  • In vitro fertilization(IVF) derived morula and blastocyst embryos were bisected by a simple method and cultured in vitro without zona pellucida And also bisected embryos were frozen-thawed and cultured in vitro) to evaluate the survival rate. The results obtained were as follows : The average number of grade I or II immature follicular oocytes recovered by slicing method per ovary was 11.9 from 142 ovaries. Following in vitro fertilization, the rates of cleavage and in vitro development to morula and blatocyst were 61.7 and 32.2% respectively. Studies on the Interrelationship between the Vestibular Semicircular Canals and the Extraocular Muscles in Rabbits and Cats (가토(家兎) 및 가묘(家猫)에 있어서 전정반규관(前庭半規管)과 외안근(外眼筋)의 상관성(相關性)에 관(關)한 연구(硏究))

  • The present experiment was carried out, in the rabbit and cat, in order to explore functional interrelationship between the vestibular semicircular canals and extraocular muscles, which are involved in the vestibulooculomotor reflex as the receptor and effector organ respectively. Semicircular canals were subjected to electrical stimulation, lymphatic fluid flow or acute freezing, and responses of the extraocular muscles were recorded in terms of changes in electromyographic activity and isometric tension. Electrical stimulation of a unilateral canal elicited contraction of the superio-medial muscle group (superior oblique, superior rectus and medial rectus muscles) in the ipsilateral eye and the inferio-lateral muscle group (inferior oblique, inferior rectus and lateral rectus muscles) in the contralateral eye. Thus a simple and distinct axiom was found in the pattern of the reflex-response of the extraocular muscles. Inhibition of the unilateral canals elicited the extraocular muscle responses contrary to those observed by excitation of the canal. Based on the present experimental results, it was demonstrated that the functional interrelations between the semicircular canals and extraocular muscles are rather equivalent in the frontal eyed cats (with binocular vision) and lateral eyed rabbits (with monocular vision). Therefore the previous thesis that the vestibuloocular relations vary from species to species awaits experimental reevaluation.

  • Cold acclimation improves freezing tolerance in plants. In higher plants, many advances have been made toward identifying the signaling and regulatory pathways that direct the low-temperature stress response; however, similar insights have not yet been gained for simple nonvascular plants, such as bryophytes. To elucidate the pathways that regulate cold acclimation in bryophytes, we used two PCR-based differential screening techniques, cDNA amplified fragment length polymorphism (cDNA-AFLP) and suppression subtractive hybridization (SSH), to isolate 510 ESTs that are differentially expressed during cold acclimation in Physcomitrella patens. We used realtime RT-PCR to further analyze expression of 29 of these transcripts during cold acclimation. Our results show that cold acclimation in the bryophyte Physcomitrella patens is not only largely similar to higher plants but also displays distinct differences, suggests significant alteration during the evolution of land plants.

  • The repair technique utilizing surface impregnant is widely used due to its simple construction procedures and economical benefit. Surface protection through this reparing technique cannot increase the bearing capacity of concrete members much but increase the durability performance and service life efficiently. In this study, fundamental tests such as water suction and permeation are performed for concrete specimens with several organic/inorgarnic impregnant and suitable impregnant is selected on the basis of the results. Finally, durability tests such as carbonation, freezing and thawing, and chloride attack are carried out for the concrete specimens with selected impregnant and it is experimentally verified that they have good resistance to deteriorations.

  • The market size of Korea's seafood industry is estimated to be about 62 trillion won ('19), of which the seafood processing industry is about 10 trillion won ('19), accounting for approximately 16% of the total market size. However, the current level of processing technology required for the development of seafood products is mainly simple primary processing, such as freezing and drying raw materials (in Jeollanam-do, which ranks first in the country in terms of seafood production, primary processors account for 86% of all seafood processors).In the future, trends in the development of processing technology for seafood will become more advanced in response to changes in domestic and international food consumption patterns. Blue food tech linked to ESG, convergence and digitalization linked to the 4th industry revolution will be further advanced in line with the national seafood industry policy direction. Through this, it is expected to establish itself as an axis of future technology management in the fisheries sector based on the large and small success stories of accumulated seafood R&D commercialization.

