화살, 남천, 차, 초피나무 4유용수종(有用樹種)의 종자(種子) 발아촉진처리(發芽促進處理)가 포지발아(圃地發芽)와 유묘(幼苗) 생장(生長)에 미치는 효과 (Effects of the Seed Treatment on Field Germination and Seedling Growth in four useful species, Euonymus alatus, Nandina domestica, Thea sinensis and Zanthoxylum piperitum)

  • 새 순(筍)이나 수피(樹皮), 열매 등(等) 식(食) 약용(藥用)으로 이용(利用)하고 있는 화살나무를 비롯한 4수종(樹種)의 종자(種子)에 대하여 포지(圃地) 발아율(發芽率)을 높이는 방법(方法)을 알아보기 위행 화살나무, 차나무, 초피나무의 종자(種子)는 10월(月) 중순(中旬)에서 11월(月) 중순(中旬) 사이에 채취(採取)하여 정선(精選)한 후(後) Pon-Pon, $H_2O_2$, $GA_3$, 종피파쇄(種皮破碎) 등(等)으로 처리(處理)하여 노천매장(露天埋藏)(119일(日)-134일(日))을 하였다가 3월(月) 29일(日) 시험포지(試驗圃地) 파종(播種)하였으며, 남천 종자(種子)는 11월(月) 하순(下旬에) 채취(採取)하여 Growth chamber에서 저장온도(貯藏溫度)($4^{\circ}C$, $25^{\circ}C$, $32^{\circ}C$), 저장기간(貯藏期間)(7일(日), 15일(日), 21일(日))별(別)로 처리(處理)한 후(後) 유리온실(溫室) 내(內)의 파종상자(播種箱子)($55{\times}45{\times}15cm$)에 파종(播種)하여 보관(保管)하던 중(中) 5월(月) 초(初)부터는 노지(露地)에서 관리(管理)하였다. 수종별(樹種別), 종자(種字) 발아촉진처리별(發芽促進處理別)로 발아(發芽) 생장(生長)한 묘목(苗木)을 파종(播種)한 당년(當年) 10월(月) 중순(中旬)에 굴취(掘取)하여 발아율(發芽率)과 유묘(幼苗) 생장(生長)을 조사(調査) 분석(分析)하였다. 종자(種子) 발아촉진처리별(發芽促進處理別) 포지(圃地) 발아율(發芽率)에서 화살나무는 $GA_3$ 처리(處理) 후(後) 노천매장(露天埋藏)한 처리구(處理區)에서 당년(當年) 발아율(發芽率) 67.1%로 일반(一般) 노천매장(露天埋藏) 처리구(處理區) 18.4%보다 높은 발아율(發芽率)을 보여 유의적(有意的)인 발아촉진(發芽促進) 효과(效果)가 인정(認定)되었다. 차나무 종자(種子)는 종피파쇄(種皮破碎)나 $H_2O_2$ 처리(處理) 후(後) 노천매장(露天埋藏)한 처리구(處理區)와 일반(一般) 노천매장처리구(露天埋藏處理區) 간(間)에 뚜렷한 차이(差異)를 보이지 않아 노천매장(露天埋藏) 만으로도 충분한 발아촉진(發芽促進) 효과(效果)가 인정(認定)되었다. 초피나무 종자(種子)의 경우 Pon-Pon 처리(處理) 후(後) 노천매장(露天埋藏)한 처리구(處理區)에서 당년(當年) 발아율(發芽率) 80.3%로 일반(一般) 노천매장(露天埋藏)한 처리구(處理區)의 12.4% 보다 유의적(有意的)인 차이(差異)가 있었다. 남천 종자(種子) $32^{\circ}C$에서 7일간(日間) 저장(貯藏)한 처리구(處理區)가 $4^{\circ}C$에서 7일간(日間) 저장(貯藏)한 처리구(處理區)보다 5개월(個月) 정도 조기(早期)에 발아(發芽)되면서 발아율(發芽率)도 높게 나타났다. 포지(圃地)에 파종(播種)한 종자(種子)의 발아최성기(發芽最盛期)는 화살나무(32일(日))>초피나무(49일(日))>차나무(83일(일))>남천(87일(日)) 순(順)이었다. 포지(圃地)에서 발아(發芽)한 실생묘목(實生苗木) 생장량(生長量)은 종자(種子) 발아촉진처리(發芽促進處理)의 효과(效果)로 발아(發芽)가 빠른 처리구(處理區)에서 양호(良好)하였다. 실생묘(實生苗)의 연(年) 생장시간(生長時間) 중(中)에서 가장 왕성(旺盛)한 생장(生長)을 보인 최대생장(最大生長) 기간(期間)은 화살나무는 종자발아(種子發芽) 후(後) 72일(日) 경(傾)인 6월(月) 하순(下旬)부터 7월(月) 중순(中旬)사이이고, 초피나무는 59일(日)째인 6월(月) 하순(下旬)부터 7월(月) 중순(中旬) 사이였으며 차나무는 54일(日)째인 8월(月) 중순(中旬)부터 9월(月) 중순(中旬)사이로 나타났다.

