Heat-Shock Protein 70 as a Tumor Antigen for in vitro Dendritic Cell Pulsing in Renal Cell Carcinoma Cases

  • Immunological functions of heat shock proteins (HSPs) have long been recognized. In this study we aimed to efficiently purify HSP70 from renal cell carcinoma and test it as a tumor antigen for pulsing dendritic cells in vitro. HSP70 was purified from renal cell carcinoma specimens by serial column chromatography on Con A-sepharose, PD-10, ADP-agarose and DEAE-cellulose, and finally subjected to fast protein liquid chromatography (FPLC). Dendritic cells derived from the adherent fraction of peripheral blood mononuclear cells were cultured in the presence of IL-4 and GM-CSF and exposed to tumor HSP70. After 24 hours, dendritic cells were phenotypically characterized by flow cytometry. T cells obtained from the non-adherent fraction of peripheral blood mononuclear cells were then co-cultured with HSP70-pulsed dendritic cells and after 3 days T cell cytotoxicity towards primary cultured renal cell carcinoma cells was examined by Cell Counting Kit-8 assay. Dendritic cells pulsed in vitro with tumor-derived HSP70 expressed higher levels of CD83, CD80, CD86 and HLA-DR maturation markers than those pulsed with tumor cell lysate and comparable to that of dendritic cells pulsed with tumor cell lysate plus TNF-${\alpha}$. Concomitantly, cytotoxic T-lymphocytes induced by HSP70-pulsed dendritic cells presented the highest cytotoxic activity. There were no significant differences when using homologous or autologous HSP70 as the tumor antigen. HSP70 can be efficiently purified by chromatography and induces in vitro dendritic cell maturation in the absence of TNF-${\alpha}$. Conspecific HSP70 may effectively be used as a tumor antigen to pulse dendritic cells in vitro.

Purification and Characterization of a Fibrinolytic Enzyme Produced by Bacillus amyloliquefaciens HC188 (Bacillus amyloliquefaciens HC188이 생산하는 혈전분해 효소의 정제 및 특성)

  • A bacterium producing a fibrinolytic enzyme was isolated from Cheonggukjang. The bacterium was identified as a strain of Bacillus amyloliquefaciens by 16S rDNA analysis and designated as B. amyloliquefaciens HC188. The optimum culture medium appeared to be one containing 0.5% (w/v) maltose and 0.5% (w/v) soytone. Bacterial growth in the optimal medium at $37^{\circ}C$ reached the stationary phase after 27 h of incubation and the fibrinolytic enzyme showed optimum activity at 24 h. The enzyme was purified by 20-80% ammonium sulfate precipitation, CM Sepharose fast flow ion exchange chromatography, and Sephacryl S-200HR column chromatography. Its specific activity was 38359.3 units/mg protein and the yield was 5.5% of the total activity of the crude extracts. The molecular weight was 24.7 kDa and the amino acids of the N-terminal sequence were AQSVPYGVSQIKAPA. The fibrinolytic enzyme activity had an optimum temperature of $40^{\circ}C$ and an optimum pH of 8.0, and the enzyme was stable in the ranges $20-40^{\circ}C$ and pH 6.0-8.0. Enzyme activity was increased by $Ca^{2+}$ and $Co^{2+}$ but inhibited by $Cu^{2+}$, EDTA, and PMSF. It is suggested that the purified enzyme is a metallo-serine protease.

Production and Characterization of Raw Starch Hydrolyzing Enzyme from Bacteria (세균에 의한 생전분 분해효소의 생성 및 특성)

  • A bacterium capable of hydrotyzing raw starch was isolated from soil, which was identified as a strain of Bacillue. The effects of culture conditions and medium compositions on the enzyme production were investigated. Among tested carbon sources, soluble starch and wheat starch were most effective for the production of the enzyme, and the level of concentration for the optimal enzyme production was 0.5%. For nitrogen sources, polypeptone was best for the enzyme production, with the level of 0.5%. The enzyme was maximally produced by cultivating the organism at medium of initial pH 6.5, and temperature of $35^{\circ}C$. The enzyme was partially purified by Sepharose CL-6B gel filtration and DEAESephacel ion-exchange chromatography. The optimal pH and temperature for the enzyme reaction were 6.5 and $70^{\circ}C$, respectively. The enzyme most stable at pH 8.0, and temperature up to $60^{\circ}C$. In kinetic studies, the k, values for corn, wheat, rice and potato starch were 1.7, 1.4,2.5 and 1.090, respectively.

