Functional Characterization of NtCDPK1 in Tobacco

  • We previously showed that NtCDPK1, a tobacco calcium-dependent protein kinase, interacts with and phosphorylates the Rpn3 regulatory subunit of the 26S proteasome, and that both NtCDPK1 and Rpn3 are mainly expressed in rapidly proliferating tissues, including shoot and root meristem. In this study, we examined NtCDPK1 expression in roots using GUS expression in transgenic Arabidopsis plants, and investigated its function in root development by generating transgenic tobacco plants carrying a sense NtCDPK1 transgene. GUS activity was first detected in roots two days after sowing. In later stages, strong GUS expression was detected in the root meristem and elongation zone, as well as the initiation sites and branch points of lateral roots. Transgenic tobacco plants in which NtCDPK1 expression was suppressed were smaller, and their root development was abnormal, with reduced lateral root formation and less elongation. These results suggest that NtCDPK1 plays a role in a signaling pathway regulating root development in tobacco.

Evaluation of MR-SENSE Reconstruction by Filtering Effect and Spatial Resolution of the Sensitivity Map for the Simulation-Based Linear Coil Array (선형적 위상배열 코일구조의 시뮬레이션을 통한 민감도지도의 공간 해상도 및 필터링 변화에 따른 MR-SENSE 영상재구성 평가)

  • Parallel imaging technique can provide several advantages for a multitude of MRI applications. Especially, in SENSE technique, sensitivity maps were always required in order to determine the reconstruction matrix, therefore, a number of difference approaches using sensitivity information from coils have been demonstrated to improve of image quality. Moreover, many filtering methods were proposed such as adaptive matched filter and nonlinear diffusion technique to optimize the suppression of background noise and to improve of image quality. In this study, we performed SENSE reconstruction using computer simulations to confirm the most suitable method for the feasibility of filtering effect and according to changing order of polynomial fit that were applied on variation of spatial resolution of sensitivity map. The image was obtained at 0.32T(Magfinder II, Genpia, Korea) MRI system using spin-echo pulse sequence(TR/TE = 500/20 ms, FOV = 300 mm, matrix = $128{\times}128$, thickness = 8 mm). For the simulation, obtained image was multiplied with four linear-array coil sensitivities which were formed of 2D-gaussian distribution and the image was complex white gaussian noise was added. Image processing was separated to apply two methods which were polynomial fitting and filtering according to spatial resolution of sensitivity map and each coil image was subsampled corresponding to reduction factor(r-factor) of 2 and 4. The results were compared to mean value of geomety factor(g-factor) and artifact power(AP) according to r-factor 2 and 4. Our results were represented while changing of spatial resolution of sensitivity map and r-factor, polynomial fit methods were represented the better results compared with general filtering methods. Although our result had limitation of computer simulation study instead of applying to experiment and coil geometric array such as linear, our method may be useful for determination of optimal sensitivity map in a linear coil array.

Improvement of active nose control in vehicle interior using a RLS algorithm (RLS 알고리즘을 이용한 승용차 내 능동소음제어의 개선)

  • While driving, the low frequency interior noise below 200Hz causes the main component that irritates the auditory acoustic sense. But these passive control methods bring out increment in cost and weight of the vehicle and result in low efficiency. Recently, various ANC(Active Noise Control) methos to suppress the low frequency noise began to launch into application. In this study, we implemented the active noise control system for passenger vehicle to cancel the engine booming noise using DSP-based control unit, 4 micorphones, and 2 speakers. We used MEFX-LMS (Multiple Error Filtered X-Least Mean Square) algorithm since it can be easily implemented in real time. Also, MEFX-RLS algorithm was taken to enhance the suppression of the harmonic components of the engine booming noise inspite of its computational complexity. The performance of two adaptive algorithms were analyzed with experimental resutls.

  • Conversion disorder is disease that anxiety is converted to physical symptom by suppression of complication unconsciously. This case is that of conversion disorder resulted from school work. The treatment of Conversion disorder is anxiety syndrome's removement. But if not possible, as a symptomatic treatment, approvement of symptom is significant, in a sense. By use of Oriental medical treatment, we could improve symptoms of two case conversion disorders. Sooner or later, the patients gained a complete recovery. So, more objective study and treatment about conversion disorder by Oriental medicine's demonstration is needed.

  • An effective and disturbance suppressible controller can be obtained by assigning the left eigenstructure (eigenvalues/left eigenvectors) of a system. However, the disturbance decouplability is governed by the right eigenstructure(eigenvalues/right eigenvectors) of the system. In this paper, in order to obtain a disturbance decouplable as well as effective and disturbance suppressible controller, the concurrent assignment scheme of the left and right eigenstructure is proposed. The biorthogonality property between the left and right modal matrices of a system well as the relations between the achievable right modal matrix and states selection matrices are used to develop the scheme. The proposed concurrent eigenstructure assignment scheme guarantees that the desired eigenvalues are achieved exactly and the desired left and right eigenvectors are assigned to the best possible(achievable) sets of eigenvectors in the least square sense, respectively. A numerical example is presented to illustrate the usefulness of the proposed scheme.

