The herb, Cajanus indicus L, is well known for its hepatoprotective action. A 43 kD protein has been isolated, purified and partially sequenced from the leaves of this herb. A number of in vivo and in vitro studies carried out in our laboratory suggest that this protein might be a major component responsible for the hepatoprotective action of the herb. Our successive studies have been designed to evaluate the potential efficacy of this protein in protecting the hepatic as well as renal tissues from the sodium fluoride (NaF) induced oxidative stress. The experimental groups of mice were exposed to NaF at a dose of 600 ppm through drinking water for one week. This exposure significantly altered the activities of the antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase (GST), glutathione reductase (GR) and the cellular metabolites such as reduced glutathione (GSH), oxidized glutathione (GSSG), total thiols, lipid peroxidation end products in liver and kidney compared to the normal mice. Intraperitoneal administration of the protein at a dose of 2 mg/kg body weight for seven days followed by NaF treatment (600 ppm for next seven days) normalized the activities of the hepato-renal antioxidant enzymes, the level of cellular metabolites and lipid peroxidation end products. Post treatment with the protein for four days showed that it could help recovering the damages after NaF administration. Time-course study suggests that the protein could stimulate the recovery of both the organs faster than natural process. Effects of a known antioxidant, vitamin E, and a non-relevant protein, bovine serum albumin (BSA) have been included in the study to validate the experimental data. Combining all, result suggests that NaF could induce severe oxidative stress both in the liver and kidney tissues in mice and the protein possessed the ability to attenuate that hepato-renal toxic effect of NaF probably via its antioxidant activity.

A cell-based assay system for monitoring NF-$textsc{k}$B activity was developed to determine the influence of activated NF-$textsc{k}$B in human HaCaT cells. The pNF-$textsc{k}$B-SEAP-NPT plasmid that permits expression of the secreted alkaline phosphatase (SEAP) reported gene in response to the NF-$textsc{k}$B activity and contains neomycin phosphotransferase (NPT) gene for the geneticin resistance in host cells was constructed and transfected into human keratinocyte cell line HaCaT. Human HaCaT transfectant cells secreted the SEAP enzyme into the culture medium in a time-dependent manner until 72h. NF-$textsc{k}$B activities were measured in the SEAP reporter gene assay using a fluorescent detection method. The treatment of HaCaT cell transfectants with known antioxidants [e.g., N-acetyl-L-cysteine and vitamin C] showed inhibition of NF-$textsc{k}$B activity in a time-and concentration-dependent manner. The phorbol 12-myristate 13-acetate (PMA) known as a stimulator of NF-$textsc{k}$B expression demonstrated that it increased NF-$textsc{k}$B activity in a time- and concentration-dependent manner. This assay system could be used to determine the quantitative measurement of NF-$textsc{k}$B activity in the human skin and allow the screening of anti-inflammatory agents from various synthetic chemicals and natural products for dermatological purpose. Hand, foot, and mouth disease (HFMD) is an acute, mostly self-limiting infection. Patients usually recover without any sequelae. However, a few cases are life threatening, especially those caused by enterovirus 71 (EV71). A 12-month-old boy was admitted to a primary hospital with high fever and vesicular lesions of the mouth, hands, and feet. After 3 days, he experienced 3 seizure episodes and was referred to our hospital. On admission, he was conscious and his chest radiograph was normal. However, 6 hours later, he suddenly lost consciousness and had developed a massive pulmonary hemorrhage that continued until his death. He experienced several more intermittent seizures, and diffuse infiltration of both lung fields was observed on chest radiography. Intravenous immunoglobulin, dexamethasone, cefotaxime, leukocyte-depleted red blood cells, fresh frozen plasma, inotropics, vitamin K, and endotracheal epinephrine were administered. The patient died 9 hours after intubation, within 3 days from fever onset. EV71 subgenotype C4a was isolated retrospectively from serum and nasopharyngeal swab by real-time reverse transcription-polymerase chain reaction. Here, we report a fatal case of EV71-associated HFMD with sudden-onset massive pulmonary hemorrhage and suspected encephalitis.

Cosmetics are primarily applied to the skin; therefore, the association of cosmetic dyes with skin diseases or inflammation is a topic of great interest. Thymic stromal lymphopoietin (TSLP) is an interleukin 7-like cytokine that activates dendritic cells to promote Th2 inflammatory immune responses. TSLP is highly expressed in keratinocytes under inflammatory conditions, which suggests that it may play a critical role in the development of skin diseases, such as atopic dermatitis. Therefore, we investigated whether cosmetic dyes influenced the production of TSLP by keratinocytes. Phloxine O, also known as D&C Red No.27, is one of the most common red synthetic pigments and is widely used in colored cosmetics. Our results showed that Phloxine O downregulated phorbol 12-myristate 13-acetate-induced production of TSLP in a murine keratinocyte cell line (PAM212). Phloxine O also suppressed TSLP expression in KCMH-1 cells, which are mouse keratinocytes that constitutively produce high levels of TSLP. To investigate the in vivo effects of Phloxine O, we induced TSLP expression in mouse ear skin by topically applying MC903, a vitamin D3 analogue that is a well-known inducer of atopic dermatitis-like symptoms. Topical application of Phloxine O prevented MC903-induced TSLP production in mouse ear skin, attenuated the acute dermatitis-like symptoms and decreased serum IgE and histamine levels in mice. Suppression of TSLP expression by Phloxine O correlated with reduced expression of OX40 ligand and Th2 cytokines in mouse ear skin. Our results showed that Phloxine O may be beneficial to prevent dermatitis by suppressing the expression of TSLP and Th2 cytokines in skin.
