  • In this study, we examined the effects of dietary 1,3-diacylglycerol (DG) compared to conventional triacylglycerol (TG) oil on the postprandial response of total and chylomicron TG, glucose, insulin, and free fatty acid (FFA). This study was conducted using a cross-over design. Ninety subjects participated in the high-fat meal tolerance test where they were randomly assigned to consume two experimental sandwiches containing mayonnaise with TG or DG oil with a seven-day interval. Blood samples were collected before ingestion and at 2, 3, 4 and 6 hr time point after ingestion and analyzed for total and chylomicron TG, glucose, insulin, FFA and phospholipid fatty acid composition. Both TG and DG ingestion had similar effects on postprandial TG response, but a different response from chylomicron TG. Compared with the TG group, TG levels were significantly lower only at 6 hr time point in the DG group. On the other hand, chylomicron TG rose steeply at 2 hr time point and decreased faster in this group. Also, the adjusted value to fasting levels was the same as the unadjusted level. Fasting levels and net differences in insulin were significantly lower at 3 hr time point where chylomicron TG levels were significantly lower in the DG group. But those of glucose and FFA in the TG and DG groups did not differ significantly. Fasting and postprandial levels of fatty acid composition in serum phospholipids in the two groups did not differ significantly. In conclusion, this study indicated that one could reduce the magnitude of postprandial lipemia without influencing glucose metabolism by consumning DG oil as a substitute for TG oil. Based on the correlation of coronary artery disease and postprandial lipemia, dietary DG ingestion might have a beneficial effect in treating such a disease. Further studies are required to clarify the long-tenn effects of dietary DG on blood lipid levels in humans.

  • We examined the effect of dietary behavior improvement on diet quality, body composition and blood lipid profiles in 68 middle-aged women. Dietary behavior intervention was consisted of counselling in the problem areas of portion control, food selection for low carbohydrate and high fiber food items, and education on the improvement of micronutrient intakes and diet quality. The subjects were divided into two groups; Improved or Not-Improved Group according to the level of changes in dietary behavior scores. After 6 months, body weight, BMI, waist-hip ratio, and visceral fat area were significantly decreased in the Improved Group compared to the Not-Improved Group. Calorie-adjusted protein, fiber, calcium, iron and vitamin C intakes were significantly increased in the Improved Group. The index of nutritional quality (INQ) and mean adequacy ratio (MAR) were increased only in the Improved Group. We observed a significantly increase in HDL-cholesterol and a decrease in total cholesterol, triglyceride and LDL-cholesterol, and an improvement of atherogenic index in the Improved Group. These results showed that dietary behavior intervention improved dietary behavior scores, and those whose dietary behavior score improved showed more improvement in diet quality, body composition, and serum profiles than those whose dietary behavior score did not improve.

  • The purpose of this study was to determine the health behaviors and nutritional status related to dyslipidemia in Korean middle-aged adults (between 50 and 64 years old) from the Korean National Health and Nutrition Examinations Survey data (2007~2010). A total of 4,721 subjects were analyzed in this study. The subjects were divided into three groups (normal, borderline, and dyslipidemia) according to serum lipid levels. Parameters included in this study were drinking and smoking, anthropometric parameters, blood and nutritional parameters. The latter parameter included food/nutrients intake. All data was adjusted by sex, region, education level, and age. General linear model and logistic regression model were used for statistical analysis. The dyslipidemia group was comprised of more men than women. By contrast, the borderline group was comprised of more women than men (p<0.001). No significant differences were observed for other general characteristics. There were more smokers and drinkers(drinking per time) in the dyslipidemia group (p<0.05). Anthropometric data showed significant difference, ie, height (p<0.05), weight, body mass index, waist circumference, percent body fat, and blood pressure were higher in the dyslipidemia group (p<0.001). Only blood urea nitrogen showed no significant difference among groups. The HbA1c (p<0.01), fasting blood glucose, GOT, GPT, creatinine levels were higher in the dyslipidemia group (p<0.001). So it is required for the management of obesity in dyslipidemia group. The dyslipidemia group ate less sea food (p<0.05). The nutrients intake of energy and protein, thiamin, riboflavin, niacin, calcium, phosphorus were lower in the dyslipidemia group (p<0.05). Therefore, to lower dyslipidemia prevalence rates, it is necessary to increase the intake of foods containing ${\omega}-3$ fatty acids. We also suggest a meal management program and nutritional education to recognize the risk of dyslipidemia, especially for people such as the individual in the borderline and dyslipidemia study groups.

