Ecological Characteristics of Cobitis pacifica (Cobitidae) in the Yeongok Stream (연곡천에 서식하는 북방종개 Cobitis pacifica (Cobitidae)의 생태적 특성)

  • Ecological characteristics of Cobitis pacifica inhabited in the Yeongok Stream, Gangwon-do, Korea was investigated from May 2006 to August 2007. The major habitat was composed of sand bottom with current velocities of $0.23{\sim}0.56m\;sec^{-1}$ and depths of $20{\sim}50cm$. The sex ratio of female to male was 1 : 0.93. It was presumed that the spawning season was the period of water temperatures of $25{\sim}27^{\circ}C$. Sexually matured individuals appeared over the two years of age. Frequency analysis of body length indicated that the group with $40{\sim}59mm$ in body length was one-year-old, the group of $60{\sim}89mm$ was two-years-old, the group of $90{\sim}109mm$ was three-years-old, and the group of over 110 mm was four-years-old. The average number of eggs in the ovaries was estimated as 809 per matured female. Main prey items were Ephemeroptera and Diptera.

  • The morphogenetically matured spermatozoa (sperm) are generated in the testes by the spermatogenesis. They travel male reproductive tract with many substances secreted from the accessory reproductive organs. One of the substances is the semenogelin (SEMG) released from the seminal vesicles that is involved in the post-testicular maturation. The expression of SEMG gene was investigated in seminal vesicle tissues of sexually matured and regressed male Syrian hamsters by reverse transcription polymerase chain reaction (RT-PCR). The SEMG gene was uniquely identified in the seminal vesicles of the matured Syrian hamsters and compared to the genes reported previously. But the expression of SEMG gene was not observed in reproductively and completely regressed testes of Syrian hamsters. These results indicate that the expressions of the SEMG gene are related to the reproductive capability in the male Syrian hamsters.

  • The population ecology (Ecological characteristics) of Squalidus chankaensis tsuchigae were investigated at Geum River from March to November, 2013. This species inhabited midstream of the river where was mostly covered with cobble and sand. The depth of the habitat was 30~60 cm and velocity of stream was 2.27~4.88 cm/sec. The sex ratio of female to male was 1 : 0.86. They were sexually mature when they were over two years of age. The spawning period was from June to July with water temperatures from $25^{\circ}C$ to $25^{\circ}C$. Sexually matured individuals appeared over the two year of age. Frequency analysis of total length indicated that the group with 55~70 mm in total length was one year old, the 71~90 mm was two years old, and the group over 91 mm was three years old. The average number of eggs in the ovaries was estimated as 2,219 per matured female. The number of matured eggs size in ovaries was 0.64~0.98 (mean: 0.85) mm. Calulation length-weight relation of S. chankaensis tsuchigae show the value of contant a as 0.0002 and b as 2.81, and condition factor (K) was 1.06 on average. The stomach contents were Cyanophyta, Chlorophyta, Bacillariophyta, Crustaceae, Copepoda, Nematota, Actithocephala, and Ephemeroptera, Hemiptera, Diptera, and Trichoptera that belong to Aquatic insect, and dormant bud of Pectinatella that belongs to Phylactolaemata. The diet of S. chankaensis tsuchigae was omnivorous, mainly vegetable feed on attached algae (filament form), and mainly animality feeded on benthic invertebrates such as Chironomidae larvae.

Glycogen distribution of germ cells and Sertoli cells of seminiferous tubules in Jindo dog (진도견 정세관의 정세포와 Sertoli 세포내 glycogen의 분포)

  • In an effort to obtain basic data of carbohydrate metabolism during spermiogenesis of the sexually-matured Jindo dog, the glycogen distribution in the testis was investigated by light and transmission electron microscopy. Periodic acid thiocarbohydrazide silver proteinate physical development(PA-TCH-SP-PD) staining method provided better results in the detection of glycogen granules from Sertoli cells and germ cells than the periodic acid schiff(PAS) staining method did. Pre-treatment of the tissue sections with ${\alpha}$-amylase elicited a significant decrease in PA-TCH-SP-PD stained granules, which suggested that the stained granules were of glycogen origin. High concentration of the glycogen granules were observed in the Sertoli cells, especially in its column, sheet-like processes, club-like processes, and tubular processes. The glycogen granules were unevenly distributed in some Sertoli cell columns. These results strongly indicated that the Sertoli cells of Jindo dogs showed vigorous activity of carbohydrate metabolism.

