Simple Method for a Cell Count of the Colonial Cyanobacterium, Microcystis sp.

  • The cell counting of colonial Microcystis spp. is a rather difficult and error-prone proposition, as this genus forms irregularly-shaped and irregularly-sized colonies, which are packed with cells. Thus, in order to facilitate a cell count, four methods of dividing the colonies into single cells were compared, including vortexing, sonication, $TiO_2$ treatment, and boiling. As a result, the boiling method was determined to generate the greatest number of single cells from a colony, and all colonies were found to have divided completely after only 6 min of treatment. Furthermore, no significant cell destruction, which might alter the actual cell density, was detected in conjunction with the boiling method (P=0.158). In order to compute the cell number more simply, the relationship between the colony size and the cell number was determined, via the boiling method. The colony volume, rather than the area or diameter was correlated more closely with the cell number ($r^2=0.727$), thereby suggesting that the cell numbers of colonial Microcystis sp. can also be estimated effectively from their volumes.

  • The polymer electrolyte membrane fuel cell (PEMFC) system was developed. In order to enhance the performance of membrane electrode assembly (MEA), the transfer printing method of the electrocatalyst layer on membrane was developed. The $H_2/O_2$ single cell with an electrode area of $50cm^2$ was fabricated and tested using 20 wt.% Pt/C as an electrocatalyst and the commercial and hand-made MEA such as Nafion 115, Hanwha, Dow, Flemion T and Gore Select. The 100-cell PEMFC stack with an active electrode area of $300cm^2$ was designed and fabricated using 40 wt.% Pt/C and 30 wt.% Pt-Ru/C as a cathode and anode electrocatalysts, respectively. The performance of PEMFC system was obtained to be 7kW (250A at 28V) and 3.5kW (70A at 50V) at $80^{\circ}C$ by flowing $H_2/air$ and methanol reformed fuel gas/air, respectively.

  • The focus of this paper is to develop a mathematical model for investigating the dynamic performance of a polymer electrolyte membrane fuel cell. The model in this work is based on physical laws having clear significance in replicating the fuel cell system and can easily be used to set up different operational strategies. Simulation results display the transient behavior of the voltage within each single cell, and also within a number of such single cells combined into a fuel cell stack system. A linear as well as a nonlinear analysis of the polymer electrolyte membrane fuel cell system(PEMFC) has been discussed in order to present a complete and comprehensive view of this kind of modeling. Also, a comparison of the two kinds of analysis has been performed. Finally, the various characteristics of the fuel cell system are plotted in order to help us understand its dynamic behavior. Results indicate that there is a considerable amount of error in the modeling process if we use a linear model of the fuel cell. Thus, the nonlinearities present in the fuel cell system should be taken into account in order to obtain a better understanding of the dynamic behavior of the fuel cell system.

Dissection of Cellular Communication between Human Primary Osteoblasts and Bone Marrow Mesenchymal Stem Cells in Osteoarthritis at Single-Cell Resolution

  • Background and Objectives: Osteoblasts are derived from bone marrow mesenchymal stem cells (BMMSCs) and play important role in bone remodeling. While our previous studies have investigated the cell subtypes and heterogeneity in osteoblasts and BMMSCs separately, cell-to-cell communications between osteoblasts and BMMSCs in vivo in humans have not been characterized. The aim of this study was to investigate the cellular communication between human primary osteoblasts and bone marrow mesenchymal stem cells. Methods and Results: To investigate the cell-to-cell communications between osteoblasts and BMMSCs and identify new cell subtypes, we performed a systematic integration analysis with our single-cell RNA sequencing (scRNA-seq) transcriptomes data from BMMSCs and osteoblasts. We successfully identified a novel preosteoblasts subtype which highly expressed ATF3, CCL2, CXCL2 and IRF1. Biological functional annotations of the transcriptomes suggested that the novel preosteoblasts subtype may inhibit osteoblasts differentiation, maintain cells to a less differentiated status and recruit osteoclasts. Ligand-receptor interaction analysis showed strong interaction between mature osteoblasts and BMMSCs. Meanwhile, we found FZD1 was highly expressed in BMMSCs of osteogenic differentiation direction. WIF1 and SFRP4, which were highly expressed in mature osteoblasts were reported to inhibit osteogenic differentiation. We speculated that WIF1 and sFRP4 expressed in mature osteoblasts inhibited the binding of FZD1 to Wnt ligand in BMMSCs, thereby further inhibiting osteogenic differentiation of BMMSCs. Conclusions: Our study provided a more systematic and comprehensive understanding of the heterogeneity of osteogenic cells. At the single cell level, this study provided insights into the cell-to-cell communications between BMMSCs and osteoblasts and mature osteoblasts may mediate negative feedback regulation of osteogenesis process.