This study was conducted to investigate the anthropometric data, bone density, serum profiles, nutrient intakes and diet quality of premenopausal and postmenopausal women. For the study, we obtained data for analysis from the combined 2008~2011 Korean National Health and Nutrition Examination Survey (KNHANES). The height and weight were 157.8 cm and 58.7 kg in premenopausal women and 155.5 cm and 58.3 kg in postmenopausal women, respectively. The obesity rate was 27.49% in premenopausal women and 34.98% in postmenopausal women (p<0.001). Total cholesterol, LDL-cholesterol, triglyceride, GOT, GPT and alkaline phosphatase in postmenopausal women were significantly higher than those in premenopausal women (p<0.001). The prevalence of osteoporosis was 0.0~0.89% in premenopausal women and 0.48~13.22% in postmenopausal women (p<0.001). In postmenopausal women, rates of hypertension, stroke, myocardial infarction, depression, and diabetes were significantly higher than those in premenopausal women. Water, fat. ash, sodium, retinol, thiamin, riboflavin and niacin intakes in premenopausal women were significantly higher than those in postmenopausal women. Water, fiber, Ca, and, K intakes were below KDRIs (Dietary Reference Intakes for Koreans) in both groups. The mean adequacy ratio (MAR) of premenopausal women was higher than that of postmenopausal women (p<0.001). The index of nutritional quality (INQ) in premenopausal women was also higher than that of postmenopausal women except iron and vitamin C. Therefore dietary guidelines and an education program should be developed for desirable improvement of health, bone density, nutrient status and dietary quality of postmenopausal women.
Literature suggests that iron deficiency anemia is prevalent among pregnant women all over the world. This study was designed to evaluate the iron status of pregnant women during the fist five months, with the intention of determining ways to reduce the prevalence of iron deficiency anemia among pregnant women in Korea. We collected dietary information and measured the biochemical status of iron in 171 pregnant women over 16 weeks of gestation (=16.7$\pm$2.34 week). Dietary intakes for 3 days were collected using the 24 hour recall and food record methods. The daily Fe intake was measured using the food frequency method. The Body Mass Index (BMI), calculated by using the pre-pregnancy weight and height, indicated that 31.3% of subjects were under-weight. We divided the subjects into normal and anemic group by using the serum ferritin levels. It appeared that the mean dietary intake of iron was 52.3% of the recommended level for pregnant women. The dietary quality evaluation showed that pregnant women ate only 58% of the recommended amount in the food groups of meat, fish, eggs, beans and milk and dairy products. The hematological indices showed that the mean Hemoglobin (Hb) was 11.9 g/dl, Hematocrit (Hct) was 35.1%, ferritin was 23.9 $ng/ml$, and transferrin was 297.3 ${\mu}g/dl$. The dietary intake of iron was significantly lower and the vitamin C intake was significantly higher in the anemic group. The pre-pregnancy BMI was significantly lower in the anemic group. Variables affecting iron intake were the Fe intake frequency index and the food group score. The Fe index showed significantly positive correlation with the pre-pregnancy food intake and the food group score. Hb showed a significantly positive correlation with the pre-pregnancy food intake. We concluded that strategies to improve iron status be implemented in the pre-pregnant stage so as to reduce the prevalence of iron deficiency and that we should stress on the importance of an adequate diet as well as the maintenance of a heathy weight.
This study was conducted to compare the nutrient intake, physical characteristics, and health status according to daily fruits and vegetables intake of adolescents using the Korean national health and nutrition examination survey for the sixth period (2013~2015). Subjects were 1,096 adolescents aged 13~18 years. The average daily intake of fruits and vegetables was $518.49{\pm}4.94g$. The two groups (insufficient, sufficient) were divided by fruits and vegetables intake (<500 g/day, ${\geq}500g/day$) and the ratio of the insufficient group was 75.9% while that of the adequate group was 24.1%. The mean of intake of energy, calcium, phosphorus, sodium, potassium, iron, thiamin, niacin, vitamin C and dietary fiber differed significantly between the insufficient and sufficient groups (P<0.01, P<0.001). However, there was no significant difference in physical characteristics between groups. The self-perceived health status was better in the sufficient group (P<0.05), while the insufficient group was aware of more stress in their daily life (P<0.05). The serum triglyceride was significantly higher in the sufficient group (P<0.05), while the total cholesterol was lower (P<0.05). As the characteristics of fruits and vegetables are different, it will be necessary to determine standards for consumption of each type. It is also necessary to consider whether a fruits and vegetables intake of 500 g would be a reasonable standard for adolescents. Nevertheless, the results of this study indicate efforts to increase fruits and vegetables intake by young people are necessary.
