  • To investigate the effects of Ixeris sonchifolia Hance diets on serum and hepatic lipid levles and enzyme activities in rats administered with ethanol chronically, Sprague-Dawley male rats were AIN-76 diet(control), control diet plus ethanol, control plus Ixeris sonchifolia Hance diet, or control plus Ixeris sonchifolia Hance diet plus ethanol for 30 days. Ixeris sonchifolia Hance diets significantly decreased the serum total cholesterol, triglyceride, LDL-cholesterol, and GOT levels that were increased due to the chronic ethanol administration. In addition, Ixeris sonchifolia Hance diets significantly decreased the liver triglyceride and total lipid levels that were increased due to the ethanol administration. The present findings, combined with previous data showing differences in the effects of cabbage diets having a high or a low level of GABA on the lipid levels and the serum Y-GPT activity of rats(Cha and OH[2000] J. Korean Soc. Food Sci. Nutr. 29, 500-505) raise the possibility that GABA in plants could have a nytraceutical role in the recovery of chronic alcohol-related diseases. • In this study, we analyzed the biochemical factors in lotus (Nelumbo nucifera) leaf, stem, and yeonjabang and their effects on serum factor levels in mice fed a high-fat (HF) diet. The loutus leaf showed $9.47{\pm}0.30%$ moisture content, $8.25{\pm}0.39%$ ash, $21.45{\pm}1.25%$ crude protein, and $2.21{\pm}0.13%$ crude fat content; the lotus stem showed $11.84{\pm}0.43%$ moisture, $10.21{\pm}0.64%$ ash, $17.55{\pm}0.92%$ crude protein, and $4.16{\pm}0.23%$ crude fat content; and the lotus yeonjabang showed $11.86{\pm}0.50%$ moisture, $6.81{\pm}0.51%$ ash, $18.71{\pm}1.02%$ crude protein, and $3.95{\pm}0.15%$ crude fat. Blood triglyceride levels were higher in the HF group ($146.43{\pm}38.81mg/dL$), and lower in the HF+yeonjabang groups ($98.00{\pm}17.18mg/dL$). In particular, blood triglyceride levels were significantly lower in the groups that had 10% dry yeonjabang powder added to the high-fat diet. The inclusion of excessive high-fat diet increased concentrations of serum insulin and leptin. Serum leptin concentrations were highest in the HF group mice ($3.00{\pm}1.35ng/dL$), whereas they were significantly lower in the HF+yeonjabang groups by $1.34{\pm}0.52ng/dL$ (p<0.05). Thus, addition of dry yeonjabang powder to the high-fat diet was more effective in regulating the levels of serum triglycerides and leptin in mice. Additional studies would help in the development of yeonjabang as a functional food.

  • This study was carried out to examine the effects of alcohol on serum lipid levels. Subjects of this study were 50 military officers living in Honsung, Korea, 25 of whom consumed alcohol on a regular basis and 25 of whom did not. All subjects were previously informed of the purpose of the study and the methods of food intake reporting and blood testing. The age distribution range of subjects was 30-49 years. Weight, height, and body mass index(BMI) were not significantly different between the two groups. Blood pressure was compared, and it was found that the drinker group had mean systolic(SBP) and diastolic blood pressures(DBP) 10mmHg higher than the non-drinker group. Nutrient intake levels of the two groups were not significantly different and were adequate with respect to the recommended dietary allowances. It was found that the drinker group ingested more calories, protein, fat and vitamn B1 than the non-drinker group, while the non-drinker group ingested more carbohydrate, calcium, iron, vitamin A, niacin and ascorbic acid. These differences were believed caused by increased caloric intake from alcohol and additional intake of meat served with drinks for the drinking group. Fourteen of the twenty-five drinkers(56%) had drank regularly for five to ten years. Most drinkers drank 2-3 times per week, with alcohol intake less than two bottles(720ml)of distilled liquor(Soju) at a time. Meat was often served with alcohol as reported by 72% of the drinker group, and was believed to contribute to raised intake of calories, protein and fat in this group. Serum lipid levels of both groups were classified as normal, but the components of total cholesterol, triglycerides were significantly higher in drinkers than non-drinkers. The correlation between triglyceride levels and alcohol intake at one sitting was shown to be significantly positive(r=0.57, p<0.01), so it was believed that triglyceride elevation was associated more with heavy drinking than light drinking. We can conclude that if alcohol is consumed with a normal diet, then serum cholesterol, triglyceride and total lipid levels are expected to increase.

