The present study was conducted to observe the effect of osmolality of glycerolated TEST-yolk glycerol extenders on post-thawing sperm kinematics of ram spermatozoa of the native Malpura breed maintained in a semi-arid tropical environment. Good quality semen obtained from adult rams was pooled, split and diluted to 1,000 million spermatozoa per ml in complete TEST-yolk-glycerol extenders of 900, 1,200, 1,500 and 1,800 mOsm/kg osmolality. Diluted semen samples were loaded in 0.25 ml straws and cooled down to $-125^{\circ}C$ freezing temperature at the rate of $-25^{\circ}C$ per minute under controlled conditions before plunging into liquid nitrogen for storage. The thawing of straws was performed at $50^{\circ}C$ in a water bath for 10 seconds and sperm kinematics of the frozen-thawed spermatozoa were assessed by a computer-assisted sperm analysis technique. Osmolality of diluent had no significant effect on post-thawing % motility, % rapid, % medium and % slow moving frozen-thawed spermatozoa but significantly (p< 0.05) affected the % linearity and % straightness. The post-thawing % motility and % rapid motile spermatozoa were highest in samples extended in diluent of 1,500 mOsm/kg osmolality and lowest in 900 mOsm/kg. The curvilinear velocity of spermatozoa was significantly (p<0.05) higher for samples extended in 1,800 mOsm/kg, compared to those in 900 and 1,200 mOsm/kg, but the effect was not significantly different to those extended in diluent of 1,500 mOsm/kg osmolality. The study indicated that ram spermatozoa could tolerate a wide osmolality range for dilution in the complete TEST-yolk-glycerol extender for their cryosurvival. The highest recovery of motile spermatozoa following thawing was achieved in samples extended in the TEST-yolk-glycerol diluent of 1,500 mOsm/kg osmolality.
The objective of this study was to assess the developmental potential in vitro produced embryos frozen-thawed with the various containers, and also examined expression and localization of heat shock protein 70 at these embryos. For the vitrification, 2-cell, 8-cell and blastocyst stage embryos produced by in vitro fertilization (IVF) and nuclear transfer (NT) were exposed the ethylene glycol 5.5 M freezing solution (EC 5.5) for 30 sec, loaded on each containers such EM grid, straw and cryo-loop, and then immediately plunged into liquid nitrogen. Thawed embryos were serially diluted in sucrose solution, each for 1 min. and cultured in CRI-aa medium. Survival rates of the vitrification production were assessed by re-expanded, hatched blastocysts. There were no differences in the survival rates of IVF using EM grid and cryo-loop. However, survival rates by straw were relatively lower than other containers. The use of cryo-loop resulted in only survival of nuclear transferred embryos (43.7%). Also, there embryos after IVF or NT were analysed by semi-quantitive reverse transcription-polymerase chain reaction (RT- PCR) methods for hsp 70 mRNA expression. Results revealed the expression of hsp 70 mRNh were higher thawed embryos than control embryos. Immunocytochemistry used to localize the hsp 70 protein in embryos. Two and 8-cell embryos derived under control condition was evenly distributed in the cytoplasm but appeared as aggregates in some frozen-thawed embryos. However, in the control, blastocysts displayed aggregate signal while Hsp70 in frozen-thawed blastocysts appeared to be more uniform In distribution. Therefore, this result suggests that the exploiting Hsp 70 in the early embryos may be role for protection of stress condition for increase viability of embryos within IVF, NT and there frozen-thawed embryos.

We compared germination efficiency for somatic embryos (SE) of Liriodendron tulipifera using semi-solid (SS), temporary immersion bioreactors (TIB), and continuous immersion bioreactors (CIB) to produce vigorous plants. The bioreactors were designed to be immersed in liquid media with plantlets with an adjustable immersion time. TIB and CIB improved germination rates up to 80.86% and 95.21%, respectively, however, CIB produced more hyperhydric plantlets than TIB. The height of plantlets in TIB was significantly higher than for those in CIB. Fresh weights of plantlets grown in CIB of were significantly lower than for those grown in TIB. The lowest chlorophyll concentration was found in in vitro plantlets from CIB. We examined abnormally developed leaves, stems, and apical zones of in vitro plantlets that were produced in CIB. Among the three types, SS showed the highest stomatal density and the shortest stomatal length in in vitro plantlets. After acclimatization, plants from CIB exhibited the lowest values in biomass, such as height, root collar diameter, leaf fresh weight, leaf length, leaf width, petiole length, petiole diameter, and leaf area. Photosynthesis and transpiration rates of ex vitro plants were not significantly different among the three culture types, but stomatal conductance was higher in TIB than in the SS and CIB. Therefore, the results suggest that TIB is the preferable bioreactor to improve in vitro plantlet regeneration of L. tulipifera. TIB-originated plants showed higher growth rate than SS and CIB after transferring to soil.

