Propagation of Bell Pepper (Capsicum annuum var. grossum) through tissue culture

  • Leaf discs and apical meristems were cultured in Murashige and Skoog (MS) medium supplemented with cytokinin and auxin at different concentrations. Callus production was observed in all tested media after six days of incubation. Callus produced in the presence of high concentration of NAA (2.0mg/1) was fragile in texture and yellow in colour. Highest callus formation was observed from leaf discs in the medium supplemented with 1.0mg/1 NAA and 0.5 mg/l BAP in dark at $25{\pm}1{\circ}C$. Percentage of callus formation was 95% and mean callus fresh weight was 654.88 43.53 mg. Shoots were induced from the callus after 4 weeks in 1/2MS medium supplemented with BAP and kinetin both at 0.5mg/1. When elongated shoots were separated and transferred into multiplication medium (MS+0.5mg/1 BAP+0.5mg/1 kinetin) multiplication rate was 6.4 after 6 weeks. Higher concentrations of BAP caused callus production at the base. Direct shoot induction was observed from apical meristems in MS medium in the presence of 0.175 mg/1 IAA + 2.25mg/1 BAP and 0.175 mg/1 IAA + 3.0 mg/1 BAP in 16 hour day at $25{\pm}1{\circ}C$. Explants (apical meristems) elongated to form a single shoot forming a callus at the base. Adventitious buds were sprouted out from the base. Percentage explants which producing shoots was 28.57 and 65.5 respectively. Multiple shoot induction was also observed in the same media. Highest multiple shoot production was observed in the presence of 0.175 mg/l IAA and 3.0mg/l BAP, Mean number of shoots per explant was 5.36 and the mean shoot length was $16.66{\pm}4.15$mm. Shoots (20 30m length) were tested for root induction. Excised shoots were transferred into rooting media, which contains different concentrations of NAA and IAA. Best rooting performance was observed in 1/2MS medium supplemented with 0.1mg/1 NAA after 10 days of incubation in 16 hr photoperiod at $25{\pm}1{\circ}C$. Mean number of roots per shoot was 6 and the mean root length was 252mm. Rooted plantlets were transferred into sterile coir dust:sand (1:4) mixture and maintained in a humid chamber for two weeks, They were gradually exposed to the natural environment. After three weeks they were transferred to pots containing coir dust:sand (1:2) mixture for further development where the 90% survival was observed.

In vitro Multiplication and Corm Production of Freesia hybrida 'Sunny Gold'

  • Freesia has been an important worldwide cut flower because of its fragrance, long vase life and the wide color range of the flower. The conventional propagation methods by seeds and corms have many disadvantages such as shorter inflorescences with fewer numbers of florets, a reduction in cut flower quality and the accumulation of plant viruses in corms by successive cultivation. Therefore, the conventional propagation systems in Freesia needs to be replaced with tissue cultures to overcome the disadvantages. This study explored an efficient multiplication protocol using the combination of plant growth regulators (PGRs) for developed cultivar 'Sunny Gold'. The combination between 6-benzylaminopurin (BA) and α-naphthalene acetic acid (NAA) did not produce new shoots but developed enlarged roots. BA only treatments and the combination between BA and kinetin treatments were effective on shoot multiplication. The highest average number of shoots was 5.3 in the presence of 3 mg/L BA and 0.5 mg/L kinetin. To produce corms and cormlets, proliferated shoots were subcultured on 1/2 Murashige and Skoog (MS) medium supplemented with 90 g/L sucrose, 1 g/L charcoal and 7 g/L plant agar and placed at 4℃ in the dark for 6 months. The small size of corms and comlets were produced. The average number of regenerated comlets was 2.75 per shoot. The results showed that shoot multiplication is more efficient than cormlet regeneration for in vitro freesia proliferation.

담양권 대나무 이용 향토음식에 관한 연구 (A study on the redeveloping traditional foods with bamboo in Damyang)

  • Damyang is famous for the bamboo and various foods. It is urgently needed that native foods in this area are studied and modernized. Developing new foods and redeveloping local economy. Bamboo shoots can be raw materials for kimchi, canned goods, salted goods, Korean cookies, bread, tea, beverage, and so on. This study aims to invent new traditional foods using local product such as bamboo shoots will contribute to boost the recipe. to explore the new method of production and to modernize the traditional dishes. In the final analysis, economic and cultural consideration would be attached in this study.

