Safety and Effects on Lipid Parameters of Rubus coreanus and Atractylodes japonica in Ovariectomized Rats

  • Defatted methanol extracts of the medicinal plants, Rubus coreanus Miq. (RC) and Atractylodes japonica Koidzumi (AJ) were added at the levels of 0.1, 0.5, or $2\%$ (w/w) to high cholesterol diets and fed to ovariectomized Sprague-Dawley female rats, weighing 212.6 $\pm$ 1.8 g for four weeks. Weight gains were lower in RC and AJ groups than the control group, but there were no changes in uterus weights. Serum levels of triglyceride decreased by 20-$27\%$ in the experimental groups fed $0.1\%$ of each extract (O.1RC and O.1AJ), compared with that of control (Ovx). Serum cholesterol levels were not changed in the RC groups but increased in the group fed $2\%$ of the AJ extract. Liver levels of cholesterol and triglyceride were reduced in both the RC and AJ groups. Microscopic observation revealed that there were no morphological alterations in liver, lung, heart, spleen and kidney tissues of the experimental groups. Plasma levels of albumin, BUN, creatinine, sodium, potassium and phosphate in the IRC and AJ groups were in normal ranges. Serum GOT and GPT activities were, however, higher in the 2.0AJ than Ovx group. These results suggest that the extracts of the Rubus coreanus Miq. and Atractylodes japonica Koidzumi at dietary levels as low as $0.1\%$ may be utilized as hypotriglyceridemic ingredients for functional foods.

The Variations of the SCE Frequency of Human Lymphocytes by Smoking Habits and Dietary Factors in College Students (남자대학생의 흡연 및 식사습관에 따른 인체 임파구 SCE 빈도 수의 변화)

  • Sister chromatid exchange(SCE) has recently become a common cytogenic assay system for detecting exposure to chemical mutagens and carcinogens. One application of SCE is the monitoring of populations believed to be exposed to such agents. A cross-sectional study of SCE frequency in peripheral blood lymphocytes from 40 college students aged 18 to 26 years was conducted. The effects of cigarette smoking alcohol and coffee consumption, dietary and environmental factors on SCE were assessed. A mean spontaneous SCE per cell for the smokers(4.88$\pm$0.17). The SCE levels of the smokers were associated with the personal smoking amount ; the observed increase in the SCE frequency correlated with the number of cigarettes smoked per day (P<0.05). There was no effect of age on SCE. There were positive linear relationship between SCE and food frequency score of meat and fish group (P<0.05) or instant food group(P<0.01) in non-smokers. But in smokers, a significant inverse association between SCE and food frequency score of green and yellow vegetable group(P<0.05). Alcohol intake produced a significant increase(P<0.01) of SCE in comparison with the mean SCE for those not drinking alcohol in combine subjects. Other dietary parameters, including coffee intake, use of artificial sweetners and processed foods, did not show any increase in SCE. SCEs were inversely related to blood glucose and serum cholesterol levels of the combine subjects. No significant correlations were found between SCE frequencise and any other hematological parameters of the subjects.

A Study on the Prediction of Parturient Syndrome in Holstein Cows (젖소에서의 산욕기질병 발생예견에 관한 연구)

  • In order to establish a method predicting susceptible cows to the parturient syndrome, various serum chemical parameters (calcium, phosphorus, Ca/P, magnesium, cholesterol, total protein, albumin, globulin, A/G, total lipid, non-esterified fatty acid(NEFA) and aspartate aminotransferase(AST)) were measured during late pregnancy and their relationships with periparturient diseases were investigated during puerpural period. The results obtained were as follows : 1. The factors affecting the prediction of susceptible cows to parturient syndrome were calcium, magnesium, total protein, globulin, A/G ratio and total lipid at 30 day antepartum and the diagnosability was 70.7%. 2. In the experimental cows producing more than 21kg of milk per day, the factors affecting the prediction of susceptible cows to parturient syndrome were calcium, NEFA and A/G ratio at 30 day antepartum and the diagnosability was 66.7%. 3. In the experimental cows calved more than 3 times, the factors affecting the perdiction of susceptible cows to parturient syndrome were calcium, total protein, albumin and NEFA at 30 day antepartum and the diagnosability was 83.3%.

The Effect of Petasites japonicus Extract on Hepatotoxicity in Rats

  • Petasites japonicus (Compositae) is a perennial herb which has been used as treatment of antitussive, expectorant, sedatives, paralysis, diurectics in folk remedies. The pharmacological studies of this natural drug have not yet estabilished. So, we examined anti-lipid peroxidative effects and liver protective effect on $CCl_4$ induced lipid peroxidation and hepatotoxicity in rats. In vivo liver lipid, MeOH Ex. revealed significant increase of anti-lipid peroxidative effects according to concentration dependently. In chemical parameters obtained from serum analysis, MeOH Ex. as a increase of medicine concentration (0.25 g/kg, 0.5 g/kg, 1.0 g/kg), GOT, GPT, AlP decreased. In 0.5 g/kg GPT administered group, there was relative in GOT, AlP. Cholesterol decreased in 1.0 g/kg and 1.0 g/kg administered group, BUN decreased relatively in 1.0 g/kg administered group.