  • We investigated the effect of Capsosiphon fulvescens (CFE) and pheophorbide a (PhA) contained in CFE on oxidative stress regarded as a factor for diabetic complication. Streptozotocin (STZ), known as an oxidative stress inducer, was intraperitoneal injected for causing diabetes. After 7 days, CFE (4 and 20 mg/kg body weight) and PhA (0.2 mg/kg body weight) were treated once a day for 9 weeks. After the sacrifice, testis tissues were collected for the experiments. We confirmed that the treatment with CFE and PhA in diabetic animals not only decreased level of lipid peroxidation and serum nitric oxide compared with the diabetes group, but also the activities of glutathione peroxidase and glutathione-S-transferase were restored remarkably. Furthermore the activity of antioxidant enzymes, catalase and superoxide dismutase, were significantly recovered. With these results, our study suggest that CFE containing PhA may prevent seminal glands damages induced by oxidative stress in diabetic condition.

  • This study was conducted to investigate the biological activity and hepatoprotective effect of DWP-04 [DDB : selenium yeast: glutathione (31.1 : 6.8 : 62.1(w/w/w)] in D-galactosamine (GaIN) intoxicated rats. The DWP-04 (50, 100 or 200 mg/kg) or its vehicle was orally administered everyday before the start of GaIN injection (400 mg/kg, ip) for two weeks and animal decapitated for 24 hrs after GaINinjected. The activities of serum enzymes, markers of liver function, were increased in the GaIN group compared to normal group and significantly lowered in the DWP-04 pretreated group than in the GaIN group. Hepatic lipid peroxide level and activities of phase 1 enzymes were significantly higher than those of GaIN group compared to normal group and lower in the DWP-04 pretreated group than in the GaIN group, and phase II enzyme activities in liver were lower in the GaIN group than in the normal group and were increased in the DWP-04 pretreated group than in the GaIN group. Total hepatic glutathione content and glutathione biosynthesis enzymes were lower in the GaIN group than in the normal group and were increased in the DWP-04 pretreated group than in the GaIN group. Therefore, the current results indicated that DWP-04 administration alleviated the GaIN-induced adverse effect through enhancing the antioxidant enzyme activities.

  • Sweet potato (Ipomoea batatas L.) is widely used in Indonesia and other countries as a traditional medicine for the treatment of diabetes mellitus (DM). The MeOH extract of white skinned sweet potatoes (WSSP) was administered orally in doses of 100 and 200 mg/kg body weight in streptozotocin (STZ)-induced diabetic rats. Experimental diabetes was induced by a single dose of STZ (45 mg/kg, i.p.) injection. Oxidative stress was measured by tissue lipid peroxide (LPO) levels, serum aspartate transaminase (AST), alanine transaminase (ALT), total triglyceride (TG), total cholesterol (TC) and by antioxidative enzymatic activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione S-transferase in the liver. An increase in blood glucose, LPO level, AST, ALT, TG and TC levels was observed in the STZ-induced diabetic rats. Administration of MeOH extract of WSSP at a dose of 200 mg/kg for two weeks caused a significant reduction in blood glucose, LPO levels, AST, ALT, TG and TC levels in the STZ-induced diabetic rats. Furthermore, oral administration of MeOH extract showed significant improvement in the activities of antioxidant enzymes (SOD, GPx, and CAT) compared to STZ-induced diabetic rats. In conclusion, the obtained results clearly indicate the role of oxidative stress in the induction of diabetes, and that the protective effects of MeOH extracts of WSSP could be used to benefit diabetic patients.

  • The purpose of the present study is to determine the occurrence of dyslipidemia and the effects of health life practices among Korean males and females. The study subjects were 4,036 persons aged at least 19 years, as selected from the health questionnaire data out of the third year data(2012) Korea National Health and Nutrition Examination Survey. They were divided into a dyslipidemia group of 2,092 subjects (male 1,042, female 1,050) and a normal group of 1,944 subjects (male 740, female 1,204) by applying the dyslipidemia treatment guidelines of the Korean Society of Lipidology and Atherosclerosis. In the present study, the SPSS 18.0 program was used to conduct t-tests, a ${\chi}^2-test$, and a logistic regression analysis of descriptive statistics. Therefore, dyslipidemia-related healthy life practice programs should apply the characteristics of Korean males and females that show differences in serum lipid concentrations and to this end, continued studies, and national-level customized education applying the effects of healthy life practices is considered necessary.