Residual effects of chlorination, in the case of distilled water as a standardized material and 0.2 ppm of seperated residual chlorine, were continued 32 hours in $0^{\circ}C$, and 25 hour in $20^{\circ}C$, of water temperature and in the case of 0.4 ppm of seperated residual chlorine were continued 47 hours in $0^{\circ}C$ and 23 hours in $20^{\circ}C$. 4. In the case of 4 ppm of seperated residual chlorine, residual effects were continued 23 hours in $5^{\circ}C$, 90 hours in $10^{\circ}C$, 78 hours in $15^{\circ}C$, and 60 hours in $20^{\circ}C$ : by the temper; lure of water, continuing residual effects of chlorination are different, so we have to car for the warm season chlorination in the hider temperature. 5. Chlorination effects of drinking waters in the case of 0.4 ppm of seperated residual chlorine were continued 237 hours in $22^{\circ}C$ water : and in the case of rechlorination as 4 ppm of residual chlorine, continued 71 hours in $22^{\circ}C$ water.
The objectives of this study were to identity antioxidant substance in heated garlic juice (HGJ). We evaluated the antioxidant activities of heated garlic juice exposed to 120, 130, and $140^{\circ}C$ for 2 hr. The HGJ was partitioned using the solvents of hexane, chloroform, ethyl acetate, butanol, and water. The ethyl acetate fraction of HGJ treated at $130^{\circ}C$ for 2 hr showed strong antioxidant activity; this extract was isolated and purified using silica gel column chromatography and semi-preparative high-performance liquid chromatography. The structure of the purified compound was determined using spectroscopic methods, i.e., ultraviolet, mass spectrometry, infrared, $^1H$ NMR, $^{13}C$ NMR, DEPT, HMBC, and HMQC. The isolated compound was identified as thiacremonone (2,4-dihydroxy-2,5-dimethyl-thiophene-3-one). Thiacremonone showed strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity, with a 50% inhibition concentration ($IC_{50}$) of $22.25{\pm}0.44\;{\mu}g/mL$, which is much higher than that of the antioxidants ascorbic acid ($30.06{\pm}0.42\;{\mu}g/mL$), ${\alpha}$-tocopherol ($71.30{\pm}0.97\;{\mu}g/mL$), and butylated hydroxyanisole ($50.54{\pm}0.94\;{\mu}g/mL$).

Organogels are semi-solid systems that consist of an apolar solvent as the liquid phase within a three-dimensional networked structure. In this study, we developed a stable and skin penetration-enhanced Lecithin Organogel (LO) containing genistein, which is one of the poorly soluble active ingredients in both polar and apolar phase. After screening of various components (type of gelators, organic and aqueous phase), hydrogenated lecithin (HL), sunflower oil (SO), dipropylene glycol (DPG), and polyethylene glycol (PEG) were mainly used in this formulation. Phase ternary diagram was employed for optimization of the composition in the LO. The formulated LO were evaluated for its organoleptic characteristics, stability, pH, rheology, phase transition temperatures, microscopic analysis and skin penetration. The optimized stable LO system can be utilized as an effective and stable cosmetic formulation that can carry poorly soluble active ingredients at high concentration for topical dermal delivery.