Extraction, purification and properties of anti-complementary polysaccharide from Arecae Pericarpium (대복피로부터 항보체 활성다당의 추출, 정제 및 그 특성)

  • To examine the characteristics of anti-complementary compounds from Arecae Pericarpium (the pericarps of Areca catechu) which showed the highest activity during our screening procedures, the extraction and purification were performed. AC-1 fraction from Arecae Pericarpium was purified by hot water extraction, methanol reflux, ethanol precipitation, dialysis and lyophilization. This compound had total sugar 48.2%, uronic acid 14.6% and protein 36.8%. Rhamnose, arabinose, mannose and galactose were found in sugar components. By cetavlon (cetyltrimethylammonium bromide) treatment AC-1 was fractionated to AC-2, AC-3 and AC-4. Among them, AC-2 showed the highest activity and yield. By periodate oxidation, AC-2 was deactivated, but had no change in activity by pronase digestion. Moreover active fractions, AC-2-IIIa and AC-2-IIIc isolated from AC-2 by two successive column chromatography using DEAE-Toyopearl $650C(Cl^-form)$ and Sephadex G-100. AC-2-IIIa was mainly made up of rhamnose, mannose, galactose and glucose, and AC-2-IIIc, mannose, galactose and glucose. These both polysaccharides were identified as homogeneous by gel filtration of Sepharose CL-4B and electrophoresis, and molecular weights of them were 120,000 and 15,000, respectively.

  • The molecular weight 30 kDa membrane protein of Toxoplusma Sondii (Toxoplasma 30 kDa) apparently conserved in most strains of T. gondii and sera of infected hosts. The present study aimed to elucidate Toxoplasmc 30 kDa as a useful diagnotic antigen for serodiagnisis of toxoplasmosis by ELISA and for induction of protective immunity. Murine spleen cells immunized with the membrane antigen of T. gondii were fused with mouse Sp2/0-Ag 14 myeloma cells. Out of 8 clones selected, five were IgG2b, the others belonged to IgG 1 and IgG2a. The 30 kDa antigen was distributed mainly on the surface membrane of tachyzoites by indirect fluorescence method. Murine peritoneal macrophages which were activated by 30 kDa antigen produced more amounts of NO2 compared with crude antigen-treated group, however there were no significant differences in toxoplamacidal activity between the two groups. Higher specificity of Toxoplosma 30 kDa antigen was recognized for serodiagnosis of toxoplasmosis than the crude antigen. From these results, ToxopLasmo 30 kDa antigen enhances the cytotoxic effect of macrophages as well as a more reliable means for the serodiagnosis of toxoplasmosis by ELISA.   • On the five interspecific protoplast fusants of Ganoderma lucidum and G. applanatum was the antitumor test performed. The fusant F-2 was selected, to examine the cultured mycelia (protein bound polysaccharide) as antitumor components. When a dose of 20 mg/kg/day of each components purifed from F-2 fusant was, i.p., injected into ICR mice, the inhibition ratio of Fr. II against the solid form of sarcoma 180 increased to 1.5 times as compared with that of their parents. When Fr. II was examined for immunopotentiation activity, it increased the amount of the superoxide anion in activated macrophages to 1.2 times and the count of hemolytic plaque forming cells in the spleen to 4.3 times as compared with that of each control group. Its chemical analysis showed 85.2% polysaccharide which consisted of glucose, galactose, mannose, fucose and xylose, and 0.39% protein of 15 amino acids. The content of hexosamine was 0.39% and the molecular weight of Fr. V was $5.6{\times}10^4$ dalton.

Gene Cloning, Nucleotide Sequence and Efficent Expression of Peptidyl proryl cis-trans Isomerase from Bacillus stearothermophilus (Bacillus stearothermophilus의 Peptidyl Prolyl cis-trans Isomerase 유전자 분리 염기배열 및 발현)

  • A PPIase gene of Bacillus stearothermophilus was screened from a genomic library by plaque hybridization using the A-1 primer as a probe. A PPIase positive plaque contained a 3.0kb insert of the chromosomal DNA. A 3.0kb fragment was subcloned into pUC18, resulting pPI1-40. A DNA fragment encoding the N-terminal portion of the PPIase in pPi-40 was amplified by polymerase chain reaction(PCR) method using the A-1 and B-2 primers. The amplified fragment was cloned into the Sma I site of pUC18 and recombinant plasmid was designated as pSN-18. The nucleotide sequence of 167bp fragment was determined. The deduced amino acid sequence of PPIase was completely matched with the determined N-terminal amino acid sequence of PPIase B. stearothermophilus. The translated protein sequence of PPIase B. stearothermophilus was compared with sequence from periplasmic PPIase from Escherichina coil ; homogies of 16 and 58%, respectively, were found. The clond PPIase gene was over-expressed in E. coil cell using pUC19 as an expression vector. The enzyme was partially purified by heat treatment and colum chromatochraphy on DEAE-Sepharose CL-6B. The molecular weight of the enzyme was dermined to be about 18.0 kDal by SDS-PAGE.

