Enhancement of DNA-mediated Energy Transfer from Ethidium to meso-Tetrakis(N-methylpyridinium-4-yl)porphyrin by Ca2+ Ion

  • The fluorescence intensity of DNA-intercalated ethidium with [ethidium]/[DNA base] being 0.005 was quenched upon the binding of another intercalating ligand, meso-tetrakis(N-methylpyridinium-4-yl)porphyrin (TMPyP). Addition of $Ca^{2+}$ enhanced the quenching efficiency. The range of separations between donor and acceptor molecules, within which total quenching occurs, was calculated using a one-dimensional resonance energy transfer mechanism to be 9.5 base-pairs or $32.3{\AA}$ in the absence of $Ca^{2+}$ ions. The distance increased to 18.7 base-pairs or about $63.6{\AA}$ in the presence $100{\mu}M$ $Ca^{2+}$. Considering that (1) $Ca^{2+}$ had little effect on the binding modes of ethidium and TMPyP, which was investigated by reduced linear dichroism and (2) spectral overlap between the emission spectrum of ethidium and the absorption spectrum of TMPyP was maintained in the presence of $Ca^{2+}$, contributions from orientation factor and spectral overlap to $Ca^{2+}$-induced enhancement in DNA mediated energy transfer was limited. Although there is no direct evidence, electron transfer along the DNA stem may accompany the observed fluorescence quenching. In this respect, DNA bound $Ca^{2+}$ act as a partially conducting medium.

Evaluation of Crack-tip Cohesive Laws for the Mode I Fracture of the Graphene from Molecular Dynamics Simulations (그래핀의 모드 I 균열에 대한 분자동역학 해석으로부터 균열 선단 응집 법칙의 평가)

  • In this paper, a novel approach to estimate cohesive laws for the mode I fracture of the graphene is presented by combining molecular dynamic simulations and an inverse algorithm based on field projection method and finite element method. The determination of crack-tip cohesive laws of the graphene based on continuum mechanics is a non-trivial inverse problem of finding unknown tractions and separations from atomic simulations. The displacements of molecular dynamic simulations in a region far away from the crack tip are transferred to finite element nodes by using moving least square approximation. Inverse analyses for extracting unknown cohesive tractions and separation behind the crack tip can be carried out by using conservation nature of the interaction J- and M-integrals with numerical auxiliary fields which are generated by systematically imposing uniform surface tractions element-by-element along the crack surfaces in finite element models. The preset method can be a very successful approach to extract crack-tip cohesive laws from molecular dynamic simulations as a scale bridging method.

Reversed-Phase Ion-Pair High Performance Liquid Chromatographic Elution Behavior of Noble Metal-Thiacrown Ether Complexes (귀금속-티아크라운에테르 착물들의 역상 이온쌍 고성능 액체크로마토그래피 용리거동)

  • In the reversed-phase ion-pair high performance liquid chromatographic (RPIP-HPLC) elution behavior of noble metal-thiacrown ether complexes, the effects of the concentration of ion-pairing reagent and kind of ligands were studied. It was found that the less the number of atoms in the ring of the thiacrown ether molecule was, the larger the selectivity was, and the elution mechanism of the complexes was explained due to the formation of ion-pair when the concentration of sodium dodecyl sulfate (SDS) in mobile phase was lower than 10 mM and due to the formation of micelle when the SDS concentration was higher than 10 mM. As a conclusion, separations of the noble metal-thiacrown ether complexes in an optimum separation condition were accomplished successfully and the method was proved to be an useful one for the separation and determination of Ag (Ⅰ) ion in a black-white photographic fixing solution.

  • Supercritical Fluid Chromatography(SFC) has become a technique for solving problems that are difficult by other chromatographic methods. However, the most widely used fluid, is no more polar than hexane. Polar samples which are difficult to be analyzed with pure supercritical C$O_2$ because of their high polarity can be separated by adding polar modifiers to supercritical C$O_2$. In this paper, a new method for monitering the mobile phase composition in modified supercritical fluid chromatography was developed. The amount of water dissolved in supercritical C$O_2$ was measured by amperometric microsensor which is made of thin film of perfluorosulfonate ionomer (PFSI). The amount of water dissolved in supercritical C$O_2$ stayed constant for a much longer time than with a saturator column. With this new mixing device, we could do good separations for insecticides and fungicides which are difficult to separate with pure C$O_2$.

  • A three dimensional numerical analysis has been conducted to analyze the effects of a pipe angle, connecting a combustion chamber and a pintle nozzle, and pintle position on pintle nozzle performance. The compressibility correction of $k-{\omega}$ SST turbulent model was implemented to precisely predict the characteristics of complex flow structures inside a supersonic pintle nozzle. Due to an 3-D asymmetric pintle nozzle configuration, complex helical flow streamlines and large flow separations were observed, which resulting in significant nozzle performance losses. As the angle of connection-tube decreases, the coefficient of performance increases and Since the flow structures are evidently changed to the pintle stroke position, the performance characteristics was analyzed.

