Inhibitory Action of the Paraquat on Superoxide Dismutase of Excherichia coli (Paraquat에 의한 Escherichia coli의 Superoxide Dismutase 활성저해)

  • Actively growin Excherichia coli(KCTC 1039) cells were treated with paraquat (1, 1'-dimethyl-4, 4'-bipyridili-um dichloride) by cultivating them in the presence of 1.0mM paraquat. The treatment was carried out with or without shaking to understand the effect of oxygen on paraquat action to thebacterial superoxide dismutase (SOd). By the treatment with vigorous shaking , population growth of the organism almostly stopped and specific activities of SOD of the cells drastically decreased. On contrast ot it, the herbicide showed only l limited inhibitory action on bacterial growth and SOD activity by stationary treatment. Proteins prepared from parquat-treated cells divided into two peaks by Sephacryl column chormatogrpahy, while proteins from the intact cells formed a single peak. Cytoplasmic proteins and plasma membrane proteins of intact cells formed separated three peaks by Sephadex G-75 column chormatography. respectively. Among them the second peak disappeared by paraquat treatment , while the third peak became more apparent. Fractions from the first and the third peak showed SOD activity. Paraquat was detected from the same fractions.

  • Mutagenic characteristics deactivation of 7, 12-dimethylbenz(a)anthraracene was studied in a nonacclimated sandy loam soil at low and neutral pH soil conditions. Soil extracts containing transformation products were separated into three fractions based on HPLC retention time(polarity). Highly polar transformation products of 7, 12-dimethylbenz(a)anthracene demonstrated a negative mutagenic response with the Ames mutagenicity assay, strain TA-100, for both low and neutral pH soils. Moderate and low polar fractions, however, induced mutagenicity for both soil samples with mutagenic ratios similar to those of the parent compound.

Effect of Proanthocyanidin-rich Extracts from Pinus radiata Bark on Termite Feeding Deterrence

  • Antioxidants are known to affect the feeding habits of termites and a good source is pine bark which contains high levels of antioxidants which can be extracted with neutral solvents. In this study procyanidins (PCs)-rich MeOH-extract and a hot-water extract (HWE) were prepared from Pinus radiata bark. HWE was further separated into crude PCs (CPCs), polymeric PCs (PPCs), and low-molecular-weight PCs (LMWPCs) fractions. The MeOH extract and these fractions were examined for termite (Reticulitermes flavipes Kollar) antifeedant activities using a no-choice test. Catechin was used as a positive control. The LMWPCs was found to be the most effective deterrent to termite feeding. The comparative efficiency of the compounds tested were LMWPCs > PPCs > CPCs > MeOH extract, with the latter being more dependent on molecular weight than on its antioxidant activity.

Free and Glycosidically Bound Volatile Components in Tobacco Leaves(Nicotiana tabacum L.) (잎담배중 유리 및 Glycoside형태로 존재하는 휘발성 향기성분)

  • Free and glycosidically bound volatiles from three green tobacco leaves(NC 82, KF 109 and Br-21) were separated by nonionic resin Amberlite XAD-2 adsorption column chromatography and election by selective solvents. Aglycones from the glycosidically bound fractions were released by enzymatic hydrolysis with almond B-glucosidase. A total of 20 components identified from free and glycosidically bound fractions, the major components were benzaldehyde, benzyl alcohol, 2-phenylethyl alcohol, 3-oxo- -ionol, 3-hydroxy-B-ionone, 3-oxo-7, 8-dihydro-n-ionol and scopoletin. Six Cl.B norisoprenoids identified in this study, which have been described to possess a characteristic tobacco aroma-enhancing effect, were not presented in free forms but rather bound glycosidic forms.

Insecticidal Activity of Extracts Isolated from Syzygium Aromaticum

  • This study separated the crude extract (70% ethanol) of and its three fractions (hexane, chloroform and ethyl acetate extracts) on the basis of polarity indexes, and examined for their insecticidal activities against aphid (Uroleucon lactucicola). For crude extraction, the 70% ethanol extract showed the best extract yield (58.0%) and insecticidal activity (69.0%) among the various concentrations tested (water, 30% ethanol, 50% ethanol, 70% ethanol and 95% ethanol). The major chemical compounds of different fractions (hexane, chloroform and ethyl acetate extracts) were identified as eugenol by head space-GC-MS analysis. The hexane extract showed the highest eugenol content (43.7%) and insecticidal activity (80.0%). The insecticidal activity is accordingly believed to be attributable to the eugenol component. This may provide a useful starting point for the development of bio-pesticides.