Antilipoperoxidant Activity of Antraquinone and Stilbene from Polygonum cuspidatum (호장으로부터 분리한 안트라퀴논 및 스틸벤 화합물의 지질과산화 저해활성)

  • Polygonum cuspidatum has been used as treatments of dermatitis, gonorrhea, inflammation, and hyperlipidaemia in traditional medicine. We examined liver protective effect on CCl$_4$ inducing hepatotoxicity and anti-oxidative activity by TBA method. Phytochemical examination of Polygonum cuspidatum led to the isolation and characterization of emodin 8-O-${\beta}$-D-glucopyranoside (compound 1), and trans-resveratrol 3-O-${\beta}$-D-glucopyranoside (compound 2). Compounds 1 and 2 enhanced the inhibition of anti-lipid peroxidative effects in liver homogenate. In chemical parameters obtained from serum analysis, compounds 1 and 2 also revealed significant decrease in hepatotoxicity. These results suggested that the antraquinone and stilbene which were isolated from Polygonum cuspidatum might be used as therapeutic agent of hepatitis.

Effect of Cyperi Rhizoma on $CCI_4$ Induced Hepatotoxicity and Lipid Peroxidation (향부자 분획물의 사염화탄소로 유도된 간장해 및 지질과산화에 미치는 영향)

  • Cyperus rotundus L. (Cyperaceae) has been used as an analgesic and antiiflammatory agent and in the treatment of menstrual disorder in folk remedies. Cyperi Rhizomata, processed and unprocessed, were extracted with methanol and fractionated with petroleum ether, chloroform. Butanol, water. The effect of unprocessed Cyperi Rhizoma and processed Cyperi Rhizoma on $CCl_4$ induced lipid peroxidation and hepatotoxity have been tested in rats. BuOH. Water fractions of unprocessed Cyperi Rhizome enhanced the inhibition of antilipid peroxidative effects in liver lipid. In chemical parameters obtained from serum analysis. Butanol fraction of unprocessed Cyperi Rhizoma showed significant decrease in hepatotoxicity. In result, unprocessed Cyperi rhizoma has significant antilipid peroxidative effect and hepatoprotective activity.

Twenty-Eight-Day Repeated Inhalation Toxicity Study of Aluminum Oxide Nanoparticles in Male Sprague-Dawley Rats

  • Aluminum oxide nanoparticles ($Al_2O_3$ NPs) are among the most widely used nanomaterials; however, relatively little information about their risk identification and assessment is available. In the present study, we aimed to investigate the potential toxicity of $Al_2O_3$ NPs following repeated inhalation exposure in male Sprague-Dawley rats. Rats were exposed to $Al_2O_3$ NPs for 28 days (5 days/week) at doses of 0, 0.2, 1, and $5mg/m^3$ using a nose-only inhalation system. During the experimental period, we evaluated the clinical signs, body weight change, hematological and serum biochemical parameters, necropsy findings, organ weight, and histopathology findings. Additionally, we analyzed the bronchoalveolar lavage fluid (BALF), including differential leukocyte counts, and aluminum contents in the major organs and blood. Aluminum contents were the highest in lung tissues and showed a dose-dependent relationship in the exposure group. Histopathology showed alveolar macrophage accumulation in the lungs of rats in the $5mg/m^3$ group during exposure and recovery. These changes tended to increase at the end of the recovery period. In the BALF analysis, total cell and neutrophil counts and lactate dehydrogenase, tumor necrosis factor-${\alpha}$, and interleukin-6 levels significantly increased in the 1 and $5mg/m^3$ groups during exposure. Under the present experimental conditions, we suggested that the no-observed-adverse-effect level of $Al_2O_3$ NPs in male rats was $1mg/m^3$, and the target organ was the lung.

Effects of Bedding Material Composition in Deep Litter Systems on Bedding Characteristics and Growth Performance of Limousin Calves

