Pentazocine의 가토혈당(家兎血糖), 혈청(血淸) Transaminase 및 Alkaline Phosphatase 활성도(活性度)에 대(對)한 작용(作用) (The Action of Pentazocine on the Blood Sugar Level, Serum Transaminase and Alkaline Phosphatase Activities in Rabbits)

  • A benzomorphan derivative, pentazocine has both opioid agonistic actions and weak narcotic antagonistic activity. In this paper, authors attempted to study the dose response of pentazocine on the blood sugar level, serum transaminase and alkaline phosphatase activity in rabbits. Eighteen rabbits were devided into 3 groups, and each group were injected with pentazocine 5mg, 15mg, and 30mg/kg respectively. And metabolic effects were investigated measuring the change of blood sugar contents, serum transaminase (S-GOT, S-GPT) activities and alkaline phosphatase activities in rabbits. The results were obtained as follows; 1. Pentazocine significantly increased the blood sugar content in rabbits. 2. Pentazocine significantly increased the serum GOT activity but in the serum GPT activity, it significantly increased in large dose. 3. Pentazocine significantly increased serum alkaline phosphatase activity.

Serum Antibody Ig G and Ig M Titers for Opisthorchis felineus Correlate with Eggs in Faeces - a Comprehensive Study in Chuvash Republic, Russia

  • The Cholangiocarcinoma is a. The risk of development of cholangiocarcinoma, generally a rare type of a liver tumor, increases during infection of Opisthorchiasis. For this reason the timely detection of Opisthorchiasis is important for Cholangiocarcinoma prevention. There are many studies which concern the detection of pathogenesis of Opisthorchis viverrini infection but a little known about Opisthorchis felineus. In this study we investigate a correlation of the eggs which are found in a faeces and are comparable with a serum Ig G and Ig M antibody level that were detected with ELISA test in a large group of patients. The result is showing positive correlation between evidence of the Opisthorchis felineus eggs that were found in a faeces and antibody Ig G and Ig M level in a serum. Moreover the combination of two methods can improve the Opisthorchiasis diagnostic: the serum antibody and faeces investigation of eggs.

Effect of edaravone in diabetes mellitus-induced nephropathy in rats

  • Edaravone, a synthetic-free radical scavenger, has been reported to reduce ischemia-reperfusion-induced renal injury by improving tubular cell function, and lowering serum creatinine and renal vascular resistance. The present study investigated the effect of edaravone in diabetes mellitus-induced nephropathy in rats. A single administration of streptozotocin (STZ, 55 mg/kg, i .p.) was employed to induce diabetes mellitus in rats. The STZ-administered diabetic rats were allowed for 10 weeks to develop nephropathy. Mean body weight, lipid alteration, renal functional and histopathology were analysed. Diabetic rats developed nephropathy as evidenced by a significant increase in serum creatinine and urea, and marked renal histopathological abnormalities like glomerulosclerosis and tubular cell degeneration. The kidney weight to body weight ratio was increased. Moreover, diabetic rats showed lipid alteration as evidenced by a significant increase in serum triglycerides and decrease in serum high-density lipoproteins. Edaravone (10 mg/kg, i .p., last 4-weeks) treatment markedly prevented the development of nephropathy in diabetic rats by reducing serum creatinine and urea and preventing renal structural abnormalities. In addition, its treatment, without significantly altering the elevated glucose level in diabetic rats, prevented diabetes mellitus-induced lipid alteration by reducing serum triglycerides and increasing serum high-density lipoproteins. Interestingly, the renoprotective effect of edaravone was comparable to that of lisinopril (5 mg/kg, p.o, 4 weeks, standard drug). Edaravone prevented renal structural and functional abnormalities and lipid alteration associated with experimental diabetes mellitus. Edaravone has a potential to prevent nephropathy without showing an anti-diabetic action, implicating its direct renoprotection in diabetic rats.