Electrophoretic and Immunological Evaluation of Secalin in Rye, Triticale, and Wheat-Rye Translocation Wheat

  • Seed storage proteins have been used for studying biochemical genetics and end-use quality aspects. We conducted enzyme-linked immunosorbent assay (ELISA) and one-dimensional SDS-PAGE (1D SDS-PAGE) to evaluate different cereal crop species and Korean wheat lines for rye secalin proteins. The antisecalin antibody showed consistent specificity for rye secalin with little cross-reactivity to gliadins. Immunological cross-reactivities measured by the ELISA technique using competition assay showed significant differences of absorbance among rye, triticale, wheat-rye translocated wheat and non-translocated wheat. The absorbance values were lowest in rye followed by triticale, translocated wheat and non-translocated wheat. The ELISA for discrimination of wheat-rye translocation on the basis of antigen-antibody reactivity showed that none of the Korean wheat lines possessed 1RS and secalin proteins. The competitive ELISA experiment demonstrated specific determination for secalin that was originated from rye chromosomal parts. The result of 1D SDS-PAGE for identifying rye secalin subunits showed all three rye specific secalin protein subunits (75 KDa, 45 KDa, and 40 KDa) for rye and triticale, and 1RS specific secalins (45 KDa and 40 KDa) for 1AL/1RS and 1BL/1RS translocated wheats. All Korean wheats were lacking 1RS of rye chromosome and secalin.

Germination Characteristics of Some Red Rice Accessions

  • Nine accessions of red rices collected from different regions were tested for germination characteristics in relation to seed maturity and several storage conditions after harvest. No red rice seeds germinated at five days after anthesis (DAA). Wanjuaengmi (long-grain red rice) seeds at 10DAA germinated up to 30% of total samples, producing only abnormal seedlings, whereas 10D-AA-seeds of normal cultivar showed only 3.3% germinability. Some red rice seeds absorbed more water than cultivated varieties, and showed less decrease in germination rate than cultivated varieties when seeds were exposed at -1, -5 or -1$0^{\circ}C$ for up to 60 hours after soaking. Red rice accessions maintained more than 95% germination when stored. indoor for 120 days. Two short-grain red rices showed about 50% germination when overwintered in the field, while other long-grain red rices and cultivated rices germinated less than 10%. Germination of seeds overwintered in clay loam soil was lower than that in loam soil, and seeds on surface germinated less than those in 1~7cm depth.

탈지들깨박 Ethanol 추출물의 항산화 효과 (Studies on Antioxidant Activity of Ethanol Extracts from Defatted Perilla Flour)

  • The antioxidant activity of ethanol extracts from defatted perilla flour was investigated by measuring peroxide value of perilla oil during storage at $45^{\circ}C$. The antioxidant activity of ethanol extracts was also compared with BHA, BHT and tocopherol. Anti-oxidant activity of ethanol extracts was also examined in corn oil and lard. The ethanol extracts contents of defatted perilla flour and the original perilla seed were 7.69 and 4.56% respectively. The antioxidant activity of ethanol extracts was superior to that of 0.02% BHT, BHA and tocopherol in the perilla oil substrate, merely in concentration of one-twentieth as much as that contained in original perilla oil seeds. The fractions of non-polar solvent (hexane and chloroform) obtained from silicic acid column chromatography are less effective than that of polar solvent as an antioxidant. Antioxidant activity of partially purified ethanol fraction is slightly inferior to that of original crude ethanol extracts. Ethanol extracts were also effective in corn oil and lard almost same as in perilla oil. The total phenolic compound contents of crude ethanol extracts and partially purified ethanol fraction were 9.3, 6.4%, respectively.

  • 양파의 저온저장중 부패방지를 위한 천연항균제 개발의 일환으로 저장양파로부터 부패세균을 분리한 후 이 균에 대한 식물추출물의 항균활성을 조사하였다. 양파의 저온저장중 과육으로부터 분리한 곰팡이를 MEA와 PDA에서 배양하면서 분생자병과 분생자의 형태적 특성을 조사한 결과 플라스크형의 phialide, 분생자병의 branching type이 simple type이며, conidial head가 columnar shape 가지고 있는 것으로 확인되어 Penicillium sp. ON-211으로 동정하였다. 또한 자생 식물은 채집하여 각종 유기용매로 추출한 후 항균활성을 조사하였다. 정향나무, 탱자나무의 잎은 chloroform 분액에서 높은 활성을 나타내었고, 측백나무 열매는 물분획에서 높은 활성을 나타내었으며, 연필향나무의 잎은 ethyl acetate 분획에서 비교적 높은 항균 활성이 나타났다.

