Morphological Characteristics of Hyphal Interaction between Grifola umbellata and its Companion Fungus

  • Morphological characteristics of hyphal interaction between Grifola umbellata (Pers. Ex Fr.) Pilat and its companion fungus which related to sclerotia formation from hyphae were investigated by external observations, light microscopy and transmission electron microscopy (TEM). External observations showed that a dense antagonism line was formed by both G. umbellata and companion fungus after their hyphae contacted each other in dual culture. Many hyphal strands emerged on the colony of G. umbellata and differentiated to sclerotia from where hyphal strands crossed. Light microscope observations revealed the process of antagonism line formation. Mature antagonism with structural differentiation, was composed of three main layers: the rind, the rind underlayer and the hypha layer. TEM observations showed that after colonies hyphal contact, a series of reactions always occurred in both G. umbellata and companion fungus. Cells in the center of antagonism line were dead. Cells of G. umbellata adjacent to the antagonism line were usually large and hollow, with unilateral thickened wall, whereas those of companion fungus were empty, with thin or thick wall. Both hyphal interaction at the antagonism line may be one of the main reasons for sclerotia of G. umbellata differentiation from hypha.

  • 2009년 9월 경상남도농업기술원 해바라기 재배포장에서 흰비단병이 발생되었다. 병징은 줄기 부분이 수침상으로 물러지고 부패되면서 병반부와 토양 표면에 흰색의 곰팡이가 솜털모양으로 생기고 갈색의 작은 둥근 균핵을 많이 형성하였다. 균핵의 크기는 1~3 mm이며 균사의 폭은 $4{\sim}8\;{\mu}m$이었다. 균사생육과 균핵형성의 적정온도는 $30^{\circ}C$이었다. 균사 특유의 clamp connection이 관찰되었다. 해바라기에 발생한 병징과 병원균의 균학적 특징 및 병원성을 검정한 결과, 이 병을 Sclerotium rolfsii에 의한 해바라기 흰비단병으로 명명하고자 제안한다.

Identification of Ciboria carunculoides RS103V, a Fungus Causing Popcorn Disease on Mulberry Fruits in Korea

  • The popcorn disease caused by sclerotia forming fungi reduces the productivity of mulberry fruits in world wide. In Korea, only two species (Ciboria shiraiana and Scleromitrula shiraiana) have been reported as the major causal organisms and their morphological features are also largely unknown. Hereby, we report the first identification of another species (i.e. Ciboria carunculoides) in Korea and detailed features of their anamorphic stage. Fungi dominantly associated with sclerotia were purely isolated from infected mulberry fruits under the microscope. PCR-amplified DNA encoding 5.8S rRNA displayed 100% similarity to Ciboria carunculoides. The anamorphic features exhibited the absence of true mycelia. Instead, very short, aseptated, branched conidiophores were directly emerged from sclerotia. Phialides were usually three in number from each conidiophore, ampuliform to navicular in shape, slightly curved and tapering towards the apex. Conidia were produced from phialides and mostly found as one celled, pear shaped, not hyaline with smooth to uneven surface walled. Diversely modified features in phialides formed pseudo-mycelial structures around the host tissue. Combined all, current study is the first report of C. carunculoides isolated in Korea and the foremost detailed description of its anamorph stage.

Occurrence of Bulb Rot of Allium victorialis var. platyphyllum Caused by Sclerotium rolfsii in Korea

  • In 2007, a bulb rot of Allium victorialis var. platyphyllum Makino caused by Sclerotium rolfsii occurred sporadically in a herb farm at Gyeongsangnam-do, Korea. The symptom initiated with water-soaking lesion and progressed into bulb rot and wilt of a whole plant. Severely infected plants were blighted and killed eventually. White mycelial mats appeared on the lesion at early stage and a number of sclerotia were formed on the bulb. The sclerotia were spherical, $1{\sim}3 mm$ and white to brown. The optimum temperature for the growth and sclerotia formation was $30^{\circ}C$ on PDA. The hyphal width was measured $4-9{\mu}m$. The typical clamp connections were observed on PDA. Based on the symptom, mycological characteristics and pathogenicity to the host plant, this fungus was identified as Sclerotium rolfsii Saccardo. This is the first report on the bulb rot of A. victorialis var. platyphyllum caused by S. rolfsii in Korea.

Occurrence of Stem Rot of Wild Aster(Aster koraiensis) Caused by Sclerotium rolfsii in Korea

  • A destructive stem rot of wild aster(Aster koraiensis) occurred sporadically some farmers' fields in Guman-myon, Kosonggun, Kyongsangnam-do in 2000. One of the most severely infected field in Kosong showed 28.6 percent of infection rate. The fungus also caused stem or crown rot and systemic wilt or blight of the plants. White mycelium spread over stems and petioles of infected plants and sclerotia formed on the old lesions and near the soil surface. The fungus showed maximum mycelial growth around $30^{\circ}C$ and did not grow under $5^{\circ}C$ and over $45^{\circ}C$ and mycelial width were $4.3{\sim}10.2{\mu}m$. Colony was white, usually many narrow mycelial stand in the aerial mycelium and formed clamp connection. Numerous sclerotia were formed on PDA at $30^{\circ}C$. The shape sclerotia were globoid and $0.8{\sim}3.0{\times}0.9{\sim}3.4$ mm in size. The fungus was isolated repeatedly from the infected tissues and confirmed its pathogenecity to wild aster and identified as Sclerotium rolfsii. This is the first report on the stem rot of wild aster caused by S. rolfsii in Korea.

