Mass Selection for Increased Lacquer Yield of Rhus verniciflua Stokes (산칠량(産漆量)이 많은 옻나무개체의 선발(選拔)에 관한 연구(硏究))

  • This study was conducted to investigate characteristics related to lacquer yield and to evaluate possibility of indirect selection of Rhus verniciflua Stokes. Twenty-four lacquer trees were randomly selected for measuring growth, urushiol content and bark thickness. Urushiol content showed a significant positive correlation with outer bark thickness and secretory canal density at 1% level, and with inner bark thickness at 5% level. Bark thickness and secretory canal density could be used as indicators for high urushiol content within bark and thus for high lacquer yield. Mass selection for high lacquer yield was made based on urushiol content within bark. Sixteen individuals with the highest urushiol content value were selected for breeding. As expected, the selected trees had thicker bark, higher density of secretory canal, better developed secretory canals compare to the non-selected lacquer trees.

  • Panax ginseng C. A. Meyer (Araliaceae) is listed in Shen Nung Pentsao Ching(神農本草經) as the remedy for replenishing the primodial qi(氣), restoring pulse, treating collapse, tonifing the spleen and lag, promoting the production of body fluid to quench thirst, tranquilizing the mind, and improving the function of brain. The prescriptions of In Sam Tang(人參揚) are also recorded in many other Chinese medical books. The identification of the age of Panax ginseng is very important in commercial market as well as in research field. However, any reports about it have not been clearly established yet. To clarify the criterion, the morphological and anatomical characteristics of the roots of various age Panax ginseng cultivated in Korea were studied. The characteristics of cork layer, secretory canal, and vessel were shown to bε useful keys to confirm the age of Panax ginseng.

  • Glandular trichomes present on the leaf surface of Drosera capensis were examined using transmission electron microscopy. A large number of stalked glands exist on the adaxial surfaces of the leaf blade. The secretory head is composed of two layers of secretory cells, one layer of middle cells, and the inner tracheids. The secretory cells contain rough endoplasmic reticulum, mitochondria, plastids, Golgi apparatus, and vacuoles. The secretory cells show prominent cell wall ingrowth, and thick cuticle restricted on the subcuticular wall. Frequently, the cuticle has some pores, canal-like structures, showing electron -dense granules being penetrated through them. Ultrastructural localization using diaminobenzidine showed the electron-dense deposits in the vacuole. No peroxidase activity was seen in the cell wall and cytolasm. The activity of peroxidase (POX) isozymes in Drosera which isoelectric point (pI) is 3.6 and some anionic POX isozymes which pIs are laid between 3.6 and 4.6 were especially increased according to the development and the formation of glandular trichomes. Also, the activity of some POX isozymes which isoelectric points are laid between 4.6 and 5.1 were increased in the regions of leaves which has trichomes.

Structure and function of the secretory ducts in Panax ginseng C.A. Mayer (인삼 분비관의 구조와 기능)

  • The distribution of the secretory ducts, fine structures of the secretory epithelial cells, and the ingredients of the metaplasmic inclusions were studied at light and electron microscopical levels in seeds, stems, leaves, and roots of ginseng. The secretory ducts occurred in the hypocotyl of the embryo, in the cortex of the roots, and also both inside and outside of each vascular bundle in the stems and leaves. Especially, it is considered that the circular layers of the secretory ducts in roots may represent their ages. The epithelial cell has well developed nucleolus, mitochondria and smooth endoplasmic reticulum. Sudanophyl and osmiophilic inclusions were found in the epithelial cytoplasm and duct lumen. But these inclusions were not observed when extracted with pyridin or alcohol. In contrast to the lumen with red color, the epithelial cells were blue in color as stained with nile blue, suggesting that the former inclusions are neutral lipid while the latter are acidic lipid. The electron density of the cell inclusions was quite high as fixed with osmium tetroxide, indicating that most of these secretory materials seem to be unsaturated lipid. Therefore, since ginseng secretory ducts are closely associated with the lipid metabolism, it should be called lipid canal or lipid duct.

Histopathologic Characterization of Metastatic Ceruminous Gland Adenocarcinoma in a Dog

  • A 13-year-old spayed female American cocker spaniel dog was submitted to an animal hospital with a mass from left ear canal and enlarged left parotid lymph node. Given one-year history of odorous exudates and chronic otitis externa, total ear canal ablation was performed. Grossly, because of the neoplastic mass in both inner and outside of annular cartilage in external ear, vertical ear canal was severely narrowed. Histologically, there were numerous proliferated glands in the ear canal mass. Many neoplastic glands contained secretory vesicles on the free margin and necrotic cellular debris. Severe multifocal necrosis and strong invasion were also observed throughout the mass. Massive metastatic foci of glandular structures originated from ceruminous gland were presented in the enlarged parotid lymph node. Neoplastic glandular epithelia contained PAS-positive diastase-resistant eosinophilic cytoplasmic granules. Immunohistochemically, the neoplastic cells showed positive reactions for cytokeratin (CK) 7 and negative for CK 5/6. Based on the clinical implication and gross findings, histopathology and immunohistochemistry, this case was diagnosed as metastatic ceruminous gland adenocarcinoma in the American cocker spaniel.

