Estimating the Important Components in Three Different Sample Types of Soybean by Near Infrared Reflectance Spectroscopy

  • This experiment was carried out to find suitable sample type for the more accurate prediction and non-destructive way in the application of near infrared reflectance spectroscopy (NIRS) technique for estimation the protein, total amino acids, and total isoflavone of soybean by comparing three different sample types, single seed, whole seeds, and milled seeds powder. The coefficient of determination in calibration ($R^2$) and coefficient of determination in cross-validation (1-VR) for three components analyzed using NIRS revealed that milled powder sample type yielded the highest, followed by single seed, and the whole seeds as the lowest. The coefficient of determination in calibration for single seed was moderately low($R^2$ 0.70-0.84), while the calibration equation developed with NIRS data scanned with whole seeds showed the lowest accuracy and reliability compared with other sample groups. The scatter plot for NIRS data versus the reference data of whole seeds showed the widest data cloud, in contrary with the milled powder type which showed flatter data cloud. By comparison of NIRS results for total isoflavone, total amino acids, and protein of soybean seeds with three sample types, the powder sample could be estimated for the most accurate prediction. However, based from the results, the use of single bean samples, without grinding the seeds and in consideration with NIRS application for more nondestructive and faster prediction, is proven to be a promising strategy for soybean component estimation using NIRS.

Effect of Density Separation after Pretreatment on Embryo Growth and Radicle Emergence of Carrot(Daucus carota L.) Seeds (당근 종자의 전 처리후 비중선이 배생장과 발아에 미치는 영향)

  • Carrot(Daucus carota L.) seeds (CV Danver 126) were primed and then separated by density differences to improve both the percentage and time of radicle emergence. Seeds for priming were soaked in aerated distilled water for 2 days (water imbibed), 25% solution of polyethylene glycol(PEG) 6000 for 6 or 10 days, salt solution of 0.2M KNO$_3$+0.1M $K_2$HPO$_4$ for 6 or 10 days, or mixed with Agro-Lig with 90% moisture content for 6 days and 70% moisture content for 6 or 10 days (SMP) at 2$0^{\circ}C$, respectively. The greatest embryo growth and the highest radicle emergence were observed from the seeds treated SMP with 90% moisture content for 6 days among the primed treatments. After the SMP treatment, the seeds were separated into density classes with a float-sink procedure using aqueous solution of Maltrin 600 with 0.02/cm$^3$ density increments. The lower density classes of the carrot seeds, the more embryo growth, the higher and the faster rates of radicle emergence were exhibited in order from 1.06 to 1.14 density classes of the carrot seeds treated SMP.

Nondestructive Classification of Viable and Non-viable Radish (Raphanus sativus L) Seeds using Hyperspectral Reflectance Imaging (초분광 반사광 영상을 이용한 무(Raphanus sativus L) 종자의 발아와 불발아 비파괴 판별)

  • Purpose: Nondestructive evaluation of seed viability is a highly demanded technique in the seed industry. In this study, hyperspectral imaging system was used for discrimination of viable and non-viable radish seeds. Method: The spectral data with the range from 400 to 1000 nm measured by hyperspectral reflectance imaging system were used. A calibration and a test models were developed by partial least square discrimination analysis (PLS-DA) for classification of viable and non-viable radish seeds. Either each data set of visible (400~750 nm) and NIR (750~1000 nm) spectra and the spectra of the combined spectral ranges were used for developing models. Results: The discrimination accuracy of calibration was 84% for visible range and 76.3% for NIR range. The discrimination accuracy of test was 84.2% for visible range and 75.8% for NIR range. The discrimination accuracies of calibration and test with full range were 92.2% and 92.5%, respectively. The resultant images based on the optimal PLS-DA model showed high performance for the discrimination of the nonviable seeds from the viable seeds with the accuracy of 95%. Conclusions: The results showed that hyperspectral reflectance imaging has good potential for discriminating nonviable radish seeds from massive amounts of viable seeds.

  • Soybean mosaic virus (SMV) infection in seeds wits investigated by an immunochemical method Seven soybean cultivars or lines Bughae No.1, KAS 662-7, Chungbugbaeg, Gwanggyo Clark, Bongeui, and Gangrim were tested using hypocotyls of germinated seeds and presence of SMV was detected in six soybean cultivars but Gangrim. The level of SMV infection in the assayed cultivars varied from 2.1 to $12.5\%$. It seemed that seed coat mottling had no correlation with seeds. SMV infection of the seeds since virus has not always been detected from the mot tled seeds SMV has not been detected in the seeds of variety Gwanggyo which showed necrotic symptoms.