Artificial Sea Ice Increasing to Mitigate Global Warming (지구 온난화 경감을 위한 인공해빙증가)

  • This study suggests a method of alleviating global warming by the increase of the Earth surface albedo through Artificial Sea ice Increasing (ASI) over the Available Freezing Areas (AFA). The method is developed based on the fact that the large sea surface area in or near the Arctic and the Antarctic has no ice even though both water and air temperatures are below zero and the artificial sea ice generation is thus available. The mean energy of $0.85Wm^{-2}$, which was suspected of adding to the earth by the global warming effect was calculated to offset at once when the sea ice area about $4.09{\times}10^6km^2$ was additionally increased. In addition, three techniques for producing ice plates on the sea surface (using ships, installation apparatus, and floating matter such as Green Cell Foam) for ASI were proposed. According to the result of simple analysis using the energy balance model, when ASI was maximally operated only for 3 months (September, October, and November) over AFA, it is expected that the annual mean temperature of earth surface would be decreased about $0.11^{\circ}C$ in the following year. On the other hand, in case of generating the artificial sea ice in all four seasons, a risk of triggering snowball earth was detected.

Effects of Triladyl-egg Yolk Diluents on the Viability of Frozen Korean Black-goat Spermatozoa from Cauda Epididymis and Electro-ejaculated Semen (Triladyl-난황 희석제가 한국 재래 흑염소의 정소상체 및 전기자극 유래 정자의 융해 후 생존성에 미치는 영향)

  • To preserve genetic materials, cryopreservation of the semen from live animals is the main technique to establish cryo-banking system which could be used for artificial insemination and embryo transfer. However, the population of Korean black goat (KBG) becomes to dwindle in number and is now faced genetic erosion by crossbreeding with non-native breeds in small KBG farms. In this study, simple freezing method was used to preserve frozen semen from KBG using spermatozoa of cauda epididymis (CE) and electro-stimulated semen (ES). The negative effects of seminal plasma on fresh sperm was confirmed using precipitation test of Triladyl egg yolk diluent and sperm viability after thawing was compared between CE and ES spermatozoa. When seminal plasma of fresh ES semen was washed with semen washing media (SWM), the rates of live sperm shown no significant difference between CE and ES spermatozoa before freezing. However, the survival rate of frozen/thawed CE sperm was higher than ES ($74.6{\pm}10.6%$ vs $53.8{\pm}5.2%$) with significant difference (p < 0.05). The results of longevity test on frozen/thawed sperm from CE showed healthier sperm than ES. Therefore, spermatozoa from CE could be used for cryo-banking system in KBG lines. The more studies are needed to increase survival rate of ES semen.

Microsurgical Training using Preserved Saphenous Vein (혈관문합 연습을 위한 보존된 복재정맥의 활용)

  • Purpose: Given that the critical nature of the microvascular anastomosis to what is often a long and difficult reconstructive operation, trainees need to have a high level of microsurgical competence before being allowed to perform microsurgery on patients. Some artificial substitutes and dead or live animal models have been used to improve manual dexterity under the operating microscope. Yet, most surgeons are not equipped with such models, so search for easy available and appropriate microsurgical practice model have been an issue. Umbilical artery, placental vessels and gastroepiploic arteries have been previously suggested as a microsurgical training model, which involves other surgical departments. The purpose of this article is to introduce that saphenous vein specimen obtained from varicose vein surgery is useful and has many advantages as training model for the practice of microvascular anastomosis. Methods: The conventional technique using perforation/inversion method with a metallic stripper is widely performed for varicose vein patients. The stripper is inserted through disconnected safeno-femoral junction and retrieved at the knee or the medial side of ankle. The length of saphenous vein specimens removed is about that of one's leg and inversed from inside out. Obtained saphenous vein specimens are re-inversed and cleansed with normal saline, to be readily available for microsurgical practice. Preserved in a squeezed wet saline gauze and refrigerated, frozen or glycerated specimens were investigated into their comparative quality for microsurgical practice. Results: Varicose vein surgery remains one of the common operations performed in the field of plastic surgery. Convenient informed consent regarding the vessel donation can be easily signed. The diameter of the obtained saphenous vein is as variable as 1.5 to 6 mm, which is already stripped, and is in sufficient length corresponding to that of patient's leg. Vessels specimens were available for microsurgical practice within 1 week period when preserved with squeezed wet saline gauze, and the preservation period could be extended monthly by freezing it. Conclusion: Saphenous vein obtained from varicose vein patients provide with variable size of vessel lumen with sufficient length. The practice can be cost effective and does not require microsurgical laboratory. Additionally there is no need of involving other surgical departments in acquiring vessel specimens. Furthermore, simple preservation method of refrigerating for a week or freezing with squeezed wet saline gauze for a month period, allow the saphenous vein obtained after varicose vein surgery as an excellent model for the microsurgical practice.

