Separation and Determination of Cadmium in Blood Serum by Solvent Extraction and Atomic Absorption Spectrophotometry (용매추출과 원자흡수분광법에 의한 혈청시료중 카드뮴의 분리 및 정량)

  • Trace amount of cadmium was quantitatively extracted with 1% Aliquat 336-MIBK from 0.2M KSCN and 0.01M-HCl. Test tubes with a screw cap stopper were used for extraction in place of conventional separating funnels and organic solvents used were less dense than water. In this analysis, 22 times concentration effect was achieved with higher selectivity without interference. The detection limit of cadmium was 0.7ppb, therefore trace cadmium was easily analyzed by this method. The proposed method was applied to the determination of cadmium in blood serum and extraction mechanism was elucidated.

Cellulose Acetate Membrane Electrophoresis of Bull Serum Protein (Cellulose Acetate막(膜) 전기영동법(電氣泳動法)에 의한 종모우(種牡牛)의 혈청단백분획(血淸蛋白分劃)에 관한 조사(調査))

  • Total protein levels of 60 bovine bull sera were treasured with refractometer and the strum proteins were fractionated by cellulose acetate membrane electorphoresis and the relative amount of each fraction was measured with automatic scanning densitometer. The sixty bovine bulls consisted of 17 Charolais bulls, 28 Holstein bulls and 15 Korean bulls. Mean total serum protein level of the total bulls was $7.50{\pm}0.50(Mean{\pm}SD)$, with the mean of Charolais $7.04{\pm}0.37g/dl$, the mean of Holstein $7.62{\pm}0.40g/dl$ and the mean of Korean $7.81{\pm}0.43g/dl$. As barbital-calcium buffer was used in fractionating the bovine serum protein with cellulose acetate membrane, electric current of 0.4 mA per centimeter width of the membrane for an hour resulted in more clear separation between ${\alpha}_1$-globulin and ${\alpha}_2$-globulin than electric current of 0.6 or 1.0 mA for an hour. However ${\alpha}_1$-globulin and ${\alpha}_2$-globulin could not be measured separately with automatic scanning densitometer. Mean serum albumin level of total bulls was $3.54{\pm}0.42g/dl$, with the mean of Charolais $3.42{\pm}0.26g/dl$, the mean of Holstein $3.69{\pm}0.38g/dl$ and the mean of Korean $3.39{\pm}0.55g/dl$. Mean serum ${\alpha}$-globulin level of the total bulls was $0.62{\pm}0.14g/dl$, with the mean of Charolais $0.61{\pm}0.07g/dl$, the mean of Holstein $0.60{\pm}0.11g/dl$ and the mean of Korean $0.67{\pm}0.23g/dl$. Mean ${\beta}$-globulin level of the total bulls was $0.85{\pm}0.21g/dl$, with the mean of Charolais $0.74{\pm}0.11g/dl$, the mean of Holstein $0.83{\pm}0.14g/dl$ and the mean of Korean $1.02{\pm}0.30g/dl$. Mean serum ${\gamma}$-globulin level of the total bulls was $2.48{\pm}0.45g/dl$ with the mean of Charolais $2.27{\pm}0.37g/dl$, the mean of Holstein $2.48{\pm}0.44g/dl$ and the mean of Korean $2.72{\pm}0.47g/dl$.

  • An ID LC/MS (isotope dilution liquid chromatography/mass spectrometry) was used as a primary method for the quantitative analysis of cholesterol in human serum. The separation of cholesterol was carried out by Thermo ODS hypersil $C^{18}$ column. The mobile phase was 100% methanol, and flow rate was $0.3m{\ell}/min$. Cholesterol and cholesterol-$3,4-13C_2$ were monitored at m/z 369.4 and 371.3, which correspond to $[M-H_2O+H]^+$ respectively. In order to verify the measurement method, NIST SRM 909b was analyzed. The results agreed well with certified values within uncertainty. The four kinds of serum certified reference material were prepared and certified. The repeatabilities of measurement were ranged from 0.1 to 0.8% (RSD), which were relatively good. The reproducibility between independent measurement run was below 0.24% (RSD). The expanded uncertainty was about 1.43% within the 95% confidence interval.

Effects of Guisinhwan on the Ovariectomized Rat Model of Postmenopausal Osteoporosis (귀신환이 난소적출 백서의 골다공증에 미치는 영향)

  • Aging is characterized by an age-dependent reduction in bone density. Pronounced bone loss by postmenopausal estrogen deficiency induces a bad effect on dynamic balance of bone metabolism. When the bone density is reduced to the level below the fracture threshold, the risk for fracture is greatly increased. The purpose of this study is to examine what are the effects of the Guisinhwan(GSH : 歸腎丸) on the ovariectomized rat model of postmenopausal osteoporosis. The results of the experiment are as follows: Body weight in control group showed significant increase in comparison with sham, but that in GSH-treated showed no change in comparison with control. The level of serum albumin in control group showed significant decrease in comparison with sham. That in GSH-treated was slightly decreased in comparison with control. The level of serum ALP activity in control group showed significant increase in comparison with sham, but that in GSH-treated was significantly decreased in comparison with control. The level of serum phosphorous, calcium, GOT, GPT, T3 did not significant change among the three groups. The level of serum estrogen in control group showed slightly decreased in comparison with sham, but that in GSH-treated showed no change in comparison with control. The level of serum T4 in control group showed significant increase in comparison with sham, but that in GSH-treated was significantly decreased in comparison with control. Trabecular bone area as well as trabecular thickness in control group showed significant decrease In comparison with sham. Those in GSH-treated showed significant increase in comparison with control. Trabecular number did not significant change among the three groups. Trabecular separation only in GSH-treated showed significant decrease in comparison with control. Osteoclast number in control group showed significant increase in comparison with sham. That in GSH-treated showed significant decrease in comparison with control. Oseoblast surface in control group showed significant increase in comparison with sham, but that in GSH-treated showed no change in comparison with control. According to the above these results, GSH has shown to be capable of preventing and curing osteoporosis caused by old-aged and postmenopause.