두릅나무 전정에 따른 연차별 새순 생산성 비교 (Effect of Pruning on Annual Aralia elata Shoot Production)

  • 두릅나무는 두릅나무과에 속하는 낙엽활엽관목으로 전국에서 재배되고 있는 고소득 작목이다. 재배의 효율성 제고 및 소득 증대를 위하여 두릅나무의 새순 수확 후 전정 시기 및 연차별 전정에 따른 생산성을 분석하였다. 두릅나무 새순 수확 직후 6일 뒤부터 2주 간격으로 5회 전정하여 최적 시기를 판단하였다. 또한 전정 효과를 규명하기 위하여 정식 후 1년 차에 지상에서 30 cm 높이로 전정하였다. 정식 후 2년 차부터 전년도 발생한 가지를 10 cm 남기고 전정하였으며, 연차별 새순 및 가지 발생 특성을 비교하였다. 그 결과 새순 수확 종료 후 6일이 지난 뒤 처음 전정한 두릅나무의 가지 수가 다른 시기보다 1.3-2.1배 많았다. 새순 특성 또한 다른 시기보다 1.2-1.7배 우수하였으며, 전정이 빠를수록 품질이 좋은 많은 양의 새순을 수확할 수 있었다. 연차별 두릅나무의 전정을 통하여 정식 후 2년 차부터 고품질의 새순(직경 약 1.5 cm, 길이 약 16 cm)을 생산할 수 있었으며, 새순 수확이 가능한 가지 발생을 유도하여 생산성을 높일 수 있었다. 또한, 연차별 전정을 통하여 새순 수확이 쉬운 150 cm 정도의 수고를 유지 시킬 수 있었다. 그 결과 두릅나무의 전정 작업은 새순 수확 후 6일 전후로 빠른 시기에 시행하는 것이 효율적이다. 또한 전정하게 되면 매년 균일한 품질의 새순을 생산할 수 있고, 본당 수확량을 증가시킬 수 있다.

Plant Regeneration through Micropropagation from Nodal Explants of Critically Endangered and Endemic Plant Exacum travancoricum Bedd

  • A rapid micropropagation protocol was established for Exacum travancoricum Bedd. The effect of two cytokinins viz. BA and kinetin were studied to evaluate the propagation of plants through nodal explants. MS medium supplemented with 13.32 ${\mu}M$ BA induced early bud break and subsequent production of multiple shoots. Rooting of shoots occurred when cultured on 1/2 strength MS medium supplemented with 14.7 ${\mu}M$ IBA. Rooted plants were acclimatized to greenhouse conditions. The propagated plants were transferred successfully to field with 65% success. As the plant was amenable to propagation in vitro, this can be employed as a tool for conservation of this critically endangered and endemic ornamental herb.

생물반응기에서 현삼의 신초 형성과 발근 (Production and Rooting of Shoots in Bioreactor Culture of Scrophularia buergeriana Miquel)

  • 본 실험은 생물반응기를 이용한 현삼 종묘의 기내생산과 기내 발근에 관여하는 요인을 검토하여 추후 종묘의 건전한 생육과 안정적 생산에 활용할 수 있는 기초자료를 얻고자 실험한 결과는 다음과 같다. 1. 생물반응기에서 현삼 배양은 MS 기본배지의 $NH_4NO$ 양을 1/4 로 줄여 413mg/ l 의 농도로 처리하고 6주간 배양하는 것이 신초의 유도에 유리하였다. 2. 생물반응기 배양시 신초의 형성과 생장에는 $NH_4NO_3$를 413mg/ l로줄인 MS 배지가 유리하였다. 3. 배지량 1.5L 의 생물반응기에서 줄기 절편의 접종량이 300개 이상일 경우 기내 식물체가 과밀화 되어 신초의 생육이 저해되고, 생물반응기 내부 및 배지 위로 노출된 신초는 괴사되었다. 따라서 신초의 생체중이 무거우면서도 신초수를 많게 하기 위해서는 적정 접종량을 300개체로 하는것이 유리하였다. 4. 신초의 발근 수와 길이생장에는 IBA 0.05mg/ l 가 효과적이었다. 5. 생물반응기에서 6주간 배양하는 것이 신초의 생육이 정상적으로 되었으며 또한 이들 신초에서 발근과 뿌리의 발육에도 유리하였다.

In vitro micropropagation of two local taro cultivars for large-scale cultivation

  • The application of traditional taro propagation methods for large-scale cultivation would be insufficient to meet the high demand for quality planting materials. Therefore, this study aimed to develop an in vitro micro-propagation technique for two local taro cultivars (cv.), Wangi and Putih. Taro cormels were collected from the Malaysian Agricultural Research and Development Institute (MARDI) germplasm (Serdang, Malaysia). Explants were taken from the shoot tip of cormels and initially cultured on Murashige and Skoog (MS) basal media for four weeks. The explants were then transferred to different multiplication media supplemented with different types and concentrations of cytokinins such as 6-benzylaminopurine (BAP ) and Thidiazuron (TDZ). Shoot production was quantified after six weeks of culture. The highest mean number of new shoots was produced by the Wangi cultivar on MS medium supplemented with 2.0 mg/l BAP (2.10 shoots), MS medium supplemented with 0.5 mg/l TDZ (2.18 shoots), and Gamborg B5 medium supplemented with 6.0 mg/l BAP (2.43 shoots). The maximum average number of the Putih cultivar shoots was obtained on MS supplemented with 2.0 mg/l BAP (3.57 shoots). MS basal media was used for root initiation, as it produced an average of 25 roots with an 11-cm length. Various types of substrate mixtures were used during acclimatization. The best acclimatization substrate for the Wangi cultivar was 100% peat soil, whereas the Putih cultivar grew optimally in a combination of peat and perlites at a 1:1 ratio. Taro plantlets require approximately 4 to 6 weeks to acclimatize before they can be transferred to the field.