  • This study was undertaken to investigate the effect of Cassia tora ethanol extract on the lipid levels in serum and liver of rats fed high cholesterol diet. Experimental rats were divided into the following 4 groups: normal diet group, high cholesterol diet group, high choleslerol-0.25% C. tora ethanol extract group and high cholesterol-0.5% C. tora ethanol extract group. After 4 weeks, rats were sacrificed and analyzed the serum lipid profiles, activities of serum alanine aminotransferase (AST), aspartate aminotransferase (ALT), hepatic glucose-6-phosphate dehydrogenase (G6PDH) and malic enzyme (ME). It was also determined the contents of total cholesterol, triglyceride and thiobarbituric acid reactive substances (TBARS) in liver. There was no difference in weight gains between experimental groups. The concentrations of serum total cholesterol, free cholesterol, triglyceride and free fatty acid were tended to be decreased in C. tora groups compared with control group. HDL-cholesterol concentration was significantly decreased in high cholesterol diet group and slightly increased by C. tora ethanol extract feeding. The contents of liver cholesterol and triglyceride were higher in high cholesterol diet group than normal group, but significantly decreased by feeding of C. tora ethanol extract. Supplementation of 0.5% C. tora extract decreased significantly the activities of hepatic G6PDH and ME. Activities of serum AST, ALT and contents of liver TBARS were tended to be increased with high cholesterol diet and reduced by C. tora ethanol extract supplementation but had not significance. These results suggest that C. tora ethanol extract may exert a lipid lowering effect in serum and liver of rats.

  • The purpose of this study was to investigate the effects of diet composition on serum leptin and lipids profiles in rats. At the baseline, seven 4-week-old Sprague-Dawley rats, male, were sacrificed and the remaining rats were divided into five groups and each group was fed one of the following five dietary regiments for 2 weeks and 6 weeks;the control diet AIN76(CAL, n=2l), high-carbonhydrate(rice)(HCR, n=2l), high-carbohydrate(flour)(HCF, n=2l), high-fat(corn oil)(HFO, n=2l), high-fat(beef tallow)(HFB, n=2l). Serum leptin was determined by a double antibody ELISA assay at the baseline(n=7), 6 week(n=35) and 10 week of age(n=70). At 6 weeks of age, the increase in the Food Efficiency Ratio(FER) was related to adipocyte hyperplasia in rats on HFB diets. The serum triglyceride, total cholesterol, and LDL-cholesterol increased significantly in HFB group, and decreased in HFO group compared to control group. The HFC group showed significant increase in serum triglyceride level compared with control group. After 2 weeks and 6 weeks, noticeably high increases in epididymal adipose tissue fat cell mass and numbers were observed with the HFB fed group. Serum leptin levels increased as body weight increased over the period of time(4weeks; 1.50$\pm$0.13ng/ml versus 10weeks; 2.08$\pm$-.13ng/ml). And this result shows that there are 193% higher in rats fed high fat-beef tallow diet than the control diet. Serum leptin levels of the HFB group(4.01$\pm$0.39mg/ml) were significantly higher than that of the HFO(2.06$\pm$0.5613ng/ml), CAL(2.08$\pm$0.1313ng/ml), HCR(2.41$\pm$0.2113ng/ml) and HCF(2.80$\pm$0.4713ng/ml) at p<0.05. The serum leptin concentration was positively correlated with the amount of epididymal fat pads(r=0.47 p<0.01), serum triglyceride(r=0.49, p<0.001), tatal cholesterol(r=0.48, p<0.001), LDL-cholesterol(r=0.58, p<0.001), atherogenic index(r=0.67, p<0.001), and inversely correlated with HDL-cholesterol(r=-0.65, p<0.001). In conclusion, the changes in composition of dietary fat and carbohydrate intake could affect changes in concentration of serum lipids and leptin. Especially, the high-fat diet with animal fat source could increase circulating leptin level. (Korean J Nutrition 34(2) : 123-131, 2001)