Pharmacognostical Studies on the Angelica species from Korea (한국산 Angelica속 식물의 생약학적 연구)

  • Angelica species from Korea has been used mainly as to promoting blood circulation, relieving pain and headache in common cold in Korea. To clarify the botanical origin of Angelica species from Korea, the anatomical characteristics of Angelica gigas, Angelica koreanum and Angelica purpuraefolia were studied. As a result, it was found the fact that the internal morphological patterns for the three Angelica species could discriminate from them by the secretory canal, xylem fiber and vascular bundle. According to these three criteria, Angelica species from Korea could be clearly distinguished so that the toxication by Angelica purpuraefolia can be prevented.

Chronic maxillary sinusitis caused by root canal overfilling of Calcipex II

  • This is a case report of chronic maxillary sinusitis caused by root canal overfilling of Calcipex II (Techno-Dent). A 60 year-old male complained of dull pain in the right maxillary molar area after complicated endodontic treatment using Calcipex II paste and was finally diagnosed with a chronic maxillary sinusitis through a clinical and radiological observation. In the biopsy examination, the periapical granuloma contained a lot of dark and translucent Calcipex II granules which were not stained with hematoxylin and eosin. They were usually engulfed by macrophages but rarely resorbed, resulting in scattering and migrating into antral mucosa. Most of the Calcipex II granules were also accumulated in the cytoplasms of secretory columnar epithelial cells, and small amount of Calcipex II granules were gradually secreted into sinus lumen by exocytosis. However, chronic granulomatous inflammation occurred without the additional recruitment of polymorphonuclear leukocytes (PMNs) and lymphocytes, and many macrophages which engulfed the Calcipex II granules were finally destroyed in the processes of cellular apoptosis. It is presumed that Calcipex II granules are likely to have a causative role to induce the granulomatous foreign body inflammation in the periapical region, and subsequently to exacerbate the chronic maxillary sinusitis in this study.

Effects of CEPA on the Secretion of Lacquer and Bark Physiology of Lactree(Rhus verniciflua Stokes) (CEPA 처리(處理)가 옻나무의 칠액분비(漆液分泌) 및 수피생리(樹皮生理)에 미치는 영향(影響))

  • This study was conducted to investigate the effects of CEPA(2-chloroethyl phosphonic acid) on bark physiology and anatomy of lactree(Rhus verniciflua Stokes). Sample trees of similar size and growth rate were selected from 7-year-old lactree plantation located in Hyengsung-kun, Kangwon-do. Lanolin pastes containing 0.1, 1, or 10% CEPA were put into the bark-removed hole made by corer(${\phi}1cm$) on the main stem at 1.2m above the ground on June 16, 1995. Five weeks after application of CEPA, bark thickness was markedly increased as a result of the increase in the amount of phloem and intercellular spaces, and correlated with the increased production of urushiol. By the application of 10% CEPA, bark thickness was increased approximately 2.5 times, and the urushiol content within bark was increased 2.8 times compared to that of untreated trees because CEPA stimulated the accumulation of urushiol within bark. Treatment of 10% CEPA also increased the size and the total number of secretory canals, and induced an increase in ray width. The phloem parenchyma cells of CEPA-treated trees were well-developed and closely packed with little intercellular space.

Observation on Oil Canals of Umbelliferous Condiment Herbs of Japanese Origin

  • This study has observed the development, structure and distribution of oil canals in mitsuba, seri, ashitaba and hamabohu, that are condiment herbs belonging to the Umbelliferae family, using a light microscope. Oil canals were found in the petioles, leaf blades, stems, roots, hypocotyls and cotyledons. Oil classified into distribution due to ring vascular bundles, as in mitsuba and seri, and distribution due to diffuse vascular bundles, as in ashitaba and hamabohu. Oil canal development in the cortex due to petiole thickening was followed by the development of collenchyma and vascular bundles. However, no vascular bundles were formed in some cases. Many oil canals were found in the periphery of the petioles. Oil canals in leaf blades were found on the adaxial and abaxial sides on the veins. Those around the main veins were larger. Steam oil canals were found in the cortex and pith in mitsuba and seri, and in the cortex and fundamental tissues around the xylem, in ashitaba and hamabohu, while those in the roots were found in the pericycle in mitsuba and seri, and in the collenchyma-like tissues and phloem in ashitaba and hamabohu. The transverse sections of oil canals were round or elliptical. The secretory cells in the cortex and pith were smaller than the neighboring parenchyma cells, while they were larger than the neighboring parenchyma cells in the phloem.

  • The fine structure of the salivary glands of the pine moth, Dendrolimus spectabilis Butler, at the last larval period is observed using light and electron microscopes. The moths have single paired tubular salivary glands which openings are connected to the oral cavity through the upper jaw. By the external morphology and its functions, the glands are subdivided into three regions which are anterior reabsorptive region, middle storage region and posterior secretory region. Along the inner canal of the salivary gland two columns of the large glandular cells are connected each other and oriented to ring-like forms. By this cellular orientation, the glands have long and large tubular structure. From anterior to posterior region large nuclei of the glands are ramified like twigs of the tree, and in the cytoplasm of the cell numerous mitochondria and vacuoles are seen. Moreover, basal plasma membranes of the gland cells are heavily infolded. The anterior region of the glands keeps several characteristics related to the reabsorption of the material from the inner cavity to the glandular cells whereas, main salivary material is synthesized and secreted through the long and convoluted posterior region. The apical plasma membranes of the cells are the most heavily invaginated at the posterior regoin, but trachea and tracheoles are distributed only at the middle and posterior regions. In the cytoplasm of the middle region Golgi complexes appeared at the vicinity of the vesicles, and at the posterior region of the salivary glands multivesicular bodies are also observed.