A total of 62 bacterial isolates were obtained from Gomsohang mud flat, Mohang mud flat, and Jeju Island, Republic of Korea. Among them, the isolate CNU114001 showed significant antagonistic activity against pathogenic fungi by dual culture method. The isolate CNU114001 was identified as Bacillus amyloliquefaciens by morphological observation and molecular data analysis, including 16SrDNA and gyraseA (gyrA) gene sequences. Antifungal substances of the isolate were extracted and purified by silica gel column chromatography, thin layer chromatography, and high performance liquid chromatography. The heat and UV ray stable compound was identified as iturin, a lipopeptide (LP). The isolate CNU114001 showed broad spectrum activity against 12 phytopathogenic fungi by dual culture method. The semi purified compound significantly inhibits the mycelial growth of pathogenic fungi (Alternaria panax, Botrytis cinera, Colletotrichum orbiculare, Penicillium digitatum, Pyricularia grisea and Sclerotinia sclerotiorum) at 200 ppm concentration. Spore germ tube elongation of Botrytis cinerea was inhibited by culture filtrate of the isolate. Crude antifungal substance showed antagonistic activity against cucumber scleotiorum rot in laboratory, and showed antagonistic activity against tomato gray mold, cucumber, and pumpkin powdery mildew in greenhouse condition.

POSCO $\#2$ Stainless steel making plant produces more than 600 thousand ton per year with a variety of products consisting of austenite and ferrite stainless steel to meet custrmers' needs since 1996. The plant has four different major processes, that are, EAF-AOD-VOD-CC to finally produce semi-product called as slab. In this study, we importantly took AOD process into consideration due to its roles such as to check and verify the final qualities through sampling inspection. But the lead-time from sampling to its verification takes five to ten minutes causing produrtivity loss as muck as the lead-time as a result. Of all indices for quality and process control the plant has, carbon ingredient in liquid type of steel is the most important since it affects in a great way to the characteristics of steel, if any problem. customers not satisfied with quality could issue a claim; therefore there is no way hut to guarantee it before delivery. in this study, to reasonably reduce lead-time ran save a cycle time and finally improve our productivity from a state-or-art alternative just such as applying statistical model based on multi-regression analysis into the A.O.D line by analyzing the statistical and technical relationship between carbon and the relevant some vital independent variables. In consequence, the model with R-square $87\%$ allowed the plant to predict, abbreviating the process in relations to sampling to verification. approximately the value of [C] so that operators could run the process line with reliability on data automatically calculated instead of actual inspection. In the future, we are going to do the best to share this type of methodology with other processes, if possible, to apply into them.

Effect of metal oxide catalysts and ionic liquids on the synthesis of methyl carbamate(MC) and dimethyl carbonate (DMC) from urea and methanol was investigated in a high pressure reaction system. In closed system, MC yield from urea and methanol reached almost 100% at reaction temperature over $150^{\circ}C$ without catalyst, whereas DMC yield of 1.5% under was obtained irrespective of catalysts used. In DMC synthesis from MC and methanol, $ZnCl_{2}$ showed the highest catalytic activity and led to the DMC yield of 16.3% under optimal conditions. Furthermore, DMC yield can be enhanced by the simultaneous application of ionic liquids with nano-sized catalysts in semi-continuous reaction system.

After recycle of spent materials has been optimised, there remains a proportion of waste which must be dealt with in the most environmentally friendly manner available. For materials such as municipal waste, clinical waste, toxic waste and special wastes such as tyres, incineration is often the most appropriate technology. The study of incineration must take a process system approach covering the following aspects: ${\bullet}$ Collection and blending of waste, ${\bullet}$ The two stage combustion process, ${\bullet}$ Quenching, scrubbing and polishing of the flue gases, ${\bullet}$ Dispersion of the flue gases and disposal of any solid or liquid effluent. The design of furnaces for the burning of a bed of material is being hampered by lack of an accurate mathematical model of the process and some semi-empirical correlations have to be used at present. The prediction of the incinerator gas phase flow is in a more advanced stage of development using computational fluid dynamics (CFD) analysis, although further validation data is still required. Unfortunately, it is not possible to scale down many aspects of waste incineration and tests on full scale incinerators are essencial. Thanks to a close relationship between SUWIC and Sheffield Heat&Power Ltd., an extended research programme has been carried out ar the Bernard Road Incinerator plant in Sheffield. This plant consists of two Municipal(35 MW) and two Clinical (5MW) Waste Incinerators which provide district heating for a large part of city. The heat is distributed as hot water to commercial, domestic ( >5000 dwelling) and industrial buildings through 30km of 14" pipes plus a smaller pipe distribution system. To improve the economics, a 6 MW generator is now being added to the system.