복합경구피임제 복용이 혈청 지질함량에 미치는 영향 (The Effect of Combined Oral Contraceptive Steroids on Serum Levels of Lipids)

  • Increased incidence of myocardial infarction, arteriosclerosis and hypertension in oral contraceptive steroids users has been recognized as serious problem. Evidence have been accumulated that oral contraceptive steroids cause an increase in serum lipid levels, mainly triglyceride as well as an increase in serum cholesterol concentration in some women. Effect of oral contraceptives on serum lipids have been predominantly confirmed in Europian and American women. It should be emphasized that effects of contraceptives may he influenced by differences in nutrition, climate, socioeconomic status and race. The present study was, therefore, attempted to determine the effect of oral contraceptive steroids on serum lipid levels in Korean women and also to demonstrate whether the duration of pill treatment may affect serum lipid levels. Ten women, who had never taken contraceptive steroids, served as a control. One hundred women on combined oral contraceptive steroids were grouped according to the duration of pill use: duration of less than one year(14 women), 1-2 years(12 women), 2-5 years(29 women), 5-10 years(33 women) and more than 10 years(13 women). Serum levels of cholesterol, phosholipid and triglyceride were measured by enzymatic method, Fiske-SubbaRow method and Sugiura method respectively. The results obtained are as follows; 1) Oral contraceptive steroids increased serum cholesterol concentration by 4.8% as compared with cholesterol value of control group. But this increase was not statistically significant. 2) As compared with control value, concentration of serum phospholipid in pill-treated women decreased by 14.3%, which was statistically significant (p<0.05). 3) Concentration of serum triglyceride in pill-treated women increased by 10.6% over control value, but this increase was not statistically significant. 4) Duration of pill-treatment did not affect serum concentrations of cholesterol, phospholipid and triglyceride. From the results mentioned above, it may be concluded that combined oral contraceptive steroids, at least used for subjects involved in this study, decreased serum phospholipid significantly, thereby influencing serum lipid concentration.

인삼(人蔘) Alkaloidal Fraction이 지질대사(脂質代謝)에 미치는 영향(影響) (Effects of Ginseng Alkaloidal Fraction on Lipid Metabolism)

  • Alkaloidal fraction, a principle of ginseng extract was administered to rats orally in the amount of 2.5 mg per day per 100 gm body weight continuously for eight weeks to assess the lipid content of the serum and liver tissue. The findings can be summarized as follows: 1. Total cholesterol contents of serum and liver tissue were found to decrease with the administration of ginseng alkaloidal fraction. 2. Phospholipid contents increased in the serum and decreased in the liver tissue with the administration of ginseng alkaloidal fraction. 3. Triglyceride contents of the serum as well as liver tissue increased with the administration of ginseng alkaloidal fraction during the first 4 weeks but decreased in the second 4 weeks to reach a level below that of the pre-experimental period.

Effect of Calcium on Estrogen and Follicle Stimulating Hormone Secretion in Rabbits

  • The effect of calcium gluconate on estrogen (estradiol) serum level as well as follicle stimulating hormone level was studied. Our results revealed that oral administration of calcium gluconate (100 mg/kg body wight) to adult non-pregnant female rabbits caused a significant increase of serum levels of estradiol and follicle stimulating hormone. On the other hand, oral contraceptive (Norminest tablets) decreased significantly follicle stimulating hormone serum level, while combined administration of calcium gluconate and oral contraceptive caused significant increase of serum level of follicle stimulating hormone compared with control values. Also, concurrent administration of calcium gluconate and Norminest tablet increased significantly the rate of conception compared with group recieved Norminest tablets only. These results indicated that combined administration of calcium and oral contraceptives must be cautiously.

  • Testosterone이 serum-free medium에서 배양한 토끼의 신장 근위세뇨관 상피세포의 세포성장과 기능에 미치는 영향을 관찰한 바 다음과 같은 결과를 얻었다. 1. 토끼의 신장 근위세뇨관 상피세포는 testosterone 1 nM의 농도에서 유의한 세포 성장 촉진 효과를 나타내었고, testosterone 10 nM이상의 농도에서는 세포성장이 억제되었다. 2. Testosterone은 serum-free medium에서 성장촉진인자의 하나인 hydrocortisone을 growth supplement로 넣어준 serum-free medium에서 토끼 신장의 근위세뇨관 상피세포의 성장을 촉진시키었다. 3. Testosterone은 hydrocortisone을 growth supplement로 넣어준 serum-free medium에서 토끼 신장의 근위세뇨관 상피세포의 성장을 촉진시키었다. 4. Testosterone은 Northern blot analysis에 의하여 확인한 토끼 신장의 근위 세뇨관 상피세포의 ${\beta}-actin$ mRNA level은 증가되었다. 이상의 결과로 미루어 보아, serum-free 그리고 hormonally defined media에서 testosterone이 토끼의 신장 근위세뇨관 상피세포의 성장 및 기능에 대하여 촉진적으로 작용하는 것은 cellular mecrofilament의 중요한 구성단백의 하나로 밝혀진 ${\beta}-actin$의 합성 증가에 기인하는 것으로 생각된다.