Objective: The purpose of this study was to identify dietary factors related to infertility in Korean women through a case-control study. Methods: The case group was composed of 236 women who had been diagnosed as infertility in hospital. The control group of 181 healthy women with children were recruited from local immunization centers. Socio-economic status, medical history, dietary intakes using food frequency questionnaire and stress were surveyed by interview. Anthropometric measurements were made and the causes of infertility were identified through medical records. Fasting blood samples were taken from subgroup of the subjects. Results: The mean age of infertile and control groups was 31.1 and 32.4 years, respectively and the difference was statistically significant. The mean Body Mass Index of infertile women was not significantly different from control women, however, Waist/ Hip Ratio and Triceps Skinfolds Thickness were significantly lower in infertile women than in control women. The dietary intake status was generally satisfactory in both groups. The intakes of energy, protein, fat, carbohydrate, retinol, vitamin B2 and niacin were lower in infertile women than in control women. The infertile women also showed lower intakes of animal foods. No differences were found between two groups in serum concentrations of albumin, hemoglobin, Fe, TIBC, total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, C3, IgA, IL-2, however, infertile women showed higher levels of Zn and IgG. The stress score was higher in infertile women. Conclusions: From the results of this study, dietary factors and nutritional status do not seem to be directly related to infertility. However, the intertile women have lower nutrient intake and lower body fat content than control women. Further researches are needed according to the causes of infertility for long term to establish the relationship between dietary factors and infertility.
To increase both iron and enhancers for iron absorption through diets should be a basic strategy to sufficiently provide increased iron for pregnancy. Previous studies reported that iron intakes of Korean pregnant women were short and their iron status deteriorated as pregnancy progressed. However, there is little data about the bioavailability of dietary iron during pregnancy. Therefore, this study was conducted to determine the changes of dietary iron intakes, its bioavailability and iron status during pregnancy longitudinally in Korean women. A total of 151 pregnant women in their first trimester of pregnancy voluntarily participated. Among them, 72 women finished the research protocol during the second trimester and 55 did it during the third trimester. Dietary intakes of total iron, both non-heme and heme iron, as well as enhancers, both MPF (meat, poulty, and fish) and vitamin C, increased significantly as pregnancy progressed. As the results, bioavailability of dietary iron and iron absorbed increased significantly as pregnancy progressed. However, the amount of iron absorbed at each trimester did not meet considerably the iron needed during pregnancy. All five indices examined in the study, Hb level, Hct, serum concentrations of ferritin and sTfR (soluble transferrin receptor), and sTfR: ferritin ratio, showed that iron status of the subjects deteriorated as pregnancy progressed. The rate of anemia of the subjects increased as pregnancy progressed although more than 80% of the subjects took iron supplements after the 20th week of pregnancy. These results imply that it is needed to provide more iron especially, heme iron and dietary enhancers to prevent the deterioration of iron status during pregnancy. Future research on bioavailability of supplemental iron should be performed to determine the iron balance precisely.
This study was intended to investigate the nutritional status between lactating and non- lactating women, especially calcium and iron. The subjects were 84 lactating women and 20 non-lactating women visiting a public health center and hospital in Daegu. Each subject was interviewed to collect the information on dietary intake for 2 consecutive days. Biochemical assessment of iron status and bone mineral density (BMD) measurement were conducted. Dietary intake of carbohydrate, potassium, Vit $B_1,\;B_2$, Vit C were significantly higher in women during lactating period (p<0.05). However, relative intake as expressed by percentage of Korean Recommended Dietary Allowances (RDA) was not significantly different between the two groups. The dietary intake of iron and calcium were 58.8%, 60.4% of Korean RDA respectively in women during lactating period. The current food habit score of these women was significantly higher than that of non-lactating women (p<0.05). When we compared the quality of nutritional status, the Index of nutritional quality (INQ) was significantly higher for vitamin $B_2$, P in lactating women than in non-lactating women (p<0.1). Mean adequacy ratio (MAR) was not significantly different between two groups. Dietary variety score (DVS) was significantly higher in women during the lactating period (p<0.05). There was no significant difference in biomarkers (Hb, Hct, Serum ferritin, Transferrin) related to iron status between the two groups. No significant difference in bone mineral density (BMD) T-score was not observed. However, it appeared that BMD of lactating women was lower than that of non-lactating women.