  • Objectives : Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Iksujisundan has been known to be effective for the treatment of diabetes. The present study was carried out to investigate the effect of Iksujisundan on renal function, peroxynitrite(ONOO-) scavenging activity and polyol pathway in streptozotocin-induced diabetic rats. Methods : The crushed Iksujisundan was extracted 3 times, each time with 3 volumes of methyl alcohol at 60$^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 87.8g. Iksujisundan extract was orally administreted at 100 mg per 1 kg of body weight for 20 days to the diabetic rats induced by streptozotocin(60mg/kg). The effects of Iksujisundan extract on the streptozotocin-induced diabetic rats were observed by measuring the serum level of glucose, insulin, lipid components, creatinine and BUN, and also the kidney levels of superoxide anion radical(${\cdot}$O2-), nitric oxide(NO) and ONOO-, and also the enzyme activities involved in the polyol pathway. Results : The effects of Iksujisundan on the streptozotocin-induced diabetic rats with regards to body weight, blood glucose and indulin levels, creatinine and BUN levels, total cholesterol and triglyceride lavels, and HDL-cholesterol levels were all shown to be good enough to prevent and cure the diabetes and its complications. Iksujisundan inhibited the generation of ${\cdot}$O2-,NO and ONOO- in the kidney of streptozotocin-induced diabetic rats. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the streptozotocin-induced diabetic rats were reversed toward natural activities. Conclusions : Iksujisundan might inhibit the development of diabetes and its complications by scavenging reactive oxygen and nitrogen species, thereby by reducing oxidative stresses and also by regulating the activities of polyol pathway enzymes, all of which could help to recover kidney function.

  • Obesity is a risk factor for numerous metabolic diseases. Recently, naturally occurring compounds that may improve obesity have received increasing attention. Xanthigen is a mixture of fucoxanthin and punicic acid derived from brown seaweed and pomegranate seed, respectively, which have been traditionally used for lipid-lowering effects in humans. In this study, we investigated whether Xanthigen attenuates high-fat diet-induced obesity in C57BL/6N mice. The mice were fed on a normal diet (ND), high-fat diet (HFD), HFD plus 1% Xanthigen or HFD plus 1% green tea extract (GTE) for 11 weeks. Food efficiency ratio (FER) and body weight were significantly reduced in mice fed HFD plus Xanthigen compared to HFD-fed mice. Consistent with the results in body weight change, Xanthigen also significantly decreased the weights of epididymal adipose tissue, retroperitoneal adipose tissue, and liver in HFD plus 1% Xanthigen-fed mice. The serum level of low-density lipoprotein (LDL)-cholesterol was significantly decreased in HFD plus Xanthigen-fed mice compared to HFD-fed mice. These results suggest that Xanthigen may be useful in the development of a functional health food for anti-obesity.

Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Gamigukihwandong-hwan has been known to be effective for the treatment of diabetes. The present study was carried out to investigate the effect of Gamigukihwandong-hwan on renal function, peroxynitrite (ONOO$^-$) scavenging activity and polyol pathway in streptozotocin-induced diabetic rats. The crushed Gamigukihwandong-hwan was extracted 3 times, each time with 3 volumes of methyl alcohol at 60$^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 74.95 g. Gamigukihwandong-hwan extract was oral-administered 100 mg per 1 kg of body weight for 20 days to the diabetic rats induced by streptozotocin (60 mg/kg). The effects of Gamigukihwandong-hwan extract on the streptozotocin-induced diabetic rats were observed by measuring the serum level of glucose, insulin, lipid components, creatinine and BUN, and also the kidney levels of superoxide anion radical (${\cdot}O_2^-$), nitric oxide (NO) and ONOO$^-$, and also the enzyme activities involved in polyol pathway. The Effects of Gamigukihwandong-hwan on the streptozotocin-induced diabetic rats with regards to body weight, blood glucose and insulin levels, creatinine and BUN levels, total cholesterol and triglyceride levels, and HDL-cholesterol levels were all shown to be good enough to cure and prevent the diabetes and its complications. Gamigukihwandong-hwan inhibited the generation of ${\cdot}O_2^-$, NO and ONOO$^-$ in the kidney of streptozotocin-induced diabetic rats. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the streptozotocin-induced diabetic rats, whereas the ones in the Gamigukihwandong-hwan administered group among the streptozotocin-induced diabetic rats were reversed toward the natural activities. Gamigukihwandong-hwan might inhibit the development of diabetic nephropathy by scavenging reactive oxygen and nitrogen species, thereby by reducing oxidative stresses and also by regulating the activities of polyol pathway enzymes, all of which could help to recover the function of kidney.