  • Ecological characteristics of Orthrias nudus from the Eoron Stream, Gangwon-do, Korea were investigated from January to December 2009. O. nudus inhabits mainly riffles of the upstream and mid-upstream stretches of Eoron Stream, which were composed of boulder and cobble bottoms. The current velocities in these habitats were 0.42~1.34 m/sec and depths were 15~46 cm. The sex ratio of female to male was 1 : 0.98. It is presumed that the spawning season is from April to June when water temperatures were $15{\sim}19.2^{\circ}C$. Sexually mature individuals appeared to be over one year of age. Frequency analysis of total length indicated that the group 58~99 mm in total length is represented by one-year-old individuals, and the group over 100 mm by two-year-old individuals. The average number of eggs in the ovaries was estimated as 4,119 per mature female. Stomach contents of O. nudus were composed of aquatic insects such as Ephemeroptera, Trichoptera and Diptera.

Measurement of the Rate of Protein Synthesis in Chickens by HPLC/MS

  • The fractional synthesis rates(FSR) were measured with 2l-wk and 3l-wk-old broiler breeder pullets and hens to investigate the effect of sexual maturity on FSR. The FSR were obtained from chicken tissues and blood samples using High-Performance Liquid Chromatography/Mass Spectrometry(HPLC/MS). A L-l-13C, 15N -leucine saline solution was infused by bolus injection as a tracer into broiler breeder pullets in the experiment. A rapid HPLC/MS method was developed to measure the isotopic enrichments of leucine in plasma, tissue samples, and eggs. The enrichments of stable isotope leucine incorporated into protein and the enrichments of the stable isotope free leucine were measured in liver, breast muscle and blood samples. Two sets of experiments were conducted. In experiment one, 2l-wk-old, sexually immature broiler breeder pullets were divided into groups of three and blood samples were collected at 20 or 30 min intervals until 1.5 h from initial injection. The pullets were sacrificed in groups of three at varying time intervals for 7 h after injection. The liver, breast muscle and blood samples were removed for analysis. The FSR were estimated to be 8.7l%/day for liver, 4.06%/day for breast muscle, and 5.08%/day for blood samples in 30 minutes after injection from the enrichment ratios. In experiment two, sexually matured 3l-wk-old broiler breeder hens were assorted into groups of three and blood samples were obtained at 20 or 30 min intervals for 2 h. The FSR for blood samples were determined. The broiler breeder hens were sacrificed in groups of three at various time intervals until 7 h after injection and liver, breast muscle and blood samples were removed for analysis. The FSR were calculated to be 5.96%/day for liver. Eggs were collected from five chickens daily for 10 days after large bolus injection. The average of total enrichments of stable isotope in egg albumin was increased by 0.064% at 4 days after injection and was back to normal in 7 days.

  • Population ecology (Ecological characteristics) of Squalidus japonicus coreanus from the Namhan River(Yangpyeong-gun Yangpyeong-eup Changde-ri), Gyeonggi-do, Korea were investigated from January to December 2011. Short barbel gudgeon inhabits habitats run of the lower river. The stream bed was mostly covered sand. Their current velocities were 0.14~1.38m/sec and depths were 80~120cm. The sex ratio of female to male was 1 : 0.82. It is presumed that the spawning season is from June to July and water temperatures were $18.5{\sim}24.3^{\circ}C$. Sexually matured individuals appeared over the two year of age. Frequency analysis of total length indicated that the group with 34~50mm in total length was one year old, the 51~74mm was two years old, and the group over 75mm was three years old. The average number of eggs in the ovaries was estimated as 1,871 per matured female. Stomach content of S. japonicus coreanus were Cyanophyta, Chlorophyta, Bacillariophyta, Rotatoria, Crustaceae, Copepoda, Ephemeroptera, Diptera, and Nematoda. omnivorous, mainly feeds on benthic invertebrates such as chironomidae larvae.

