Influence of Soil and Forage Minerals on Buffalo (Bubalus bubalis) Parturient Haemoglobinuria

  • The present study was carried out to investigate the serum minerals profile in buffaloes (Bubalus bubalis) suffering from parturient haemoglobinuria (PHU) along with minerals profile of soils and fodders from the disease prone areas and their interrelationships. Serum samples were collected from 60 each of healthy and PHU affected buffaloes randomly selected from field cases. Serum samples were collected from each animal. Fifty composite soil samples were collected where PHU was prevalent. Fifty samples of fodders including leaves and stems being fed to the diseased buffaloes were collected. The difference in the levels of calcium and potassium between upper and lower soil surface of disease prone areas under study were statistically non-significant. The mean values of phosphorous, copper, iron, selenium and molybdenum in upper soil surface were significantly (p<0.05) higher than in lower soil surface. None of the fodders offered to the diseased animals met the dietary requirements of phosphorus and copper whereas none of the fodders was deficient in potassium, iron and selenium rather were having excess of potassium, iron and selenium. The concentration of calcium was adequate in lucerne, berseem, sarson and sorghum, while maize, sugarcane and wheat straw did not meet the required levels for dairy animals. Molybdenum contents in all fodders were adequate to meet the dietary requirements of the dairy buffaloes. Serum phosphorus, copper and selenium were significantly (p<0.001) lower whereas potassium, iron and molybdenum in buffaloes suffering from PHU were significantly (p<0.001) higher than in healthy buffaloes. It was concluded that phosphorous deficient soils play a major role by transferring this deficiency to plants and ultimately reaching to animals where hypophosphataemia is a consistent finding.

Mineral Status of Soils and Forages in Southwestern Punjab-Pakistan: Micro-minerals

  • This study was conducted to evaluate the trace elemental nutritive values of soil and forages collected from southwestern part of the province of Punjab, Pakistan. Soil and forage samples were collected fortnightly for two seasons. The concentrations of some trace minerals varied greatly among seasons and sampling periods. Seasonal effects were found in all soil micro-minerals except zinc, while forage iron, zinc, and selenium were affected by seasonal changes. Sampling periods effects were observed in all soil minerals and in forage copper, iron, zinc, and manganese only. All soil mineral levels except cobalt and selenium were sufficiently high to meet the requirements of plants for normal growth during both seasons. In contrast, soil Co and Se levels were severely deficient during both seasons and considered inadequate for plant growth. Soil Fe, Zn, Co, and Se levels were higher, and Cu and Mn lower during winter than those during summer. Forage Zn levels during summer were at marginal deficient levels, and in contrast, all other forage micro-minerals were within the required range for ruminants during both seasons. Although forage mico-minerals were within the range required by the ruminants, they were not sufficiently high to prevent the predisposition to various diseases caused by nutrient deficiency. Consequently, grazing animals at this location need continued mineral supplementation of these elements with a mixtures of high bio-availability rather than of high micro-mineral contents to support optimum ruminant productivity.

Effects of Intraruminal Soluble Glass Bolus on Blood Selenium and Plasma Mineral Level of Grazing Does under Backyard Conditions in Selected Areas in Nueva Ecija, Philippines

  • Soluble glass bolus with selenium (Se), copper (Cu) and cobalt (Co) was administered intraruminally to Philippine grazing does fed under backyard farming conditions to determine its effect on blood mineral status. Forty-five does were dosed with SGB intraruminally every 6 months, whereas 15 were without SGB during this experimental period of 10 months. Blood of does in both treatment groups were collected every other month and blood Se, plasma Cu, Ca, P and Mg were determined in this study. All does did not show clinical Se, Cu, Ca, P or Mg deficiency during this experimental period. Selenium concentration of treated does increased (p<0.01) after beginning of this experiment, whereas the level of control does decreased slowly (p<0.01). Two months after SGB administration, all treated does had higher blood Se than the lower limit of $20{\mu}g/l$ suggested by NRC (1981), whereas some control does had lower blood Se concentration than the lower limit of $20{\mu}g/l$. On the other hand, plasma Cu concentration of treated does started to increase more remarkably than control does 4 months after this experiment had started although the difference was insignificant. There were no significant differences between plasma Ca, P or Mg concentration of does in both treatment groups. Soluble glass bolus had no harmful effects on plasma macro mineral concentrations of grazing does. This study showed that SGB was available as mineral supplement to improve imbalances of selected mineral of grazing goats in the traditional backyard conditions in Luzon Island, Philippines.