Effect of Gibberellin Acid on Embryo Development and Germination of Dicentra spectabilis (L.) Lem. Seeds

  • This study was carried out to develop the seed propagation method of Dicenta spectabilis (L.) Lem. which is an ornamental plant native to Korea. In the previous studies, it was found that the seeds of D. spectabilis were morphophysiologically dormant (MPD), and high and low temperature of stratification were continuously required for the embryo growth and germination of the seeds. Especially, it was most effective to store for 1 month at $20^{\circ}C$ and then to transfer to $4^{\circ}C$. The treatment of $GA_3$ was carried out to promote embryo development and germination. The seeds were submerged in 100, 200 or $500mg{\cdot}L^{-1}$ $GA_3$ for 72 hours and then stored at various conditions as follow. The temperature conditions disposed of this experiment were 1 month at 10, 15, 20, and $25^{\circ}C$ or 2, 4, 8, and 12 months at $4^{\circ}C$, respectively. As a result, the length of embryo and germination rate of the seeds were the best when stored at $4^{\circ}C$ for 8 months after $500mg{\cdot}L^{-1}$ $GA_3$ treatment. Besides, when the seeds stored at $4^{\circ}C$, significant differences in embryo length and germination rate were shown with $GA_3$ concentration and storage period. It was also proved that high-concentration of $GA_3$ could replace the high temperature and could promote germination. Consequentially, the D. spectabilis seeds were classified into intermediate simple levels among MPD types.

Prosopis juliflora invasion and environmental factors on density of soil seed bank in Afar Region, Northeast Ethiopia

  • The aims of the study were to analyze (1) the effects of Prosopis juliflora (Prosopis) on the spatial distribution and soil seed banks (SSB) diversity and density, (2) the effects of environmental factors on SSB diversity and density (number of seeds in the soil per unit area), and (3) the effects of animal fecal droppings on SSB diversity, density, and dispersal. Aboveground vegetation data were collected from different Prosopis-infested habitats from quadrats (20 × 20 m) in Prosopis thickets, Prosopis + native species stand, non-invaded woodlands, and open grazing lands. In each Prosopis-infested habitats, soil samples were collected from the litter layer and three successive soil layer, i.e., 0-3 cm, 3-6 cm, and 6-9 cm. Seeds from soil samples and animal fecal matter were separated in the green house using the seedling emergence technique. Invasion of Prosopis had significant effects on the soil seed bank diversity. Results revealed that the mean value of the Shannon diversity of non-invaded woodlands was being higher by 19.2%, 18.5%, and 11.0% than Prosopis thickets; Prosopis + native species stand and open grazing lands, respectively. The seed diversity and richness, recovered from 6-9-cm-deep layer were the highest. On the other hand, the density of Prosopis seeds was the highest in the litter layer. About 156 of seeds/kg (92.9%) of seeds were germinated from cattle fecal matter. However, in a small proportion of seedlings, 12 of seeds/kg (7.1%) were germinated from shot fecal matter. Thus, as the seeds in the soil were low in the study areas, in situ and ex situ conservation of original plants and reseeding of persistent grass species such as Cynodon dactylon, Cenchrus ciliaris, Chrysopogon plumulosus, and Brachiaria ramosa are recommended.

Comparison of Sodium Hypochiorite and Potassium Hydroxide as Seed Treatment for Stimulating Germination of Korean Lawngrass (Zoysia japonica Steud.) Seed (한국잔디종자의 발아촉진을 위한 Sodium Hypochlorite와 Potassium Hydroxide 처리효과의 비교)

  • Research was conducted to obtain easy and secure methods for promoting germination of Korean lawngrass (Zoysia japonic a Steud.) seed. The effect of sodium hypochiorite (NaOCl) treatment on germination of seeds was compared with that of potassium hydroxide (KOH) treatment under conditions of duration of seed storage after harvest and temperature of treatment. Emergence of seedlings from soil and micro-flora infection rate among seed treatments were investigated. 1. The promotion of germination was much more evident in seeds treated with 4% NaOCl for 8 and 10 hours than in seeds treated with 25% or 30% KOH for 30 and 40 minutes. Longer durations of storage before seed treatment in KOH than in NaOCl were required to initiate enough germination. 2. Differences in temperature of treatment with NaOCT did not have much influence on germination of seeds and treatment temperature of 15˚C was better than that of 200˚C and 300˚C for promoting germination. But seed treatment with KOH significantly promoted germination with increasing temperature of treatment from I 5˚C to 20˚C and 30˚C. 3. GA$_3$ treatment enhanced germination in NaOCl-pretreated seeds at early stage of imbibition and in-creased about 10% germination after 10 days of imbibition in KOH-pretreated seeds. 4. NaOC1 treatments significantly decreased the rate of the infection of microflora in seed samples and enhaced emergence of seedlings from soil compared with KOH treatment. 5. NaOC1 treatment had advantage over KOH treatment with respect to ease of preparing, securing and handling in stimulating germinating of Korean lawngrass seeds.