Purification and Characterization of Exo-xylanase from Escherichia coli Cells Harboring the Recombinant Plasmid pMGl (재조합 균주 Escherichia coli가 생산하는 Bacillus stearothermophilus Exo-xylanase의 정제 및 특성)

  • Exo-xylanase encoded by the xylA gene of Bacillus stearothermoPhillus was produced from Escherichia coli ]M109 carrying a recombinant plasmid pMGL Synthesis of the enzyme was observed to be cell-associated, and about 94% of the enzyme synthesized was located in the cytoplasmic region. The maximum production was attained when the E. coli strain was grown at $37^{\circ}C$ for 8 hours on the medium containing 0.5% fructose, 1.0% tryptone, 1.0% sodium chloride, and 0.5% yeast extract. The exo-xylanase was purified to homogeneity using a combination of salting out with ammonium sulfate, DEAE-Sepharose CL-6B ion exchange chromatography, Sephadex G-IOO gel filtration, and Sephadex G-150 gel filtration. The' purified enzyme was most active at pH 6.0 and $45^{\circ}C$. $Ca^{2+}$ and $Co^{2+}$ activated the exo-xylanase activity by about 20% while $Ag^{2+}$, $Fe^{2+}$, $Mg^{2+}$ and $Zn^{2+}$ inhibited the enzyme activity by up to 60%. The $K_m$, value on p-nitrophenyl-$\beta$-D-xylanopyranoside was 2.75 mM. The enzyme had a pI value of 4.7. The estimated molecular weight of the native protein was 200,000 daL SDS-polyacrylamide gel electrophoresis analysis suggested that the native enzyme was a trimer composed of three identical 66,000 da!. polypeptides. The purified enzyme efficiently converted all the xylo-oligosaccharides tested to xylose. It was also confirmed that the enzyme split xylans in an exo-manner even though the degree of hydrolysis was fairly low. The xylanolytic enzyme was, therefore, classified to be one of the few bacterial exo-xylanases lacking transferase activity.

Structural Properties of Cold Water Extractable ${\alpha}-D-glucan$ in Rice Flours (쌀가루 냉수추출 ${\alpha}-D-glucan$의 분자구조적 특성)

  • The structural properties of cold water extractable ${\alpha}-D-glucan$ in rice flours obtained by various milling methods were investigated. The blue value, ${\lambda}_{max}$ values of the iodine complex and ${\beta}-amylolysis$ limit of the cold water extractable ${\alpha}-D-glucan$ were in the range of $0.026{\sim}0.030,\;518{\sim}522\;nm$ and $52.7{\sim}59.6%$, respectively, indicating these materials were composed mainly of amylopectin-like polymer. The gel chromatography on Sepharose CL-2B indicated that the cold water extractable ${\alpha}-D-glucan$ had lower molecular weight but wider molecular weight distribution than that of the original rice amylopectin. The Sephadex G-50 gel chromatography showed that the unit chain distribution of cold water extratable ${\alpha}-D-glucan$ after debranching with pullulanase was similar to that of rice amylopectin. And the ion chromatography of the distribution pattern of the chain length below D.P.30 in the debranched ${\alpha}-D-glucan$ indicated only blade-milled flour was similar to the amylopectin, but in the amount of unit chain below D.P.10, all the ${\alpha}-D-glucan$ in rice flours was higher than that of amylopectin.

Extraction and Chemical Composition of Soluble Polysaccharide from Green Laver, Enteromorpha prolifera (가시파래 수용성 다당의 추출 및 화학적 조성)

  • Soluble polysaccharide (SP) from green layer, Enteromorpba prolifera was extracted 3 times with distilled water at 100$^{\circ}C$ for 2 hrs and fractionated with cetylpyridinium chloride (CPC) and ion exchange chromatography (DEAE Separose CL-6B). The SP amounted to $23.7\%$ of the dry seaweed weight and contained $68.8\%$ carbohydrate. It was mainly constituted of rhamnose, glucose, xylose, sulfate and uronic acid and was fractionated with CPC into three (CPC-S, CPC-PS, CPC-PP) tractions. The major acid fraction CPC-PS accounted for $10.2\%$ of the dry algal weight. CPC-PS was further fractionated on DEAE Sepharose CL-6B into Fr-1 ($8.0\%$), Fr-2 ($35.8\%$), Fr-3 ($23.7\%$) fractions. The Fr-3 fraction contained $2.2\%$ protein, $21.4\%$ sulfate, $15.3\%$ uronic acid, and $72.4\%$ polysacchnrides composed of rhamnose, xylose and glucose. The Fr-2 fraction, which was richer in uronic acid ($17.5\%$) and poorer in sulfate ($19.0\%$) and total sugar ($68.8\%$) than the Fr-3, had a sugar composition close to that of Fr-3. The average molecular weights of Fr-2 and Fr-3 were 510,000 and 830,000 daltons, respectively. Fr-3 turned out to be homogeneous by cellulose acetate electrophoresis.