한약재내의 미량원소의 의의와 치료효과에 대한 고찰 (Therapeutical Effect with Trace Elements in Herbal Medicine)

  • Trace element are involved in enzymatic activities, immunological reactions. physiological mechanisms. Deficiency in some trace elements, such as iron and iodine. is still an important health problem, The role of trace elements deficiency is suspected in various clinical situations and is now confirmed by well designed supplementation studies. However, the importance of trace elements as chinese herbal constituents is not sufficiently appreciated by the oriental medical profession, although in recent years a significant increase of new finding on their essential character in chinese herbal medicine occurred. It is well known that herbal medicine contains a variety of trace elements which would show therapeutic effects with active components in herbal medicine . In china, recent work showed some positive correlation between trace element and traditional chinese medicine (TCM) in terms of therapeutic effects even if their role in therapeutic effects is still obscure. In korea, not much attention has been on the therapeutic importance of trace element contained in herbal medicine Here, the therapeutic effects of trace element in TCM were reviewed and summarized. 1. Iron, copper, zinc and manganese are mainly contained in TCM. In addtion, chromium, magnesium, molybdenum, nickel, alminium, cobalt, arsenic and selenium has been studied for their therapeutic effects 2. Zinc, is decreased in patients who have deficiency of kindney(腎虛) and chronic disease. Fe is decreased in patients who have deficiency of blood(血虛). However copper is increased in patients who have chronic disease and hepatic disease.3 Iron concentration is high in herbs used for tonifying and nourishing yin or blood(補陰補血藥) Zinc concentration is high also in herb used for tonifying kidiney and vital essence(補腎補精藥). In addition. copper concentration Is high in herb used for replenishing qi(補氣藥) 4 In herbal drugs, the therapeutic substances in TCM are not only organic but also inorganic. It seems that trace elements would be one of components in herb for its therapeutic effects. This indicates that therapeutic effects of TCM should be extended not only to herb itself, bur also to trace elements contained in herb.

MINERAL NUTRITION OF GRAZING SHEEP IN NORTHERN CHINA II. SELENIUM, COPPER, MOLYBDENUM, IRON AND ZINC IN PASTURE, FEED SUPPLEMENTS AND SHEEP

  • This study determined the concentrations of micro-minerals in pastures, in feed supplements and in grazing, reproducing ewes, at different times during the year, at three farms in Northern China. Samples were collected 5 to 8 times during the year and analysed for selenium, copper, iron, molybdenum and zinc. On two farms selenium concentrations in both pastures and animal tissues were low for part of the year. The lowest concentrations in pasture (< $30{\mu}g/kg$ DM) and liver (< $100{\mu}g/kg$ wet weight) indicated that productivity of the sheep may be reduced by a deficiency of this element. On one farm copper concentrations in the lever were in the liver were in the deficient range (< $5{\mu}g/kg$ wet weight) for part of the year. It is likely that this is a result of high intakes of iron from pasture (up to 4.5 g Fe/kg DM) and soil, as indicated by high concentrations of iron in faeces (up to 7 g Fe/kg DM). Molybdenum intake is unlikely to have had much influence on copper absorption because pasture concentrations of this element were not unusually high (1 to 5 mg/kg DM). Zinc in pastures on two farms was below 10 mg/kg DM for part of the year. On one of these farms, the concentration of zinc in faeces was below 30 mg/kg DM throughout the year and this is consistent with zinc intakes of 7 to 15 mg/kg. Despite these low intakes, the concentratons of zinc in plasma were consistently above deficient levels. No clinical signs of deficiencies of any of the elements studied were observed.

Cell Proliferation and Motility Are Inhibited by G1 Phase Arrest in 15-kDa Selenoprotein-Deficient Chang Liver Cells

  • The 15-kDa selenoprotein (Sep15) is a selenoprotein residing in the lumen of the endoplasmic reticulum (ER) and implicated in quality control of protein folding. Herein, we established an inducible RNAi cell line that targets Sep15 mRNA in Chang liver cells. RNAi-induced Sep15 deficiency led to inhibition of cell proliferation, whereas cell growth was resumed after removal of the knockdown inducer. Sep15-deficient cells were arrested at the G1 phase by upregulating p21 and p27, and these cells were also characterized by ER stress. In addition, Sep15 deficiency led to the relocation of focal adhesions to the periphery of the cell basement and to the decrease of the migratory and invasive ability. All these changes were reversible depending on Sep15 status. Rescuing the knockdown state by expressing a silent mutant Sep15 mRNA that is resistant to siRNA also reversed the phenotypic changes. Our results suggest that SEP15 plays important roles in the regulation of the G1 phase during the cell cycle as well as in cell motility in Chang liver cells, and that this selenoprotein offers a novel functional link between the cell cycle and cell motility.

