Utilization and Isolation of new active substances from Sericulture Related MaterialsII. Development of an oral antihyperglycemic agent from silkworm powder

  • Since 1992, Sericulture & Entomology Research Institute(NSERI) and Kyung Hee University group screened an activity of lowering blood-glucose levels with silkworm related materials such as silkworm larvae powder, dried feces, pupae and silkworm moth in other to guide laymans to rational and educated utilization of silkworm-related materials for the treatment of diabetes mellitus. In experiments examining several silkworms in different stages and prepared in different conditions, a freeze dried silkworm powder of 5th instar 3rd day showed a higher glucose lowering activity about 20% than heat dried matured silkworm powder. Among the three dosage of 500mg, 830mg and 1,160mg in ate preliminary clinical trial, the 830mg exhibited a significant effect on postprandial blood glucose level and did and did not cause any hypoglycemic side effect. In the blood glucose lowering activity of mulberry and silkworm varieties, the Yongcheonppong and Samkwangjam showed the highest activity for lowering blood glucose levels. In experiments to see the difference in blood-glucose lowering activity between either male and female or larvae and pupae, activity of larvae was higher than that of pupae and the male was higher than female. The heating dry and artificial diet showed lower than mulberry diet and normal freeze dry of the 5th instar 3rd day. Among the sericultural products, larvae showed the highest activity. We find out the fact that effect of silkworm powder attributed to the inhibition of ${\alpha}$-glucohydrolase catalyzed reaction in the small intestine.

1-Deoxynojirimycin Content and Blood Glucose-Lowering Effect of Silkworm (Bombyx mori) Extract Powder

  • We investigated the 1-deoxynojirimycin (1-DNJ) content of extracts from silkworm larvae at each developmental stage within three silkworm varieties. We also compared the content of the following polyhydroxylated alkaloids in the silkworm extracts: 1-DNJ, fagomine, and 1,4-dideoxy-1,4-imino-d-arabinitol (DAB). In addition, we evaluated the glucose-lowering effects of silkworm extract powder in db/db mice. The 1-DNJ content was the highest in Yeonnokjam $5^{th}$ instar $3^{rd}$ d larvae and Hansaengjam $5^{th}$ instar $3^{rd}$ d larvae, which contained 18.4 mg/100 g dry weight and 18.3 mg/100 g dry weight, respectively. The $5^{th}$ instar $3^{rd}$ d larvae exhibited a higher content of 1-DNJ than that of $5^{th}$ instar $5^{th}$ d larvae among all varieties. The glucose-lowering effects of silkworm extracts and Yeonnokjam powder were tested on db/db mice, and the blood glucose levels were found to decrease significantly in the YR70 group. Silkworm extracts (180 mg/kg, 90 mg/kg, 45 mg/kg, and 22.5 mg/kg) and acarbose (50 mg/kg) were administered orally for 4 wk. Changes in water intake were not statistically significant between control and silkworm extract-treated groups. Compared to the control group, blood glucose levels in the silkworm extract powder-treated group decreased in the 22.5 mg/kg/d group after being administered for 4 wk. This decrease was statistically significant. Furthermore, biochemical changes in the AST(Aspartate aminotransferase), ALT(Alanine aminotransferase), TCHO(Total Cholesterol), TG(Triglyceride), LDL(Low density lipoprotein), and HDL(High density lipoprotein) levels in blood were not observed. However, statistically significant decreases in blood GLU in the 22.5 mg/kg/d group compared to that of the control group occurred. In addition, the epididymal fat weight of the silkworm extract powder-treated group decreased significantly in both the 22.5 mg/kg/d group and 180 mg/kg/d group compared to that of the control group, but there were no statistically significant changes in perirenal fat weight. These results demonstrate that silkworm extracts inhibit changes in blood glucose levels in model diabetic mice.

Biology of a predatory bug Eocanthecona furcellata Wolff (Hemiptera : Pentatomidae) on Vapourer tussock moth larvae: a major pest of tasar silkworm food plants

  • Tropical Tasar culture is forest and agro-based activity that covers agricultural and industrial activity. Tasar silk is produced by the silkworm, Antheraea mylitta which is polyphagous in nature. A large number of pests are reported to attack both the host plant as well as silkworm. As the rearing is conducted outdoor silkworms are exposed to attack of various pests during their life cycle. Eocanthecona furcellata (Hemiptera: Pentatomidae) is the major predator of tasar silkworm during young age rearing. Both nymphs and adults of the stink bug suck the heamolymph of silkworm leading to death. The present work was undertaken to study the life cycle of E. furcellata on larvae of vapourer tussock moth which is a major pest of tasar host plants. The incubation period, nymphal duration and total developmental period from egg to adult was $8.0{\pm}0.45d$, $16.0{\pm}3.24$ and $22.0{\pm}4.20d$, respectively. Pre-oviposition, oviposition and longevity of male and female stink bug was $3.20{\pm}0.38$, $17.60{\pm}1.86$, $42.40{\pm}1.94$, $37.00{\pm}3.18d$, respectively. Analysis of the data revealed no significant difference in life cycle of the predator when provided silkworm and vapourer tussock moth larvae as hosts. Which clearly indicates that E. furcellata can multiply on vapourer tussock moth larvae during non rearing periods. Adoption of recommended pest control measures for defoliators of tasar host plants during non-rearing periods will help in reducing the incidence of host plant pests and also in reducing the incidence of stink bug during rearing season. During the study it was also observed that larvae semilooper, boll headed caterpillar and nymphs of leaf hopper serve as alternate/secondary hosts to E. furcellata during non rearing seasons.

