to the groups. As a result of the first value assessment on classified biotope types, there are 16 types of natural rivers which is full of vegetation as a I class. Also it is analysed as 9 types of IIclass, 14 of IIIclass, 8 of IVclass, and 4 of Vclass. In particular, in light of a wildlife habitat, EB, in case of broad-leaved tree of mixed forest assessed as a II class, was classified into Iclass which is one-step upgraded as a final class with the analysis as there is a structural characteristic (more than 71% of low density, 50 years of age-class). As a result of second assessment, it is analysed that there are 17 special sites (1a,1b) and 33 special sites (2a, 2b, 2c) respectively for preservation of species and biotope. Particularly, in case of the No. 27 space, it was assessed that it has the value of about medium (IIIclass) level, but its value was upgraded with the on-spot detailed investigation that most of Aristolochia contorta, designated as a rare plant by Ministry of Environment, is growing. It is regarded that the above-mentioned research result on evaluation of biotope preservation value is expected to provide very important basic materials for future district unit plans and smooth integration with landscape ecology plans and eco-friendly space development.

Background: In order to obtain a deeper understanding of an incident, it needs to be investigated to "peel back the layers" and examine both immediate and underlying failures that contributed to the event itself. One of the key elements of an effective accident investigation is recording the scene for future reference. In recent years, however, there have been major advances in survey technology, which have provided the ability to capture scenes in three dimension to an unprecedented level of detail, using laser scanners. Methods: A case study involving a fatal incident was surveyed using three-dimensional laser scanning, and subsequently recreated through virtual and physical models. The created models were then utilized in both accident investigation and legal process, to explore the technologies used in this setting. Results: Benefits include explanation of the event and environment, incident reconstruction, preservation of evidence, reducing the need for site visits, and testing of theories. Drawbacks include limited technology within courtrooms, confusion caused by models, cost, and personal interpretation and acceptance in the data. Conclusion: Laser scanning surveys can be of considerable use in jury trials, for example, in case the location supports the use of a high-definition survey, or an object has to be altered after the accident and it has a specific influence on the case and needs to be recorded. However, consideration has to be made in its application and to ensure a fair trial, with emphasis being placed on the facts of the case and personal interpretation controlled.