식이내 Selenium과 Vitamin E가 Alcohol을 섭취한 흰쥐의 간 지질 과산화에 관련된 효소의 활성에 미치는 영향 (The Effect of Selenium and Vitamin E on Activity of Enzyme Related to the Lipid Peroxidation in Rat with Alcohol Administration)

  • 본 실험은 식이내 Se와 vitamin E 수준이 알코올을 섭취한 흰쥐의 간 지질과산화에 관련된 효소의 활성에 미치는 영향을 살펴보고자 한 것이다. 이를 위해 평균체중이 58~62g인 Sprague-Dawley계의 숫쥐 72마리를 Se의 투여량 (0mg, 0.4mg, 10mg/kg diet)과 vitamin E 투여량 (0mg, 150mg/kg diet) 및 알코올 섭취 여부에 따라 12군으로 구분하여 7주간 사육 하였다. 알코올섭취는 사육 3주째부터 급수용 물에 10%로 맞추어 투여하여 제한 없이 먹게 하였다. 혈장중의 ${\gamma}$-GTP합성은 알코올 섭취군이 비섭취군 보다 높았고, Se의 과잉(HSe) 및 결핍된군(LSe)이 정상군(CSe) 보다 높았으며 알코올 섭취시 Se과 vitamin E의 결핍은 ${\gamma}$-GTP량의 상승에 더 큰 영향을 미쳤다. 혈장 GOT는 알코올섭취군이 비섭취군에 비해서 높은 경향을 나타내었다. 그리고 혈장 GPT 활성은 알코올 섭취군이 비섭취군 보다 약간 높은 경향이었고, Se이 결핍된 군에서의 알코올 섭취의 영향은 다른 군에서의 알코올섭취 영향보다 GPT의 상승에 더 큰 영향을 미쳤다. GSH-Px의 활성은 Se이 결핍된 LSe군은 HSe군과 CSe군에 비해서 유의적으로 낮았다. Cytosol fraction의 GSH-Px 활성은 알코올 섭취군에서 약간 낮은 값이었고 Se이 과잉 및 결핍된 HSe군과 LSe군은 CSe군에 비해서 약 2배정도 낮은 값을 나타내었다. HSe군의 혈장내 Se과 cytosol fraction GSH-Px의 상관관계는 negative 상관관계를 보였고 (r=-0.662, p< 0.001) L-군은 positive 상관관계를 보였다(r=0.640, p<0.001). Microtome fraction에서 GSH S-transferase의 활성은 알코올 섭취군에서 약간 높은 경향이었고, LSe군이 다른군에 비해서 유의적으로 높았으며, cytosol fraction에서도 LSe, CSe, HSe군 순서로 높았고, vitamin E 비섭취군은 섭취군 보다 높은 경향을 나타내었고, 알코올 섭취시 Se과 vitamin E결핍은 GSH S-transferase를 더욱 증가시켰다. Mitochondria의 catalase 활성은 HSe군은 CSe군 보다 낮은 경향이었으나 Se을 결핍시킨 LSe군은 오히려 높은 경향을 나타내었다. 간 cytosol fraction의 SOD는 각 군간에 큰 변화가 없었고 cytochrome P-450은 알코올 섭취군이 높았으며 Se을 과잉으로 섭취한 HSe군에서 유의적으로 낮았다. 결론적으로 Se 와 vitamin E의 결핍은 지질과산화에 관련된 효소의 활성을 높혀 간 지질 과산화를 촉진하고 더우기 알코올의 섭취시에는 그 영향이 더욱 두드러진 것으로 보인다.

식이중의 Methionine첨가수준이 흰쥐의 체내 지질 과산화와 간조직 형태에 미치는 영향 (Effects of Dietary Methionine Level on Lipid Peroxidation and Hepatic Morphology in Rat)

  • 본 실험에서는 항산화제로 보고된 vitamin E와 Se이 부족된 상태에서 과산화반응의 원인물질이 되는 고도불포화지방산을 섭취시켰을때 여러수준의 methionine 이 생체내 지질과산화 반응에 미치는 영향을 알아보기 위하여 85g 내외의 Spraque-Dawley종 숫쥐 40마리를 5군으로 나누어 12주간 사육한 후 지질과산화물 함량, glutathione peroxidase활성도 및 전자현미경학적 조사를 하였으며, 그 결과 체중증가, 사료효율은 대조군과 비교했을때 MF군이 유의적으로 낮았고, 간의 중량 비대현상이 나타났다. 그란 0.3%-0.6%수준의 methionine공급에 의해서 호전될 수 있었다. 혈장과 간mitochondria 분획에서의 지질과산화물 함량은 대조군과 비교했을대 MF군이 유의적으로 높았는데, 0.6% methionine 공급에 의해서 저하시킬 수 있었다. 적혈구와 간mitochondria 분획에서의 GSH-Px 활성도는 MF준이 대조군에 비해서 유의적으로 낮았으나 3M군과 6M군에 비해서 높은 경향을 나타내었다. 간세포의 전자현미경조사에서는 MF군이 정상군에 비해서 지방소적, mitochondria의 swelling 및 융합, 그리고 disse강내 microvilli의 파괴가 관찰되었으나 methionine의 공급에 의해 다소 호전되었다. 그러나 정상적인 간세포 형태를 나타내지는 못하였다. 본 연구결과는 vitamin E와 Se의 부족 및 고도 불포화지방산을 섭취시켰을때 지질과산화물형성이 증가되며, 조직학적 변화를 유발하였다. 그러나 methionine에 의해서 이들 다소 억제시킬 수가 있었으며, 본 실험에서는 0.6%수준에서 비교적 양호하게 나타났다. 또한 methionine 단독으로서의 항산화력 증진 보다는 항산화제로서 관련된 영양소와의 상호작용에 의해 그 효과가 더욱 더 증대 될 수 있다고 활 수 있으며, 따라서 보다 균형적인 영양소 섭취에 의해서 지질과 산화의 유해한 반응으로부터 보호작용을 나타낼 수 있을 것으로 사료된다.