Hepatoprotective Activity of Thespesia populnea Bark Extracts against Carbon Tetrachloride-Induced Liver Toxicity in Rats

  • In the present study, Thespesia populnea (Malvaceae) bark was extracted with methanol and water. The extracts were vacuum dried to yield the respective methanol (MET) and aqueous extract (AET). The extracts were evaluated for hepatoprotective activity against carbon tetrachloride $(CCl_{4})$ induced liver damage at 2 dose levels (250 and 500 mg/kg). The biochemical parameters observed in serum were total bilirubin, alkaline phosphatase (ALP), serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT) levels and total protein. Aspartate transaminase (AST), alanine transaminase (ALT) and total protein levels in liver were also evaluated. Histopathological study on the liver tissue was also performed. The extracts exhibited dose dependent reduction in total bilirubin, ALP SGOT, SGPT, AST, ALT and increase in total protein (serum and liver) levels. The extracts also exhibited only mild hepatocytic damage compared to the $CCl_{4}$ Treated group. MET was found to exhibit higher hepatoprotection than AET.

Comparative Effects on Secretion of LH, FSH, Prolactin, and Testosterone by Chronic and Direct Hypothalamic Administration of Nonylphenol to Adult Male Rats

  • Nonylphenol (NP) is a widespread environmental pollutant that has been shown to exert both toxic and estrogenic effects on mammalian cells. As the effects of NP on the reproductive system of adult male vertebrates are virtually unknown, we investigated not only the changes of reproductive hormone secretion in serum after chronic exposure to NP but also, in order to identify the site of its action, the reproductive hormone secretion in serum 48 hours after microinfusion of NP within hypothalamic preoptic area (POA). In the chronic exposure, the luteinizing hormone (LH), follicle stimulating hormone (FSH), and testosterone in serum were decreased but prolactin (PRL) concentrations were increased. The LH, FSH, and testosterone in serum were decreased through the direct infusion of NP into POA, while there was no difference in mean serum prolactin between NP and control groups. These observations suggest that NP as endocrine disruptor has modulatory effects on hypothalamo-pituitary-gonadal axis and that the site of action of NP could be hypothalamic POA.

  • Steroid hormone의 하나인 ${\beta}-estradiol$이 serum-free medium에서 배양한 토끼의 신장 근위세뇨관 상피세포의 세포성장과 기능에 미치는 영향을 관찰한 바 다음과 같은 결과를 얻었다 1. Serum-free medium에서 토끼의 신장 근위세뇨관 상피세포는 ${\beta}-estradiol$ 1 nM의 농도에서 유의한 세포 성장 촉진 효과를 나타내었고, ${\beta}-estradiol$ 10 nM이상의 농도에서는 세포성장이 억제되었다. 2. ${\beta}-Estradiol$은 serum-free medium에서 성장촉진인자의 하나인 hydrocortisone을 뺀 조건하에서 세포 성장을 증가시키었다. 3. ${\beta}-Estradiol$은 hydrocortisone을 growth supplement로 넣어준 serum-free medium에서 토끼 신장의 근위세뇨관 상피세포의 성장을 촉진시키었다. 4. ${\beta}-Estradiol$은 Northern blot analysis에 의하여 확인한 alpha I (IV) collagen mRNA level에는 별다른 변화를 보이지 않으나, ${\beta}-actin$mRNA level은 증가되었다. 이상의 결과로 미루어 보아, serum-free 그리고 hormonally defined media에서 ${\beta}-estradiol이$ 토끼의 신장 근위세뇨관 상피세포의 성장 및 기능에 대하여 촉진적으로 작용하는 것은 cellular microfilament의 중요한 구성단백의 하나로 밝혀진 ${\beta}-actin$의 합성 증가에 기인하는 것으로 생각된다.