Production techniques to Improve the Quality of Steamed and Freeze-Dried Mature Silkworm Larval Powder

  • Cooked mature silkworms have received increasing attention as a new source of income for sericultural farms. Quality improvement of these worms as the end-product requires prevention of the following: inclusion of unfertilized silkworm eggs, mixing with impurities, producing undercooked or overcooked mature larvae, and mixing spewed silk substance. Preventing unfertilized eggs and impurities and reducing undercooked or overcooked mature larvae can be achieved by sericultural farms. However, the prevention of spewed silk substance requires the development of new techniques. This study developed new techniques for optimum cold storage temperature, soaking, and hydro-thermal treatment to minimize the spewing of mature larvae prior to steaming: when larvae mature after 7 days on the $5^{th}$ instar, they should be soaked in water at room temperature of $25^{\circ}C$ immediately upon collection and stored at $8^{\circ}C$ for less than 3 days. They should be soaked in boiling water for approximately 10 s immediately before steaming to kill the larvae and cooked with steam. This method allows for the production of high-quality mature larvae without spewed silk substance. These new techniques will considerably improve the production of well-cooked quality mature larvae by minimizing silk substance spewed out from the larvae.

Influence of Different Environmental Conditions on Cocoon Parameters and Their Effects on Reeling Performance of Bivoltine Hybrids of Silkworm, Bombyx mori. L.

  • Three newly authorized bivoltine silkworm hybrids namely, $CSR2{\times}CSR4$ (productive single hybrid), $(CSR6{\times}CSR26){\times}(CSR2{\times}CSR27)$ (productive double hybrid) and $CSR18{\times}CSR19$ (robust single hybrid) were chosen for the present study. These hybrids were subjected to different temperature and humidity treatments i.e., $25{\pm}$1^{\circ}C and RH $65{\pm}5%$ (control), $30{\pm}1^{\circ}C$, with combinations of low relative humidity (RH $65{\pm}5%$) and high RH ($85{\pm}5%$) at different stages during rearing and spinning of silkworm larvae. The larvae of after 3rd moult were subjected to different thermal and humidity stress till the assessment of cocoon traits. The comparative rearing and reeling performance clearly indicated that the deleterious effect of high temperature and high RH was more pronounced for the majority of traits such as cocoon uniformity, cocoon weight, shell weight, shell percentage, reelability, filament length, raw silk percentage raw silk recovery denier and waste percentage on silk weight than other temperature and RH treatments and this effect was almost similar for all three silkworm hybrids studied. The present investigation clearly indicate that the deleterious effect of high temperature and high RH was more pronounced on rearing and spinning of silkworm larvae than other temperature and RH treatments and similar effect was noticed for all the three silkworm hybrids studied. The cocoon characters can be improved by providing ideal environmental conditions even during spinning stage of larvae affected with high temperature and RH. The study also suggest that high temperature and low humidity has greater effect during rearing stage than spinning stage.

인공사료의 White Ginseng 첨가가 누에의 성장에 미치는 영향 (Effect of Dietary White Ginseng on Larval Growth of Silkworm, Bombyx mori)

  • 인공사료육에 있어서 White ginseng이 누에의 성장에 미치는 영향을 알기 위해서, 애기누에 및 큰누에들을 4수준의 Ginseng extract를 함유한 인공사료로 사육하여 다음과 같은 결과를 얻었다. 1. Ginseng extract를 함유한 인공사료를 먹은 누에의 체중은 46.6 mg ginseng extract level, 139.9mg ginseng extract level, 279.8mg ginseng extract level의 순으로 무거웠다. 특히 누에의 체중증가는 46.6mg ginseng extract level에 있어서 대조에 있어서 보다 상당히 많았다. 따라서, 인공사료육에 있어서 알맞은 양의 White ginseng은 누에의 성장을 양호하게 하는 효과가 있다고 생각된다. 2. Ginseng extract를 함유한 인공사료를 먹은 누에의 혈당량은 함유하지 않은 인공사료를 먹은 누에보다 현저하게 감소하였다.