Genomic and evolutionary analysis with gluten proteins of major food crops in the Triticeae tribe

  • Prolamins are the main seed storage proteins in cereals. Gluten proteins seem to be prolamins because their primary structure have the meaningful quantity of proline and glutamine amino acid residues. Gluten proteins are found in crops such as wheat (Triticum aestivum), barley (Hordeum vulgare), and rye (Secale cereale) which are major food crops in the Triticeae tribe. Glutenin and gliadin, hordein, and secalin are typical gluten proteins found in wheat, barley, and rye, respectively. Gluten affect grain quality so that many researches, such as isolation or characterization of their genes, have been carried out. To improve the quality of grains in the Triticeae tribe, it is necessary to understand the relationship within their gluten proteins and their evolutionary changes. The sequences of nucleotides and amino acids of gluten protein including glutenins, gliadins, hordeins, and secalins were retrieved from NCBI (https://www.ncbi.nlm.nih.gov/) and Uniprot (http://www.uniprot.org/). The sequence analysis and the phylogenetic analysis of gluten proteins were performed with various website tools. The results demonstrated that gluten proteins were grouped with their homology and were mostly corresponded with the previous reports. However, some genes were moved, duplicated, or disappeared as evolutionary process. The obtained data will encourage the breeding programs of wheat, barley, rye, and other crops in the Triticeae tribe.

Expression of Nutritionally Well-balanced Protein, AmA1, in Saccharomyces cerevisiae

  • Food yeast, Saccharomyces cerevisiae, is a safe organism with a long history of use for the production of biomass rich in high quality proteins and vitamins. AmA1, a seed storage albumin from Amaranthus hypochondriacus, has a well-balanced amino acid composition and high levels of essential amino acids and offers the possibility of further improving food animal feed additives. In order to find an effective means of expressing AmA1 in yeast, the gene was cloned into an episomal shuttle vector. Four different promoters were tested: the glyceraldehyde-3-phosphate dehydrogenase promoter, galactose dehydrogenase 10 promoter, alcohol dehydrogenase II promoter, and a hybrid ADH2-GPD promoter. The recombinant AmA1 genes were then introduced into the yeast Saccharomyces cerevisiae 2805. Northern and Western blot analyses of the yeast under appropriate conditions revealed that AmA1 was expressed by all four promoters at varying levels. An enzyme-linked immunosorbent assay demonstrated that the amount of AmA1 protein in the recombinant yeast was 1.3-4.3% of the total soluble proteins. The highest expression level was obtained from the hybrid ADH2-GPD promoter.

  • This study was carried out to understand and analyze the cooking and processing methods presented in CHE MIN YO SUL (chapter 82~86). The result from the study are as follows; 1. Byung was made from dough of wheat flour, glutinous rice flour, small green peas flour an rice of millet etc. by steaming, roasting, deep fat frying or sauted, or it had eaten Byung which was made from egg or seagull's egg without cereal was simillar to Korea's fried egg cake. 2. Jong and Youl were steamed product made from mixture of millet and rice which were wrapped with leaf of Julpool or bamboo's leaf. It's shape was simillar to Korea's Song-Pyun but different from materials and cooking method. 3. Jamyong's cooking method was boild the mixture of white rice soup, Myong-Jup, and salt. and then pour the rised Bal. 4. Ye and Lo were boiling product made from new oat and appricot seed flour. It was simillar to sweet drink made from rice, and put them in the jar for storage.

  • We report on the successful fabrication of ZnO nanorod (NR)/polystyrene (PS) nanosphere hybrid nanostructure by combining drop coating and hydrothermal methods. Especially, by adopting an atomic layer deposition method for seed layer formation, very uniform ZnO NR structure is grown on the complicated PS surfaces. By using zinc nitrate hexahydrate $[Zn(NO_3)_2{\cdot}6H_2O]$ and hexamine $[(CH_2)_6N_4]$ as sources for Zn and O in hydrothermal process, hexagonal shaped single crystal ZnO NRs are synthesized without dissolution of PS in hydrothermal solution. X-ray diffraction results show that the ZnO NRs are grown along c-axis with single crystalline structure and there is no trace of impurities or unintentionally formed intermetallic compounds. Photoluminescence spectrum measured at room temperature for the ZnO NRs on flat Si and PS show typical two emission bands, which are corresponding to the band-edge and deep level emissions in ZnO crystal. Based on these structural and optical investigations, we confirm that the ZnO NRs can be grown well even on the complicated PS surface morphology to form the chestnut-shaped hybrid nanostructures for the energy generation and storage applications.

  • 대두 발아종 오존수 처리에 의한 배유세포와 산성 인산호효소의 변화에 관해서 전자현미경으로 조사한 결과는 다음과 같다. 산성 인산화효소의 활성은 발달하고 있는 종자 배유의 여러 소기관에 나타났는데, 효소의 활성은 0.5 ppm 오존수를 처리한 후 12시간 배양중에 나타났다. 오존수를 처리한 후 배유가 분화함에 따라 산성 인산화효소의 반응산물은 액포내에 축적되는 것으로 나타나는데, 이것은 산성 인산화효소가 세포간 저장물질의 분해와 이동에 관여하는 것으로 여겨진다.