  • 2006년에서 2008년까지 진주에서 재배중인 페튜니아에서 병이 발생하였다. 병징은 줄기와 잎이 수침상으로 물러지고 썩으면서 갈색으로 변하며 시들어 말라 죽었다. 줄기, 지제부 및 토양 표면에 솜털모양의 흰색 균사체가 생기고 $1{\sim}3\;mm$ 크기의 둥근 갈색 균핵을 많이 형성하였다. PDA배지 상에서 병원균의 균사 폭은 $4{\sim}8\;{\mu}m$이며 clamp connection이 형성되었고 균사생육과 균핵형성 적온은 $30^{\circ}C$이었다. 페튜니아에서 발생한 병징과 병원균의 균학적 특징 및 병원성을 기초로 본 병을 Sclerotium rolfsii Saccardo에 의한 페튜니아 흰비단병으로 명명하고자 제안한다.

  • In July 2001, a destructive stem rot of tatarian aster(Aster taturicus) was occurred sporadically in exhibition farm of Gyeongsangnam-do Agricultural Research and Extension Services, in Hamyang, Korea. The fungus also caused collar and crown rot and systemic wilt or blight of whole plant. White mycelium spread over stems and petioles of infected plants and sclerotia formed on the old lesions and near the soil surface. The fungus showed maximum mycelial growth was obtained around $30^{\circ}C$ but did not grow below $5^{\circ}C$ or above $45^{\circ}C$. The mycelial width ranges $4.2{\sim}10.4{\mu}m$. and the color is white, usually many narrow mycelial stand grow in the aerial mycelium and formed clamp connection. Numerous sclerotia were formed in artificial media like PDA at $30^{\circ}C$. The shape of sclerotia were sphere and $1.0{\sim}3.2{\mu}m$ in diameter. The fungus was isolated repeatedly from the infected, tissues and confirmed its pathogenecity to aster and identified as Sclerotium rolfsii. This is the first report that Sclerotium rolfsii causes stem rot of tatarian aster in Korea.

樹種, 接種量 및 接種方法이 茯笭 菌絲生長과 結笭에 미치는 영향 (Effect of Tree Species, Inoculation Amount and Inoculation Methods on Mycelium Growth and Sclerotia Formatino of Poria cocos Wolf)

  • When medicinal mushroom, Poria cocos, is cultured , inoculation method of spawn is cross slice inoculation of which the both sides of pine tree were peeled and spawn of P.cocos was inoculated. However, this method required lots of inoculation amount. This study was carried out to improve the culturing method of P. cocos. A good growth of P.cocos was observed in MCM(mushroom complete medium), showing proper mycelia growth and density. In inoculation amount, conventional method(cross slice inoculation) requires 20 bottles of spawn. In contrast, short log method required 8 bottles of spawn and drilling inoculation method 2~3 bottles, which could save by 60% and 85-90% respectively. In the selectrion of tree species, pine and larch had better condition for spawn culture and sclerotia formation condition.In terms of yield , pine was 33.7kg/3.3$m^2$. In the yield of pine, conventional method was 23.4kg/3.3$m^2$, drilling inoculation 29.4kg/3.3$m^2$, short log inoculation 31.7kg/3.3$m^2$, therefore drilling inoculation could increase by 25% and short log inoculation 35%, In addition, management cost was also saved.

복령의 인공 재배법 개선과 항산화활성에 관한 연구 (Studies on Improvement of Artificial Cultivation and Antioxidative Activity of Poria cocos)

  • 복령의 인공재배기술을 개선하고 복령추출물의 생리활성을 검토하고자 본 연구를 수행하였다. 종균의 종류 및 접종법에 따른 검토결과, 종목 양면접종법이 균핵형성정도, 균핵 갯수, 균핵 생산수율 및 개당 평균 중량에서 가장 좋았으며, 톱밥 양면접종법이 균핵형성 및 생성수율에서 가장 낮았다 복령의 품질과 생산성을 종합하여 판단할 때, 최적 종균 및 접종방법은 종목 양면접종 > 종목 절단면접종 > 균핵 양면접종 > 톱밥 양면접종의 순이었다. 복령균핵으로부터 분리한 생리활성물질인 조 복령당의 함량은 83%이었고, 농도가 증가함에 따라 상대점도가 상승하였으며, 염의 첨가 농도에는 큰 영향을 받지 않았다. 항산화 활성(EDA, POV, TBA)을 측정한 결과, TBA가의 경우에는 천연 항산화제인 비타민 E와 유사한 활성을 나타내었으나, POV와 EDA에 의한 항산화활성 측정 결과에서는 BHT나 비타민 C보다 낮은 활성을 나타내었다.