In vitro and in vivo Transient Expression in Insect Cells Mediated by the Cationic Liposome DDAB/DOPE

  • Cationic liposomes complexed with DNA have been extensively utilized for the delivery of reporter or therapeutic genes both in culture and in vivo. We investigated and determined the optimum conditions of a cationic liposome, composed of dimethyldioctadecy-lammonium bromide (DDAB) and dioleoyl phosphati-dylethanolamine UOPE), mediated a reporter plasmid expressing luciferase into insect cell lines (Sf-21 and Bm-N) and silkworm larvae. Together the data demonstrated that Bombyx mori nuclear polyhedrosis virus (BmNPV) genomic DNA (128 kb) was successfully transfected into Bm-5 cells using this liposome. These results suggest that DDAB/DOPE liposome will be useful as delivery agents for gene transfer to insect cells both in vitro and in vivo.

Improvement of Predictive and Corrective Inspection Methods to Control Nosema bombycis Infection in the Silkworm, Bombyx mori

  • The microsporidian infection with Nosema bombycis, reconfirmed its high virulence and transovarial tranmissibility, however, the characteristic symptom of the spots like pepper grains on the diseased larval skin was no more recognized by present varieties of the silkworm. Transovarial transmission rate detected from moth was above 90% in dead eggs or dead larvae in the rearing by mulberry leaves, 80% in the newly hatched larvae starved to death. Transovarially transmitted N. bombycis was easily observed from dead eggs and larvae, and were suggested an individual inspection of a few of dead eggs for detection of the pathogenic spores. The progeny population provided indicative factors on the sampling of predictive and corrective inspection. The higher concentration of N. bombycis spores included in the hindabdormal part of infected moth, applicative on the simple method of indivisual moth inspection. For the predictive inspection of growned 5th-instar larvae, N. bombycis infection was detectable without microscopic observation by the unique symptom of turbid milky-white spots on the silk gland. Inspection of the meconia artificially discharged from silkworm moth, was also succesful of microscopic observation before crossing, without killing or homogenize the moths. The results provided a basis of rational methods for the inspection of N. bombycis infection of the silkworm.

아세틸화와 숙시닐화한 번데기 농축단백질의 기능성 (Functional Properties of Acetylated and Succinylated Silkworm Larvae Protein Concentrates)

  • 번데기 농축단백질을 무수초산 및 호박산으로 숙시닐화 및 아세틸화시켜, 식품단백질원으로 이용가능성을 검토하기 위해 몇가지 중요한 기능성을 측정하여 다음과 같은 결과를 얻었다. 1. 번데기농축단백질이 아세틸화율은 82.0%, 숙시닐화율은 88.0%였으며, 아실화되는 과정에서 번데기 농축단백질은 밝은 상아빛으로 부드러웠으며, 특유한 냄새도 제거되었다. 2. 번데기 농축단백질은 pH 5.0에서 최소의 용해도를 나타내었으나 에세틸화 및 숙시닐화한 단백질은 pH 4.5에서 최소를 나타내었으며, 아실화에 의하여 용해도가 증가되었고 특히 숙시닐화는 약5배의 증가를 보였다. 3. 번데기 농축단백질의 겉보기 밀도와 수분흡수력은 아세틸화와 숙시닐화에 의해서 높은 증가를 나타내었다. 4. 번데기 농축단백질이 유화활성, 유화안정성, 유화용량은 아세틸화한 단백질에서는 유의성이 인정되지 않았으나 숙시닐화한 단백질은 높은 유의성을 나타내었다. 5. 번데기 농축단백질의 포말성은 숙시닐화한 단백질에서 30%의 증가 현상을 나타내었으며 포말안정성은 아세틸화 및 숙시닐화에 의해서 3시간, 8시간으로 향상되었다. 6. 각 농도별 점도는 아세틸화한 단백질에서는 별 영향이 없었으나 숙시닐화는 점도가 많이 증가되었으며 4∼5%에서 증가 현상이 가장 크게 나타났다.

1984년도 추잠기 잠병발생 실태조사 (Survey of Silkworm Disease Occurrence in Autumn Rearing Season, 1984)

  • 전국 621개 양잠시범부락의 84가을 누에병 발생율과 그 원인을 조사한 결과는 다음과 같다. 1. 누에 사육 기간중 누에병 발생율은 약 17%이었다. 2. 누에병 종류별 발생율은 농병(38.5%), 경화병 (20.3%), 연화병(17.9%) 및 쉬파리병 (9.4%)의 순이었다. 3. 누에병은 종잠기보다 장잠기 이후에 주로 발생하였다. 4. 누에병 발생의 주원인은 기상환경불량, 농약피해 및 소독불량이었고, 엽질불량, 사육기술부족 및 공해는 큰 문제가 되지 않았다.
