Immunochemical Studies on Expression of Quinoproteins in Escherichia coli

  • An immunochemical method has been develooped as the most sensitive tool for studying the expression of quinoproteins containing pyrroloquinoline qinone(PQQ) in E. coli. The PQQ was conjugated to bovine serum albumin (BSA), and the conjugant was purified by using a $KwikSep^{TM}$ dextran desalting column chromatography. The PQQ-BSA conjugant was immunized to rabbits, and the IgG fractions of the antisera were purified. The most sensitive antibody against PQQ-BSA conjugant recognized some nanogram quantity of the antigen on the blot, but had little cross reactivity with BSA. Using this batch of the antibody, all the immunochemical assays of quinoproteins in E. coli were preformed. Some six different PQQ-specfic spots were detected by Western blot analysis of the soluble proteins in E. coli were performed. Their molecular weights on the blot were estimated to be about 100-, 90-, 72-, 58-, 52-, and 50kDa. Their pI values fell in the range from 4.8 to 5.5. These results stronly suggest that quinoproteins are present in E. coli, and that the protein moieties were covalently bound to PQQ.

대장균의 내열성장독소 측정법개발을 위한 단세포군항체의 생산 (Production of the Monoclonal Antibodies to the Escherichia coli Heat-Stable Enterotoxin)

  • Monoclonal antibody to the Escherichia coli heat-stable enterotoxin(ST) was produced to develop a rapid and convenient diagnostic method to the ST. The toxin was purified from culture supernatant of enterotoxigenic E. coli O148H28($ST^+/LT^+$) and conjugated to bovine serum albumin(BSA). The ST-BSA conjugate was used to immunize BALB/c mice and the immune spleen cells from these mice were fused with $P3{\times}63$ Ag8.V653 plasmacytoma cells. Hybridomas were screened by ELISA and positive hybridomas were cloned by limiting dilution. Finally, one stable clone (AS36) specific to ST was selected for further growth and characterization. Antibody titers of culture supernatant and ascitic fluid from BALB/c mice were 1:1,024 and 1:20,480 respectively in ELISA. The isotype and subclass of monoclonal antibody was IgG1 in sandwich ELISA. To test the neutralizing effect of monoclonal antibody on toxin activity of ST, mixture of ascitic fluid and ST was assayed by infant mouse assay and this monoclonel antibody was proved to be a neutralizing antibody. The titer of ascitic fluid which completely neutralized biological activity of 4 units of ST was 1:4. Purified ST was quantitatively measured by competitive ELISA and minimum amount of ST detectable by this assay was 250pg, which was an amount six-fold smaller than that detectable by infant mouse assay. Four reference strains of enterotoxigenic E. coli from WHO were detected by competitive ELISA and highly specific, sensitive and reproducible result was obtained.

Latex 응집반응과 간접형광항체법을 이용한 개 톡소플라즈마병의 혈청학적 진단 (Serodiagnosis of canine toxoplasmosis by latex agglutination and indirect fluorescent antibody test)

  • This study was conducted to determine the serum antibodies against toxoplasma in the artificially infected dogs, pet and street dogs by latex agglutination (LA) and indirect fluorescent antibody (IFA) test. LA test was carried out with commercial Toxo-MT kit (Eiken chemical Co.) and IFA test was carried out with rabbit-anti-dog IgG labelled with FITC (Cappel Co.) and toxo-antigen slides prepared in laboratory. The results obtained were as follows ; 1. Antibodies to Toxoplasma gondii in the artificially infected dogs were detected firstly at the Day 8 in IFA and Day 9 in LA test after inoculation. Positive antibody reactions by these tests were declined gradually afterward but maintained up to 12 weeks. 2. In LA test serum antibody titers in 310 test sera were shown as 10 cases(32%) in 1 : 32.5(1.0%) in 1 : 64, 4(1.3%) in 1 : 128 and 2(0.7%) in 1 : 256. In IFA test serum antibody titers 310 test sera were shown as 17 cases(5.5%) in 1 : 64, 8(2.6%) in 1 : 128 and 5(1.6%) in 1 : 256. 3. In the total of 310 sera from pet and street dogs toxoplasma antibody positive rates were 21 cases (6.8%) by LA and 30 cases (9.7%) by IFA test and the positive detection rates between these two groups by LA and IFA test were not significant(p<0.05). 4. In the total of 115 sera from pet dogs toxoplasma antibody positive rates were 12 cases(10.4%) by LA and 15(13.0%) by IFA test. And in the 195 street dogs the positive rates were 9 cases(4.6%) by LA and 15(7.7%) by IFA test. Also, the positive detection rates between these two groups by LA and IFA test were not significant(p<0.05). 5. Agreement of reactivity between LA and IFA test for 310 sera was 91.3% in total of 283 cases consisting of 12 cases(3.9%) of both LA and IFA positive and 271 cases(87.4%) of LA and IFA negative. 6. LA test was almostly equivalent to the IFA test in producibility and proved to be a simple tool for the screening of toxoplasma antibody in laboratory.