Synthesis of $H_2$-Permselective Silica Films by Chemical Vapor Deposition (화학증착(CVD)에 의한 선택적 수소 투과성 실리카막의 제조)

  • Hydrogen-permselective silica membranes were synthesized within tim walls of porous Vycor tubes by chemical vapor depostion of $SiO_2$. Film deposition was carried out using $SiCl_4$ hydrolysis either in the oppm shag reactants or in the one-sided geometry. At temperatures above $600^{\circ}C$ the permeation rate of hydrogen thorough the silica films varied between 0.01 and $025cm^3(STP)/cm^2-min-atm$ depending on the reaction geometry and the $H_2 : N_2$ permeation ratio was about 1000. Permeation rates of both $H_2$ and $N_2$ increased with increasing temperature. The silica membranes produced by one-sided deposition have higher hydrogen permmeation rates than those produced by the opposing reactants geometry although the membranes formed in an opposing reactants geometry were relatively stable during the heat treatment or after exposure to ambient air. These membranes can be applied to high temperature gas separations or membrane reactors once the film deposition process is optimized to get high permeability as well as good stability.

Statistics on Radiation Field Waveforms Associated with Multiple Intracloud Lightning Discharges

  • This paper presents the detailed statistics on radiation field signatures associated with multiple-intracloud lightning discharges. A transient signal recording system was used to measure the electric and magnetic fields produced by lightning flashes. The measurements were made in th summers of 1995 through 1999, and the location of the observation station was in Inchon on the coast of the Yellow Sea in Korea(37$^{\circ}$25'N, 126$^{\circ}$ 39'E). Most of lightning flashes typically contains between two and five strokes. The individual intracloud stork radiation fields were the bipolar pulse. On the average, the ratio of the peak of the second stroke to the first stroke peak was 75.1$\pm$40.1% for the negative, and a fraction of the subsequent stroke peaks were higher than the first stroke peak. The greater the number of the subsequent stroke order, less time separations between strokes were produced. The mean of the depth of the dip was 81.2$\pm$27.9% for the positive polarity and 75.9$\pm$24.4% for the negative. The depth of the dip increased for the positive bipolar pulses and decreased for the negative as the number of the stroke order increased.

Numerical Analysis of Reflection Characteristics of Perforated Breakwater with a Resonant Channel (공진수로 내장형 유공방파제의 반사특성에 관한 수치해석 연구)

  • In this study, a new concept perforated breakwater is proposed, which is having resonant channels. In the channel, perforated plate is installed for dissipating wave energy induced by flow separations. The breakwater has two advantages compared with conventional perforated breakwater having wave chamber with slotted walls. One is easy to control the target wave condition for dissipating wave energy, and the other is having the high structural safety because the structural members are not exposed to impact waves, directly. To evaluate wave reflection characteristics of the proposed breakwater, numerical experiment was carried out by using Galerkin's finite element model based on the linear potential theory. The results indicated that considerable energy dissipation occurs near the resonant period of channel, and wave reflection characteristics are affected by channel shape, location and opening ratio.

A Novel Fluoroscopic View for Positioning the AO Clavicle Hook Plate Decreases Its Associated in situ Complications

  • Background: The goal of this study was to evaluate whether a modified fluoroscopic technique for positioning a hook plate affected the clinical results of treating Neer type II distal clavicle fractures and Rockwood type V acromioclavicular (AC) joint separations with this device. Methods: The study was a retrospective consecutive case series with data analysis. Sixty-four patients with a Neer type II distal clavicle fracture or a Rockwood type V AC joint injury treated between March 2009 and June 2013 were divided into 2 groups: traditional fluoroscopic technique (traditional view, 31 patients) or modified fluoroscopic technique ('hook' view, 33 patients). A visual analogue scale (VAS) score, the modified University of California-Los Angeles (UCLA) shoulder scale score, and radiographic osteolysis were the main outcome measures. Results: The traditional group included a significantly larger number of patients with acromial osteolysis than the hook view group: 23 patients (74.2%) vs. 11 patients (33.3%), respectively (p=0.01). Before plate removal, the hook group reported less pain and higher UCLA shoulder scale scores than the traditional group: average VAS score, 1.55 vs. 2.26, respectively; average UCLA score, 30.88 vs. 27.06, respectively. However, there was no significant difference after plate removal. Conclusions: The hook view allows more accurate bending of the hook plate around the contour of the acromion, resulting in decreased osteolysis, decreased pain, and better function with the plate in situ.

Clinical and Radiological Outcomes of Acute Acromioclavicular Joint Dislocation: Comparison of Hook Plate Fixation with Single Tight Rope Technique

  • Background: This study was conducted to compare the clinical and radiological outcomes of the locking hook plate fixation (HP) technique and the single tight rope (TR) technique applied for acute high-grade acromioclavicular (AC) joint separations. Methods: Between 2009 and 2014, 135 consecutive patients with acute AC joint separation Rockwood types III, IV, and V were subjected to surgical reconstruction. One hundred fourteen patients (84.4%) were available for retrospective evaluation. Of them, 62 and 52 were treated using the single TR group and clavicular HP group techniques, respectively. The visual analogue scale, Constant, American Shoulder and Elbow Surgeons (ASES), and Taft scores were used for clinical assessment. Postoperative shoulder range of motion was also assessed. An anteroposterior radiograph of the coracoclavicular distance (CCD) was obtained to evaluate the radiographic signs of recurrence. Results: The TR group patients had better Constant, ASES, and Taft scores than the HP group patients. The loss of reduction in terms of the CCD did not differ between groups. Subacromial osteolysis was observed in 34.6% of the cases in the HP group. However, there were no significant differences in the clinical outcomes between the patients with and without osteolysis in the HP group. Subcoracoid osteolysis, drill tunnel widening, and metal displacement were observed in 3.2%, 22.6%, and 4.8% of the cases in the TR group, respectively. Conclusions: The single TR technique was relatively more effective at treating acute high-grade AC joint injuries than the HP fixation technique (level of evidence: therapeutic; retrospective comparative study, Level III).