Adsorption of Antibiotics on Serum Albumin Nanoparticle (혈청 알부민 나노입자를 이용한 항생제 흡착)

  • Antibiotics are compounds broadly used to treat patients with infectious diseases and to enhance productivity in agriculture, fisheries, and livestock industries. However, due to the overuse of antibiotics and their low biodegradability, a substantial amount of antibiotics is leaking into the sewer, subsequently resulting in pollution and the emergence of antibiotic-resistant bacteria. This study explores biodegradable serum albumin's potential as an adsorbent to remove antibiotics from water. Serum albumin is a natural blood protein that transports various metabolites and hormones to all tissues' extravascular spaces. While serum albumin is highly water-soluble, it has intrinsic binding sites which readily accommodate ionic, hydrophilic, or hydrophobic molecules, rendering it a good building block for a nano-adsorbent. To induce coacervation, a desolvating agent, ethanol, was added dropwise into the aqueous albumin solution, resulting in dehydration and liquid-liquid phase separation of albumins into albumin nanoparticles within a size range of 150 ~ 170 nm. The addition of glutaraldehyde as a cross-linker improved the size stability and homogeneity of albumin nanoparticles. Adsorption of amoxicillin antibiotics on albumin nanoparticles was dependent upon glutaraldehyde concentration used in desolvation and pH during adsorption. The maximum adsorption capacity measured by spectrophotometry was found to be 12.4 micrograms of amoxicillin per milligram of albumin nanoparticle. These results demonstrate serum albumin's potential as a building block for fabricating a natural nano-adsorbent to remove antibiotics from water.

  • A han transfers her serum immunoglobulin G to the agg (IgY) and gives immunity to her offspring. Therefore, The hen agg can be an effective supplier of a large amount of antigen specific antibody that accumulates in the egg yolk. Antigen specific antibody has been widely used for immunological analysis in the field of diagnosis as well as pure scientific research. The production and separation technology of IgY is demonstrated in the present study.

  • Increase in the hydrophilicity (HPCT) of polysulfone (PS) membrane and subsequently decrease in fouling can be achieved by surface modification of PS based membranes. Therefore, in this work, ultrafiltration membranes with increased HPCT were prepared using the enantiomeric tartaric acid (D-TA) and racemic tartaric acid (DL-TA). Phase inversion technique was used for the preparation of polyethylene glycol and TA blended PS membrane. Morphological analysis of the fabricated membranes was done by scanning electron microscope and atomic force microscopy. Bovine serum albumin (BSA) solution was taken for finding the permeation and rejection behavior of prepared membranes. Maximum BSA rejection was increased by 70.5% for the modified membrane.

Separation of Lactoferrin from Model Whey Protein Mixture by Reverse Micelles Formed by Cationic Surfactant

  • The selective extraction behavior of lactoferrin (Lf) from whey protein mixture was examined using reverse micelles formed by the cationic surfactant, cetyldimethylammonium bromide (CDAB). The major whey proteins, including ${\beta}$-lactoglobulin, ${\alpha}$-lactalbumin and bovine serum albumin, were solubilized from aqueous phase to organic phase while Lf was recovered in the aqueous phase. The solubilization behaviors of the proteins were manipulated by the process parameters such as the pH and salt concentration of the aqueous phase and the surfactant concentration in the organic phase. Efficient forward extraction was achieved with sodium borate buffer (50 mM, pH 9) containing 50 mM KCl and organic phase containing 100 mM CDAB. Based on SDS-PAGE and densitometry, about 96% of the initial Lf remained in the aqueous phase after forward extraction. The dialyzed Lf fully maintained its bacteriostatic activity against E. coli O157:H7.

  • For development of $EMIT-T_{3}$ assay, the conjugation of 3, 5, 3'-triiodothyroformic acid NHS ester to G6PDH was attempted in various reaction conditions. Up to now, the best conjugation condition was the ratio of $T_{3}$-NHS:G6PDH=100 in 25% carbitol-Tris buffer at pH 9, $0^{\circ}C$ during overnight. The obtained $T_{3}$-G6PDH conjugates usually had 20% residual enzyme activity which was inhibited by 40-70% with various $anti-T_{3}$ antibodies. Utilizing the conjugate I and an antibody (S2633G), a useful standard curve for $T_{3}$ assay was obtained in the range of 0 to 5ng/ml with 499 EMIT units of separation.

Synthesis of Size Controllable Silk Fibroin Microparticles and Their Stability on Different Solutions

  • Silk fibroin microparticles were fabricated using a phase separation technique between silk fibroin solution and polyvinyl alcohol. We found that the concentration of polyvinyl alcohol determines the size of microparticles. The mean diameter of the silk fibroin microparticles varied from 3.48 ㎛ to 4.05 ㎛. The silk fibroin microparticle size increased as a function of the concentration of PVA in aqueous silk solution. The resulting silk fibroin microparticles have narrow size distribution (i.e. monodisperse) and smooth/spherical surface. Also, we studied the effects of mouse serum, sodium phosphate buffer (PBS), and pH on the stability of the silk fibroin microparticles. Overall, we demonstrated the simple method to fabricate and to control the silk fibroin microparticles that makes our silk microparticles to be usable for a potential drug delivery carrier.