  • An American Cocker Spaniel (3-year-old, intact female, 6.0 kg) was referred to the Veterinary Medical Teaching Hospital of Chungnam National University for evaluation of pustules and crusts in the periocular region, dorsal and ventral region of the trunk, and digits. Complete blood count (CBC) revealed leukocytosis with mature neutrophilia, and a serum biochemistry profile revealed hypoalbuminemia. Tape strip tests identified numerous neutrophils and acatholytic cells. Histopathology identified intraepithelial pustules with neutrophils and acantholytic keratinocytes. Definitive diagnosis of pemphigus foliaceus (PF) was made by direct immunofluorescence (DIF) test with goat anti-canine IgG antibody. The human intravenous immunoglobulin (IVIG) was administered at a rate of 15 ml/h over 6 hours for 4 days. After that, the dog was maintained on prednisolone (2.2 mg/kg, PO, SID) and azathioprine (2.0 m/kg, PO, SID). An infusion of IVIG (0.5 g/kg) was repeated 3 days after 4 weeks. After 10 weeks, the dog showed the remarkable regression of lesions.

Increased Antitumor Immunity of Mouse GM-CSF in Mouse Colon Tumor (CT-26) Model

  • Oncolytic vaccinia virus is an engineered vaccinia virus that selectively destroys cancer cells and induces tumor immune response. Oncolytic vaccinia expressing mouse GM-CSF showed cytotoxic activity against various kinds of cancer cells when oncolytic vaccinia virus expressing human GM-CSF and mouse GM-CSF is intravenously administered in the mouse CT26 colon tumor model. Cancer cells treated with isolated immunoglobulin G from the serum with complement showed these cytotoxic activity and complement observed dose-dependent cytotoxic effect. These results suggest that oncolytic vaccinia virus expressing mouse GM-CSF can increase oncolytic vaccinia virus by inducing anticancer antibody in a mouse tumor model. Further studies are needed on antitumor immunity of GM-CSF.

  • 효소면역측정법에 의한 aflatoxin $B_1(AFB_1)$의 정량법을 개발하기 위하여, 항체를 생산.정제하여 분석법을 확립하고 직접법과 간접법(direct/indirect competitive ELISA)의 특성 및 문제점을 비교. 검토하였다. Bovine serum albumin(BSA)을 carrier protein으로 한 $AFB_1$-1-(O-carboxymethyl)oxime -BSA를 토끼에 면역하여 항$AFB_1$항혈청을 생산하였다. 정량 침강반응에 의하여 항혈청으로부터 항BSA항체를 제거하고 황산암모늄 침전법 및 EDAE-Sephadex A-50 이온교환 크로마토그래피를 통하여 순도 높은 IgG항체를 정제하여 항$AFB_1$항체를 사용하였다.

효소면역측정법을 이용한 스파르가눔증의 혈청학적 진단 (Serological Diagnosis of Human Sparganosis by means of micro-ELISA)

  • 외과적으로 충체를 적출하여 진단한 스파르가눔증 환자 7명으로부터 혈청을 수집하고 혈청내 스파르가눔 특이 IgG 항체가를 호소면역 측정 법으로 측정하였다. 스파르가눔 항원은 유혈목이에 자쳔 감염된 충체를 갈아 생리 식염수로 추출한 것으로 단백질 함량은 Smgyxnl이었다. 효소면역측정법은 McLaren 등(1978)의 방법에 따라 실시하였다. 스파르가눔중 환자 혈청이 폐흡충 항원 또는 유구낭미충 항원에 교차반응이 있는지 여부도 측정하였다. 또한 스파르가눔 항원에 대해서 정상인, 케흡충중 환자, 간홉충 감염자, 유구낭미충증 환자와 무구조충 감염자 등 모두 71명에 대해서도 교차반응을 나타내는지 를 관찰하고자 효소면역 측정 법을 실시하였다. 그 결과를 요약하면 다음과 같다. 1. 스파르가늠증 환자 7명중 석회화한 스파르가눔에 감염되어 있었고 수술후 1개월에 혈청을 수집한 1례를 제외하면 모두 홉광도 0.3 이상을 나타내었다. 양성판정기준을 흠광도 0.25로 하였을 때 효소면역 측정 법의 민감도(sensitivity)는 85.7이 었다. 2. 기타 기생충 감염자와 정상인 71명중에서 스파르가눔 항원에 대한 특이항체 양성자는 무구조충감염자 3명으로 특이도(specificity)는 97.5%이었다. 3. 스파르가눔중 환자 혈청은 폐흡충 항원 및 유구낭미충 항원에 대해서 항체가는 낮은 값을 보이고 있어 교차 반응은 발견할 수 없었다. 4. 중추신경계를 침범한 스파르가눔중 환자 2례에서 뇌척수액을 희석하지 않은 상태로 효소면역 측정 법을 실시하였던 바 1례는 폐흡충 및 유구낭미충 항원에 대해 교차반응 없이 스파르가눔항원에 대해 높은 항체가를 보인데 비해 다른 1례에서는 스파르가눔 항원 및 유구낭미충 항원에 대해 비슷한 항체가를 나타내고 있었다. 이상의 결과에서 효소면역 측정 법을 이용하여 스파르가눔중을 혈청학적으로 진단하는 경우 매우 특이하고 민감한 결과를 얻을 수 있을 것으로 생각하였다. 스파르가눔중의 수술전 진단이 실제로 어려운 것이 현실이므로 혈청학적 잖단은 역학적 유해조사나 형태학적으로 감별이 어려운 병리소견을 보인 경우 등에서 보조적인 진단법으로 이용할 수 있을 것으로 생각한다.