Evaluation of Moisture Susceptibility of Porous Hot Mixed Asphalt Mixtures with Hydrated Lime using Non-destructive Impact Test (비파괴 충격파 시험을 통한 소석회 첨가 투수성 가열 아스팔트 혼합물의 수분민감성 평가)

  • PURPOSES: It is theoretically well known all over the world, that porous hot mixed asphalt (HMA) with hydrated Lime improves moisture and rutting resistance, and reduces pothole occurrence frequency, as well as the life cycle cost (LCC). METHODS : Addictive in the two different formations of the liquid anti-stripping Agent and powder Hydrated-Lime was applied in this investigation in order to obtain relatively clear results according to their types and conditions. Firstly, the moisture conditions were set, and applied to the porous HMA mixtures with hydrated lime (anti-stripping agent). Next, it was followed by a non-destructive test with the application of three freeze-thaw cycles, which were individually carried out thrice to compare the results of the dynamic moduli. Lastly, the hydrated lime effect related to moisture sensibility to porous HMA has been verified through the analysis of the modulus results regarding the change rate of dynamic modulus per n-cycle. RESULTS: It is clear from this investigation, that the dynamic modulus is inversely proportional to the change in temperature, as the graph representing the rigidity of the thermorheologically simple (TRS) material showed gradual decline of the dynamic modulus with the increase in temperature. CONCLUSIONS: The porous HMA mixture with the anti-stripping agent (hydrated Lime) has been found to be more moisture resistant to freezing and thawing than the normal porous HMA mixture. It is clear that the hydrated lime helps the HMA mixture to improve its fatigue resistance.

Bioavailability of Tripotassium Dicitrato Bismuthate by ICP-MS in Human Volunteers (ICP-MS를 사용한 구연산비스마스칼륨 (Tripotassium dicitrato bismuthate)의 생체이용률 측정)

  • This study was aimed to establish analytical method of Bi to develop a guideline of the bioequivalence test of tripotassium dicitrato bismuthate (TDB). For this purpose, a simple, specific and sensitive inductively coupled plasma-mass spectrometry (ICP/MS) method were developed and validated in human plasma. Various concentrations of bismuth standard solution (0-25ng/mL) were prepared with distilled water and human blank plasma. To 10mL of the volumetric flasks, 2mL of blank plasma was added with 8ml of distilled water. Bi standard solution was added to prepare the calibration samples and injected into ICP-MS. The plasma samples obtained from volunteers given 3 tablets of bismuth (total 900mg as TDB) were analyzed as described above. As a result, the coefficients of variation were <20% in quantitation limit (0.2 ng/mL) and <15% at the rest of concentrations. The stability test by repeated freezing-thawing cycles showed that the samples were stable only for 24hr. The stability tested for samples with a short-term period of storage at room temperature and pre-treatment prior to the analysis showed very stable over 24hr. In 8 healthy Korean subjects received Denol tablets at the dose of 900mg bismuth, AUC, $C_{max},\;T_{max}$ and half-life $(t_{1/2})$ were determined to be $198.33{\pm}173.78 ng{\cdot}hr/mL,\;64.48{\pm}27.06 ng/mL,\;0.52{\pm}0.21 hr,\;and\;5.15{\pm}2.67 hr$, respectively, from the plasma bismuth concentration-time curves. In conclusion, the method was suitable for the determination of bismuth in human plasma samples and could be applied to bioequivalence test of bismuth tablet.