Dichotomy of the Galactic Halo as Revealed by Carbon-Enhanced Metal-Poor Giants

  • We present distinct chemical and kinematic properties associated with the inner and outer halos of the Milky Way, as identified by metal-poor stars from the Sloan Digital Sky Survey. In particular, using carbon-enhance metal-poor (CEMP) giants, we first map out the fractions of CEMP-no stars (without strongly enhanced neutron-capture elements) and CEMP-s stars (with a large enhancement of s-process elements) in the inner- and outer-halo populations, separated by their spatial distribution of carbonicity ([C/Fe]). The CEMP-no and CEMP-s objects are classified by their different levels of absolute carbon abundances, A(C). We investigate characteristics of rotational velocity and orbital eccentricity for these sub-classes within the halo populations. Distinct kinematic features and fractions between CEMP-no and CEMP-s stars identified in each halo region will provide important clues on the origin of the dichotomy of the Galactic halo.

Separation of Antioxidant compounds from Persimmon Leaves (감잎(시엽(枾葉))의 항산화 성분 분리)

  • This study was carried out to separate and identify the antioxidative substances in persimmon leaves. The antioxidative substances in persimmon leaves were extracted by methanol. The extract was fractionnated by SEP-PAK cartridge colum. From these results five fractions(F-I${\sim}$V) were obtained. Antioxidative activity of each fractions was examined by the DPPH methord. The F-II, III and IV showed antioxidative activity and among them F-II and F-III showed the strongest. Five frations were separated by TLC using ethylacetate : chloroform : formic acid : $H_2O$(8 : 1 : 1 : 1 v /v) as the solvent. From these results were obtained spots of Rf 0.71, 0.35 and 0.25. This spots were scraped from the plate and extracted by methanol. The extracts thuse obtained were used for examination of identify by TLC, UV /VIS-spectrophotometer and HPLC. Among them spot of Rf 0.71 were demonstrated to catechin and the spots of Rf 0.35 and 0.25 was suggested to polyphenol substances.

  • Colloidal stability is one of crucial factors for many applications of nanodiamond. Despite recent development, nanodiamonds available on the market often exhibit a high impurity content, wide size distribution of aggregates and low resistance to sedimentation. In the current study, four commercial nanodiamond powders synthesized by detonation synthesis were surface modified and then separated with respect to the size into six fractions by centrifugation. The fractions were evaluated by zeta potential, particle size distribution and elemental composition. The results showed that the modified nanodiamonds form stable colloidal suspensions without sedimentation for a long time.

A Sperm Factor Inducing Second Polar Body Formation in Mouse Secondayy Oocyte

  • A sperm factor(s) for oocyte activation during fertilization has not been clearly identified. In this study to elucidate an oocyte activation factor(s), mouse sperm were sonicated and ultra-filtered with a 30 kilo-daltons (KD) cutoff membrane and the ultra-filtrate was then sequentially fractionated over Suporose 12 column and Superdex column, The recovered fractions were micro-injected into Mⅱmouse oocytes and second polar body formation (PBF) was examined. Suporose fraction RV2.10 prepared from sperm extract significantly increased PBF. Of Superdex fractions re-separated from Suporose fraction RV2.10, fraction RV2.12 also had the strongest PBF activity. By analyzing with micro-reverse phase column (URPC), the Superdex fraction RV2.12 appeared to be glutamic acid. In microinjection test, glutamic acid significantly increased PBF. This study suggests that glutamic acid should be a type of sperm factor for second polar body formation related to oocyte activation.

대두가수분해물로부터 새로운 항혈전성 펩타이드, SSGE와 DEE의 분리

  • A soy protein hydrolysate was found to inhibit rat platelet aggregation induced by ADP, an aggregating agent. To find out its principal antiplatelet peptide(s), the soy protein hydrolysate was separated successively by gel filtration chromatography, revere-phase HPLC, and cation exchange HPLC. During the course of separation, we observed that most fractions had antiplatelet effects, which suggests that most peptides have some degree of antiplatelet effect. Following the inhibitory fractions, we purified and identified two new peptides, SSGE and DEE, by LC-electrospray ionization MS and peptide equencing. Both peptides were highly hydrophilic. The concentrations to obtain 50% inhibition ($IC_50$) of the aggregation intensity were approximately $\458muM$ and $\485muM$, respectively, for SSGE and DEE.