Histological Studies on Gonad and Germ Cell Development of Diploid and Triploid Mud Loach (Misgurnus mizolepis) (2배체와 3배체 미꾸라지(Misgurnus mizolepis)의 생식소 발달)

  • A Study was conducted by using a histological method to examine gonadal development in diploid and triploid mud loach, Misgurnus mizolepis. Shape of ovaries from both groups looked like a single mass however, size of triploid ovary was significantly smaller than that of diploid ovary. Testis of triploid was also similar that of diploid in shape, but it was smaller than that of diploid. Ovarian development and oogenesis in diploid were significantly more rapid than those process in triploid, and first matured eggs were observed 100-days after hatching (SL, 5.68cm). Triploid ovary from hatching to 9-month-old had external appearance of undeveloped gonad and a few of oocytes of perinucleolus stage. However, normal matured eggs like those of female diploid were observed in one 5year-old triploid fish ovary examined. Although testicular development and sperrnatogenesis of diploid were prosperous sexually, testicular development of triploid were rather retarded compare to their male diploid counterpart in spermatogenesis.

Spawning Ecology of Sarcocheilichthys variegatus wakiyae (Cyprinidae) in Jicheon Stream, Korea (지천에 서식하는 참중고기 Sarcocheilichthys variegatus wakiyae (Cyprinidae)의 산란생태)

  • The study on the spawning ecology of Sarcocheilichthys variegatus wakiyae at Jicheon stream, Korea was investigated from March 2016 to February, 2017. The sex ratio of male to female was 1 : 0.97 and there was no significant difference. The spawning season was from the end of April to the mid-July, and the peak of spawning period was from the beginning of May to the end of June. The average water temperature at this time was $18^{\circ}C$. According to frequency analysis of total length, it is presumed that the group with a total length of less than 51~70 mm was one year old, the 71~100 mm was two years old, and the group over 101 mm was three years old. They sexually matured when they were over two years. The average number of eggs in the ovaries was estimated as $428{\pm}194per$ matured female. The mature eggs were yellowish, oval, and had a mean diameter of $2.38{\pm}0.25mm$ and a short diameter of $1.98{\pm}0.20mm$.

Sexually Matured Female Mice showed Higher γ-Linolenic Acid (GLA) in the Muscle Tissue

  • The sexual maturation occurred by the changes of steroid hormones was known to sex-dependently and/or age-dependently regulate the lipid metabolism in various animal species. Our current study demonstrates that lipid and its functional fatty acids can be changed depending on the status of sexual maturation. Of the functional fatty acids, ${\gamma}$-linolenic acid (GLA; 18:3n-6) is an important factor for maintaining human health. The purpose of our study was to investigate the level of GLA in mice with different stages of sexual maturation. To this end, the longissimus muscle (LM) of immature (3-week-old) and mature (7-week-old) female mice was analysed for the fatty acid composition by gas chromatography. Furthermore, both gene and protein level of ${\Delta}6$ desaturase (FADS2) which is involved in GLA metabolism by real time PCR and Western blotting, respectively. Mature females showed greater (P<0.05) serum $17{\beta}$-estradiol (E2) level and LM GLA contents than immature group. The mRNA and protein levels of FADS2, which converts precursor linoleic acid into GLA, were higher (P<0.05) in mature female mice than in immature mice. In conclusion, these results show that sexual maturation of female mice induces GLA and FADS2 contents in LM.