비타민 E와 불포화 지방과의 관계 - 들깨유(油)를 중심으로 한 동물의 비교 연구 - (Relationship between Vitamin E and Polyunsaturated Fat - A comparative animal study emphasizing perilla seed oil as a fat constituent -)

  • Perilla (frutescens) seed oil, which is widely used as a source of vegetable oil in Korea, contains a strikingly large amount (58.4% of total fatty acids) of polyunsaturated linolenic acid (18 : 3) which is one of the essential fatty acids. Our hypothesis was that vitamin E contained in this oil would not be enough to prevent peroxidation of this polyunsaturated oil. A comparative study was carried out using rats and chicks devided into seven groups with various diet combinations emphasizing fat sources for the period of four weeks. The level of fat in each diet was 15% and animals were fed ad libitum. Various diet combinations were as follows; perilla seed oil and sesame seed oil with and without vitamin E supplementation, tallow as a saturated fat source and perilla seed hull group (10% at the expense of carbohydrate). The fat constituents of control group were consisted of 50% vegetable oil and 50% animal fat. A few important findings are as follows: 1. Rats fed perilla seed oil lost their hair focally around the neck and suffered from a bad skin lesion at the same place. In chicks, yellow pigmentation both of feather and of skin was clearly observed only in groups fed perilla seed oil with or without vitamin E supplementation. The basis of biochemical mechanisms of this phenomena remains as an important research interest. 2. The mean value for hematocrit was significantly lower for the chicks fed perilla seed oil than for those fed control diet. This result seems to be attributable to the effect on the red cell membrane known as peroxidation-hemolysis of vitamin E deficiency. 3. The serum cholesterol level was higher for the rats fed perilla seed oil than for those fed control diet, whereas in chicks the group fed perilla seed oil showed lower value than the control group indicating that different animal species could vary in their responses to the same diet. 4. In pathological examinations, the sign of hepatic fibrosis was seen in the perilla seed hull group and it was noticeable that the level of hepatic RNA was significantly increased in the rat recovering from vitamin E deficiency. It is hoped that more detailed studies on perilla seed oil and hulls will soon be carried out in many aspects especially i) at various levels of fat in the diet, ii) in relation to dietary selenium level and iii) to find an optimum level of dietary essential fatty acids in terms of P/S ratio using various animal species. In the mean time, the public should be informed to preserve this particular oil with care to minimize fatty acid oxidation and should be discouraged from overconsuming this oil. MINERAL STATUS OF GRAZING PHILIPPINE GOATS II. THE NUTRITION OF SELENIUM, COPPER AND ZINC OF GOATS IN LUZON ISLAND

  • Nutritional status of trace minerals (Se, Cu and Zn) in goats grazed on the native pasture was investigated during 2 years in Luzon Island, Republic of the Philippines. Three regions (South, Central and North) were objected to collect the samples of blood of goats and forages on the pasture. Se content of major species forages in the pasture was $24.7{\mu}g/kg$ DM, and was clearly lower than the value ($200{\mu}g/kg$ DM) required commonly for ruminant feed. The Cu and Zn contents of all forage samples ranged from 7.6 to 24.3 and 11.4 to 50.6 mg/kg DM, respectively, and these values almost exceeded the dietary level required for sheep. The blood Se levels in about 55% of goats grazed alone were under the normal range ($20{\mu}g/l$), but it in goats fed some concentrates as a supplement were almost within a normal ranges ($20-200{\mu}g/l$), though the Cu content of forages almost exceeded the dietary level required commonly. The 4-13% of plasma samples of goats without supplement showed the Zn level below the lower limit (0.6 mg/l) of normal range. The Zn deficiency was mostly improved by the addition of a small amount of concentrates. From these results, it will be necessary to study about unstable Cu status of grazed goats in Luzon Island regarding an interaction of Cu and other elements known to relate to the malabsorption of dietary Cu.