Identification of a novel triterpene saponin from Panax ginseng seeds, pseudoginsenoside RT8, and its antiinflammatory activity

  • Background: Panax ginseng Meyer (Araliaceae) is a highly valued medicinal plant in Asian regions, especially in Korea, China, and Japan. Chemical and biological studies on P. ginseng have focused primarily on its roots, whereas the seeds remain poorly understood. This study explores the phytochemical and biological properties of compounds from P. ginseng seeds. Methods: P. ginseng seeds were extracted with methanol, and 16 compounds were isolated using various chromatographic methods. The chemical structures of the isolates were determined by spectroscopic data. Antiinflammatory activities were evaluated for triterpene and steroidal saponins using lipopolysaccharide-stimulated RAW264.7 macrophages and THP-1 monocyte leukemia cells. Results: Phytochemical investigation of P. ginseng seeds led to the isolation of a novel triterpene saponin, pseudoginsenoside RT8, along with 15 known compounds. Pseudoginsenoside RT8 exhibited more potent antiinflammatory activity than the other saponins, attenuating lipopolysaccharide-mediated induction of proinflammatory genes such as interleukin-1β, interleukin-6, inducible nitric oxide synthase, cyclooxygenase-2, and matrix metalloproteinase-9, and suppressed reactive oxygen species and nitric oxide generation in a dose-dependent manner. Conclusion: These findings indicate that pseudoginsenoside RT8 has a pharmaceutical potential as an antiinflammatory agent and that P. ginseng seeds are a good natural source for discovering novel bioactive molecules.

Anticancer Effects of the Extracts of Oriental Melon (Cucumis melo L. var makuwa Makino) Seeds (참외(Cucumis melo L. var makuwa Makino) 종자 추출물의 항암 활성)

  • The objective of this study was to investigate the anticancer effects of the extracts of oriental melon seeds. Various solvent extracts of oriental melon seeds were prepared and their anticancer effects were examined using in vitro MTT and CV assays. The anticancer effects of various extracts of oriental melon seeds were also examined in five human cancer cell lines including A549, AGS, HT-29, MCF-7 and HepG2. The ethanol extract of heated oriental melon seeds showed the potent cytotoxic effects especially against mouse hepatoma cell line(Hepa1c1c7), human hepatoma cell line(HepG2) and human breast cancer cell line(MCF-7). These data suggest that oriental melon seeds can be a promising anticancer agent against human liver and breast cancer.

Effect of Red Pepper Seeds Powder on Lipid Composition in Rats Fed High-Fat.High-Cholesterol Diets

  • The purpose of the present study was to evaluate the effects of red pepper seeds powder on lipid metabolism in rats fed high fat high cholesterol diet. Rats were divided into five experimental groups: normal diet group, high fat high cholesterol diet group, high fat high cholesterol diet with 5% red pepper seeds powder supplemented group (SA group), high fat high cholesterol diet with 10% red pepper seeds powder supplemented group (SB group) and high fat high cholesterol diet with 15% red peeper seeds powder supplemented group (SC group). The serum triglyceride (TG) and cholesterol contents, and LDL-cholesterol and atherogenic index (AI) of the red pepper seed powder supplemented groups were significantly decreased compared to the HF group. The serum HDL-cholesterol contents of the red pepper seed powder supplemented groups were increased compared to the HF group. However, there was no significant difference in the serum HDL-cholesterol among all experimental groups. The hepatic TG and cholesterol contents of the red pepper seed powder supplemented groups were significantly decreased compared to the HF group. The fecal total cholesterol and triglyceride contents of the red pepper seeds powder supplemented groups were significantly increased compared to the HF group. These results suggest that supplementation of red pepper seed powder may have a pronounced impact on markers of lipid metabolism in serum and liver of rats fed high fat high cholesterol diets.