  • In order to investigate the protective effect of Samduhaejungtang on the liver injury of rats induced by $CCl_4$ and d-galactosamine, the serum transaminase(GOT& GPT), alkaline phosphatase(ALP), lactic dehydrogenase(LDH) for enzyme activities and triglyceride, total bilirubin amounts for serum component were measured. All animals were divided into 4 groups, those were normal group(untreated), control group(treated with vehicle 0.9% Saline solution), sample Ⅰ group(1000mg/kg administrated), sample Ⅱ group(2000mg/kg administrated). The results were as follows : 1. The inhibitory effects of the serum GOT activities in rats induced by $CCl_4$ were noted in both sample Ⅰ group(p<0.01) and sample Ⅱ group(p<0.001). In serum GPT activities, sample Ⅱ group(p<0.05) only showed the inhibitory effects. 2. The inhibitory effects of the serum ALP activities in rats induced by $CCl_4$ were noted in both sample Ⅰ group(p<0.01) and sample Ⅱ group(p<0.001). 3. The inhibitory effects of the serum LDH activities in rats induced by $CCl_4$ were noted in sample Ⅱ group(p<0.05). 4. The increases of the serum triglyceride content level in rats induced by $CCl_4$ were inhibited in both sample Ⅰ group(p<0.01) and sample Ⅱ group(p<0.001). 5. The increases of the serum total bilirubin content level in rats induced by $CCl_4$ were inhibited in both sample Ⅰ group(p<0.01) and sample Ⅱ group(p<0.001). 6. The inhibitory effects of the serum GOT& GPT activities in rats induced by d-galactosamine were noted in sample Ⅱ group(p<0.001), but sample Ⅰ group was not recongnized. 7. The significantly inhibitory effects of in the serum LDH activities in rats induced by d-galactosamine were note in sample Ⅱ group(p<0.05). 8. The increases of the serum triglyceride content level in rats induced by d-galactosamine were inhibited in sample Ⅱ group(p<0.01). 9. The increases of the serum total bilirubin content level in rats induced by d-galactosamine were inhibited in sample Ⅱ group(p<0.05). According to the above results, it is considered that Samduhaejungtang has protective effect against liver injury in rats induced by $CCl_4$ and d-galactosamine.

  • Effect of Gamisamul-tang(GS) on body weight, uterine weight, splenial weight, serum and estrogen receptor-α (ER-α) in kidney were investigated in ovariectomized(OVX) rats. Female Sprague -Dawley rats with a body weight of 220 g were undergone ovariectomy or sham operation. The ovariectomy caused significant decrease in serum levels of HDL-cholesterol and in uterine weight compared to those of sham-operated animals. while caused significant increase in serum levels of total cholesterol, LDL-cholesterol, triglyceride, body weight and in splenial weight. In contrast, administration of OVX rats with the GS significantly elevated serum levels of HDL-cholesterol and uterine weight, but significantly depressed serum levels of total cholesterol, LDL-cholesterol, triglyceride, body weight and in splenial weight. ER-α localized in the kidney vessel endothelial cells and its expression was increased by ovariectomy and decreased by administration of OVX rats with the GS. Our results suggest that a possible protective effect of as against hyperlipidemia in postmenopausal cardiovascular disease and as increase the estrogen-estrogen receptor romp/ex in the kidney vessel endothelial cells.