A Novel Type of Non-coding RNA, nc886, Implicated in Tumor Sensing and Suppression

  • nc886 (=vtRNA2-1, pre-miR-886, or CBL3) is a newly identified non-coding RNA (ncRNA) that represses the activity of protein kinase R (PKR). nc886 is transcribed by RNA polymerase III (Pol III) and is intriguingly the first case of a Pol III gene whose expression is silenced by CpG DNA hypermethylation in several types of cancer. PKR is a sensor protein that recognizes evading viruses and induces apoptosis to eliminate infected cells. Like viral infection, nc886 silencing activates PKR and induces apoptosis. Thus, the significance of the nc886:PKR pathway in cancer is to sense and eliminate pre-malignant cells, which is analogous to PKR's role in cellular innate immunity. Beyond this tumor sensing role, nc886 plays a putative tumor suppressor role as supported by experimental evidence. Collectively, nc886 provides a novel example how epigenetic silencing of a ncRNA contributes to tumorigenesis by controlling the activity of its protein ligand.

Psychological Changes of Undergraduate using Color Programs and Entertainment Programs (색채프로그램과 예능프로그램을 활용한 대학생의 심리변화)

  • This study will be conducted to verify that color programs and entertainment programs affect undergraduates' psychology(Obsessive-compulsive, anxiety and hostility). The results of this study can be summarized as follows. First, the experimental group's psychology(Obsessive-compulsive, anxiety and hostility) participated in color programs than did all the pre-investigation levels decreased post-mortem examination, statistically significant at 95% appeared to be. Second, after watching entertainment programs, pass through cognition process of color, undergraduates' color representation worked perception of red and blue respectively. This results is that the reason of white and the suppression of black what to do now with a sense of responsibility can be considered as a change process. In conclusion, after watching entertainment programs take advantage of color programs that a positive impact on undergraduates' psychological state, as they can provide bright colors with a variety of ways that make sense to suggest.

The Role of AiiA, a Quorum-Quenching Enzyme from Bacillus thuringiensis, on the Rhizosphere Competence

  • Bacteria sense their population density and coordinate the expression of target genes, including virulence factors in Gram-negative bacteria, by the N-acylhomoserine lactones (AHLs)-dependent quorum sensing (QS) mechanism. In contrast, several soil bacteria are able to interfere with QS by enzymatic degradation of AHLs, referred to as quorum quenching. A potent AHL-degrading enzyme, AiiA, from Bacillus thuringiensis has been reported to effectively attenuate the virulence of bacteria by quorum quenching. However, little is known about the role of AiiA in B. thuringiensis itself. In the present study, an aiiA-defective mutant was generated to investigate the role of AHA in rhizosphere competence in the root system of pepper. The aiiA mutant showed no detectable AHL¬-egrading activity and was less effective for suppression of soft-rot symptom caused by Erwinia carotovora on the potato slice. On the pepper root, the survival rate of the aiiA mutant significantly decreased over time compared with that of wild type. Interestingly, viable cell count analysis revealed that the bacterial number and composition of E. carotovora were not different between treatments of wild type and the aiiA mutant. These results provide evidence that AHA can play an important role in rhizosphere competentce of B. thuringiensis and bacterial quorum quenching to Gram-negative bacteria without changing bacterial number or composition.

Experiences of Families in the Intensive Care Unit: Interactions with Health Care Providers (중환자실 환자 가족의 경험: 의료인들과의 상호작용)

  • Purpose: The purpose of the study was to provide deep understanding of the reported experiences of families with their loved one in the intensive care unit (ICU), focusing on interactions with healthcare providers. Methods: The data were collected by individual interviews of eleven participants. The transcribed data were analyzed using qualitative content analysis to identify major themes and sub-themes that represented the experiences of families. Results: Five themes and 13 sub-themes emerged. "Captive of patients' delayed death: Fear and anxiety" describes psychological problems arising when the family member became critically ill enough to warrant being admitted to the ICU. "Families as the weak: Suppression and resistance" describes interpersonal difficulties arising due to lack of information and trust with healthcare providers. "Deprivation of authority and duty as families: Helplessness" illustrate situational barriers in attempting to protect and support family member. "Re-establishment of trust relationship with healthcare providers: Gratitude and appreciation" describes how they satisfied with themselves by regaining trust relationship. Lastly, "Acceptance of reality through direct care participation: Relief and peace" illustrates peace of mind by gaining sense of reality through active direct care participation. Conclusion: This study demonstrates the positive and negative experiences of families with ICU patients. The results will be useful in developing family-centered nursing interventions.

New Hairpin RNAi Vector with Brassica rapa ssp. pekinensis Intron for Gene Silencing in Plants

  • Homology-specific transcriptional and post-transcriptional silencing, an intrinsic mechanism of gene regulation in most eukaryotes, can be induced by anti-sense, co-suppression, or hairpin-based double-stranded RNA. Hairpin-based RNA interference (RNAi) has been applied to analyze gene function and genetically modify crops. However, RNAi vector construction usually requires high-cost cloning steps and large amounts of time, or involves methods that are protected by intellectual property rights. We describe a more effective method for generating intron-spliced RNAi constructs. To produce intron-spliced hairpin RNA, an RNAi cassette was ligated with the first intron and splicing sequences of the Brassica rapa ssp. pekinensis histone deacetylase 1 gene. This method requires a single ligation of the PCR-amplified target gene to SpeI-NcoI and SacI-BglII enzyme sites to create a gene-specific silencing construct. We named the resulting binary vector system pKHi and verified its functionality by constructing a vector to silence DIHYDROFLAVONOL 4-REDUCTASE (DFR), transforming it into tobacco plants, and confirming DFR gene-silencing via PCR, RT-qPCR, and analysis of the accumulation of small interfering RNAs. Reduction of anthocyanin biosynthesis was also confirmed by analyzing flower color of the transgenic tobacco plants. This study demonstrates that small interfering RNAs generated through the pKHi vector system can efficiently silence target genes and could be used in developing genetically modified crops.