The apical bud as a novel explant for high-frequency in vitro plantlet regeneration of Perilla frutescens L. Britton

  • In this study, we established an in vitro regeneration system to maximize the recovery of leafy perilla (Perilla frutescens L. Britton) plantlets as part of developing a molecular biotechnology-based metabolic engineering program for this crop plant. Hypocotyl segments including the apical buds were used as explants for the direct production of shoots without an interim callus phase. The number of shoots produced from the apical buds peaked within 3-4 weeks, and the shoots were subsequently cultured on Murashige and Skoog (MS) media supplemented with 2 mg $1^{-1}$ benzylaminopurine (BA). Spontaneous rhizogenesis was observed after 7-10 days of culture on MS media without hormonal additives. The rooted shoots developed into normal plants in soil after hardening on distilled water for 3-4 days. The average plantlet regeneration frequency was higher for the apical buds (64.33%) than for the top (15.66%), middle (4%), and basal (1.33%) segments of the hypocotyls. This regeneration system demonstrates a capacity for high-frequency plantlet recovery and thus should be considered for use in the genetic manipulation of leafy perilla.

High Frequency Induction of Multiple Shoots from Nodal Explants of Vitex negundo L. Using Sodium Sulphate

  • The effect of sodium sulphate on shoot induction and multiple shoot formation from nodal explants of Vitex negundo L. was tested on Murashige and Skoog's (MS) medium fortified with different auxins, cytokinins and sucrose. Highest percentage $(97.78\%)$ of explants for shoot induction and multiple shoot (20.68/explant) production were observed in the combination treatment of $N^6-Benzyl$ adenine (BA) $(17.80\;{\mu}M/L)$, ${\alpha}-Naphthalene$ acetic acid (NAA) $(2.15\;{\mu}M/L)$ and $5\%$ sucrose supplemented with 100 mg/L sodium sulphate. In vitro raised shoots were rooted on the half-strength MS medium fortified with different concentrations of NAA, Indole-3-acetic acid (IAA), and Indole-3-butyric acid (IBA) alone and in combinations. Among the treatments, $4.90\;{\mu}M/L$ of IBA was found most effective $(95.56\%)$ in inducing roots. The rooted plantlets were shifted to glasshouse for acclimatization and later transferred to the field with cent percent survival. Furthermore, in vitro flowering was observed in the shoots cultured on MS medium supplemented with BA $(8.90\;{mu}M/L)$ and NAA $(1.61\;{\mu}M/L)$.

포도 캠벨얼리 품종의 2차지를 이용한 과실생산 (Berry Production Using Secondary Shoots in 'Campbell Early' Grapevines)

  • 포도 '캠벨얼리'에 있어서 2차지에 결실되도록 하기 위해서 만개후 13일부터 10일 간격으로 신초의 기부로부터 3, 6, 9마디의 눈을 각각 남기고 하계절단 처리를 한 결과, 액아 발아는 7~8일, 발아 후 개화까지는 19~25일 소요되었다. 하계 절단처리시기별 2차지의 화수수는 만개 후 13일 절단처리가 2.8개, 만개 후 23일 및 33일 절단처리가 3.2개로 차이가 없었다. 1차지의 기부에서 3번째 마디의 눈에서 발달한 2차지의 화수수 2.0~2.4개, 화수장 9.3~10.4cm 정도로 발달이 미약했으나, 6번째와 9번째 마디의 눈에서 발생한 2차지의 화수수 3.1~3.8, 화수장 12~14.9cm였고, 특히 6번째 마디의 눈에서 발생한 2차지에서 화수발달이 가장 충실하였다. 만개 후 13일에 절단 처리한 6, 9번째 눈에서 발생된 2차지의 수확기 과실형태 및 특성은 1차지의 과실과 차이가 없었으며, 나머지 만개 후 23일이나 33일 처리는 2차지의 과실 형태는 양호하였으나 성숙기간이 짧아 품질이 낮았다. 따라서 2차지의 과실 결실을 위한 하계 전정의 적기는 만개후 13일이고, 절단하는 마디의 위치는 1차지의 기부로부터 6번째 마디임이 구명되었다.