Effects of Chromium Propionate Supplementation on Growth Performance, Serum Traits and Immune Response in Weaned Pigs

  • This study investigated the effects of chromium propionate on growth performance, serum traits and immune response in weaned pigs. Twenty-four 4 wk-old crossbred weanling pigs (initial body weight about 9.52${\pm}$0.48 kg) were randomly allotted into one of two groups, a control group (basal diet), chromium propionate group (diet supplemented with 200 ${\mu}g$ $kg^{-1}$ (ppb) of chromium propionate). This experiment was conducted over nine weeks. Escherichia coli lipopolysaccharide (LPS) 100 ${\mu}g$ $kg^{-1}$ BW was used as the stress-inducing agent in the middle (4 wks) and final (8 wks) periods. The experimental results indicated that chromium propionate had no effect on growth performance (p>0.05). Chromium propionate supplementation reduced the percentage of LDL+VLDL (low and very low-density lipoprotein) and increased HDL (high-density lipoprotein), but did not affect other serum traits. Pigs supplemented with chromium propionate had higher antibody titers specific for sheep red blood cells (SRBC) and serum total globulin relative to the control during the final period (p<0.05). A challenge with LPS increased white blood cells in the chromium propionate group in both experimental periods (p<0.05). The chromium propionate group exhibited higher IgG and $\gamma$-globulin than the control during the middle experimental period (p<0.05). Moreover, the PHA (phytohemagglutinin) challenge result in the chromium propionate group was better than the control group (p=0.056). Greater neutrophil activity was displayed than in the control (p<0.05). This suggests that chromium propionate supplementation benefited the weaned pigs in lipoprotein and immune response.

Prevalence of Toxoplasma gondii in Dogs in Zhanjiang, Southern China

  • Toxoplasmosis, caused by Toxoplasma gondii, is a parasitic zoonosis with worldwide distribution. The present study investigated the prevalence of T. gondii in dogs in Zhanjiang city, southern China, using both serological and molecular detection. A total of 364 serum samples and 432 liver tissue samples were collected from the slaughter house between December 2012 and January 2013 and were examined for T. gondii IgG antibody by ELISA and T. gondii DNA by semi-nested PCR based on B1 gene, respectively. The overall seroprevalence of T. gondii IgG antibody was 51.9%, and T. gondii DNA was detected in 37 of 432 (8.6%) liver tissue samples. These positive DNA samples were analyzed by PCR-RFLP at 3'- and 5'-SAG2. Only 8 samples gave the PCR-RFLP data, and they were all classified as type I, which may suggest that the T. gondii isolates from dogs in Zhanjiang city may represent type I or type I variant. This study revealed the high prevalence of T. gondii infection in dogs in Zhanjiang city, southern China. Integrated measures should be taken to prevent and control toxoplasmosis in dogs in this area for public health concern.

Autogenous toxoid-bacterin treatment for Staphylococcus aureus subclinical mastitis in lactating cows

  • To evaluate clinical effects of autogenous toxoid-bacterin treatment for Staphylococcus aureus subclinical mastitis in lactating cows, 22 cows which had at least one S aureus infected quarter were selected from one S aureus prevalent dairy farm, of which 11 cows were injected their own autogenous toxoid-bacterin and the others were maintain as noninjected control. In the toxoid-bacterin injected group, 27% of infected quarters were cured during the 12 weeks trials as compared to 5% in the control group. New intramammary infections with S aureus were only detected in three-quarters of the control group. Mean IgG antibody titers against S aureus somatic antigens and $\alpha$-toxin in serum and milk were significantly increased in the toxoid-bacterin injected group (p < 0.05) and remained higher than those of the control group which showed no significant changes (p < 0.05). From 3 weeks after second injection (at 7 week), mean S aureus CFU/ml in milk samples from previously infected quarters with S aureus of the toxoid-bacterin injected group was lower than that from preinjection state (p < 0.05). In the toxoid-bacterin injected group, significant decrease of mean SCC was detected from milk samples from previously infected quarters with S aureus from week 7 to week 10 (p < 0.05). These data suggested that autogenous toxoid-bacterin treatment aganist S aureus subclinical mastitis in lactating cows might increase the cure rate of the infections, reduce the severity of the infections and also prevent occurrence of the new infections.