  • Effects of hyperthermia on the induction of DNA single strand breaks and replication inhibition were studied in bleomycin-treated CHO-K$_1$ cells by alkaline elution and alkaline sucrose gradient sedimentation. Bleomycin-induced DNA single strand breaks of DNA were dose-and time-dependently increased, and these strand breaks of DNA were gradually rejoined as post-incubation time passed. Treatment with hyperthermia alone did not affect the induction of DNA single strand breaks. However, pre-exposure of cells to hyperthermia followed by bleomycin treatment greatly increased the single strand breaks, and also reduced the rejoining processes of bleomycin-induced DNA single strand breaks. Bleomycin selectively inhibited the replicon initiation. The combined treatment with hyperthermia and bleomycin markedly potentiated the nonspecific inhibition of replication.

  • For low-cost embedded EEPROM, in this paper, single polysilicon EEPROM and n-channel high-voltage LDMOST device are developed in a $0.25{\mu}m$ standard CMOS logic process. Using these devices developed, the EEPROM chip is fabricated. The fabricated EEPROM chip is composed of 1 Kbit single polysilicon EEPROM away and high voltage driver circuits. The program and erase characteristics of the fabricated EEPROM chip are evaluated using 'STA-EL421C'. The fabricated n-channel high-voltage LDMOST device operation voltage is over 10 V and threshold voltage window between program and erase states of the memory cell is about 2.0 V.

  • We have designed a single gap transflective liquid crystal display (LCD) driven by a fringe electric field, in which the +LC (${\Delta}{\varepsilon}$=7.4, rubbing angle= $80^{\circ}$) is homogeneously aligned in the initial state. This device is a problem that the voltage-dependent transmittance and reflectance curves do not match each other. Thus a dual driving circuit is required. This study shows that optimization of the rubbing angle in the transmissive and reflective regions solves this problem so that the transflective display with a single cell gap and single gamma curve for reflective and transmissive region is possible.

  • As a fuel cell converts the chemical energy of the fuel cell into electrical energy by electrochemical reaction, the fuel cell system is uniquely integrated technique including fuel processor, fuel cell stack, power conditioning system. The residential fuel cell-PCS(Power Conditioning System) needs to convert efficiently the DC current produced by the fuel cell into AC current using single-phase DC-AC inverter. A single-phase DC-AC inverter has naturally low frequency ripple which is twice frequency of the output current. This low frequency(120Hz) ripple reduces the efficiency of the fuel cell. This paper presents notch filter with IP voltage controller to reject specific 120Hz current ripple in single-phase inverter. The notch filter is designed that suppress just only specific frequency component and no phase delay. Finally, the proposed notch filter design method has been verified with computer simulation and experimentation.

  • Subsequently, Part I [1], which was about the single-cell model, a composite thin-walled beam with a multi-cell of chord-wise asymmetric cross-section, was selected in this study. Moreover, the theoretical dynamic characteristics of the model were analyzed. For this analysis, mathematical modeling was performed by considering the warping restraint effects, transverse shear effects, taper ratio and cross-section ratio. Similar to part I, the mass, stiffness coefficients and Eigen frequencies of the multi-cell section considered were investigated. In particular, the comparison between the multi-cell and single-cell sections and the effects of the cross-section ratio and taper ratio of the model on the Eigen frequencies were analyzed. However, the results compared when the asymmetry of the section was considered and warping function were not corrected.

  • The genus Chattonella belonging to the class raphidophyceae, is a harmful algal bloom species. Recently, its occurrence has been increasing and expanding along the Korean coast. Species identification of the genus Chattonella only by morphological observation is difficult due to the lack of rigid cell walls. In this study, the morphological characteristics and genetic affinity of Chattonella sp. isolated from Deungnyang Bay in 2017 were examined. We carried out single-cell isolation from field samples then sequenced three different areas using the single-cell PCR method: 1) parts of ribosomal operon, the large subunit (LSU) of the rDNA, 2) the chloroplast-encoded subunit psaA of Photosystem I, and 3) rbcL encoding the large subunit of the Rubisco gene. The cells were morphologically very similar to the general genus Chattonella ($74.0{\pm}10.1{\mu}m$ in length, $33.1{\pm}3.6{\mu}m$ in width). The three partial gene sequences were insufficient to justify distinction at the species rank. However, they clustered at 99-100 % sequence similarity with C. marina, C. marina var. antiqua and C. marina var. ovata.