  • The objective of this study was to evaluate the effects of different litter mixture compositions on bedding system temperature, pH and volatile fatty acid and ammonia-N ($NH_3$-N) content, and the serum physico-chemical parameters and growth indices of calves. Thirty-two Limousin calves ($280{\pm}20kg$) were randomly assigned to four groups (n = 8 for each group) according to the bedding system used: i) control with soil only (CTR); ii) mixture with 50% paddy hulls (PH), 30% saw dusts (SD), 10% peat moss (PM) and 10% corn cobs (CC) (TRT1); iii) mixture with 15% PH, 15% SD, 10% PM, 40% CC, and 20% corn stover (CS) (TRT2); iv) mixture with 30% PH, 10% PM, 40% CC, and 20% CS (TRT3). The litter material combinations of different treatments were based on the cost of bedding system materials in China. The cost of four treatments from low to high: Control$NH_3$-N level (271.83 to 894.72 mg/kg) was lowest for TRT1 (p<0.0001) and highest for TRT2 (p<0.0001). The acetate, propionate and butyrate levels were highest for the control group (p<0.0001). In all the groups, the pH value (6.90 to 9.09) increased at the beginning and later remained stable at below 9.09. The temperature of deep litter increased at the first week and reached the maximum ($42.1^{\circ}C$) on day 38. 3,5,3'-Triiodothyronine ($T_3$) levels in the TRT1 group animals (p<0.0001) were lower than those in the control and TRT2 animals. 3,5,3',5'-Tetraiodothyronine ($T_4$) in the TRT1 group (p = 0.006) was lower than that in the other treatment groups. Cortisol (COR) in the control and TRT1 group was lower (p<0.0001) than that in the TRT2 and TRT3 groups. Corticosterone (CORt) in the control group was higher (p<0.0001) than that in the treatment groups. The findings indicate that the deep litter bedding systems provided better conditions for animal health and growth performance compared with the control system. Furthermore, the litter composition of TRT1 was found to be optimal among the three treatment groups.

Bioequivalence of LG Clarithromycin Tablet to Klaricid Tablet (Clarithromycin 250 mg) (클래리시드 정(클래리스로마이신 250mg)에 대한 LG클래리스로마이신 정의 생물학적 동등성)

  • Bioequivalence of two clarithromycin tablets, the $Klaricid^{TM}$ (Ciba-Geigy Korea Ltd., Seoul, Korea) and the LG clarithromycin (LG Chemical Co., Ltd., Seoul, Korea), was evaluated according to the Korean Guidelines for Bioequivalence Test (KGBT 1998). Sixteen normal male volunteers $(20{\sim}26\;years\;old)$ were randomly divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After one tablet containing 250 mg of clarithromycin was orally administered, blood sample was taken at predetennined time intervals, and the concentrations of clarithromycin in serum were detennined using HPLC method with electrochemical detector. The pharmacokinetic parameters $(AUC_t,\;C_{max}\;and\; T_{max})$ were calculated and ANOVA was utilized for the statistical analysis of parameters. The results showed that the differences in $AUC_t$, $C_{max}$, and $T_{max}$ between two tablets based on $Klaricid^{TM}$ tablet were 4.06%,2.67% and -9.70%, respectively. The powers $(1-{\beta})$ for $AUC_t$, $C_{max}$ and $T_{max}$ were 83.53%, 92.34% and 96.64%, respectively. Detectable differences $({\Delta})$ and 90 % confidence intervals $(a=0.05) $were all less than ${\pm}20%$. All the parameters above met the criteria of KGBT 1998, indicating that LG clarithromycin tablet is bioequivalent to $Klaricid^{TM}$ tablet.

Cognitive Improvement Effects of Krill Oil in a Scopolamine-induced Mice Model (Scopolamine 유도 인지 저하 마우스 모델에서 크릴 오일의 인지 개선 효과)

  • A previous study showed that krill oil improved recognition and memory through anti-oxidative effects in an amyloid β model, but the authors noted that further investigations are necessary of alterations to neurotransmitters' states and of serum lipid profile improvements related to serum lipid peroxidation. Accordingly, in this study, ICR mice were pre-treated intraperitoneally with scopolamine prior to induced neurotransmission impairment, and the effects of krill oil provision on their capabilities of cognition were tested by performing a passive avoidance test (PAT), water maze test (WMT), and novel object recognition test. Then, parameters including the acetylcholine (ACh) concentration, acetylcholinesterase activity (AChE), lipid peroxidation, serum lipid levels, and nerve cell proliferation were investigated. The results showed that krill oil improved the mice's abilities in recognition and memory as the times taken to complete the PAT and WMT were reduced compared to the mice in a comparison scopolamine-treated group. Krill oil produced an increased concentration of Ach, and this was accompanied by a decrease in AChE. As shown in a scopolamine-treated SH-SY5Y cell line, krill oil reduced the activity of AChE. Moreover, the suppression of lipid peroxidation-reflected in the finding that malondialdehyde was decreased with krill oil provision-is speculated to affect the recorded serum triglyceride and cholesterol decreases and LDL cholesterol increase. The intake of krill oil was also found to produce an improvement in brain-derived neurotrophic factor expression by stimulating the activation of cyclic AMP response element binding protein in the brain tissue. Overall, the current results imply that the provision of krill oil raises the cognition and memory by elevating neurotransmitters and by improving the serum lipid profile and nerve cell proliferation, which occur as lipid peroxidation is suppressed in the brain tissue.
